Optimizing Single Nuclei Sequencing of Brain Samples From Space Flown Mice Across Age and Strain

The NASA GeneLab Sample Processing Laboratory offers high-throughput sequencing services to NASA-funded space biology researchers. Space biology studies have specific challenges such as low sample numbers, introducing susceptibility to batch effects from sample handling. These issues are compounded by complex protocols such as single-nuclei isolation and sequencing, which has recently become an attractive methodology for assessing the cellular diversity within spaceflight samples. High quality single-nuclei sequencing requires reproducible protocols to dissociate tissue and generate clean suspension of intact single nuclei. Producing single-nuclei suspension from brain tissue is particularly challenging due to cell type heterogeneity and the myelin sheath that carries over into the nuclei suspension as debris. Current procedures tend to be time consuming and sometimes include steps that can alter gene expression and create cell-type bias. Commercially available nuclei isolation kits, such as the 10X Genomics nuclei isolation kit, offers a streamlined way to process samples for nuclei isolation, thereby minimizing batch effects and enabling reproducibility. In this study, we report on the performance of the 10X Genomics nuclei isolation kit and Chromium Next GEM Single Cell Multiome ATAC + Gene Expression kit to generate sequencing libraries from space-flown mouse brain samples. Single nuclei sequencing was performed on frozen mouse brain tissue from two spaceflight missions, Rodent Research-10 (RR-10) and RR Reference Mission-2 (RRRM-2). RR-10 mice were female B6129SF2/J, euthanized at 18-19 weeks whereas RRRM-2 mice were female C57BL/6NTac, euthanized at 20 or 37 weeks. Sequencing data was processed using standard GeneLab data processing pipelines. We report evaluation of the performance of the 10X Genomics nuclei isolation kit for spaceflight samples from mouse brain, and evaluation of reproducibility across different mouse strains and age groups. We also report preliminary scientific results including cell type inference, cell clustering, and differentially expressed genes and pathways between spaceflight and ground control samples.

Long-term effects of acute irradiation and isolation on crew age-matched mice

The future of space missions beyond low-Earth orbit presents exciting opportunities but may come with formidable health challenges. Astronauts face a unique combination of space stressors capable of exerting effects on the central nervous system (CNS) with possible sex-dependent differences. The interaction of these combined spaceflight stressors may induce oxidative stress, thereby altering brain integrity and behavioral performance. Understanding these changes is critical for safeguarding astronaut health and enabling successful exploration. By understanding the physiological responses to the combined effects of ionizing radiation (IR) and social isolation in the brain through immunohistochemistry (IHC), proteomics and cytokine expression analyses, we aim to identify CNS pathways ripe for countermeasure development to mitigate adverse brain changes with future deep space exploration. Here, we study the combined effects of simulated 5-ion galactic cosmic radiation (GCRsim) at acute dosage (5, 15, and 50 cGy) and social isolation on crew age-matched male and female mice at 124 days post-irradiation. We conducted analyses of hippocampal cytokine biomarkers and proteomic profiling on hippocampal lysates. Our findings reveal sex-specific regulation of hippocampal cytokines and proteomic profiles. Ongoing immunohistochemistry findings on the hippocampus will provide qualitative information regarding key protein expression and cellular changes such as microglial activation, blood brain barrier integrity, and neuroinflammation. With future plans for long-duration space missions, such as Mars exploration, understanding CNS changes over extended periods in mice can provide insights into potential long-term health impacts on astronauts. This project will provide us with valuable insights into sex-differences in neurobiological processes and disease mechanisms impacting men and women in space.

