VPS45 is required for both diffuse and tip growth of Arabidopsis thaliana cells

Introduction VPS45 belongs to the Sec1/Munc18 family of proteins, which interact with and regulate Qa-SNARE function during membrane fusion. We have shown previously that Arabidopsis thaliana VPS45 interacts with the SYP61/SYP41/VTI12 SNARE complex, which locates on the trans -Golgi network (TGN). It is required for SYP41 stability, and it functions in cargo trafficking to the vacuole and in cell expansion. It is also required for correct auxin distribution during gravitropism and lateral root growth. Results As vps45 knockout mutation is lethal in Arabidopsis, we identified a mutant, vps45-3 , with a point mutation in the VPS45 gene causing a serine 284-to-phenylalanine substitution. The VPS45-3 protein is stable and maintains interaction with SYP61 and SYP41. However, vps45-3 plants display severe growth defects with significantly reduced organ and cell size, similar to vps45 RNAi transgenic lines that have reduced VPS45 protein levels. Root hair and pollen tube elongation, both processes of tip growth, are highly compromised in vps45-3 . Mutant root hairs are shorter and thicker than those of wild-type plants, and are wavy. These root hairs have vacuolar defects, containing many small vacuoles, compared with WT root hairs with a single large vacuole occupying much of the cell volume. Pollen tubes were also significantly shorter in vps45-3 compared to WT. Discussion We thus show that VPS45 is essential for proper tip growth and propose that the observed vacuolar defects lead to loss of the turgor pressure needed for tip growth.

Inhibited Surface Diffusion in Nanoporous Multi-Principal Element Alloy Thin Films Prepared by Vacuum Thermal Dealloying

Nanoporous structures with 3D interconnected networks are traditionally made by dealloying a binary precursor. Certain approaches for fabricating these materials have been applied to refractory multi-principal element alloys (RMPEAs), which can be suitable candidates for high-temperature applications. In this study, nanoporous refractory multi-principal element alloys (np-RMPEAs) were fabricated from magnesium-based thin films (VMoNbTaMg) that had been prepared by magnetron sputtering. Vacuum thermal dealloying (VTD), which involves sublimation of a higher vapor pressure element, is a novel technique for synthesizing nanoporous refractory elements that are prone to oxidation. When VMoNbTaMg was heated under vacuum, a nanoporous structure was created by the sublimation of the highest vapor pressure element (Mg). X-ray photoelectron spectroscopy depth profiling indicated significantly less ligament oxidation during VTD as compared to traditional dealloying methods. Furthermore, np-RMPEAs exhibited outstanding stability against coarsening, retaining smaller ligaments (~25 nm) at elevated temperature (700 °C) for a prolonged period (48 h).

Systematically Measuring Ultra-diffuse Galaxies (SMUDGes). V. The Complete SMUDGes Catalog and the Nature of Ultradiffuse Galaxies

We present the completed catalog of ultradiffuse galaxy (UDG) candidates (7070 objects) from our search of the DR9 Legacy Survey images, including distance and total mass estimates for 1529 and 1436 galaxies, respectively, that we provide and describe in detail. From the sample with estimated distances, we obtain a sample of 585 UDGs (μ 0, g ≥ 24 mag arcsec –2 and r e ≥ 1.5 kpc) over 20,000 square degrees of sky in various environments. We conclude that UDGs in our sample are limited to 10 10 ≲ M h /M ⊙ ≲ 10 11.5 and are on average a factor of 1.5–7 deficient in stars relative to the general population of galaxies of the same total mass. That factor increases with increasing galaxy size and mass up to a factor of ~10 when the total mass of the UDG increases beyond M h = 10 11 M ⊙ . We do not find evidence that this factor has a dependence on the UDGs large-scale environment.

Cosmological Neutron Stars Produce Diffuse Axion X-Ray Signatures

Axion-like particles can be abundantly produced through scattering processes in the cores of neutron stars (NSs). If they are ultralight ($m_a \lesssim 10^{-4}$ eV), then they can efficiently convert to detectable photons in the external NS magnetospheres, and if they are heavy ($m_a \gtrsim 1$ eV), then they can decay into photons before reaching Earth. In this work, we search for the resulting X-ray signatures from both of these channels summing over the $\textit{cosmological}$ NS population. We compare the predicted axion-induced X-ray signal to the cosmic X-ray background today as measured by a number of instruments such as NuSTAR, HEAO, Swift, and INTEGRAL. We model the axion-induced signal using NS cooling simulations and magnetic field evolution models. We find no evidence for axions and derive strong constraints for both ultralight and heavy axion scenarios, covering new parameter space for the axion-photon and axion-nucleon couplings. Our results rule out the axion-explanation of the Magnificent Seven X-ray excess from nearby isolated NSs.

Combining machine-learned and empirical force fields with the parareal algorithm: application to the diffusion of atomistic defects

We numerically investigate an adaptive version of the parareal algorithm in the context of molecular dynamics. This adaptive variant has been originally introduced in [1]. We focus here on test cases of physical interest where the dynamics of the system is modelled by the Langevin equation and is simulated using the molecular dynamics software LAMMPS. In this work, the parareal algorithm uses a family of machine-learning spectral neighbor analysis potentials (SNAP) as fine, reference, potentials and embedded-atom method potentials (EAM) as coarse potentials. We consider a self-interstitial atom in a tungsten lattice and compute the average residence time of the system in metastable states. Our numerical results demonstrate significant computational gains using the adaptive parareal algorithm in comparison to a sequential integration of the Langevin dynamics. We also identify a large regime of numerical parameters for which statistical accuracy is reached without being a consequence of trajectorial accuracy.
