Energy Snapshot - Micronesia

This profile provides a snapshot of the energy landscape of the Federated States of Micronesia (FSM), a sovereign nation and U.S.-associated state in the western Pacific Ocean. The FSM is made up of more than 600 islands, which presents a significant challenge of delivering electricity to people living on outer islands. The country is striving to overcome electricity access needs, reduce high energy costs, and ensure energy security. Currently, almost all of the electricity produced in Micronesia is dependent upon imported petroleum based fossil fuels, with some solar photovoltaic systems in operation.

Tolyporphins A–R, unusual tetrapyrrole macrocycles in a cyanobacterium from Micronesia, assessed quantitatively from the culture HT-58-2

The ‘pigments of life’ family has been enlarged by the discovery of the tetrapyrrole macrocycles dubbed tolyporphins, which are present in a cyanobacterial culture (HT-58-2). Of the 18 known tolyporphins, fourteen (A–J and L–O) are dioxobacteriochlorins, three (K, Q and R) are oxochlorins, and one (P) is a porphyrin. The members with a given chromophore absorb similarly but not identically with each other, presenting the problem of absorption spectral aliasing. Tolyporphins G and H are isomers, as are tolyporphins L–O, given the nature of the peripheral ( C -glycoside, –OH, or –OAc) substituents, presenting the (isobaric) problem of mass spectrometric analysis. The distribution of tolyporphin members is known to change over time and under different growth conditions, however, the complexity of the tolyporphins mixture presents challenges toward characterization of the underlying factors that alter such distributions. Liquid chromatography-mass spectrometry with dynamic multiple reaction monitoring (LC-MS-dMRM) was employed to characterize the distribution of tolyporphins under distinct growth media, using two semisynthetic standards, tolyporphin A O,O -dibutyrate and tolyporphin D O,O,O,O -tetrabutyrate. Growth in media containing both ammonium and nitrate salts versus nitrate alone affords increased production of tolyporphins. The absorption spectra of tolyporphins A–R have been reevaluated with regards to molar absorption coefficients, are compared with those of selected tetrapyrrole macrocycles to better understand the effects of chromophores and substituents, and are made available here in print and digital forms for comparison with isolated tolyporphins and mixtures thereof. Together, the work provides the foundation for quantitative assessment of these unusual natural products.

Fluorescence Assay for Tolyporphins Amidst Abundant Chlorophyll in Crude Cyanobacterial Extracts

Abstract Tolyporphins are distinctive tetrapyrrole natural products found singularly in a filamentous cyanobacterial‐microbial holobiont (termed HT‐58‐2) from Micronesia. The absorption and fluorescence features of tolyporphins resemble those of chlorophyll a , complicating direct analysis of culture samples. Treatment of the crude (unfractionated) organic extract (CH 2 Cl 2 /2‐propanol, 1:1) of HT‐58‐2 cultures with NaBH 4 in methanol causes reduction of the peripheral ketone auxochromes, whereupon tolyporphins (predominantly 7,17‐dioxobacteriochlorins) exhibit a bathochromic shift ( λ abs ˜ 676 → ˜ 700 nm) and chlorophyll a (a 13 1 ‐oxochlorin) exhibits a hypsochromic shift ( λ abs 665 → 634 nm). Fluorescence excitation spectroscopy (at 368 and 491 nm with λ em 710 nm) enabled detection of reduced tolyporphins amidst abundant reduced chlorophyll a (1:19 ratio), a detection sensitivity >5 times that without reduction. The resulting assay combines simple sample preparation from non‐axenic cultures at microscale quantities (2 mL, 2 μ m ), absence of any fractionation procedures, and fluorescence detection. Tolyporphins were readily detected in cultures of HT‐58‐2 at reasonable growth periods in the absence of environmental stressors, which was not possible previously.

Energy Snapshot - Palau

This profile provides a snapshot of the energy landscape of Palau, an independent island nation geographically located in the Micronesia region. Over 97% of the island's electricity production is dependent on imported fossil fuels, primarily diesel. Palau is aiming for 45% renewable energy generation by 2025, and is striving to overcome technological, financial, and institutional capacity challenges to meet this goal.

Energy Snapshot - Marshall Islands

This profile provides a snapshot of the energy landscape of the Republic of the Marshall Islands, an island country and a United States associated state near the equator in the Pacific Ocean. Geographically, the country is part of the larger island group of Micronesia. The Marshall Islands electricity rates for residential customers average $0.36 U.S. dollars (USD) per kilowatt-hour (kWh), nearly 3 times the average U.S. residential rate of $0.13 USD/kWh.
