Metabolic and evolutionary responses of Clostridium thermocellum to genetic interventions aimed at improving ethanol production

Engineering efforts targeted at increasing ethanol by modifying the central fermentative metabolism of Clostridium thermocellum have been variably successful. Here, we aim to understand this variation by a multifaceted approach including genomic and transcriptomic analysis combined with chemostat cultivation and high solids cellulose fermentation. Three strain lineages comprising 16 strains total were examined. Two strain lineages in which genes involved in pathways leading to organic acids and/or sporulation had been knocked out resulted in four end-strains after adaptive laboratory evolution (ALE). A third strain lineage recapitulated mutations involving adhE that occurred spontaneously in some of the engineered strains.

MVP-CTL Fellowship for Suicide Intervention (CRADA Final Report)

Research conducted with Crisis Text Line (CTL) served as an important stepping stone in the group’s ongoing work of developing natural language processing (NLP) methodologies to discover indicators of suicide factors in the U.S. veteran population. With rising cases of suicide, detecting risk factors among veterans is of growing concern. Socio-economic factors are often under-reported along structured medical variables, and extracting information through NLP methods may improve the sensitivity of suicide prediction models. The research explored through the CTL collaboration provided insights on suicide crisis detection and de-escalation, experience in applying scalable data analysis on real world datasets, contributions to study designs for detecting rare-event and produced transferable methods applicable to the domain of veteran suicide mitigation. Tools developed during this research directly benefits ongoing work on the Million Veteran Project (MVP) collaboration with the Department of Veteran Affairs (VA).

Interventions to Disrupt Coronavirus Disease Transmission at a University, Wisconsin, USA, August–October 2020

University settings have demonstrated potential for coronavirus disease (COVID-19) outbreaks; they combine congregate living, substantial social activity, and a young population predisposed to mild illness. Using genomic and epidemiologic data, we describe a COVID-19 outbreak at the University of Wisconsin-Madison, Madison, Wisconsin, USA. During August-October 2020, a total of 3,485 students, including 856/6,162 students living in dormitories, tested positive. Case counts began rising during move-in week, August 25–31, 2020, then rose rapidly during September 1–11, 2020. The university initiated multiple prevention efforts, including quarantining 2 dormitories; a subsequent decline in cases was observed. Genomic surveillance of cases from Dane County, in which the university is located, did not find evidence of transmission from a large cluster of cases in the 2 quarantined dorms during the outbreak. Coordinated implementation of prevention measures can reduce COVID-19 spread in university settings and may limit spillover to the surrounding community.

Postbiotic Gamma-Aminobutyric Acid and Camel Milk Intervention as Innovative Trends Against Hyperglycemia and Hyperlipidemia in Streptozotocin-Induced C57BL/6J Diabetic Mice

Diabetes is a serious disease that threatens human health worldwide. The study hypothesis is to investigate the novel trends that may aid in the prevention of diabetic complications. Camel milk was presented as traditional functional food, and Lactobacillus brevis KLDS 1.0727 and KLDS 1.0373 strains were shown to synthesize postbiotic Gamma-aminobutyric acid as a potential food additive, which can therapeutically intervene against hyperglycemia and hyperlipidemia in streptozotocin-induced C 57 BL/6J mice. During a four-week timeframe, body weight and postprandial blood glucose levels were monitored. Post-euthanasia, blood plasma was obtained to investigate hyperlipidemia, insulin concentrations, liver, and renal functions. The liver, pancreas, kidney, and spleen underwent histopathological examinations. The results demonstrated that KLDS 1.0727 and KLDS 1.0373 ( LAC S1 , LAC S2 ) and camel milk treatments all had a significant influence on hypoglycemic activity, as evidenced by reduced postprandial blood glucose levels. LAC S1 , LAC S2 , and camel milk therapy significantly reduced blood hypolipidemic, and some liver enzymes such as (alanine aminotransferase and aspartate transaminase) levels. Therefore, we recommend consuming camel milk regularly and expanding its use with fermented foods containing L. brevis , one of the probiotics capable of producing gamma-aminobutyric acid (GABA) as future food additives that can improve human health and reduce the prevalence of several diseases disorders.

An Exercise Prescription Intervention Program with Periodic Ergometric Grading

A long term exercise prescription type of physical conditioning program has been available to executive personnel of the NASA Manned Spacecraft Center for the past two years. Periodic ergometric testing with a heart rate controlled, automatically programmed, bicycle ergometer is used to follow the individual's progress and appropriately alter his exercise prescription from time to time. Such a program appears feasible, and acceptance is excellent, dropout rates small and periodic testing participation good. Subjects training diligently can maintain satisfactory levels of conditioning.

Intervention into a turbulent urban situation: A case study

The application is reported of NASA management philosophy and techniques within New Castle County, Delaware, to meet actual problems of community violence. It resulted in restructuring the county approach to problems of this nature, and development of a comprehensive system for planning, based on the NASA planning process. The method involved federal, state, and local resources with community representatives in solving the problems. The concept of a turbulent environment is presented with parallels drawn between NASA management experience and problems of management within an urban arena.

Design of a Vehicle Based Intervention System to Prevent Ozone Loss

This project was designed to be completed over a period of three years. Overall project goals were: (1) To understand the processes that contribute to stratospheric ozone loss; (2) To determine the best prevention scheme for loss; (3) To design a delivery vehicle to accomplish the prevention scheme. The 1994-1995 design objectives included: (1) To review the results of the 1993-1994 design team, including a reevaluation of the major assumptions and criteria selected to choose a vehicle; (2) To evaluate preliminary vehicle concepts and perform quantitative trade studies to select the optimal vehicle concept.

Design of a Vehicle-Based Intervention System to Prevent Ozone Loss

Reduced quantities of ozone in the atmosphere allow greater levels of ultraviolet (UV) radiation to reach the earth's surface. The 1992/1993 project goals for the Virginia Tech Senior Design Team were to 1) understand the processes which contribute to stratospheric ozone loss, 2) examine ways to prevent ozone loss, and 3) define the requirements for an implementation vehicle to carry out the prevention scheme. A scheme proposed by R.J. Cicerone, el al late in 1991 was selected because of its supporting research and economic feasibility. This scheme uses hydrocarbon injected into the Antarctic ozone hole to form stable compounds with free chlorine, thus reducing ozone depletion. A study of the hydrocarbon injection requirements determined that 130 aircraft traveling Mach 2.4 at a maximum altitude of 66,000 ft. would provide the most economic approach to preventing ozone loss. Each aircraft would require an 8,000 nm. range and be able to carry 35,000 lbs. of propane. The propane would be stored in a three-tank high pressure system. Modularity and multi-role functionality were selected to be key design features. Missions originate from airports located in South America and Australia.

Human Adaptation Genetic Response Suites: Toward New Interventions and Countermeasures for Spaceflight

Genetic response suites in human lymphocytes in response to microgravity are important to identify and further study in order to augment human physiological adaptation to novel environments. Emerging technologies, such as DNA micro array profiling, have the potential to identify novel genes that are involved in mediating adaptation to these environments. These genes may prove to be therapeutically valuable as new targets for countermeasures, or as predictive biomarkers of response to these new environments. Human lymphocytes cultured in lg and microgravity analog culture were analyzed for their differential gene expression response. Different groups of genes related to the immune response, cardiovascular system and stress response were then analyzed. Analysis of cells from multiple donors reveals a small shared set that are likely to be essential to adaptation. These three groups focus on human adaptation to new environments. The shared set contains genes related to T cell activation, immune response and stress response to analog microgravity.
