Cloning and expression vectors and systems

Vectors for cloning, maintaining and expressing a wide range of coding sequences in inducible T7 expression systems in Escherichia coli expression hosts are disclosed herein. Target genes that can be stably maintained and expressed include those that specify proteins that are highly toxic to the host cell. Different configurations of vectors and expression hosts provide different rates of transcription and translation of target genes and therefore different rates of accumulation of target proteins. Methods for cloning by asymmetric ligation and co-expression of more than one target protein in a single vector are also disclosed, as are variants of BL21(DE3) having lower basal transcription by T7 RNA polymerase.

Comparative investigation of iterative solutions of the all-frequency stable formulation and its vector-potential-only variation

An all-frequency stable formulation has been proposed and applied to solve low-frequency and multiscale electromagnetic problems to avoid the low-frequency breakdown catastrophe of the traditional vector wave equation. Based on the potential representation of the fields, this formulation involves vector and scalar potentials, as well as an auxiliary potential to enforce the gauge condition. In this paper, such a formulation is first simplified to involve vector potential only. Their iterative solutions are then sought by using an incomplete LU preconditioner and an iterative solver. The iterative performance of both formulations are investigated in a comparative study.

Search for single-production of vector-like quarks decaying into Wb in the fully hadronic final state in pp collisions at $\sqrt{s}$ = 13 TeV with the ATLAS detector

A search for T and Y vector-like quarks produced in proton-proton collisions at a centre-of-mass energy of 13 TeV and decaying into Wb in the fully hadronic final state is presented. The search uses 139 fb −1 of data collected by the ATLAS detector at the LHC from 2015 to 2018. The final state is characterised by a hadronically decaying W boson with large Lorentz boost and a b-tagged jet, which are used to reconstruct the invariant mass of the vector-like quark candidate. The main background is QCD multijet production, which is estimated using a data-driven method. Upon finding no significant excess in data, mass limits at 95% confidence level are obtained as a function of the global coupling parameter, κ. The observed lower limits on the masses of Y quarks with κ = 0.5 and κ = 0.7 are 2.0 TeV and 2.4 TeV, respectively. For T quarks, the observed mass limits are 1.4 TeV for κ = 0.5 and 1.9 TeV for κ = 0.7.

Vector boson scattering and anomalous quartic couplings in final states with ℓνqq or ℓℓqq plus jets using proton-proton collisions at $\sqrt{s}=13$ TeV

A measurement is presented of the electroweak vector boson scattering production of ZV (V = W, Z) boson pairs associated with two jets in proton-proton collisions at a center-of-mass energy of 13 TeV. The data, corresponding to an integrated luminosity of 138 fb −1 , were collected at the CERN LHC with the CMS detector during the 2016–2018 data-taking period. The analysis targets final states with a pair of isolated electrons or muons from Z boson decays and three or four jets, depending on the momentum of the vector boson that decays into quarks. Signal strength is measured for events characterized by a large invariant mass of two forward jets with a wide pseudorapidity gap between them. The electroweak production of ZV in association with two jets is measured with an observed (expected) significance of 1.3 (1.8) standard deviations. A combination of the analyses of ZV channel and the previously published WV channel in the lepton plus jets final state places constraints on effective field theory parameters that describe anomalous electroweak production of WW, WZ, and ZZ boson pairs in association with two jets. Several world best limits are set on anomalous quartic gauge couplings in terms of dimension-8 standard model effective field theory operators.

Search for single production of vector-like T quarks decaying into Ht or Zt in pp collisions at $\sqrt{s}$ = 13 TeV with the ATLAS detector

This paper describes a search for the single production of an up-type vector-like quark (T) decaying as T → Ht or T → Zt. The search utilises a dataset of pp collisions at $\sqrt{s}$ = 13 TeV collected with the ATLAS detector during the 2015–2018 data-taking period of the Large Hadron Collider, corresponding to an integrated luminosity of 139 fb ₋1 . Data are analysed in final states containing a single lepton with multiple jets and b-jets. The presence of boosted heavy resonances in the event is exploited to discriminate the signal from the Standard Model background. No significant excess above the Standard Model expectation is observed, and 95% CL upper limits are set on the production cross section of T quarks in different decay channels. The results are interpreted in several benchmark scenarios to set limits on the mass and universal coupling strength (κ) of the vector-like quark. For singlet T quarks, κ values above 0.53 are excluded for all masses below 2.3 TeV. At a mass of 1.6 TeV, κ values as low as 0.35 are excluded. For T quarks in the doublet scenario, where the production cross section is much lower, κ values above 0.72 are excluded for all masses below 1.7 TeV, and this exclusion is extended to κ above 0.55 for low masses around 1.0 TeV.

