Radiographic Imaging and Tomography

Radiographic imaging and tomography (RadIT) come in many flavors such as X-ray imaging and tomography (IT), proton IT, neutron IT, muon IT, neutrino IT, etc. We identify five RadIT themes: Physics, Sources, Detectors, Methods, and Data Science, which are integral parts of image interpretation and three-dimensional (3D) tomographic reconstruction. Traditionally, RadIT have been driven by medicine, non-destructive testing, material sciences, and security applications. The latest thrusts of growth come from automation, machine vision, additive manufacturing and virtual reality (‘metaverse’). The five RadIT themes parallel their counterparts in optical IT. Synergies among different forms of RadIT and with optical IT motivate further advances towards multi-modal IT and quantum IT.

Data Assimilation using Non-invasive Monte Carlo Sensitivity Analysis of Reactor Kinetics Parameters [Slides]

Sensitivity coefficients of prompt neutron decay constant (α) and effective delayed neutron fraction (β eff ) to Pu-239 nuclear data were calculated using a newly available tool (ACEtk). Uncertainty calculations showed the following trends: (1) The prompt neutron decay constant can be used to reduce nuclear data-induced uncertainty in Pu-239(n,f) and (2) the effective delayed neutron fraction can be used to effectively reduce nuclear data-induced uncertainty in Pu-239 total fission $\tilde{v}$. Sensitivity/uncertainty analysis can be used to determine optimal experiment/detector setup to identify compensating errors in nuclear data. Upcoming work includes investigating other nuclide-reaction pairs and response sensitivities, such as Δ$\rho$, neutron leakage spectra, and reaction rate measurements to constrain nuclear data of interest.

LANSCE Facility Longevity Assessment

This LANSCE Facility Longevity Assessment examined 34 total systems critical to the operation of the LANSCE accelerator complex. These systems are primarily owned, operated, and maintained by AOT Division. Based on the criteria used in this assessment, there are 12 AOT Division systems that are potential high risk as related to longterm LANSCE operations. These systems may also impact the success of LAMP. Also summarized in this report are the facility activities planned over the next decade. These include general facility maintenance and infrastructure upgrades, as well as planned electrical system upgrades. The LANSCE Facility Operations (LFO) Division is responsible for this activity scope and budget planning. Most of the planned activities are general in nature and should have little impact on accelerator operations or LAMP activities as currently envisioned. However, close coordination with between AOT, LFO, and LAMP would be prudent. A next step should be to develop an actionable plan to address the high-risk systems. Potential criteria for developing such a plan could include: (1) Evaluating the global impact of the system (operations, program, safety, etc.), (2) Ranking by impact (ranked now by risk), (3) Estimating cost to implement the corrective actions (Is there already a plan or not?), and (4) Estimating the time required for corrective actions. An alternative could include integration of the assessment data into both the Asset Management System and the Maintenance Analytics Risk Register.

Electrical Fault Detection, Power Quality, Distributed Energy Resource Use Cases, and Cyber Event Applications with the Cyber Grid Guard System Using Distributed Ledger Technology

Electrical utilities continue to deploy more intelligent electronic devices (IEDs) inside and outside electrical substation and are associated with distributed energy resources (DERs). The integrity and confidentiality of data from IEDs is crucial, and distributed ledger technology (DLT) could improve the resilience of microgrids by helping to make these data more secure. The most popular applications using blockchain technology for electrical utilities is in the field is based on energy trading. However, the dynamism of the penetration of customer owned DERs and the deployment of sensors with IEDs have led to the identification of new applications using DLT that are focused on other areas, such as monitoring, operation and management of the grid and its assets. In addition, the majority of studies on electrical grid applications with blockchain were validated with software simulations. Although general monitoring of power systems for using DLT could be evaluated in operational electric grids, other DLT research applications such as defense against cyber-attacks and/or electrical fault detection are not likely to be performed in a real infrastructure because of possible risks to the network/equipment security. This report summarizes the application of power system applications using distributed ledger technology (DLT), providing a secure DLT framework for collecting data from IEDs like power meters and protective relays inside and outside of an electrical substation and/or between two different electrical utilities. In this study, the use case scenarios were created and assessed for different power system application by using DLT. The electrical fault detection for faulted phases (1), power quality monitoring of phase voltage magnitudes, frequency levels and load power factor (2), DERs use case monitoring (3), and cyber-event applications (4) were performed in a test bed with a Cyber-Grid Guard (CGG) system using DLT. It had a real-time simulator with power meters and protective relays in-the-loop. The first section of this report presents a literature review of power system applications using blockchain at research level. The second section shows the theory and equations used on this report. The third section shows the description of the test bed, equipment, architecture, and electrical grid diagrams. The fourth section shows the experimental models and use case scenarios that were performed for the electrical fault detection, power quality, DERs use case, and cyber event applications with the CGG system using DLT. The fifth section shows the results collected from the tests based on comparing the time stamped events of the analog signals from the IEDs, DLT computer and real time simulator. The sixth section performed the discussion of the results for the use case scenarios. Finally, section seven presents the conclusions for this report were presented.

