A Refined Method to Translate Solar Data Quality Assessment Flags to Estimated Measurement Uncertainty

Integrating solar resource uncertainties due to radiometer measurement performance and operational data quality assessment can provide improved estimates of economic bankability, system design performance, and compliance of solar energy conversion systems. Estimating radiometer measurement uncertainty is an established procedure consistent with recognized best practices and international guidelines. SERI QC is a robust solar data quality assessment software tool that has been in continuous use for more than three decades. This report, the fourth of six for the Data Quality and Uncertainty Integration Project, presents a refined algorithm description for software to translate solar resource data quality assessment results into estimated uncertainty values in a Solar Resource Operational Uncertainty Integrator (SROUI) application. This algorithm requires three-component solar irradiance measurements - global horizontal irradiance, direct normal irradiance, and diffuse horizontal irradiance - collected at 1- to 60-minute intervals, as described in the previous deliverables. The development of this report as Deliverable 6.4 was an iterative process that included reviews and feedback from the project team on initial drafts designed to refine how the new software could best support determining solar resource data uncertainty. The results of this effort will contribute to the final software system development by National Renewable Energy Laboratory staff.

Fortalecimiento de la Electrificación Rural en Honduras: Una Evaluación Integrada de PV+BESS y Usos Productivos de la Electricidad en Gracias a Dios, Honduras [Empowering Rural Electrification in Honduras: An Integrated Assessment of PV/BESS and Productive Uses of Electricity in Gracias a Dios] (Spanish Translation)

Honduras faces significant challenges in its energy sector, particularly in rural areas where access to reliable and affordable electricity remains limited. The flagship rural electrification initiative for Honduras Secretary of Energy (SEN) is the Politica de Acceso Universal a la Electricidad (PAUEH - Universal Electricity Access Policy), a key solution for addressing this energy poverty is the deployment of more than 1700 distributed solar and hybrid mini-grid solutions. In late 2023 as a first step towards supporting SEN's electrification efforts, the National Renewable Energy Laboratory (NREL) developed a literature review of SEN electrification policy documents and conducted a series of technical capacity-building workshops with SEN and other energy sector stakeholders in Honduras focused on using NREL's open-source REopt tool to conduct techno-economic assessments and develop least-cost optimizations for potential solar + storage mini-grid systems. Building on some initial capacity building on mini-grid modeling, the National Renewable Energy Laboratory (NREL) worked with SEN to develop a detailed techno-economic assessment for electrifying two schools, a healthcare clinic, and a hospital in a hypothetical community within the department of Gracias a Dios. The analysis also evaluates the business case for cold storage productive use of energy (PUE) applications for the fisheries value chain and how the incorporation of these PUE loads potentially impacts both the viability of the PV+BESS solutions as well as local economic development. By assessing the potential for deployment of integrated PV/BESS systems to both support critical community services like education and healthcare, as well as potential for downstream enterprise and economic development, this analysis represents a first step that can help to inform specific strategies for development of pilot PV+BESS projects aligned with national priorities and sector level planning under PAUEH. The Art of Automation: Translating Electron Microscopy Workflows Into Automated Processes

Acquiring data using a scanning transmission electron microscope (STEM) is a complex, multi-step process. The intricacy of the process depends on the type of sample, composition of the material, desired results of the experiment, resolution requirement and other experimental factors. Each experiment presents unique complications, such as sample drift and contamination, that the microscopist must consider when acquiring data. All these challenges are handled fluidly and expertly by experienced microscopists, but to reach new levels of innovation in material development, including greater reproducibility, throughput, and precision, the automation of these workflows is essential. The initial phase of this work involved translating intuition-based workflows into discrete, programmable steps. Some common key stages in STEM workflows are the initial tuning, scanning the sample for areas of interest, and then acquiring the data. Each stage can be broken further into specific parameter adjustments, such as aberration correction and dwell time optimization, depending on the experiment. When deconstructing various experiments each step was assessed for automation feasibility based on the amount of real time operator decisions. There are steps that lend themselves to automation more readily than others, such as course focusing and sample screening, but there is potential for full automation of all stages with time. As an initial step, an automated montage routine was developed, allowing for the efficient acquisition of large portions of the sample without requiring continuous intervention from the operator. The automation of this small process of the procedure demonstrates the value of this capability. A major challenge in automation arises from discrepancies between commanded, reported and actual stage movements. Using systematic tests, stage movement was quantified. This error can be corrected algorithmically for more accurate workflows in the future. Expanding automation capabilities would result in larger, more efficient data acquisition which allows for more robust statistical analysis. Additionally, this work lays the groundwork for a closed loop system where machine learning algorithms would intake automatically acquired data and make real time decisions. By progressively automating this instrument, this work establishes the foundation for fully automated experimentation in transmission electron microscopy.

