Omics-Lethal Human Viruses, Ebola Experiment EHUVEC001

The purpose of this experiment was to evaluate the human host response to Zaire Ebola wild-type virus and mutant virus infection in VP30 expression background. Samples were obtained from human umbilical cord endothelial cells (HUVEC) infected with wild-type Zaire Ebola virus in the ΔVP30 background (deltaVP30-WT) and mutant lacking the mucin domain (deltaVP30-deltamucin) encoding a glycoprotein lacking the mucin domain for mRNA transcriptome expression analysis. Secondary host-associated viral dataset downloads contain one or more statistically processed (normalization data transformation) quantitative dataset collections resulting in qualitative expression analyses of primary host-pathogen experimental study designs. Transcriptomics dataset downloads have a direct relationship to a primary sample submission corresponding to a specific Ebola virus infection.

Physicochemical and biological characterization of a bispecific antibody in a CrossMab/KIH format that targets EGFR and VEGF-A

Introduction Bispecific antibodies (BsAbs) are a class of antibody therapeutics engineered in various molecular formats to bind two distinct antigens and potentially mediate multiple biological effects. These molecular formats are tailored to mediate specific mechanisms of action and possess unique physicochemical and biological properties that are necessary to assure product quality. In ovarian cancer (OC), both EGFR- and VEGF-A-mediated signaling pathways are often upregulated and cooperate to promote tumor growth and angiogenesis. Thus, inhibiting of EGFR- and VEGF-A pathways with a BsAb may provide synergistic anti-tumor activity. Methods Using publicly available sequences and applying immunoglobulin domain crossover (CrossMab) and knobs-into-holes (KIH) technologies, we generated a BsAb to simultaneously bind EGFR and VEGF-A (designated as anti-EGFR/VEGF-A BsAb). This BsAb served as a model for physiochemical and biological characterization of quality attributes that would be critical for the BsAb’s mechanisms of action. Our goal was to gain fundamental insights into BsAbs designed to target a receptor with one arm and a soluble ligand with the other, to support bioassay development and inform quality control strategies. Results Our data demonstrated that the CrossMab/KIH platform successfully produced a correctly assembled BsAb during cell culture. Characterization confirmed that the anti-EGFR/VEGF-A BsAb bound both EGFR and VEGF-A with comparable activity and affinity to the respective parental monoclonal antibodies. Functionally, the BsAb disrupted both EGF/EGFR and VEGF-A/VEGFR2 signaling pathways in OC and human umbilical vein endothelial cell (HUVEC) models. Furthermore, the BsAb effectively blocked angiogenic signaling driven by VEGF-A secreted from OC cells in a paracrine manner. Discussion Based on the combinatorial mechanism of action and our characterization findings, we concluded that two or more bioassays may be needed to accurately assess the activity of both arms of this type of BsAb.

Defining Infrastructure Feasibility for Hub-Scale Offshore Atlantic Carbon Storage in the Northeastern United States

In the Northeast U.S., deep rock formations along the Atlantic outer continental shelf may have the potential to sequester 150–1136 million metric tons of CO2. However, the design and infrastructure necessary to develop offshore carbon storage in this region is not well defined because there has been little oil and gas exploration and no commercial production. Consequently, an infrastructure feasibility design was completed for a hub-scale offshore CO2 storage system along the Northeast U.S. Atlantic. The design included development of a detailed, site-specific geological model for a location near the Great Stone Dome geological structure in the Baltimore Canyon Trough off the coast of Delaware, Maryland, and New Jersey. A field injection system topology design was completed to portray a design with eight wells in two clusters connected by central manifolds. Reservoir simulations were completed for the injection system that showed the hub may be able to inject 17 million metric tons (MMT) of CO2 per year for thirty years, but injection rates varied substantially across the eight wells. A CO2 pipeline design determined feasible routes from the east coast shoreline to the injection field. Finally, the CO2 injection system design included subsea injection trees, manifolds, and power umbilicals. This is the first study to define large-scale carbon storage design and infrastructure options for the offshore Atlantic, which can help to progress this region towards field characterization and early-mover deployment for future decarbonization in the region.

Experiment D016: Power tool evaluation

Power tool evaluation was not attempted during the Gemini 11 mission because of the premature termination of the on-umbilical extravehicular activity (EVA). The experiment had been scheduled for the Gemini 8 mission, but early mission termination prevented completion of the experiment. However, the objective, equipment description, and experimental precedures are described briefly in this report.

Regenerable thermal control and carbon dioxide control techniques for use in advanced extravehicular protective systems

The most promising closed CO2 control concept identified by this study is the solid pellet, Mg(OH2)2 system. Two promising approaches to closed thermal control were identified. The AHS system uses modular fusible heat sinks, with a contingency evaporative mode, to allow maximum EVA mobility. The AHS/refrigerator top-off subsystem requires an umbilical to minimize expendables, but less EVA time is used to operate the system, since there is no requirement to change modules. Both of these subsystems are thought to be practical solutions to the problem of providing closed heat rejection for an EVA system.

Atmospheric electricity criteria guidelines for use in aerospace vehicle development

Lightning has always been of concern for aerospace vehicle ground activities. The unexpected triggering of lightning discharges by the Apollo 12 space vehicle shortly after launch and the more recent repeated lightning strikes to the launch umbilical tower while the Apollo 15 space vehicle was being readied for launch have renewed interest in studies of atmospheric electricity as it relates to space vehicle missions. The material presented reflects some of the results of these studies with regard to updating the current criteria guidelines.

Physiology of chimpanzees in orbit. Part 2: Interface document

Interface requirements are presented for the design and development of an earth orbiting experiment to be known as POCO, Physiology of Chimpanzees in Orbit. The POCO experiment may be designed to operate within an orbiting space station (provided artificial gravity measures are not employed), a Saturn 4-B workshop, an Apollo command module or service module, a Saturn-1B spacecraft LM adapter, or aboard one of the presently conceived appendages connected by an umbilical to a space station. This document sets forth the experiment definition and requirements and describes the hardware under development to accomplish these objectives.

Skylab Rescue Space Vehicle OAT No. 1 Plugs in Test

A test is described which demonstrates the compatibility of the Skylab Rescue Space Vehicle systems, the ground support equipment, and off-site support facilities by proceeding through a simulated launch countdown, liftoff, and flight. The functions of propellant loading, umbilical ejection, holddown arm release, service arm retraction, liftoff, and inflight separation are simulated. An external power source supplies transfer power to internal, and instrument unit commands are simulated by the digital command system. The test outline is presented along with a list of references, intercommunications information, radio frequency matrix, and interface control chart.
