Recent advances and accomplishments of artificial intelligence (AI) and deep generative models have established their usefulness in medicinal applications, especially in drug discovery and development. To correctly apply AI, the developer and user face questions such as which protocols to consider, which factors to scrutinize, and how the deep generative models can integrate the relevant disciplines. This review summarizes classical and newly developed AI approaches, providing an updated and accessible guide to the broad computational drug discovery and development community. We introduce deep generative models from different standpoints and describe the theoretical frameworks for representing chemical and biological structures and their applications. We discuss the data and technical challenges and highlight future directions of multimodal deep generative models for accelerating drug discovery.

Inverse mapping of properties to composition through generative modeling for designing molten salts

Generative modeling (GM) has been increasingly used for the inverse design and optimization of materials, yet its application to molten salt mixtures remains unexplored despite how a successful approach to the inverse design of molten salts would contribute to efficiently exploiting their customizability and unlocking their advantages in applications, such as energy production and energy storage. This work presents a workflow for the inverse design of molten salts with targeted density values, addressing the challenge of representing these complex mixtures in GM. A dataset of critically evaluated molten salt densities is used to train a variational autoencoder coupled with a predictive deep neural network, which then can be used to generate new molten salt compositions with desired density values. The effectiveness of the approach is demonstrated by designing mixtures with distinct densities and validating the predicted values using ab initio molecular dynamics simulations.

Infrared Computer-Generated Holograms: Design and Application for the WFIRST Grism Using Wavelength-Tuning Interferometry

Interferometers using computer-generated holograms (CGHs) have become the industry standard to accurately measure aspheric optics. The CGH is a diffractive optical element that can create a phase or amplitude distribution and can be manufactured with low uncertainty using modern lithographic techniques. However, these CGHs have conventionally been used with visible light and piezo-shifting interferometers. Testing the performance of transmissive optics in the infrared requires infrared CGHs and an infrared interferometer. Such an instrument is used in this investigation, which introduces its phase shift via wavelength-tuning. A procedure on how to design and manufacture infrared CGHs and how these were successfully used to model and measure the Wide-Field Infrared Survey Telescope grism elements is provided. Additionally, the paper provides a parametric model, simulation results, and calculations of the errors and measurements that come about when interferometers introduce a phase variation via wavelength-tuning interferometry to measure precision aspheres.

Alloy Design Data Generated for B2-Ordered Compounds

Developing alloys based on ordered compounds is significantly more complicated than developing designs based on disordered materials. In ordered compounds, the major constituent elements reside on particular sublattices. Therefore, the addition of a ternary element to a binary-ordered compound is complicated by the manner in which the ternary addition is made (at the expense of which binary component). When ternary additions are substituted for the wrong constituent, the physical and mechanical properties usually degrade. In some cases the resulting degradation in properties can be quite severe. For example, adding alloying additions to NiAl in the wrong combination (i.e., alloying additions that prefer the Al sublattice but are added at the expense of Ni) will severely embrittle the alloy to the point that it can literally fall apart during processing on cooling from the molten state. Consequently, alloying additions that strongly prefer one sublattice over another should always be added at the expense of that component during alloy development. Elements that have a very weak preference for a sublattice can usually be safely added at the expense of either element and will accommodate any deviation from stoichiometry by filling in for the deficient component. Unfortunately, this type of information is not known beforehand for most ordered systems. Therefore, a computational survey study, using a recently developed quantum approximate method, was undertaken at the NASA Glenn Research Center to determine the preferred site occupancy of ternary alloying additions to 12 different B2-ordered compounds including NiAl, FeAl, CoAl, CoFe, CoHf, CoTi, FeTi, RuAl, RuSi, RuHf, RuTi, and RuZr. Some of these compounds are potential high temperature structural alloys; others are used in thin-film magnetic and other electronic applications. The results are summarized. The italicized elements represent the previous sum total alloying information known and verify the computational method used to establish the table. Details of the computational procedures used to determine the preferred site occupancy can be found in reference 2. As further substantiation of the validity of the technique, and its extension to even more complicated systems, it was applied to two simultaneous alloying additions in an ordered alloy.

