An investigation of a sterile access technique for the repair and adjustment of sterile spacecraft

A description is presented of a unique system for the sterilization and sterile repair of spacecraft and the results of a test program designed to assess the biological integrity and engineering reliability of the system. This trailer-mounted system, designated the model assembly sterilizer for testing (MAST), is capable of the dry-heat sterilization of spacecraft and/or components less than 2.3 meters in diameter at temperatures up to 433 K and the steam sterilization of components less than 0.724 meter in diameter. Sterile access to spacecraft is provided by two tunnel suits, called the bioisolator suit systems (BISS), which are contiguous with the walls of the sterilization chambers. The test program was designed primarily to verify the biological and engineering reliability of the MAST system by processing simulated space hardware. Each test cycle simulated the initial sterilization of a spacecraft, sterile repair of a failed component, removal of the spacecraft from the MAST for mating with the bus, and a sterile recycle repair.

Replacement of Cobalt in Medical Device Sterilization: Current Trends, Opportunities and Barriers to Adoption of X-ray and E-Beam Within the Medical Device Sterilization Market

Radioactive sources, specifically Co-60, are crucially important to the medical device sterilization industry; however, their benefits must be balanced against the need to ensure that the materials are secure. Accelerator-based sterilization methods, including X-ray and E-Beam, can in many cases sufficiently replace Co-60 and ethylene oxide, the dominant modalities currently in use; however, a lack of hands-on experience with accelerators, regulations, and a lack of capacity have hampered faster adoption of these technologies. The market is likely to change, however, given public pressures against ethylene oxide and concerns over the cost and long-term supply chain robustness of Co-60. This may provide an opportunity for growth for accelerator capabilities. Current research at Sandia National Laboratories and Argonne National Laboratory, on behalf of the Department of Energy’s National Nuclear Security Administration Office of Radiological Security, is focused on developing an understanding of the medical device sterilization marketplace, the different modalities that are used to sterilize goods, and the drivers that affect the actors within it. Research at Pacific Northwest National Laboratory, in conjunction with the medical device sterilization industry, has focused on expanding the industry’s understanding of the capabilities of X-ray sterilization and developing data that can inform the industry’s consideration of a transition towards accelerator-based devices.

Sterilization of N95 Respirators via Gamma Radiation: Comparison of Post-sterilization Efficacy

This study evaluated gamma irradiation for sterilization and reuse of two models of N95 respirators after gamma radiation sterilization as a method to increase availability of N95 respirators during a shortage. The Sandia National Laboratories Gamma Irradiation Facility was used to irradiate two different models of N95 filtering facepiece respirators at doses ranging from 0 kGy(tissue) to 50 kGy(tissue). The following tests were used to determine the efficacy of the respirator after irradiation sterilization: Ambient Aerosol Condensation Nuclei Counter Quantitative Fit Test, tensile test, strain cycling, oscillatory dynamic mechanical analysis, microscopic image analysis of fiber layers, and electrostatic field measurements. Both of the respirator models exhibited statistically significant changes after gamma irradiation as shown by the Quantitative Fit Test, electrostatic testing and the aerosol testing. The change in electrostatic charge of the filter was correlated with a reduction in capturing particles near the 200 nm size by approximately 40-50%. Both tested respirators showed statistically significant changes associated with gamma sterilization. However, our results indicate that choices in materials and manufacturing methods to achieve N95 filtration lead to different magnitudes of damage when exposed to gamma radiation at sterilization relevant doses. This damage results in lower filtration performance. While our sample size (2 different types of respirators) was small, we did observe a change in electrostatic properties on a filter layer that coincided with the failure on the Quantitative Fit Test and reduction in aerosol filtering efficiency. Key Words: N95 respirators, respirators, airborne transmission, pandemic prevention, COVID-19, gamma sterilization

Primordial black hole sterile neutrinogenesis: sterile neutrino dark matter production independent of couplings

Abstract Sterile neutrinos (ν s s) are well-motivated and actively searched for hypothetical neutral particles that would mix with the Standard Model active neutrinos. They are considered prime warm dark matter (DM) candidates, typically when their mass is in the keV range, although they can also be hot or cold DM components. We discuss in detail the characteristics and phenomenology ofν s s that minimally couple only to active neutrinos and are produced in the evaporation of early Universe primordial black holes (PBHs), a process we called “PBH sterile neutrinogenesis”. Contrary to the previously studiedν s production mechanisms, this novel mechanism does not depend on the active-sterile mixing. The resultingν s s have a distinctive spectrum and are produced with larger energies than in typical scenarios. This characteristic enablesν s s to be WDM in the unusual 0.3 MeV to 0.3 TeV mass range, if PBHs do not matter-dominate the Universe before evaporating. When PBHs matter-dominate before evaporating, the possible coincidence of induced gravitational waves associated with PBH evaporation and astrophysical X-ray observations fromν s decays constitutes a distinct signature of our scenario.