Search for Higgs boson pair production with one associated vector boson in proton-proton collisions at $ \sqrt{s} $ = 13 TeV

A search for Higgs boson pair (HH) production in association with a vector boson V (W or Z boson) is presented. The search is based on proton-proton collision data at a center-of-mass energy of 13 TeV, collected with the CMS detector at the LHC, corresponding to an integrated luminosity of 138 fb$^{−1}$. Both hadronic and leptonic decays of V bosons are used. The leptons considered are electrons, muons, and neutrinos. The HH production is searched for in the $ \textrm{b}\overline{\textrm{b}}\textrm{b}\overline{\textrm{b}} $ decay channel. An observed (expected) upper limit at 95% confidence level of VHH production cross section is set at 294 (124) times the standard model prediction. Constraints are also set on the modifiers of the Higgs boson trilinear self-coupling, k$_{λ}$, assuming k$_{2V}$ = 1, and vice versa on the coupling of two Higgs bosons with two vector bosons, k$_{2V}$. The observed (expected) 95% confidence intervals of these coupling modifiers are −37.7 < k$_{λ}$ < 37.2 (−30.1 < k$_{λ}$ < 28.9) and −12.2 < k$_{2V}$ < 13.5 (−7.2 < k$_{2V}$ < 8.9), respectively.[graphic not available: see fulltext]

Search for single vector-like $B$ quark production and decay via $B$ → $bH$($b\overline{b}$) in $pp$ collisions at $\sqrt{s}$ = 13 TeV with the ATLAS detector

A search is presented for single production of a vector-like B quark decaying into a Standard Model b-quark and a Standard Model Higgs boson, which decays into a $b\overline{b}$ pair. The search is carried out in 139 fb -1 of $\sqrt{s}$ = 13 TeV proton-proton collision data collected by the ATLAS detector at the LHC between 2015 and 2018. No significant deviation from the Standard Model background prediction is observed, and mass-dependent exclusion limits at the 95% confidence level are set on the resonance production cross-section in several theoretical scenarios determined by the couplings c W , c Z and c H between the B quark and the Standard Model W, Z and Higgs bosons, respectively. For a vector-like B occurring as an isospin singlet, the search excludes values of c W greater than 0.45 for a B resonance mass (m B ) between 1.0 and 1.2 TeV. For 1.2 TeV < m B < 2.0 TeV, c W values larger than 0.50–0.65 are excluded. If the B occurs as part of a (B, Y) doublet, the smallest excluded c Z coupling values range between 0.3 and 0.5 across the investigated resonance mass range 1.0 TeV < m B < 2.0 TeV.

Observation of Vector Spin Seebeck Effect in a Noncollinear Antiferromagnet

Spintronic phenomena to date have been established in magnets with collinear moments, where the spin injection through the spin Seebeck effect (SSE) is always along the out-of-plane direction. Here, we report the observation of a vector SSE in a noncollinear antiferromagnet (AF) LuFeO 3 , where temperature gradient along the out-of-plane and also the in-plane directions can both inject a pure spin current and generate a voltage in the heavy metal via the inverse spin Hall effect (ISHE). We show that the thermovoltages are due to the magnetization from canted spins in LuFeO 3 . Furthermore, in contrast to the challenges of generating, manipulating, and detecting spin current in collinear AFs, the vector SSE in LuFeO 3 is readily viable in zero magnetic field and can be controlled by a small magnetic field of about 150 Oe at room temperature. Furthermore, the noncollinear AFs expand new realms for exploring spin phenomena and provide a new route to low-field antiferromagnetic spin caloritronics and magnonics.

Vector bosons and jets in proton collisions

Events with vector bosons produced in association with jets have been extensively studied at hadron colliders and provide high-accuracy tests of the standard model. A good understanding of these processes is of paramount importance for precision Higgs physics, as well as for searches for new physics. In particular, associated production of γ, W, or Z bosons with light-flavor and heavy-flavor jets is a powerful tool for testing perturbative QCD calculations, Monte Carlo event generators, and can also constrain the parametrizations used to describe the parton content of the proton. Furthermore, events with a W or Z boson produced with two well-separated jets can be used to distinguish between electroweak and strong production mechanisms, and to set limits on contributions of physics beyond the standard model. This review summarizes the historical theoretical developments and the state-of-the-art in the modeling of vector-boson-plus-jet physics while focusing on experimental results by the LHC collaborations in run 1 and run 2 and including comparisons with recent measurements at the Tevatron.