X-Ray and Near-Infrared Observations of the Middle-aged Pulsar B1055-52, Its Multiwavelength Spectrum, and Proper Motion

Previous observations of the middle-aged γ-ray, X-ray, and radio pulsar B1055–52 indicated some peculiarities, such as a suspected changing of the X-ray flux and spectral parameters, a large excess of the alleged thermal component of the UV spectrum over the Rayleigh–Jeans extension of the X-ray thermal spectrum, and a possible double break in the nonthermal spectral component between the optical and X-ray bands. We observed PSR B1055–52 with the XMM-Newton observatory in X-rays and the Hubble Space Telescope in near-infrared (NIR). The analysis of the XMM-Newton observations does not support the notion of long-term changes in the X-ray flux and broadband X-ray spectrum of the pulsar. Using an observing mode less affected by background noise than the previous XMM-Newton observations, we constrain the power-law (PL) spectral index as X 0.57 0.25 0.26 a = - - + (Fν ∝ ν α ) in the energy band 3–10 keV. From the NIR–optical data we obtain a PL slope αO = −0.24 ± 0.10 for the color index E(B − V ) = 0.03 mag. The slopes and fluxes of the NIR–optical and X-ray nonthermal spectra suggest that the NIR through X-ray emission can be described by the same PL and is generated by the same mechanism, unlike the pulsar’s γ-ray emission. The excess of the UV thermal component over the extension of the X-ray thermal component became smaller but did not disappear, indicating a nonuniformity of the bulk surface temperature. The NIR data also enable us to measure the proper motion accurately, with values μα = 47.5 ± 0.7 mas yr−1 and μδ = −8.7 ± 0.7 mas yr−1 .

Impact of Tree Age and Anatomical Fraction on Forest Residue Pyrolysis and Hydrotreating

Utilization of cost-advantaged biomass and waste resources to produce clean, domestic biofuels will be a key factor in decarbonizing transportation in the United States. This study investigates how the distribution of anatomical fractions (stem wood, bark, needles, branches) from pine forest residues of different ages impacts pyrolysis and hydrotreating processes, including product composition, carbon efficiency, economics, and overall sustainability. Commercially sourced 13- and 23-year-old loblolly pine residues were chosen to represent available resources; whole tree thinnings, and tops and branches from merchantable timber, respectively. Detailed characterization of the residue samples, product distributions, bio-oil, char, and gas characterization data are presented for 13 experiments, including the individual fractions, blends, and air-classified samples to reduce ash content. Pyrolysis mass balances averaged 97 +/- 3%. As expected, bio-oil yields were lower for the bark, needles, and blends with high proportions of these (53-56% dry basis), and highest for the debarked stem wood (67-71% dry basis). Lower yields were attributed to the high ash content of the needles, but also to the high lignin content in the bark (>50%), which is hypothesized to have led to increased char formation during pyrolysis. Higher-oil yield was generally correlated with total volatiles, hydrogen, oxygen, glucan, and mannan; somewhat correlated with xylan and acetyl content; and negatively correlated with total carbon, fixed carbon, nitrogen, sulfur, and extractives. Air classification of the residues was effective in reducing ash content (1.5% to 0.64%), extractives (~7% to 2%), and sulfur (0.04% to 0.02%), resulting in a small increase in yield, a significant increase in the GC-detectable proportion of oil (27% to 34%) and a net economic benefit. These experimental results were used to conduct technoeconomic and life cycle analyses, which are also summarized here.

Aspects of Crystal Growth and Atomic-Scale Characterization of U/Th Age-Dating Particles

Trace analysis of particulate material collected on environmental swipe samples continues to be one of the cornerstones of the IAEA’s process of verifying member state compliance as part of the Non-Proliferation Treaty (NPT). While complementary to traditional bulk analysis, the ability to measure individual particles can be far more impactful from a radiochronometry perspective. The 234U/230Th radiochronometer is being evaluated as a means of establishing model ages for discrete particles with sufficient confidence. To support the extension of current capabilities, reference materials with a suitable particle form factor and well-documented purification date are actively sought for development with large geometry secondary ion mass spectroscopy.

