Promoter-dependent and -independent activation of insulin-like growth factor binding protein-5 gene expression by prostaglandin E2 in primary rat osteoblasts

Insulin-like growth factor (IGF) action is mediated by high affinity cell surface IGF receptors and modulated by a family of secreted IGF binding proteins (IGFBPs). IGFBP-5, the most conserved of six IGFBPs characterized to date, uniquely potentiates the anabolic actions of IGF-I for skeletal cells. In osteoblasts, IGFBP-5 production is stimulated by prostaglandin E2 (PGE2), a local factor that mediates certain effects induced by parathyroid hormone, cytokines such as interleukin-1 and transforming growth factor-beta, and mechanical strain. In this study, we show that transcriptional and post-transcriptional events initiated by PGE2 collaborate to enhance IGFBP-5 gene expression in primary fetal rat osteoblast cultures. PGE2 treatment stimulated up to a 7-fold rise in steady-state levels of IGFBP-5 mRNA throughout 32 h of incubation. Analysis of nascent IGFBP-5 mRNA suggested that PGE2 had only a modest stimulatory effect on IGFBP-5 gene transcription, and transient transfection studies with IGFBP-5 promoter-reporter genes confirmed that PGE2 enhanced promoter activity by approximately 2-fold. Similar stimulatory effects were seen with forskolin. A DNA fragment with only 51 base pairs of the 5'-flanking sequence retained hormonal responsiveness, which may be mediated by a binding site for transcription factor AP-2 located at positions -44 to -36 in the proximal IGFBP-5 promoter. Incubation of osteoblasts with the mRNA transcriptional inhibitor 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole demonstrated that PGE2 enhanced IGFBP-5 mRNA stability by 2-fold, increasing the t1/2 from 9 to 18 h. The effects of PGE2 on steady-state IGFBP-5 transcripts were abrogated by preincubating cells with cycloheximide, indicating that the effects of PGE2 on both gene transcription and mRNA stability required ongoing protein synthesis. Therefore, both promoter-dependent and -independent pathways converge to enhance IGFBP-5 gene expression in response to PGE2 in osteoblasts.

Persistent Layers of Enhanced Gravity Wave Dissipation in the Upper Mesosphere Revealed From SABER Observations

Dissipation of gravity waves (GWs) and resultant momentum flux deposition and eddy diffusion significantly influence the atmospheric structure and circulation. Using the temperature profiles measured by the SABER instrument (2002–2021), we derive the global GW action and its scale height (HAm). Comparing to the background scale height (H), we identify a persistent layer of enhanced GW dissipation (HAm/H > 2) in the upper mesosphere. The enhanced dissipation is primarily centered at 80–85 km with a vertical range of ∼8–20 km. At middle and high latitudes, the enhanced dissipation extends down to ∼60 km and is located above the eastward (westward) wind peak in winter (summer). At low latitudes, enhanced dissipation occurs around the wind reversals. The possible mechanisms of enhanced dissipation include wave refraction, wind filtering, and reduced static stability. The persistent layer of enhanced dissipation identified here provide observational evidence for GW dissipation constrains in models.

Increased Stability in Crystal Growth Kinetics in Response to Bulk Transport Enhancement

High-resolution interferometry on facets of lysozyme crystals growing under steady external conditions revealed intrinsic kinetics instabilities that occur as fluctuations in the spacing of growth steps. The amplitude of these fluctuations is reduced by forced solution flow. This increased stability is interpreted in terms of the ratio of the characteristic rates of lattice incorporation and bulk transport. Apparently a steady state is approached as an asymptote of a continuous family of unsteady states.