Comparative Effects of Radiation Sterilization Modality on Medical PVC Tubing

As the production volume of single-use, polymer-based medical devices increases each year, so does the need for throughput capacity to sterilize the devices. Around 80% of radiation-sterilized devices are processed using gamma-ray radiation from industrial cobalt-60 sources. Supply chain, security, and disposal concerns are prompting manufacturers and contract sterilizers to explore alternatives to irradiation using cobalt-60 gamma-rays, such as electron beam (E-beam) and X-ray radiation. Advantages of these ‘machine’ sources over radioisotope-based sources are that they do not decay, a characteristic that requires cobalt-60 to be replaced over time, they can be turned off when not in use, and they do not represent the same level of concern regarding misuse. One of the barriers to direct transition from gamma-ray sterilization to E-beam or X-ray sterilization is concern over the relatively unknown effects of sterilization dose of these alternative modalities on the performance of medical device polymers. Commercially available medical devices sterilized with gamma-ray irradiation exhibit acceptable device function, acceptable mechanical performance, and acceptable discoloration. To be viable, alternate sterilization methods must not only achieve acceptable sterilization, but the process must also result in a product with functional, mechanical, and visual effects acceptable to customers and the regulator. Flexible polyvinyl chloride (PVC) is a plastic material that is commonly found in single-use medical devices and is known for its sensitivity to radiation processing. For instance, transparent PVC is known to yellow upon irradiation. To encouraging sterilization alternatives to cobalt-60 gamma-ray, we directly compared the effects of gamma-ray, E-beam, and X-ray radiation exposure at similar sterilization-relevant doses on key properties of medical grade PVC tubing.

Elucidating the mechanisms of enhanced lignin bioconversion by an alkali sterilization strategy

Biological lignin valorization represents an emerging green approach to upgrade lignin for sustainable and economic biorefineries. However, lignin generally exhibits poor water solubility and inhomogeneous distribution in an aqueous medium, significantly limiting its bioconversion efficiency. Herein, we develop a novel alkali sterilization strategy to effectively enhance the dispersion and fermentation performance of lignin substrates. The colloidal particle size and molecular structure variations of lignin during the sterilization were thoroughly investigated to reveal the mechanisms of enhanced fermentation performance. Results showed that alkali sterilization achieved a completely aseptic effect when mixing lignin medium at an initial pH of 12.7 for 24 h. Dynamic light scattering (DLS) analysis demonstrated that the hydrodynamic volume of colloidal lignin particles decreased by 96.3% by alkali sterilization compared with the conventional thermal sterilization. Moreover, lignin characterizations by nuclear magnetic resonance (NMR) spectroscopy and gel permeation chromatography (GPC) suggested that alkali sterilization modified the lignin molecular structure by generating 50% more hydrophilic carboxyl groups, reducing the weight-average molecular weight (M w ) by 23.0%, and narrowing the molar-mass dispersity (D M ) by 23.8%. The generation of lignin substrates with more uniform distribution and lower molecular weight improved Rhodococcus opacus PD630 cell growth and metabolism. Microbial cell amount, lignin degradation, and lipid production in alkali sterilized medium increased by 309%, 30.3%, and 48.3%, respectively, compared to those in thermally sterilized medium. These results clearly demonstrated that alkali sterilization dramatically improved the lignin bioconversion performance. Furthermore, this work presents a facile and effective sterilization strategy to overcome inhomogeneous lignin distribution in aqueous fermentation media, showing great potentials as a platform technique for promoting biological lignin valorization.