LWR Aging Management for Life Beyond 80

While several decades of materials degradation research enabled the current US fleet of Light Water Reactors (LWR) to plan to produce electrical power through extended operations up to 80 years of plant life, it now seems appropriate to evaluate what issues, methods, and timelines need to be considered to meet the expected electric power demands for life beyond eighty (LBE) years. To address these questions, it will be necessary to review the electrical capacity and the projected LWR fleet capacity including new builds and advanced reactors. This effort should begin with a materials degradation assessment focused on known and possible unknown issues using a reduced Expanded Materials Degradation Assessment [1], which was employed to identify knowledge gaps for a second license renewal (60-80 yrs.). The focus of the LBE assessment should include: Establishing a timeline to initiate assessments of possible materials aging issues and to ensure sufficient time to evaluate materials degradation research gaps; assessing components and materials to be evaluated, e.g., metals (reactor pressure vessels and alloys within and outside the pressure boundary), concrete, cables, as well as mitigation methods, advanced monitoring, and validation with ex-service materials; initiating a LBE research plan focused on developing an expanded mechanistic understanding of materials degradation and refined models through Codes and Standards evaluation for use by the nuclear industry; and maintaining and strengthening the nuclear materials human knowledge base to address new and as yet unknown degradation modes.

Ambient Aging of Epitaxial and Polycrystalline Rhenium Thin Films

Extending the lifetime of superconducting qubits requires materials that are resistant to surface oxidation. Rhenium has emerged as one such promising candidate that forms a sub-nanometer oxide. Grain boundaries are known to promote material degradation, and thus, we compare epitaxial (single crystal, no grain boundaries) and polycrystalline (many grain boundaries) rhenium thin films after three years of ambient environmental exposure. Epitaxial rhenium remained free of undesired particles, while polycrystalline rhenium developed surface oxide crystallites. Surface characterization using scanning electron microscopy, energy dispersive x-ray spectroscopy, and x-ray photoelectron spectroscopy reveals localized oxygen and nitrogen enrichment, together with a Re⁷⁺ oxidation state in the polycrystalline surface particles, consistent with the proposed formation of ammonium perrhenate. Epitaxial rhenium demonstrated greater resistance to aging and oxidation, making it a more promising candidate for superconducting qubits. Ongoing cleaning and XPS depth profiling will further determine the identity and origin of the surface crystallites.

Aging Effect Analysis of PV Inverter Semiconductors for Ancillary Services Support

PV inverters can provide reactive power while generating active power. An ongoing microgrid implementation at Duke Energy actively engages non-utility PVs to generate/absorb reactive power in support of ancillary services to increase microgrid resiliency during extreme events. PV systems are requested to provide reactive power support: 1) in response to grid voltage variation to better regulate the local voltage; or 2) in response to utility incentives, such as following Transactive Energy System (TES) incentives. However, providing ancillary services might shorten the lifetime expectation of PV inverter semiconductors. This paper summarizes the potential impacts on a PV inverter semiconductor's lifetime when providing ancillary services. The analysis presented in this research work shows that providing reactive power support will increase the mean junction temperature and the junction temperature variation of the inverter diodes. This increased junction temperature will eventually lead to shorter diode lifetime. The lifetime estimation of semiconductors is briefly reviewed. The power losses of PV inverter semiconductors are derived as a support analysis to the junction temperature calculation. In addition, the impact of the filtering inductor on the semiconductor current distribution is discussed. The theoretical analysis presented in this research work is supported by simulation results.

Aging studies of Dual functional materials for CO 2 direct air capture with in situ methanation under simulated ambient conditions: Ru thrifting for cost reduction

Dual function materials (DFMs) comprised of 0.25 %Ru, 6.1 %Na 2 O/γ-Al 2 O 3 //monolith were evaluated for about 250 hours time-on-stream (TOS) at various simulated ambient climate capture conditions followed by temperature swing methanation to 280 °C. Herein this paper focuses on the impact of thrifting Ru to low levels and the impact on performance. Results showed both stable CO 2 capture capacity and CH 4 production with 0.25 % Ru DFM deposited on a ceramic monolith. The CO 2 conversion to CH 4 production was decreased slightly by the Ru decrease from 1 % to 0.25 %. The capture capacity decreased since a lower Ru content reduces the complete decomposition of the Na 2 CO 3 precursor producing fewer active adsorption sites (“Na 2 O”). The deployment of a low Ru DFM monolith would significantly decrease the overall capital cost and give more potential for a large-scale direct air capture and methanation (DACM) application.