PMR-15/Layered Silicate Nanocomposites For Improved Thermal Stability And Mechanical Properties

Montmorillonite clay was organically modified by co-exchange of an aromatic diamine and a primary alkyl amine. The clay was dispersed into a PMR (Polymerization of Monomer Reactants)-15 matrix and the glass transition temperature and thermal oxidative stability of the resulting nanocomposites were evaluated. PMR-15/ silicate nanocomposites were also investigated as a matrix material for carbon fabric reinforced composites. Dispersion of the organically modified silicate into the PMR-15 matrix enhanced the thermal oxidative stability, the flexural strength, flexural modulus, and interlaminar shear strength of the polymer matrix composite.

Enhanced Engine Control for Emergency Operation

C-MAPSS40k engine simulation has been developed and is available to the public. The authenticity of the engine performance and controller enabled the development of realistic enhanced control modes through controller modification alone. Use of enhanced control modes improved stability and control of an impaired aircraft. - Fast Response is useful for manual manipulation of the throttles - Use of Fast Response improved stability as part of a yaw rate feedback system. - Use of Overthrust shortened takeoff distance, but was generally useful in flight, too. Initial lack of pilot familiarity resulted in discomfort, especially with yaw rate feedback, but that was the only drawback, overall the pilot found the enhanced modes very helpful.

Reliable Optical Pump Architecture for Highly Coherent Lasers Used in Space Metrology Applications

The design and initial demonstration of a laser pump module (LPM) incorporating single-mode, grating-stabilized 808-nm diode lasers and a low-loss, high-port-count optical combiner are completed. The purpose of the developed LPM is to reliably pump an Nd:YAG crystal in the laser head (LH), which serves as the optical metrology source for SIMLite mission. Using the narrow-linewidth, single-mode laser diodes enables placement of the pump power near Nd adsorption peak, which enhances pumping efficiency. Grating stabilization allows for stable pump spectra as diode operating temperature and bias current change. The low-loss, high-port-count optical combiner enables efficient combining of tens of pumps. Overall, the module supports 5+ years of continuous operation at 2 W of pump power with reliability approaching 100 percent. The LPM consists of a laser diode farm (LDF) and a pump beam combiner (PBC). An array of 807- to 808-nm fiber-pigtailed laser diodes makes up the LDF. A Bragg grating in each 5- m core single-mode (SM) fiber pigtail acts to stabilize the lasing spectra over a range of diode operating conditions. These commercially available single-mode laser diodes can deliver up to 150 mW of optical power. The outputs from the multiple pumps in the LDF are routed to the PBC, which is a 37-input by 1-output all-fiber device. The input ports consist of 5- m core SM fiber, while the output port consists of 105- m core, 0.15 NA (numerical aperture) multi-mode (MM) fiber. The combiner is fabricated by fusing the 37 input fibers while simultaneously tapering the fused region. At the completion of this process, the MM fiber is spliced to the end of the adiabatic taper, and, for protection, the combiner is sheathed by a capillary tube. A compact and robust metal housing was designed and fabricated to protect the PBC during space deployment.

Flexible Launch Vehicle Stability Analysis Using Steady and Unsteady Computational Fluid Dynamics

Launch vehicles frequently experience a reduced stability margin through the transonic Mach number range. This reduced stability margin can be caused by the aerodynamic undamping one of the lower-frequency flexible or rigid body modes. Analysis of the behavior of a flexible vehicle is routinely performed with quasi-steady aerodynamic line loads derived from steady rigid aerodynamics. However, a quasi-steady aeroelastic stability analysis can be unconservative at the critical Mach numbers, where experiment or unsteady computational aeroelastic analysis show a reduced or even negative aerodynamic damping.Amethod of enhancing the quasi-steady aeroelastic stability analysis of a launch vehicle with unsteady aerodynamics is developed that uses unsteady computational fluid dynamics to compute the response of selected lower-frequency modes. The response is contained in a time history of the vehicle line loads. A proper orthogonal decomposition of the unsteady aerodynamic line-load response is used to reduce the scale of data volume and system identification is used to derive the aerodynamic stiffness, damping, and mass matrices. The results are compared with the damping and frequency computed from unsteady computational aeroelasticity and from a quasi-steady analysis. The results show that incorporating unsteady aerodynamics in this way brings the enhanced quasi-steady aeroelastic stability analysis into close agreement with the unsteady computational aeroelastic results.