Advancing the Oxygen Generation Assembly Design to Increase Reliability and Reduce Costs for a Future Long Duration Mission

The state-of-the-art Oxygen Generation Assembly (OGA) has been reliably producing breathing oxygen for the crew aboard the International Space Station (ISS) for over eight years. Lessons learned from operating the ISS OGA have led to proposing incremental improvements to advance the baseline design for use in a future long duration mission. These improvements are intended to reduce system weight, crew maintenance time and resupply mass from Earth while increasing reliability. The proposed improvements include replacing the cell stack membrane material, deleting the nitrogen purge equipment, replacing the hydrogen sensors, deleting the wastewater interface, replacing the hydrogen dome and redesigning the cell stack power supply. The development work to date will be discussed and forward work will be outlined. Additionally, a redesigned system architecture will be proposed.

Design criteria for and applications of the vortex generator mixing principal

There are at this date numerable projects, in various Divisions of U.A.C., either in progress or contemplated, which are employing the vortex generator mixing principle. This principle is defined as the method which utilises tip vortices generated by low aspect ratio airfoils for the purpose of transverse mixing of the flow of a confined fluid medium where the axis of the vortex is parallel to the direction of flow. To date, vortex generator design criteria have not been published. Therefore, for the assistance of those individuals concerned and in the interest of the vortex mixing principle, the following material (subject to modifications) is presented in this memorandum report.

Sequence-based generative AI design of versatile tryptophan synthases

Enzymes are powerful and sustainable catalysts, but their widespread application is limited by the difficulty of identifying functional starting points for optimization, creating a major bottleneck in early- stage biocatalyst discovery. Designing libraries of such starting enzymes remains particularly challenging. Here, we use the GenSLM protein language model to generate novel β-subunit of tryptophan synthase (TrpB) enzymes that express in Escherichia coli and are both stable and catalytically active. Many generated TrpBs also display significant substrate promiscuity, outperforming their natural counterparts on non-native substrates. Some even surpass laboratory-evolved TrpBs. Comparison of the most-active and most-promiscuous generated TrpB to its closest natural homolog confirms that the enhanced versatility is absent from the natural enzyme, highlighting the creative potential of generative models. These results demonstrate that the generated TrpBs not only preserve natural structure and function but also acquire non-natural properties, establishing generative models as powerful tools for biocatalyst discovery and engineering.

Conceptual Design of Environmentally Friendly Rotorcraft - A Comparison of NASA and ONERA Approaches

In 2011, a task was initiated under the US-French Project Agreement on rotorcraft studies to collaborate on design methodologies for environmentally friendly rotorcraft. This paper summarizes the efforts of that collaboration. The French and US aerospace agencies, ONERA and NASA, have their own software toolsets and approaches to rotorcraft design. The first step of this research effort was to understand how rotorcraft impact the environment, with the initial focus on air pollution. Second, similar baseline helicopters were developed for a passenger transport mission, using NASA and ONERA rotorcraft design software tools. Comparisons were made between the designs generated by the two tools. Finally, rotorcraft designs were generated targeting reduced environmental impact. The results show that a rotorcraft design that targets reduced environmental impact can be significantly different than one that targets traditional cost drivers, such as fuel burn and empty weight.

Steam Generators: Thermal Hydraulic Measurements and Instrumentation

Steam generators are integral components of both conventional and advanced reactors utilizing steam power cycles, as they influence the power conversion performance. Steam generator designs are highly influenced by the type of reactor coolant used, which mainly governs the different control, monitoring, and safety system actuation requirements. Therefore, the present work reviews the thermal hydraulics instrumentation within steam generators designed for pressurized water reactors (PWRs), high-temperature gas-cooled reactors (HTGRs), sodium fast reactors (SFRs), and molten salt cooled reactors (MSRs). Emphasis is given to measurement techniques that are either unique or particularly important to specific reactor types. Lastly, a discussion is provided on the instrumentation and measured parameters. The work identifies standard parameters of interest and is expected to aid the development of test beds and novel measurements techniques.
