FCIC Task X - Project Management and Consortium Overview

The objective of the FCIC Program Management Task is to provide scientific direction and leadership to the nine participating labs, and to provide robust project management to ensure robust operational planning and execution. The FCIC directly supports the BETO portfolio by focusing on feedstock variability across the bioenergy value chain, and the Project Management Task ensures the smooth operation of the FCIC, making sure that the 9 independent projects with different objectives have a common focus - understanding and mitigating the impacts of feedstock variability across the value chain.

Overview and Lessons From the Preclinical Chemoradiotherapy Testing Consortium

In the current molecular-targeted cancer treatment era, many new agents are being developed so that optimizing therapy with a combination of radiation and drugs is complex. The use of emerging laboratory technologies to further biological understanding of drug-radiation mechanisms of action will enhance the efficiency of the progression from preclinical studies to clinical trials. In 2017, the National Cancer Institute (NCI) solicited proposals through PAR 16-111 to conduct preclinical research combining targeted anticancer agents in the Cancer Therapy Evaluation Program's portfolio with chemoradiation.

Brain ageing in schizophrenia: evidence from 26 international cohorts via the ENIGMA Schizophrenia consortium

Schizophrenia (SZ) is associated with an increased risk of life-long cognitive impairments, age-related chronic disease, and premature mortality. We investigated evidence for advanced brain ageing in adult SZ patients, and whether this was associated with clinical characteristics in a prospective meta-analytic study conducted by the ENIGMA Schizophrenia Working Group. The study included data from 26 cohorts worldwide, with a total of 2803 SZ patients (mean age 34.2 years; range 18–72 years; 67% male) and 2598 healthy controls (mean age 33.8 years, range 18–73 years, 55% male). Brain-predicted age was individually estimated using a model trained on independent data based on 68 measures of cortical thickness and surface area, 7 subcortical volumes, lateral ventricular volumes and total intracranial volume, all derived from T1-weighted brain magnetic resonance imaging (MRI) scans. Deviations from a healthy brain ageing trajectory were assessed by the difference between brain-predicted age and chronological age (brain-predicted age difference [brain-PAD]). On average, SZ patients showed a higher brain-PAD of +3.55 years (95% CI: 2.91, 4.19; I 2 = 57.53%) compared to controls, after adjusting for age, sex and site (Cohen’s d = 0.48). Among SZ patients, brain-PAD was not associated with specific clinical characteristics (age of onset, duration of illness, symptom severity, or antipsychotic use and dose). This large-scale collaborative study suggests advanced structural brain ageing in SZ. Longitudinal studies of SZ and a range of mental and somatic health outcomes will help to further evaluate the clinical implications of increased brain-PAD and its ability to be influenced by interventions.

Mapping microhabitats of lignocellulose decomposition by a microbial consortium

The leaf-cutter ant fungal garden ecosystem is a naturally evolved model system for efficient plant biomass degradation. Degradation processes mediated by the symbiotic fungus Leucoagaricus gongylophorus are difficult to characterize due to dynamic metabolisms and spatial complexity of the system. Herein, we performed microscale imaging across 12-µm-thick adjacent sections of Atta cephalotes fungal gardens and applied a metabolome-informed proteome imaging approach to map lignin degradation. This approach combines two spatial multiomics mass spectrometry modalities that enabled us to visualize colocalized metabolites and proteins across and through the fungal garden. Spatially profiled metabolites revealed an accumulation of lignin-related products, outlining morphologically unique lignin microhabitats. Metaproteomic analyses of these microhabitats revealed carbohydrate-degrading enzymes, indicating a prominent fungal role in lignocellulose decomposition. Integration of metabolome-informed proteome imaging data provides a comprehensive view of underlying biological pathways to inform our understanding of metabolic fungal pathways in plant matter degradation within the micrometer-scale environment.