Accelerating the identification of novel secondary metabolites in bioenergy plant root exudates using MicroED

Small molecule metabolites drive inter- and intraspecies communication and dependencies in diverse biological systems, yet a large proportion of these important chemical compounds remain uncharacterized in plants and microbes. Approximately 90% of the metabolites in root exudate profiles are unknown compounds, despite the importance of root exudate composition in plant-microbe interactions. We need advanced analytical capabilities that will support rapid discovery and structural elucidation of metabolites from biological samples that may be limited in quantity and high in complexity. To fill this gap, this project aimed to develop an integrated workflow involving metabolite extraction, separation, and crystallization from plant root exudates followed by characterization using nuclear magnetic resonance (NMR) spectroscopy, mass spectrometry, and microcrystal electron diffraction (MicroED). Using crude root exudates from sorghum, this project successfully developed higher throughput exudate fractionation strategies to obtain pure compounds for crystallization and identified crystals in multiple fractions that diffracted. Additional efforts to increase the throughput of high-quality crystal generation for MicroED, such as crystallization screening and crystallization chaperone exploration, will be needed to further advance root exudate metabolite identification. The overall optimized sample preparation process can then be integrated with the existing data collection and data analysis pipelines for MicroED at PNNL to facilitate more rapid natural product discovery.

Sovereign Credit Rating Processes Adapted to Critical Infrastructure Cyber Risk Assessment

United States critical infrastructure entities are increasingly targeted by motivated and capable threat actors and must be prepared to assess and treat a diverse range of cyber risks. Consequently, this necessitates some form of analytical process to evaluate risks and inform cyber security investment decisions. A potential solution for structuring cyber risk evaluation exists within the field of sovereign credit ratings – where agencies employ mature approaches that integrate quantitative and qualitative data to produce a singular value of assessment. Adapting such approaches, we present a novel criterion and methodology for measuring and communicating the likelihood element of cyber risk. The methodology is composed of three sequential phases: a quantitative baseline organized by distinct capability frames, a bounded qualitative adjustment per frame, and a greater-bounded qualitative adjustment spanning the entire process. The process culminates in publication of a cyber capability rating that communicates a critical infrastructure entity’s ability and willingness to mitigate discontinuous function due to cyberattack.

Characterization and Quantification of Radiation-Induced Clusters/Precipitates in RPV Steels Using STEM-EDS and Machine Learning

