Accuracy of Tau Positron Emission Tomography as a Prognostic Marker in Preclinical and Prodromal Alzheimer Disease: A Head-to-Head Comparison Against Amyloid Positron Emission Tomography and Magnetic Resonance Imaging

Importance: Tau positron emission tomography (PET) tracers have proven useful for the differential diagnosis of dementia, but their utility for predicting cognitive change is unclear. Objective: To examine the prognostic accuracy of baseline fluorine 18 ( 18 F)-flortaucipir and [18F]RO948 (tau) PET in individuals across the Alzheimer disease (AD) clinical spectrum and to perform a head-to-head comparison against established magnetic resonance imaging (MRI) and amyloid PET markers. Design, setting, and participants: This prognostic study collected data from 8 cohorts in South Korea, Sweden, and the US from June 1, 2014, to February 28, 2021, with a mean (SD) follow-up of 1.9 (0.8) years. A total of 1431 participants were recruited from memory clinics, clinical trials, or cohort studies; 673 were cognitively unimpaired (CU group; 253 [37.6%] positive for amyloid-β [Aβ]), 443 had mild cognitive impairment (MCI group; 271 [61.2%] positive for Aβ), and 315 had a clinical diagnosis of AD dementia (315 [100%] positive for Aβ). Exposures: [ 18 F]Flortaucipir PET in the discovery cohort (n = 1135) or [ 18 F]RO948 PET in the replication cohort (n = 296), T1-weighted MRI (n = 1431), and amyloid PET (n = 1329) at baseline and repeated Mini-Mental State Examination (MMSE) evaluation. Main outcomes and measures: Baseline [ 18 F]flortaucipir/[ 18 F]RO948 PET retention within a temporal region of interest, MRI-based AD-signature cortical thickness, and amyloid PET Centiloids were used to predict changes in MMSE using linear mixed-effects models adjusted for age, sex, education, and cohort. Mediation/interaction analyses tested whether associations between baseline tau PET and cognitive change were mediated by baseline MRI measures and whether age, sex, and APOE genotype modified these associations. Results: Among 1431 participants, the mean (SD) age was 71.2 (8.8) years; 751 (52.5%) were male. Findings for [ 18 F]flortaucipir PET predicted longitudinal changes in MMSE, and effect sizes were stronger than for AD-signature cortical thickness and amyloid PET across all participants (R 2 , 0.35 [tau PET] vs 0.24 [MRI] vs 0.17 [amyloid PET]; P < .001, bootstrapped for difference) in the Aβ-positive MCI group (R2, 0.25 [tau PET] vs 0.15 [MRI] vs 0.07 [amyloid PET]; P < .001, bootstrapped for difference) and in the Aβ-positive CU group (R 2 , 0.16 [tau PET] vs 0.08 [MRI] vs 0.08 [amyloid PET]; P < .001, bootstrapped for difference). These findings were replicated in the [ 18 F]RO948 PET cohort. MRI mediated the association between [ 18 F]flortaucipir PET and MMSE in the groups with AD dementia (33.4% [95% CI, 15.5%-60.0%] of the total effect) and Aβ-positive MCI (13.6% [95% CI, 0.0%-28.0%] of the total effect), but not the Aβ-positive CU group (3.7% [95% CI, -17.5% to 39.0%]; P = .71). Age (t = -2.28; P = .02), but not sex (t = 0.92; P = .36) or APOE genotype (t = 1.06; P = .29) modified the association between baseline [ 18 F]flortaucipir PET and cognitive change, such that older individuals showed faster cognitive decline at similar tau PET levels. Conclusions and relevance: The findings of this prognostic study suggest that tau PET is a promising tool for predicting cognitive change that is superior to amyloid PET and MRI and may support the prognostic process in preclinical and prodromal stages of AD.

On the Origin of Room-Temperature Amplified Spontaneous Emission in CsPbBr 3 Single Crystals

CsPbBr 3 is an all-inorganic halide perovskite with excellent photoluminescence (PL) properties for laser applications. Amplified spontaneous emission (ASE) is a prerequisite for lasing and typically observed from low-dimensional CsPbBr3 nanostructures, where quantum confinement enhances ASE. However, a gain medium for lasing should be prepared into a robust bulk form that works under intense light illumination. Here, we demonstrate that bulk CsPbBr 3 single crystals exhibit highly efficient ASE with a threshold of 46 MWcm -2 at 520 nm, if PL reabsorption via the indirect Rashba gap is properly suppressed by thickness control. Based on a series of spectroscopic and microscopic measurements, we show that this below-the-gap absorption can significantly alter the PL feature and even the apparent color of the crystal depending on the crystal size. Our results show that a thin CsPbBr 3 single crystal can be utilized for lasing applications when engineered into a submillimeter thickness for effective light–matter interaction.

