Untargeted GC-MS Metabolic Profiling of Anaerobic Gut Fungi Reveals Putative Terpenoids and Strain-Specific Metabolites

Background/Objectives: Anaerobic gut fungi (Neocallimastigomycota) are biotechnologically relevant, lignocellulose-degrading microbes with under-explored biosynthetic potential for secondary metabolites. Untargeted metabolomic profiling with gas chromatography–mass spectrometry (GC-MS) was applied to two gut fungal strains, Anaeromyces robustus and Caecomyces churrovis, to establish a foundational metabolomic dataset to identify metabolites and provide insights into gut fungal metabolic capabilities. Methods: Gut fungi were cultured anaerobically in rumen-fluid-based media with a soluble substrate (cellobiose), and metabolites were extracted using the Metabolite, Protein, and Lipid Extraction (MPLEx) method, enabling metabolomic and proteomic analysis from the same cell samples. Samples were derivatized and analyzed via GC-MS, followed by compound identification by spectral matching to reference databases, molecular networking, and statistical analyses. Results: Distinct metabolites were identified between A. robustus and C. churrovis, including 2,3-dihydroxyisovaleric acid produced by A. robustus and maltotriitol, maltotriose, and melibiose produced by C. churrovis. C. churrovis may polymerize maltotriose to form an extracellular polysaccharide, like pullulan. GC-MS profiling potentially captured sufficiently volatile products of proteomically detected, putative non-ribosomal peptide synthetases and polyketide synthases of A. robustus and C. churrovis. The triterpene squalene and triterpenoid tetrahymanol were putatively identified in A. robustus and C. churrovis. Their conserved, predicted biosynthetic genes—squalene synthase and squalene tetrahymanol cyclase—were identified in A. robustus, C. churrovis, and other anaerobic gut fungal genera. Conclusions: This study provides a foundational, untargeted metabolomic dataset to unmask gut fungal metabolic pathways and biosynthetic potential and to prioritize future efforts for compound isolation and identification.

The Gold Standard: Accurate Stellar and Planetary Parameters for Eight Kepler M Dwarf Systems Enabled by Parallaxes

We report parallaxes and proper motions from the Hawaii Infrared Parallax Program for eight nearby M dwarf stars with transiting exoplanets discovered by Kepler. We combine our directly measured distances with mass–luminosity and radius–luminosity relationships to significantly improve constraints on the host stars’ properties. Our astrometry enables the identification of wide stellar companions to the planet hosts. Within our limited sample, all the multi-transiting planet hosts (three of three) appear to be single stars, while nearly all (four of five) of the systems with a single detected planet have wide stellar companions. By applying strict priors on average stellar density from our updated radius and mass in our transit fitting analysis, we measure the eccentricity probability distributions for each transiting planet. Planets in single-star systems tend to have smaller eccentricities than those in binaries, although this difference is not significant in our small sample. In the case of Kepler-42bcd, where the eccentricities are known to be ≃0, we demonstrate that such systems can serve as powerful tests of M dwarf evolutionary models by working in L{sub ∗}--ρ{sub ∗} space. The transit-fit density for Kepler-42bcd is inconsistent with model predictions at 2.1σ (22%), but matches more empirical estimates at 0.2σ (2%), consistent with earlier results showing model radii of M dwarfs are underinflated. Gaia will provide high-precision parallaxes for the entire Kepler M dwarf sample, and the Transiting Exoplanet Survey Satellite will identify more planets transiting nearby, late-type stars, enabling significant improvements in our understanding of the eccentricity distribution of small planets and the parameters of late-type dwarfs.

Magnetic Inflation and Stellar Mass. I. Revised Parameters for the Component Stars of the Kepler Low-mass Eclipsing Binary T-Cyg1-12664

Several low-mass eclipsing binary stars show larger than expected radii for their measured mass, metallicity, and age. One proposed mechanism for this radius inflation involves inhibited internal convection and starspots caused by strong magnetic fields. One particular eclipsing binary, T-Cyg1-12664, has proven confounding to this scenario. Çakırlı et al. measured a radius for the secondary component that is twice as large as model predictions for stars with the same mass and age, but a primary mass that is consistent with predictions. Iglesias-Marzoa et al. independently measured the radii and masses of the component stars and found that the radius of the secondary is not in fact inflated with respect to models, but that the primary is, which is consistent with the inhibited convection scenario. However, in their mass determinations, Iglesias-Marzoa et al. lacked independent radial velocity measurements for the secondary component due to the star’s faintness at optical wavelengths. The secondary component is especially interesting, as its purported mass is near the transition from partially convective to a fully convective interior. In this article, we independently determined the masses and radii of the component stars of T-Cyg1-12664 using archival Kepler data and radial velocity measurements of both component stars obtained with IGRINS on the Discovery Channel Telescope and NIRSPEC and HIRES on the Keck Telescopes. We show that neither of the component stars is inflated with respect to models. Our results are broadly consistent with modern stellar evolutionary models for main-sequence M dwarf stars and do not require inhibited convection by magnetic fields to account for the stellar radii.

