The narrative arc: Revealing core narrative structures through text analysis

Scholars across disciplines have long debated the existence of a common structure that underlies narratives. Using computer-based language analysis methods, several structural and psychological categories of language were measured across ~40,000 traditional narratives (e.g., novels and movie scripts) and ~20,000 nontraditional narratives (science reporting in newspaper articles, TED talks, and Supreme Court opinions). Across traditional narratives, a consistent underlying story structure emerged that revealed three primary processes: staging, plot progression, and cognitive tension. No evidence emerged to indicate that adherence to normative story structures was related to the popularity of the story. Last, analysis of fact-driven texts revealed structures that differed from story-based narratives.

High pressure–derived nonsymmetrical [Cu 2 O] 2+ core for room-temperature methane hydroxylation

Nonsymmetrical oxygen-bridged binuclear copper centers have been proposed and modeled as intermediates and transition states in several C–H oxidation pathways, leading to the postulation that structural dissymmetry enhances the reactivity of the bridging oxygen. However, experimentally characterizing the structure and reactivity of these transient species is remarkably challenging. Here, we report the high-pressure synthesis of a metastable nonsymmetrical dicopper-μ-oxo compound with exceptional reactivity toward the mono-oxygenation of aliphatic C–H bonds. The nonequivalent coordination environment of copper stabilizes localized mixed valency and greatly enhances the hydrogen atom abstraction activity of the bridging oxygen, enabling room-temperature hydroxylation of methane under pressure. These findings highlight the role of dissymmetry in the reactivity of binuclear copper centers and demonstrate precise control of molecular structures by mechanical means.

A Rhodopseudomonas strain with a substantially smaller genome retains the core metabolic versatility of its genus

ABSTRACT Rhodopseudomonas are a group of phototrophic microbes with a marked metabolic versatility and flexibility that underpins their potential use in the production of value-added products, bioremediation, and plant growth promotion. Members of this group have an average genome size of about 5.5 Mb, but two closely related strains have genome sizes of about 4.0 Mb. To identify the types of genes missing in a reduced genome strain, we compared strain DSM127 with other Rhodopseudomonas isolates at the genomic and phenotypic levels. We found that DSM127 can grow as well as other members of the Rhodopseudomonas genus and retains most of their metabolic versatility, but it has many fewer genes associated with high-affinity transport of nutrients, iron uptake, nitrogen metabolism, and biodegradation of aromatic compounds. This analysis indicates genes that can be deleted in genome reduction campaigns and suggests that DSM127 could be a favorable choice for biotechnology applications using Rhodopseudomonas or as a strain that can be engineered further to reside in a specialized natural environment. IMPORTANCE Rhodopseudomonas are a cohort of phototrophic bacteria with broad metabolic versatility. Members of this group are present in diverse soil and water environments, and some strains are found associated with plants and have plant growth-promoting activity. Motivated by the idea that it may be possible to design bacteria with reduced genomes that can survive well only in a specific environment or that may be more metabolically efficient, we compared Rhodopseudomonas strains with typical genome sizes of about 5.5 Mb to a strain with a reduced genome size of 4.0 Mb. From this, we concluded that metabolic versatility is part of the identity of the Rhodopseudomonas group, but high-affinity transport genes and genes of apparent redundant function can be dispensed with.

The Azospirillum brasilense Core Chemotaxis Proteins CheA1 and CheA4 Link Chemotaxis Signaling with Nitrogen Metabolism

Bacterial chemotaxis affords motile bacteria the ability to navigate the environment to locate niches for growth and survival. At the molecular level, chemotaxis depends on chemoreceptor signaling arrays that interact with cytoplasmic proteins to control the direction of movement. In Azospirillum brasilense , chemotaxis is mediated by two distinct chemotaxis pathways: Che1 and Che4. Both Che1 and Che4 are critical in the A. brasilense free-living and plant-associated lifestyles. Here, we use whole-cell proteomics and metabolomics to characterize the role of chemotaxis in A. brasilense physiology. We found that mutants lacking CheA1 or CheA4 or both are affected in nonchemotaxis functions, including major changes in transcription, signaling transport, and cell metabolism. We identify specific effects of CheA1 and CheA4 on nitrogen metabolism, including nitrate assimilation and nitrogen fixation, that may depend, at least, on the transcriptional control of rpoN , which encodes RpoN, a global regulator of metabolism, including nitrogen. Consistent with proteomics, the abundance of several nitrogenous compounds (purines, pyrimidines, and amino acids) changed in the metabolomes of the chemotaxis mutants relative to the parental strain. Further, we uncover novel, and yet uncharacterized, layers of transcriptional and posttranscriptional control of nitrogen metabolism regulators. Together, our data reveal roles for CheA1 and CheA4 in linking chemotaxis and nitrogen metabolism, likely through control of global regulatory networks.