Probing mass orderings in presence of a very light sterile neutrino in a liquid argon detector

Results from experiments like LSND and MiniBooNE hint towards the possible presence of an extra eV scale sterile neutrino. The addition of such a neutrino will significantly impact the standard three flavor neutrino oscillations. In particular, it can give rise to additional degeneracies due to additional sterile parameters. For an eV scale sterile neutrino, the cosmological constraints dictate that the sterile state is heavier than the three active states. However, for lower masses of sterile neutrinos, the sterile state can be lighter than one and/or more of the three states. In such cases, the mass ordering of the sterile neutrinos also becomes unknown, along with the mass ordering of the active states. In this paper, we explore the mass ordering sensitivity in the presence of a sterile neutrino assuming the mass squared difference |Δ 41 | to be in the range 10 –4 –1 eV 2 . We study (i) how the ordering of the active states, i.e. the determination of the sign of Δ 31 gets affected by the presence of a sterile neutrino in the above mass range, (ii) the possible determination of the sign of Δ 41 for Δ 41 in the range 10 –4 –0.1 eV 2 . This analysis is done in the context of a liquid argon detector using beam neutrinos traveling a distance of 1300 km and atmospheric neutrinos that propagate through a distance ranging from 10 – 10000 km, allowing resonant matter effects. Apart from presenting separate results from these sources, we also do a combined study and probe the synergy between these two in giving an enhanced sensitivity.

System for sterilizing objects

A system for producing a stream of humidified sterilizing gas for sterilizing objects such as the water systems of space vehicles and the like includes a source of sterilant gas which is fed to a mixing chamber which has inlet and outlet ports. The level of the water only partially fills the mixing chamber so as to provide an empty space adjacent the top of the chamber. A heater is provided for heating the water in the chamber so as to produce a humidified atmosphere. The sterilant gas is fed through an arcuate shaped tubular member connected to the inlet port of the mixing chamber for producing a vortex type of flow of sterilant gas into the chamber for humidification. A tubular member extends from the mixing chamber for supplying the humidified sterilant gas to the object for being sterilized. Scrubbers are provided for removing the sterilant gas after use.

Ultrashort Pulse Laser Surface Processing Techniques for Sterilization of Metal Surfaces for Planetary Protection

To prevent forward contamination from microbes aboard spacecraft intended to search for extraterrestrial life, there is a need for effective sterilization methods. However, current techniques are both time-consuming and expensive. For example, dry heat sterilization requires removal from the assembly site and several days of treatment. Furthermore, some components such as optics and electronics are not compatible with current sterilization techniques. Here, we report the latest results in our development of a novel femtosecond laser processing technique for the rapid sterilization of spacecraft hardware. Femtosecond lasers produce extremely high photon fluxes (10^29 photons/sec*cm^2, ~0.03 J/cm^2) in extremely short pulses, which can inactivate even stress-tolerant microbial spores with minimal damage to the spacecraft surface. Aluminum coupons were inoculated with specific densities of Bacillus subtilis bacterial endospores. These coupons were treated with various laser illumination parameters. Afterward, metal coupon samples were assayed for viable spores using a polyvinyl alcohol (PVA) peel, serial dilution, and plating for colony-forming units (CFU). Results indicate that with high enough energy density and pulse counts, most bacterial spores are inactivated with minimal damage to the metal. The sterilization is dependent on both the fluence and pulse count. In addition, femtosecond pulses are more effective than longer pulses for inactivation. These experiments have consistently achieved 4-log reduction in viable spores. Sterilization has been achieved on both flat metal coupons and non-flat surfaces with microchannels, with a slight reduction in sterilization efficiency on the uneven surface. The application of air flow during laser processing was also investigated as a way to remove spores that are dislodged from the surface by the laser illumination, which would contribute to the reduction of spacecraft bioburden. With laser processing technology rapidly evolving, our results support the possibility of an extremely rapid, in-situ surface sterilization method for use in spacecraft assembly clean rooms.

Neutrinoless double beta decay versus other probes of heavy sterile neutrinos

We make a comparative study of the neutrinoless double beta decay constraints on heavy sterile neutrinos versus other direct and indirect constraints from both lepton number conserving and violating processes, as a sensitive probe of the extent of lepton number violation and possible interference effects in the sterile sector. We introduce a phenomenological parametrisation of the simplified one-generation seesaw model with one active and two sterile neutrino states in terms of experimentally measurable quantities, such as active-sterile neutrino mixing angles, CP phases, masses and mass splittings. This simple parametrisation enables us to analytically derive a spectrum of possible scenarios between the canonical seesaw with purely Majorana heavy neutrinos and inverse seesaw with pseudo-Dirac ones. We then go on to constrain the simplified parameters of this model from various experiments at the energy, intensity and cosmic frontiers. We emphasise that the constraints from lepton number violating processes strongly depend on the mass splitting between the two sterile states and the relative CP phase between them. This is particularly relevant for neutrinoless double beta decay, which is weakened for small mass splitting and opposite CP parities between the sterile states. On the other hand, neutrinoless double beta decay is especially sensitive for Majorana sterile neutrinos with masses around 0.1-10 GeV.

