In-flight positional and energy use data set of a DJI Matrice 100 quadcopter for small package delivery

Abstract We autonomously directed a small quadcopter package delivery Uncrewed Aerial Vehicle (UAV) or “drone” to take off, fly a specified route, and land for a total of 209 flights while varying a set of operational parameters. The vehicle was equipped with onboard sensors, including GPS, IMU, voltage and current sensors, and an ultrasonic anemometer, to collect high-resolution data on the inertial states, wind speed, and power consumption. Operational parameters, such as commanded ground speed, payload, and cruise altitude, were varied for each flight. This large data set has a total flight time of 10 hours and 45 minutes and was collected from April to October of 2019 covering a total distance of approximately 65 kilometers. The data collected were validated by comparing flights with similar operational parameters. We believe these data will be of great interest to the research and industrial communities, who can use the data to improve UAV designs, safety, and energy efficiency, as well as advance the physical understanding of in-flight operations for package delivery drones.

Packaged delivery of CRISPR–Cas9 ribonucleoproteins accelerates genome editing

Effective genome editing requires a sufficient dose of CRISPR–Cas9 ribonucleoproteins (RNPs) to enter the target cell while minimizing immune responses, off-target editing, and cytotoxicity. Clinical use of Cas9 RNPs currently entails electroporation into cells ex vivo, but no systematic comparison of this method to packaged RNP delivery has been made. Here we compared two delivery strategies, electroporation and enveloped delivery vehicles (EDVs), to investigate the Cas9 dosage requirements for genome editing. Using fluorescence correlation spectroscopy, we determined that >1300 Cas9 RNPs per nucleus are typically required for productive genome editing. EDV-mediated editing was >30-fold more efficient than electroporation, and editing occurs at least 2-fold faster for EDV delivery at comparable total Cas9 RNP doses. We hypothesize that differences in efficacy between these methods result in part from the increased duration of RNP nuclear residence resulting from EDV delivery. Our results directly compare RNP delivery strategies, showing that packaged delivery could dramatically reduce the amount of CRISPR–Cas9 RNPs required for experimental or clinical genome editing.

Medium and Heavy Duty Vehicle Powertrain Electrification and Fleet Demonstration

The Contract Award Grant # EE0007513 dated 2 June, 2016 is for the design, development and demonstration of medium duty package delivery vehicles that will result in a 100% increase in fuel economy as measured in miles per gallon. The project was proposed in 3 phases over two budget periods. The Phases were: 1. Modeling and Simulation – This phase was completed and formed the basis of the specifications for the design and build of the physical systems. The modeling and simulation was conducted by AVL and used real world package delivery routes that AVL mapped. The motors, battery pack, range extender and balance of plant support systems were modeled and iterated until a system with the desired fuel economy improvement was realized. Early design of the major subsystems using the actual vehicle space constraints was also completed. This phase was successfully completed in May of 2017. 2. 1st Build and FE Confirmation - This phase was completed and resulted in the final design, release, procurement and build of all major vehicle systems for commissioning into the first vehicle. The work was a partnered effort between McLaren and AVL. McLaren built the eAxle system while AVL built the range extender and integrated the range extender, battery and balance of plant sub-systems into the first vehicle with a control system to make it operate. Prior to the end of this phase a real world HIL system test gave evidence that the fuel economy improvement would be achieved in the real world. This phase was exited with a 7 month delay due mostly to procurement and supply base issues and ended in May, 2018. 3. Final Build and Demonstration - This phase included the final build and commissioning of the four program vehicles and the operation of a demonstration on a set of routes near San Diego California. During this phase, a multitude of issues arose in different systems in the vehicle. These issue resolution exercises were quite complex and caused repeated delays but were all successfully completed. It should also be noted that an early commercialization analysis resulted in the conclusion that the complexity of the system would not make it cost effective, leading to the start of a follow-on design that used newly available component technologies to specify a more cost effective system. This design will be repurposed and put into demonstration on a separately (non-DOE) funded program. It was decided and agreed with DOE that this phase should be considered completed. The knowledge and know-how gained on this program has been incorporated into and transferred to the new program. This phase and the entire program was considered closed on June 1, 2021.

Medium-Duty Delivery Truck Integrated Bidirectional Wireless Power Transfer System With Grid and Stationary Energy Storage System Connectivity

Electric vehicles (EVs) can provide power to the grid or buildings similar to distributed energy resources (DER) for energy balancing applications or optimizing the operation of the microgrids in harmony with the other DER assets. This article presents the operating modes of a bidirectional wireless power transfer (WPT) system designed for a medium-duty package delivery vehicle. The WPT system designed for this study can transfer 20 kW of power across 11 in of air gap using custom-designed double-D (DD) couplers with LCC–LCC tuning networks. The proposed system utilizes a 480-V three-phase grid connection, a plug-in hybrid delivery truck with bidirectional WPT, and a stationary energy storage system (SESS) that can be connected to the primary-side dc link. Due to the differences in primary and secondary dc bus voltages, and considering the voltage of the SESS, asymmetric voltage gains were used in the system. Sensitivity analyses of this system with respect to these voltage levels are presented. Five different operating modes of the grid, SESS, and the EV battery are investigated with experimental results. Control algorithms are described for grid-to-vehicle (G2V) and vehicle-to-grid (V2G) operating modes. A bidirectional WPT system is operated with a power factor of 0.99 on the grid side in every operating mode. The EV battery was charged with 20.3 kW with an overall efficiency of 93.02% in the G2V operating mode. Finally, in the V2G operating mode, the WPT system provided 12.82 kW of power back to the grid with an overall efficiency of 89.08%.

Droning on to Delivery: Examining the Energy Impacts of Using Drones for Moving Goods

The demand for fast, localized delivery has grown significantly in recent years. Whether for cheesy snacks, prepared food, medical solutions, or business deliveries, fast and efficient delivery is increasingly a demand and differentiator. Drone delivery in the freight sector offers to revolutionize last-mile logistics and improve services. This research analyzes the impacts of drone energy for delivery operations, aiming to compare various types of drones (large and small, rotary and VTOL) and various types of business methods. The project executes novel open-air and laboratory-based testing to look at the impacts of weights, operations, temperatures and weather conditions. The study combines this real-world experimental data with fleet optimization mathematical models to assess energy consumption of different scenarios as well as examine the minimum fleet size and additional battery requirements. These fleet optimization models compare energy between types of deployments with existing delivery methods. The model was also extended to look at mixed fleets of aerial drones and ground vehicles to accommodate different restrictions on drones or when weather prohibits their use. The analysis provided insights into factors such as drone design, payload weight, flight distance, weather conditions and operational parameters and impacts of each. It showed how to combine different types of vehicles to reduce energy and improve services. And it showed that drone speed, routing restrictions, and unfavorable weather significantly influence energy consumption. Our open experiment data and optimization models can assist stakeholders and industry in understanding drone package delivery and offers keys to improving deployment.