A Formalization of Core Why3 in Coq

Intermediate verification languages like Why3 and Boogie have made it much easier to build program verifiers, transforming the process into a logic compilation problem rather than a proof automation one. Why3 in particular implements a rich logic for program specification with polymorphism, algebraic data types, recursive functions and predicates, and inductive predicates; it translates this logic to over a dozen solvers and proof assistants. Accordingly, it serves as a backend for many tools, including Frama-C, EasyCrypt, and GNATProve for Ada SPARK. But how can we be sure that these tools are correct? The alternate foundational approach, taken by tools like VST and CakeML, provides strong guarantees by implementing the entire toolchain in a proof assistant, but these tools are harder to build and cannot directly take advantage of SMT solver automation. As a first step toward enabling automated tools with similar foundational guarantees, we give a formal semantics in Coq for the logic fragment of Why3. We show that our semantics are useful by giving a correct-by-construction natural deduction proof system for this logic, using this proof system to verify parts of Why3's standard library, and proving sound two of Why3's transformations used to convert terms and formulas into the simpler logics supported by the backend solvers.

Research on Deformation Law of Deep Foundation Pit of Station in Core Region of Saturated Soft Loess Based on Monitoring

Saturated soft loess has a large pore structure, high compressibility, low strength, fluid plastic state, and poor engineering properties. It is still one of the key problems that engineering needs to solve. In order to study the influence of deep foundation pit excavation in the saturated soft loess area on the deformation of foundation pits, the deformation laws of ground settlement, enclosure structure, and supporting axial force were clarified based on the field tests on the deformation characteristics of deep foundation pits in areas with the high-water level in saturated soft loess, combined with geological conditions and on-site construction procedures. The results indicate the following: water supply and construction process were found to be the main factor in changing the surface settlement curve of deep foundation pits in saturated soft loess; increasing the construction speed of the pit bottom floor, inverted braces, floor frame beams, and sidewall frame beams to close the structure, which is conducive to restraining the deformation of the continuous underground wall and foundation pits in similar areas. In the initial stage of support layout, the axial force of steel support tends to increase too fast or even exceed the standard control value. Therefore, a reasonable preadding axial force is an effective means to control the deformation of the continuous underground wall and the axial force of the steel support. The current research results may provide a reference for constructing deep foundation pits in similar areas.

A core lighting curriculum for university students and lighting professionals

Here, in collaboration with a group of lighting professionals, learning outcomes were defined, prioritized, organized and mapped to a three-course sequence of lighting courses within a Bachelor of Science in Architectural Engineering degree programme. Syllabi and educational exercises were developed to support the learning outcomes – including classroom activities, homework assignments and design projects. The learning exercises balance the technical foundations of applied illuminating engineering with the artistic aspects of applied lighting design and are intended to promote significant and lasting learning by providing students with education that is useful and relevant to current lighting practice. The process for identifying and prioritizing lighting content is described, a process that could be adapted by other lighting educators to other pedagogical contexts. The syllabi and learning exercises are disseminated for reuse or adaptation, or for self-study by independent learners.

Data for Characterization of the Ghd8 Flowering Time Gene in a Mini-Core Collection of Miscanthus sinensis

The optimal flowering time for bioenergy crop miscanthus is essential for environmental adaptability and biomass accumulation. However, little is known about how genes controlling flowering in other grasses contribute to flowering regulation in miscanthus. Here, we report on the sequence characterization and gene expression of Miscanthus sinensisGhd8 , a transcription factor encoding a HAP3/NF-YB DNA-binding domain, which has been identified as a major quantitative trait locus in rice, with pleiotropic effects on grain yield, heading date and plant height. In M. sinensis , we identified two homoeologous loci, MsiGhd8A located on chromosome 13 and MsiGhd8B on chromosome 7, with one on each of this paleo-allotetraploid species’ subgenomes. A total of 46 alleles and 28 predicted protein sequence types were identified in 12 wild-collected accessions. Several variants of MsiGhd8 showed a geographic and latitudinal distribution. Quantitative real-time PCR revealed that MsiGhd8 expressed under both long days and short days, and MsiGhd8B showed a significantly higher expression than MsiGhd8A . The comparison between flowering time and gene expression indicated that MsiGhd8B affected flowering time in response to day length for some accessions. This study provides insight into the conserved function of Ghd8 in the Poaceae, and is an important initial step in elucidating the flowering regulatory network of Miscanthus.