Sterile neutrinos and neutrinoless double beta decay in effective field theory

We investigate neutrinoless double beta decay (0νββ) in the presence of sterile neutrinos with Majorana mass terms. These gauge-singlet fields are allowed to interact with Standard-Model (SM) fields via renormalizable Yukawa couplings as well as higher-dimensional gauge-invariant operators up to dimension seven in the Standard Model Effective Field Theory extended with sterile neutrinos. At the GeV scale, we use Chiral effective field theory involving sterile neutrinos to connect the operators at the level of quarks and gluons to hadronic interactions involving pions and nucleons. This allows us to derive an expression for 0νββ rates for various isotopes in terms of phase-space factors, hadronic low-energy constants, nuclear matrix elements, the neutrino masses, and the Wilson coefficients of higher-dimensional operators. The required hadronic low-energy constants and nuclear matrix elements depend on the neutrino masses, for which we obtain interpolation formulae grounded in QCD and chiral perturbation theory that improve existing formulae that are only valid in a small regime of neutrino masses. The resulting framework can be used directly to assess the impact of 0νββ experiments on scenarios with light sterile neutrinos and should prove useful in global analyses of sterile-neutrino searches. We per- form several phenomenological studies of 0νββ in the presence of sterile neutrinos with and without higher-dimensional operators. We find that non-standard interactions involving sterile neutrinos have a dramatic impact on 0νββ phenomenology, and next-generation experiments can probe such interactions up to scales of $\mathcal O$ (100) TeV.

Targeted seed EMS mutagenesis reveals a basic helix–loop–helix transcription factor underlying male sterility in sorghum

Abstract Forward genetic screens of mutant populations are fundamental for functional genomics studies. However, isolating independent mutant alleles to molecularly identify causal genes is challenging in species recalcitrant to genetic manipulation. Here, we demonstrate that classic seed ethyl methanesulfonate (EMS) mutagenesis coupled with genome sequencing can overcome this limitation in sorghum. We used this method to generate new mutant alleles of sorghum MALE STERILE 8 (MS8) and identified the causal locus for the ms8 phenotype as Sobic.004G270900, which encodes the sorghum ortholog of maize bhlh122, a basic helix–loop–helix (bHLH) transcription factor required for male fertility in maize. Bulked segregant analysis mapped ms8-1 to a region on chromosome 4 containing Sobic.004G270900. Seeds from heterozygous MS8/ms8-1 plants were mutagenized and screened for chimeric inflorescences containing sectors with white, sterile anthers resembling the ms8-1 homozygous phenotype. DNA sequencing of sterile and fertile sectors from a single chimeric inflorescence revealed two mutations in Sobic.004G270900 within the sterile sector, but not the fertile sector. Isolation of this loss-of-function allele (ms8-2) established Sobic.004G270900 as the causative locus for male sterility in the ms8 mutant. We generated additional alleles of MS8 in a different genetic background using CRISPR/Cas9-based gene editing, where deletions in Sobic.004G270900 also resulted in male sterility. Our work identified a gene underlying male sterility in sorghum and provides a novel and straightforward genetic tool for researchers who lack access to advanced transformation facilities to validate gene candidates. Unlike gene editing, no prior knowledge of candidate genes is required for targeted seed EMS mutagenesis to aid identification of causal loci.

A Search for Sterile Neutrino Dark Matter (Final Report)

Decades of dark matter searches with increasingly higher sensitivity have so far not been able to detect the most-favored hypothetical dark matter particles. This has increased interest in sterile neutrinos with masses in the kilo-electronvolt (keV) range as possible dark matter candidates. We have established a and new and world-leading approach to search for keV-scale sterile neutrinos with superconducting radiation detectors. For this, we have implanted radioactive beryllium-7 (Be-7) nuclei into our superconducting tunnel junction (STJ) detectors and measured the recoil energy of the lithium-7 (Li-7) daughter nucleus with high accuracy. If the associated electron neutrino wave function contains some admixture of a keV sterile flavor, the Li-7 recoil energy will be shifted and produce an additional peak in the spectrum whose energy depends on the sterile neutrino mass. While we have not observed any evidence for sterile neutrinos in these precision decay experiments, the experiment has increased the sensitivity of sterile neutrino searches by a factor of ~10 over competing approaches in the mass range from ~10 to ~800 keV. The next generation of STJ detectors that we have developed as part of this project will be able further increase the sensitivity of the sterile neutrino search by at least another two orders of magnitude.