Low-mass vector-meson production at forward rapidity in 𝑝 + 𝑝 and Au + Au collisions at $\sqrt{𝑠_{𝑁⁢𝑁}}$ = 200 GeV

The PHENIX experiment at the Relativistic Heavy Ion Collider has measured low-mass vector-meson (𝜔+𝜌 and 𝜙) production through the dimuon decay channel at forward rapidity (1.2 < |y| < 2.2) in 𝑝 + 𝑝 and Au + Au collisions at $\sqrt{𝑠_{𝑁⁢𝑁}}$ = 200 GeV. The low-mass vector-meson yield and nuclear-modification factor were measured as a function of the average number of participating nucleons, ⟨𝑁 part ⟩, and the transverse momentum 𝑝 𝑇 . These results were compared with those obtained via the kaon decay channel in a similar 𝑝 𝑇 range at midrapidity. The nuclear-modification factors in both rapidity regions are consistent within the uncertainties. A comparison of the 𝜔 + 𝜌 and 𝐽/𝜓 mesons reveals that the light and heavy flavors are consistently suppressed across both 𝑝 𝑇 and ⟨𝑁 part ⟩. Finally, in contrast, the 𝜙 meson displays a nuclear-modification factor consistent with unity, suggesting strangeness enhancement in the medium formed.

A Six-Phase Space Vector PWM Technique for Wide Bandgap Device-Based Three-Level Inverters

This article presents a space vector pulsewidth modulation (SVPWM) technique for wide bandgap (WBG) device-based three-level (3L) six-phase inverters. Implementation of the SVPWM techniques for any six-phase inverter is complex for its high number of possible voltage vectors. Different six-phase load configurations are also possible depending on the angle between two consecutive phases and neutral point connections. The implementation techniques change depending on the load configuration. Moreover, a common neutral point for all six phases leads to circulating currents. Capacitor voltage balancing is also needed for 3L inverters. Capacitor voltage balancing and addressing the circulating current problem further increases the implementation complexity. In this article, a generalized SVPWM technique is proposed for the six-phase 3L inverters that is implementable using two three-phase 3L SVPWM blocks. The SVPWM technique also ensures dc bus capacitor voltage balancing and minimizes circulating current. Here, the SVPWM technique is experimentally verified using a GaN-based 3L six-phase active neutral point clamped (ANPC) inverter prototype, and the results are presented in this article.

Lost in transduction: Critical considerations when using viral vectors

The application of retroviral vectors in the laboratory requires considerations that often go overlooked but are often easy to circumvent. Here, we discuss the relationship between the observed transduction efficiency of a cell population and per-cell viral insertions—and describe how differential cell-type susceptibilities can confound results. We consider the math underlying this problem and review an alternative approach to the commonly used “multiplicity of infection” (MOI) method of titering and using viral vectors in the biomedical research laboratory.

Sparse matrix‐vector and matrix‐multivector products for the truncated SVD on graphics processors

Summary Many practical algorithms for numerical rank computations implement an iterative procedure that involves repeated multiplications of a vector, or a collection of vectors, with both a sparse matrix and its transpose. Unfortunately, the realization of these sparse products on current high performance libraries often deliver much lower arithmetic throughput when the matrix involved in the product is transposed. In this work, we propose a hybrid sparse matrix layout, named CSRC, that combines the flexibility of some well‐known sparse formats to offer a number of appealing properties: (1) CSRC can be obtained at low cost from the popular CSR (compressed sparse row) format; (2) CSRC has similar storage requirements as CSR; and especially, (3) the implementation of the sparse product kernels delivers high performance for both the direct product and its transposed variant on modern graphics accelerators thanks to a significant reduction of atomic operations compared to a conventional implementation based on CSR. This solution thus renders considerably higher performance when integrated into an iterative algorithm for the truncated singular value decomposition (SVD), such as the randomized SVD or, as demonstrated in the experimental results, the block Golub–Kahan–Lanczos algorithm.

Ribavirin targets sugar transporter 6 to suppress acquisition and transmission of rice stripe tenuivirus by its vector Laodelphax striatellus

Abstract BACKGROUND Rice stripe tenuivirus (RSV) is one of the most destructive pathogens of rice and other cereal crops. The virus is transmitted by the small brown planthopper (SBPH, Laodelphax striatellus ) in a circulative‐propagative manner. Thus, blocking transmission by the insect vector would provide an effective strategy to prevent epidemic outbreaks of the disease. RESULTS In this study, we explored the effect of ribavirin on acquisition and transmission of the virus by specifically inhibiting the expression of sugar transporter 6 (LsSt‐6), which was recently reported as a key vector component for RSV transmission. Ribavirin at the highest concentration tested (250 μmol L ‐1 ) significantly reduced RSV acquisition and transmission efficiency by SBPHs through inhibiting LsSt‐6 messenger RNA (mRNA) level. Survival of the model insect Spodoptera frugiperda cell line (Sf9) was 95.0 ± 2.2 and 85.6 ± 2.1% after exposure to 250 μmol L ‐1 ribavirin or 8‐azaguanine, respectively. Further study confirmed that 250 μmol L ‐1 ribavirin also significantly reduced LsSt‐6 mRNA and protein levels in Sf9 cells. However, 8‐azaguanine did not significantly inhibit viral infectivity and LsSt‐6 mRNA levels in SBPH or the Sf9 cell line. CONCLUSION This result provides evidence that ribavirin has the potential to disrupt LsSt‐6 expression but not others like viral RNAs to prevent acquiring RSV, which leads to less viral accumulation in SBPH tissues and thereby lower transmission efficiency. © 2020 Society of Chemical Industry