The Gravity Collective: A Search for the Electromagnetic Counterpart to the Neutron Star–Black Hole Merger GW190814

We present optical follow-up imaging obtained with the Katzman Automatic Imaging Telescope, Las Cumbres Observatory Global Telescope Network, Nickel Telescope, Swope Telescope, and Thacher Telescope of the LIGO/Virgo gravitational wave (GW) signal from the neutron star–black hole (NSBH) merger GW190814. We searched the GW190814 localization region (19 deg 2 for the 90th percentile best localization), covering a total of 51 deg 2 and 94.6% of the two-dimensional localization region. Analyzing the properties of 189 transients that we consider as candidate counterparts to the NSBH merger, including their localizations, discovery times from merger, optical spectra, likely host galaxy redshifts, and photometric evolution, we conclude that none of these objects are likely to be associated with GW190814. Based on this finding, we consider the likely optical properties of an electromagnetic counterpart to GW190814, including possible kilonovae and short gamma-ray burst afterglows. Using the joint limits from our follow-up imaging, we conclude that a counterpart with an r-band decline rate of 0.68 mag day -1 , similar to the kilonova AT 2017gfo, could peak at an absolute magnitude of at most -17.8 mag (50% confidence). Our data are not constraining for "red" kilonovae and rule out "blue" kilonovae with M > 0.5 M ⊙ (30% confidence). We strongly rule out all known types of short gamma-ray burst afterglows with viewing angles <17° assuming an initial jet opening angle of ~5$^°_∙$2 and explosion energies and circumburst densities similar to afterglows explored in the literature. Lastly, we explore the possibility that GW190814 merged in the disk of an active galactic nucleus, of which we find four in the localization region, but we do not find any candidate counterparts among these sources.

The Gravity Collective: A Comprehensive Analysis of the Electromagnetic Search for the Binary Neutron Star Merger GW190425

We present an ultraviolet to infrared search for the electromagnetic (EM) counterpart to GW190425, the second ever binary neutron star merger discovered by the LIGO-Virgo-KAGRA Collaboration. GW190425 was more distant and had a larger localization area than GW170817, so we use a new tool, Teglon, to redistribute the GW190425 localization probability in the context of galaxy catalogs within the final localization volume. We derive a 90th percentile area of 6688 deg 2 , a ∼1.5× improvement relative to the LIGO/Virgo map, and show how Teglon provides an order-of-magnitude boost to the search efficiency of small (≤1 deg 2 ) field-of-view instruments. We combine our data with a large, publicly reported imaging data set, covering 9078.59 deg 2 of unique area and 48.13% of the LIGO/Virgo-assigned localization probability, to calculate the most comprehensive kilonova (KN), short gamma-ray burst (sGRB) afterglow, and model-independent constraints on the EM emission from a hypothetical counterpart to GW190425 to date under the assumption that no counterpart was found in these data. If the counterpart were similar to AT 2017gfo, there would be a 28.4% chance of it being detected in the combined data set. We are relatively insensitive to an on-axis sGRB, and rule out a generic transient with a similar peak luminosity and decline rate as AT 2017gfo to 30% confidence. Finally, across our new imaging and publicly reported data, we find 28 candidate optical counterparts that we cannot rule out as being associated with GW190425, finding that four such counterparts discovered within the localization volume and within 5 days of merger exhibit luminosities consistent with a KN.

Comparison of ablators for the polar direct drive exploding pusher platform

We examine the performance of pure boron, boron carbide, high density carbon, and boron nitride ablators in the polar direct drive exploding pusher (PDXP) platform. The platform uses the polar direct drive conguration at the National Ignition Facility to drive high ion temperatures in a room temperature capsule and has potential applications for plasma physics studies and as a neutron source. The higher tensile strength of these materials compared to plastic enables a thinner ablator to support higher gas pressures, which could help optimize its performance for plasma physics experiments, while ablators containing boron enable the possiblity of collecting addtional data to constrain models of the platform. Applying recently developed and experimentally validated equation of state models for the boron materials, we examine the performance of these materials as ablators in 2D simulations, with particular focus on changes to the ablator and gas areal density, as well as the predicted symmetry of the inherently 2D implosion.