Over the operational lifespan of a nuclear reactor, reactor pressure vessel (RPV) steels are subjected to significant neutron irradiation, resulting in complex microstructural changes and the consequent degradation of mechanical properties. Various physically motivated correlation models have been developed to predict neutron irradiation-induced embrittlement of RPVs under different irradiation conditions. However, the efficient and accurate characterizations and quantification of radiation-induced clusters in RPVs are still challenging, which will affect the precision of the predictive models for embrittlement of RPV components. In the DOE Visiting Faculty Program (VFP) research work at Oak Ridge National Lab (ORNL), I integrate machine learning to aid Scanning Transmission Electron Microscopy – Energy Dispersive X-ray Spectroscopy (STEM-EDS) analyses, which improve the characterization and quantification of radiation-induced clusters in RPV steels, thereby enabling more accurate predictions of material behavior under irradiation. The surveillance base- and welded- RPV steels were annealed at various temperatures of 340 °C, 450 °C and 500 °C for up to 168 hours, respectively. Afterwards, I have characterized radiation-induced clusters using advanced STEM-EDS techniques and subsequently applying machine learning algorithms to analyze and refine STEM-EDS datasets, enhancing the quantification of clusters compositions and distributions. In the end, an efficient workflow for integrating STEM-EDS data analysis with machine learning to address challenges including noise reduction has been developed. The completion of this VFP work will support bridge critical gaps in the accurate quantification of radiation-induced clusters in RPV steels using STEM-EDS and support the development of more precise models for predicting RPV embrittlement in the Light Water Reactor Sustainability program supported by Department of Energy and enhancing the collaboration between ORNL and Alred University. The outcome of the VFP project will leverage a few research papers submission to peer-reviewed journals in the relevant scientific field and a few oral presentations at national and international conferences.

Prediction of non-intuitive metabolic targets with bayesian metabolic control analysis to improve 3-hydroxypropionic acid production in Aspergillus niger

Development of efficient bioconversion processes is limited by the ability to predictably improve metabolic flux. Here we deployed Bayesian Metabolic Control Analysis as a platform to integrate multi-omics data with metabolic modeling and evaluated its ability to predict genetic interventions that improve metabolic flux. Global Metabolomics and proteomics data was collected from 17 Aspergillus niger strains engineered to produce the platform biochemical 3-hydroxypropionic acid from which seven actional genetic interventions were predicted from significant flux control coefficients. Of the suggested genetic interventions, two were present within the intuitively designed strains used for training (malonic semialdehyde dehydrogenase and pyruvate carboxylase) while five predicted targets were present within non-intuitive areas of the metabolic network including 5-formyltetrahydrofolate deformylase and four mitochondrial enzymes, alcohol dehydrogenase, succinyl-CoA ligase, aspartate aminotransferase, and malate dehydrogenase. Six of the targets were validated in the highest performing 3-HP strain used for multi-omics data generation which contained a prior disruption of the highest scoring target malonic semialdehyde dehydrogenase. Predicted directional perturbation of five of the six tested targets significantly improved titer and rate of 3-HP production and two significantly improved yield. The greatest improvements were observed following disruption of the non-intuitive target succinyl-CoA ligase which increased titer by 39% and yield by 29% (to 20.4 g/L 3-HP and 0.31 g 3-HP/g glucose) over the strains used for training. This study demonstrates the utility of Bayesian Metabolic Control Analysis and highlights the ability to predict meaningful genetic targets in unexpected areas of metabolism to improve engineered strains for bioconversion.

RNA-seq and metabolomic analyses of beneficial plant phenol biochemical pathways in red alder

Red alder ( Alnus rubra ) has highly desirable wood, dye pigment, and (traditional) medicinal properties which have been capitalized on for thousands of years, including by Pacific West Coast Native Americans. A rapidly growing tree species native to North American western coastal and riparian regions, it undergoes symbiosis with actinobacterium Frankia via their nitrogen-fixing root nodules. Red alder’s desirable properties are, however, largely attributed to its bioactive plant phenol metabolites, including for plant defense, for its attractive wood and bark coloration, and various beneficial medicinal properties. Integrated transcriptome and metabolome data analyses were carried out using buds, leaves, stems, roots, and root nodules from greenhouse grown red alder saplings with samples collected during different time-points (Spring, Summer, and Fall) of the growing season. Pollen and catkins were collected from field grown mature trees. Overall plant phenol biochemical pathways operative in red alder were determined, with a particular emphasis on potentially identifying candidates for the long unknown gateway entry points to the proanthocyanidin (PA) and ellagitannin metabolic classes, as well as in gaining better understanding of the biochemical basis of diarylheptanoid formation, i.e. that help define red alder’s varied medicinal uses, and its extensive wood and dye usage.