An extensible vector toolkit and parts library for advanced engineering of plant genomes

Abstract Plant biotechnology is rife with new advances in transformation and genome engineering techniques. A common requirement for delivery and coordinated expression in plant cells, however, places the design and assembly of transformation constructs at a crucial juncture as desired reagent suites grow more complex. Modular cloning principles have simplified some aspects of vector design, yet many important components remain unavailable or poorly adapted for rapid implementation in biotechnology research. Here, we describe a universal Golden Gate cloning toolkit for vector construction. The toolkit chassis is compatible with the widely accepted Phytobrick standard for genetic parts, and supports assembly of arbitrarily complex T‐DNAs through improved capacity, positional flexibility, and extensibility in comparison to extant kits. We also provision a substantial library of newly adapted Phytobricks, including regulatory elements for monocot and dicot gene expression, and coding sequences for genes of interest such as reporters, developmental regulators, and site‐specific recombinases. Finally, we use a series of dual‐luciferase assays to measure contributions to expression from promoters, terminators, and from cross‐cassette interactions attributable to enhancer elements in certain promoters. Taken together, these publicly available cloning resources can greatly accelerate the testing and deployment of new tools for plant engineering.

Moments of axial-vector GPD from lattice QCD: quark helicity, orbital angular momentum, and spin-orbit correlation

In this work, we present a lattice QCD calculation of the Mellin moments of the twist-2 axial-vector generalized parton distribution (GPD), $\overset{\sim }{H}\left(x,\xi, t\right)$ , at zero skewness, ξ, with multiple values of the momentum transfer, t. Our analysis employs the short-distance factorization framework on ratio-scheme renormalized quasi-GPD matrix elements. The calculations are based on an N f = 2 + 1 + 1 twisted mass fermions ensemble with clover improvement, a lattice spacing of a = 0.093 fm, and a pion mass of m π = 260 MeV. We consider both the iso-vector and iso-scalar cases, utilizing next-to-leading-order perturbative matching while omitting the disconnected contributions and gluon mixing in the iso-scalar case. For the first time, we determine the Mellin moments of $\overset{\sim }{H}$ up to the fifth order. From these moments, we discuss the quark helicity and orbital angular momentum contributions to the nucleon spin, as well as the spin-orbit correlations of the quarks. Additionally, we perform a Fourier transform over the momentum transfer, which allows us to explore the spin structure in the impact-parameter space.

Long-lived vectors from electromagnetic cascades at SHiP

We simulate dark-vector, V, production from electromagnetic cascades at the recently approved SHiP experiment. The cascades (initiated by photons from π$^{0}$ → γγ) can lead to 3–4 orders of magnitude increase of the event rate relative to using primary production alone. We provide new SHiP sensitivity projections for dark photons and electrophilic gauge bosons, which are significantly improved compared to previous literature. The main gain in sensitivity occurs for long-lived dark vectors with masses below ~ 50 − 300 MeV. The dominant production mode in this parameter space is low-energy annihilation e$^{+}$e$^{−}$ → V (γ). This motivates a detailed study of backgrounds and efficiencies in the SHiP experiment for sub-GeV signals.

Test of CP-invariance of the Higgs boson in vector-boson fusion production and in its decay into four leptons

A search for CP violation in the decay kinematics and vector-boson fusion production of the Higgs boson is performed in the H → ZZ * → 4 ℓ ( ℓ = e, μ ) decay channel. The results are based on proton-proton collision data produced at the LHC at a centre-of-mass energy of 13 TeV and recorded by the ATLAS detector from 2015 to 2018, corresponding to an integrated luminosity of 139 fb –1 . Matrix element-based optimal observables are used to constrain CP-odd couplings beyond the Standard Model in the framework of Standard Model effective field theory expressed in the Warsaw and Higgs bases. Differential fiducial cross-section measurements of the optimal observables are also performed, and a new fiducial cross-section measurement for vector-boson-fusion production is provided. All measurements are in agreement with the Standard Model prediction of a CP-even Higgs boson.