A precision star tracker utilizing advanced techniques and materials

An image dissector star tracker has been developed which operates in the photon counting mode making it possible to utilize all digital electronics. A unique pulse processing circuit allows bright stars to be tracked as well as dim stars. Thermal mechanical stability has been greatly enhanced by fabricating a housing with graphite/epoxy composite material with a linear coefficient of thermal expansion near zero. Test results indicate the +10 Mv stars can be acquired and tracked, while position variation with star intensity is less than 2 arc seconds from 2.5 Mv to +10 Mv. The noise equivalent angle for a +8 Mv star is 3 arc seconds. Polynominal correction for remaining cross-coupling and nonlinearity reduced error over total 1 x 1 deg field to 1.5 arc seconds.

Plasma transport in the auroral return current region

The classical and anomalous transport properties of a multifluid plasma consisting of H(+), O(+), and electron populations in the presence of auroral field-aligned return currents are investigated, using a multimoment fluid model with anomalous transport coefficients. The macroscopic effects of the electrostatic ion cyclotron (EIC) instability and of an EIC-related anomalous resistivity mechanism which heats the electrons are included in the present version of the model. The responses of the outflowing polar wind plasma to the application of current, with and without instabilities, are exhibited. The simulations show that the electron drift velocity corresponding to a return current of 0.65 micro-A/sq m is above the threshold for EIC waves. Downward electron heat flow competes with upward convection and adiabatic effects to determine the direction of the electron temperature anisotropy. Resistive electron heating lowers the critical drift velocity for marginal EIC stability and leads to enhanced ion heating.

A new approach to adaptive control of manipulators

An approach in which the manipulator inverse is used as a feedforward controller is employed in the adaptive control of manipulators in order to achieve trajectory tracking by the joint angles. The desired trajectory is applied as an input to the feedforward controller, and the controller output is used as the driving torque for the manipulator. An adaptive algorithm obtained from MRAC theory is used to update the controller gains to cope with variations in the manipulator inverse due to changes of the operating point. An adaptive feedback controller and an auxiliary signal enhance closed-loop stability and achieve faster adaptation. Simulation results demonstrate the effectiveness of the proposed control scheme for different reference trajectories, and despite large variations in the payload.

Adaptive control of robotic manipulators

The author presents a novel approach to adaptive control of manipulators to achieve trajectory tracking by the joint angles. The central concept in this approach is the utilization of the manipulator inverse as a feedforward controller. The desired trajectory is applied as an input to the feedforward controller which behaves as the inverse of the manipulator at any operating point; the controller output is used as the driving torque for the manipulator. The controller gains are then updated by an adaptation algorithm derived from MRAC (model reference adaptive control) theory to cope with variations in the manipulator inverse due to changes of the operating point. An adaptive feedback controller and an auxiliary signal are also used to enhance closed-loop stability and to achieve faster adaptation. The proposed control scheme is computationally fast and does not require a priori knowledge of the complex dynamic model or the parameter values of the manipulator or the payload.

PMR Resin Compositions For High Temperatures

Report describes experiments to identify polymer matrix resins suitable for making graphite-fiber laminates used at 700 degree F (371 degree C) in such applications as aircraft engines to achieve higher thrust-to-weight ratios. Two particular high-molecular-weight formulations of PMR (polymerization of monomer reactants) resins most promising. PMR compositions of higher FMW exhibit enhanced thermo-oxidative stability. Formation of high-quality laminates with these compositions requires use of curing pressures higher than those suitable for compositions of lower FMW.

