scifi-ATAC-seq: massive-scale single-cell chromatin accessibility sequencing using combinatorial fluidic indexing

Abstract Single-cell ATAC-seq has emerged as a powerful approach for revealing candidate cis-regulatory elements genome-wide at cell-type resolution. However, current single-cell methods suffer from limited throughput and high costs. Here, we present a novel technique called scifi-ATAC-seq, single-cell combinatorial fluidic indexing ATAC-sequencing, which combines a barcoded Tn5 pre-indexing step with droplet-based single-cell ATAC-seq using the 10X Genomics platform. With scifi-ATAC-seq, up to 200,000 nuclei across multiple samples can be indexed in a single emulsion reaction, representing an approximately 20-fold increase in throughput compared to the standard 10X Genomics workflow.

Unattended Monitoring of Uranium Holdup with Scintillating Fibers

This work evaluated the use of scintillating fiber (SciFi) radiation sensors as a cost-effective, unattended monitoring technique for uranium holdup accumulation in reprocessing equipment. With ruggedized packaging and an outer diameter of less than 1 cm, scintillating fibers offer deployment flexibility. Additionally, plastic scintillating fibers coupled with standard optical fibers provide an affordable option to monitor locations kilometers away from a localized control room or data acquisition (DAQ) system. These combinations of characteristics along with their relatively low manufacturing costs make scintillating fibers ideal for installation at key measurement points such as pipes, valves, and filtration systems.

An Imaging Calorimeter for Access-Concept Study

A mission concept study to define the "Advanced Cosmic-ray Composition Experiment for Space Station (ACCESS)" was sponsored by the National Aeronautics and Space Administration (NASA). The ACCESS instrument complement contains a transition radiation detector and an ionization calorimeter to measure tile spectrum of protons, helium, and heavier nuclei up to approximately 10(exp 15) eV to search for the limit of S/N shock wave acceleration, or evidence for other explanations of the spectra. Several calorimeter configurations have been studied, including the "baseline" totally active bismuth germanate instrument and sampling calorimeters utilizing various detectors. The Imaging Calorimeter for ACCESS (ICA) concept comprises a carbon target and a calorimeter using a high atomic number absorber sampled approximately each radiation length (rl) by thin scintillating fiber (SCIFI) detectors. The main features and options of the ICA instrument configuration are described in this paper. Since direct calibration is not possible over most of the energy range, the best approach must be decided from simulations of calorimeter performance extrapolated from CERN calibrations at 0.375 TeV. This paper presents results from the ICA simulations study.

TRIPWIRE – Deployment and Field-Testing Report

TRIPWIRE is a multi-modal system for containment verification in inaccessible radiological and nuclear waste repositories. The sensing modes are focused on radiation, electromagnetic and vibration. The radiation detection component has been developed over the last three years at Idaho National Laboratory (INL). This development has focused on the fabrication, characterization, and testing of long-length (greater than 10-m) plastic scintillating fibers. This fiscal year, the focus of TRIPWIRE has been on field testing and demonstration of the radiation sensing component in a relevant environment. After reviewing the static storage facilities at INL, the Radioactive Scrap Waste Facility (RSWF) south storage area outside the Materials and Fuels Complex (MFC) was selected due to its periodic changes in configuration and inventory. This report summarizes the research outcomes for the work performed in fiscal year (FY) 2023.