Large-scale analysis of structural brain asymmetries in schizophrenia via the ENIGMA consortium

Left–right asymmetry is an important organizing feature of the healthy brain that may be altered in schizophrenia, but most studies have used relatively small samples and heterogeneous approaches, resulting in equivocal findings. We carried out the largest case–control study of structural brain asymmetries in schizophrenia, with MRI data from 5,080 affected individuals and 6,015 controls across 46 datasets, using a single image analysis protocol. Asymmetry indexes were calculated for global and regional cortical thickness, surface area, and subcortical volume measures. Differences of asymmetry were calculated between affected individuals and controls per dataset, and effect sizes were meta-analyzed across datasets. Small average case–control differences were observed for thickness asymmetries of the rostral anterior cingulate and the middle temporal gyrus, both driven by thinner left-hemispheric cortices in schizophrenia. Analyses of these asymmetries with respect to the use of antipsychotic medication and other clinical variables did not show any significant associations. Assessment of age- and sex-specific effects revealed a stronger average leftward asymmetry of pallidum volume between older cases and controls. Case–control differences in a multivariate context were assessed in a subset of the data (N = 2,029), which revealed that 7% of the variance across all structural asymmetries was explained by case–control status. Subtle case–control differences of brain macrostructural asymmetry may reflect differences at the molecular, cytoarchitectonic, or circuit levels that have functional relevance for the disorder. Reduced left middle temporal cortical thickness is consistent with altered left-hemisphere language network organization in schizophrenia.

The U.S. High-Performance Computing Consortium in the Fight Against COVID-19

U.S. computing leaders, including Department of Energy National Laboratories, have partnered with universities, government agencies, and the private sector to research responses to COVID-19, providing an unprecedented collection of resources that include some of the fastest computers in the world. For HPC users, these leadership machines will drive the AI to accelerate the discovery of promising treatments, enable at-scale simulations to understand the virus’s protein structure and attack mechanisms, and help inform policymakers to deploy resources effectively.

Heliostat Consortium: Gap Analysis on State of the Art in Wind Load Design

Wind loads are a major driver of heliostat cost. Standardized methods and tools are needed for a more detailed understanding of the static and dynamic loads of a heliostat design. This will enable cost reduction of wind-dependent heliostats to avoid unnecessarily conservative heliostat designs and increase field efficiency and reliability to reduce the risk of component failures due to high-wind events. Gaps related to wind load include lack of site characterization for wind measurements, insufficient critical load cases for heliostat design, insufficient understanding of turbulence impacts on heliostat tracking error, lack of knowledge on wind load under various heliostat array configurations, and underexplored heliostat field wind-load reduction and operating strategies. Recommended pathway forward is to develop wind load and site characterization guidelines for heliostat design and develop heliostat field wind-load models with optical performance impacts.

NA-22 Write for Consortium Efforts

We are asked to look into the improvements for the measurement of Pu of a variety of material types (MT): MT41 (Pu-242, 20 – 60% enriched), MT42 (Pu-242 > 60% enrichment), and MT-82 (Np-237). Many of these are legacy items that were packaged years ago (Fig 1&2), and when the items are re-packaged for long-term storage, they must be re-measured using either NDA (nondestructive analysis methods or radiochemical methods. It is of interest to improve the NDA and radiochemistry to reduce the un-certainty in the measurements, as in the case of larger items (250 g to 2 kg items) the uncertainty in measurements makes it difficult to certify the mass of the material down to the 0.5 g accountability requirement per DOE orders by only doing NDA measurement. It would be of interest to look into methods to improve the detection of these materials, which would not only benefit the weapon manufacturing community, but also has the ability to significantly improve the non-proliferation community.

ElectroCat (Electrocatalysis Consortium) [Slides]

ElectroCat was created as part of the USDOE Energy Materials Network in February 2016. The mission of the collaboration is to develop and implement PGM-free catalysts and electrodes by streamlining access to unique synthesis and characterization tools across national labs, developing missing strategic capabilities, and curating a public database of information. This presentation provides an overview of their work.