National Emission Standards for Hazardous Air Pollutants – Radionuclide Emissions (CY 2019)

The U.S. Department of Energy (DOE), National Nuclear Security Administration Nevada Field Office (NNSA/NFO) operates the Nevada National Security Site (NNSS) and the North Las Vegas Facility (NLVF). From 1951 through 1992, the NNSS was the continental testing location for U.S. nuclear weapons. The release of radionuclides from NNSS activities has been monitored since the initiation of atmospheric testing. After 1962, testing was limited to underground detonations, which greatly reduced radiation exposure to the public. Since the end of nuclear testing in 1992, radiation monitoring has focused on detecting airborne radionuclides from historically contaminated soils because this source dominates the potential offsite dose. These radionuclides are derived from re-suspension of soil (primarily by wind) and emission of tritium-contaminated soil moisture through evapotranspiration. Low amounts of legacy-related tritium are also emitted to air at the NLVF, an NNSS support complex in North Las Vegas.

Compact far ultraviolet emission source with rich spectral emission 1150-3100 A

The article describes a compact hollow Pt cathode emission source for the far UV, developed for use as a high-resolution wavelength standard in laboratory work or on spacecraft (specifically, the NASA International UV Explorer - IUE). The source is small, rugged, lightweight, spectrally rich, bright in the 1150-3200 A region, features long service life, low operating voltage, and lower power drain, and stably emits a large number of very sharp lines with a spectrum lending itself to projection onto the focal plane of a spectrograph. The source has successfully passed exacting environmental tests, and serves as a transfer standard for absolute sensitivity calibration of spectrometric instruments.

Broad emission features in QSOs and active galactic nuclei. II - New observations and theory of Fe II and H I emission

Seven QSOs of low to intermediate redshift are characterized on the basis of: 320-850-nm spectra obtained with the UVITS spectrograph and image-dissector scanner at the f/9 Cassegrain focus of the 2.7-m reflecting telescope at McDonald Observatory; improved model calculations for the Fe II, Mg II, and Balmer lines and the Balmer continuum; and the theoretical results of Netzer and Wills (1983). The data are presented in tables, spectra, and difference spectra and described in detail. Fe II is found to be the largest contributor to the emission line spectrum, with strong UV Fe II lines in all QSOs (regardless of the strength of the optical Fe II lines) and an abundance ratio suggesting a factor-of-three overabundance of Fe relative to Mg and probably H. A discrepancy between the observed Fe II strength and the predictions of dustless standard photoionization models is noted, and the addition of a high-density component to the models is suggested.

The IR emission features - Emission from PAH molecules and amorphous carbon particles

Given the current understanding of polycyclic aromatic hydrocarbons (PAHs), the spectroscopic data suggest that are at least two components which contribute to the interstellar emission spectrum: (1) free molecule-sized PAHs producing the narrow features and (2) amorphous carbon particles (which are primarily composed of an irregular 'lattice' of PAHs) contributing to the broad underlying components. An exact treatment of the IR fluorescence from highly vibrationally excited large molecules demonstrates that species containing between 20 and 30 carbon atoms are responsible for the narrow features, although the spectra match more closely with the spectra of amorphous carbon particles. It is concluded that, since little is known about the spectroscopic properties of free PAHs and PAH clusters, much laboratory work is required along with an observational program focusing on the spatial characteristics of the spectra.

Medium resolution studies of extreme ultraviolet emission from N2 by electron impact - The effect of predissociation on the emission cross section of the b 1Pi(u) state

The electron impact induced fluorescence spectrum of N2 is measured in the 102-134 nm range at a 0.05 nm spectral resolution. The spectral measurements provide the emission cross sections of the transitions of the b 1Pi(u)-X 1Sigma(+)g Birge-Hopfield I-band system. The structure and vibrational population distribution of this system are strongly affected by a configuration interaction of the valence b 1Pi(u) and Rydberg c 1Pi(u) and o 1Pi(u) states. The excitation function (0-400 eV) for the b-X (1,2) transition is measured and a modified Born approximation analytic model is applied to calculate the oscillator strength for the b 1Pi(u)-X band system. With the exception of the v-prime = 1 level, vibrational levels of the b 1Pi(u) state predissociate with a branching ratio of between 0.95 and 1.00. Predissociation of the b 1Pi(u) state contributes approximately six percent of the total dissociation cross section of N2 by electron impact at 100 eV.

Three-dimensional structured shocks in AM Herculis-type systems. II - Cyclotron emission from ridge-shaped emission regions

The properties of cyclotron emission from accretion shocks in AM Herculis binaries are studied which result from nonuniform accretion onto linearly extended regions on the surface of the magnetic white dwarf. The resulting ridge-shaped postshock structures yield cyclotron spectra with prominent harmonic features over a wide range of viewing aspects, even for high total accretion rates, in contrast to what has been found previously for axisymmetric accretion shocks.