Improved PMR Polyimides For Heat-Stable Laminates

Second-generation PMR-type polyimides (PMR-II polyimides) of enhanced thermo-oxidative stability prepared by substitution of para-aminostyrene (PAS) end caps for nadic-ester (NE) end caps used in prior PMR-II polyimides. Laminates unidirectionally reinforced with graphite fibers and made with PAS-capped resins exhibited thermo-oxidative stabilities significantly greater than those of similar laminates made with NE-capped PMR-II resins. One new laminate exhibited high retention of weight and strength after 1,000 h of exposure to air at 371 degrees C.

A reference model for scientific information interchange

This paper presents an overview of an Information Interchange Reference Model (IIRM) currently being developed by individuals participating in the Consultative Committee for Space Data Systems (CCSDS) Panel 2, the Planetary Data Systems (PDS), and the Committee on Earth Observing Satellites (CEOS). This is an ongoing research activity and is not an official position by these bodies. This reference model provides a framework for describing and assessing current and proposed methodologies for information interchange within and among the space agencies. It is hoped that this model will improve interoperability between the various methodologies. As such, this model attempts to address key information interchange issues as seen by the producers and users of space-related data and to put them into a coherent framework. Information is understood as the knowledge (e.g., the scientific content) represented by data. Therefore, concern is not primarily on mechanisms for transferring data from user to user (e.g., compact disk read-only memory (CD-ROM), wide-area networks, optical tape, and so forth) but on how information is encoded as data and how the information content is maintained with minimal loss or distortion during transmittal. The model assumes open systems, which means that the protocols or methods used should be fully described and the descriptions publicly available. Ideally these protocols are promoted by recognized standards organizations using processes that permit involvement by those most likely to be affected, thereby enhancing the protocol's stability and the likelihood of wide support.

Efficient Sequential Attitude Estimation from Vector Observations

This paper presents a computationally efficient, sequential method for attitude matrix estimation using gyro and vector measurements. The method is based on a recently introduced, minimal-parameter third-order method for solving the orthogonal matrix differential equation in R(sup n). In the three-dimensional case, these third-order attitude parameters can be interpreted as temporal-integrals of the body-frame angular velocity components. A nonlinear algorithm is developed, which uses this minimal set of three parameters to estimate the nine-parameter direction-cosine matrix. Having an extremely simple kinematic equation, these parameters render the resulting estimator highly computationally efficient. An orthogonalization procedure, incorporated into the measurement processing stage, enhances the accuracy and stability of the resulting algorithm, yet retains reasonable simplicity. The performance of the estimator is demonstrated via a Monte Carlo simulation study.

Preservation Methods Utilized for Space Food

Food for manned space flight has been provided by NASA-Johnson Space Center since 1962. The various mission scenarios and space craft designs dictated the type of food preservation methodologies required to meet mission objectives. The preservation techniques used in space flight include freeze-dehydration, thermostabilization, irradiation, freezing and moisture adjustment. Innovative packaging material and techniques enhanced the shelf-stability of the food items. Future space voyages may include extended duration exploration missions requiring new packaging materials and advanced preservation techniques to meet mission goals of up to 5-year shelf-life foods.

Electrochemical Reconstitution of Biomolecules for Applications as Electrocatalysts for the Bionanofuel Cell

Platinum-cored ferritins were synthesized as electrocatalysts by electrochemical biomineralization of immobilized apoferritin with platinum. The platinum cored ferritin was fabricated by exposing the immobilized apoferritin to platinum ions at a reduction potential. On the platinum-cored ferritin, oxygen is reduced to water with four protons and four electrons generated from the anode. The ferritin acts as a nano-scale template, a biocompatible cage, and a separator between the nanoparticles. This results in a smaller catalyst loading of the electrodes for fuel cells or other electrochemical devices. In addition, the catalytic activity of the ferritin-stabilized platinum nanoparticles is enhanced by the large surface area and particle size phenomena. The work presented herein details the immobilization of ferritin with various surface modifications, the electrochemical biomineralization of ferritin with different inorganic cores, and the fabrication of self-assembled 2-D arrays with thiolated ferritin.