Translating advances in microbial bioproduction to sustainable biotechnology

Advances in synthetic biology have radically changed our ability to rewire microorganisms and significantly improved the scalable production of a vast array of drop-in biopolymers and biofuels. The success of a drop-in bioproduct is contingent on market competition with petrochemical analogues and weighted upon relative economic and environmental metrics. While the quantification of comparative trade-offs is critical for accurate process-level decision making, the translation of industrial ecology to synthetic biology is often ambiguous and assessment accuracy has proven challenging. In this review, we explore strategies for evaluating industrial biotechnology through life cycle and techno-economic assessment, then contextualize how recent developments in synthetic biology have improved process viability by expanding feedstock availability and the productivity of microbes. By juxtaposing biological and industrial constraints, we highlight major obstacles between the disparate disciplines that hinder accurate process evaluation. The convergence of these disciplines is crucial in shifting towards carbon neutrality and a circular bioeconomy.

CrossMP: Enabling Cross-Modality Translation between Single-Cell RNA-Seq and Single-Cell ATAC-Seq through Web-Based Portal

In recent years, there has been a growing interest in profiling multiomic modalities within individual cells simultaneously. One such example is integrating combined single-cell RNA sequencing (scRNA-seq) data and single-cell transposase-accessible chromatin sequencing (scATAC-seq) data. Integrated analysis of diverse modalities has helped researchers make more accurate predictions and gain a more comprehensive understanding than with single-modality analysis. However, generating such multimodal data is technically challenging and expensive, leading to limited availability of single-cell co-assay data. Here, we propose a model for cross-modal prediction between the transcriptome and chromatin profiles in single cells. Our model is based on a deep neural network architecture that learns the latent representations from the source modality and then predicts the target modality. It demonstrates reliable performance in accurately translating between these modalities across multiple paired human scATAC-seq and scRNA-seq datasets. Additionally, we developed CrossMP, a web-based portal allowing researchers to upload their single-cell modality data through an interactive web interface and predict the other type of modality data, using high-performance computing resources plugged at the backend.

Myths of German Graphite in World War II, with Original Translations

We re-examine the common narrative that a 1941 experimental error by physicists Walther Bothe and Peter Jensen led Germany to abandon graphite as a reactor moderator during World War II. We first detail the history of both German and American graphite experiments, noting that the Americans faced similar setbacks but succeeded only at the end of a costly 18-month graphite purification program. We then use Monte Carlo N-Particle simulations to reconstruct Bothe's 1941 experiment. We find the thermal absorption cross section of Bothe's Siemens electrographite to be 12.2 mb, in contrast to his reported 7.9 mb. This discrepancy arises because the neutrons in Bothe's experiment did not reach thermal equilibrium, leading to an underestimation of neutron absorption. Additionally, despite misconceptions that the Germans were unaware of boron impurities, we share evidence that Wilhelm Hanle accurately measured boron and cadmium impurities in the electrographite. To support our findings, we provide 9 excerpted and 3 complete English translations of classified wartime reports by Heisenberg, Joos, Bothe, Jensen, Höcker, Hanle, and Kremer. Our work aims to illuminate the rational constraints behind Germany's decision to forgo graphite moderation.

Altering translation allows E. coli to overcome chemically stabilized G-quadruplexes

To investigate the effects of stabilizing G-quadruplexes on Escherichia coli, cells were grown in the presence of the G-quadruplex stabilizing compound, NMM, and RNA-seq was performed. This was done for the control cell strain (MG1655 dtolC) as well as a strain with a decrease in a translation elongation factor (MG1655 dtolC tufA::kan). TPM values were used for differential gene expression analysis between each growth condition. All samples were grown and sequenced in triplicate.

CCAMP: An Integrated Translation and Optimization Framework for OpenACC and OpenMP

Heterogeneous computing and exploration into specialized accelerators are inevitable in current and future supercomputers. Although this diversity of devices is promising for performance, the array of architectures presents programming challenges. High-level programming strategies have emerged to face these challenges, such as the OpenMP offloading model and OpenACC. The varying levels of support for these standards, however, within vendor-specific and open-source tools, as well as the lack of performance portability across devices, have prevented the standards from achieving their goals. To address these shortcomings, we present CCAMP, an OpenMP and OpenACC interoperable framework. CCAMP provides two primary facilities: language translation between the two standards and device-specific directive optimization within each standard. We show that by using the CCAMP framework, programmers can easily transplant non-portable code into new ecosystems for new architectures. Additionally, by using CCAMP device-specific directive optimizations, users can achieve optimized performance across architectures using a single source code.

Beam Loss Monitoring with Fixed and Translating Scintillation Detectors Along the Fermilab Drift-tube Linac

The Fermilab Linac is a roughly 145 meter linear accelerator that accelerates H- beam from 750 keV to 400 MeV and provides beam for the Booster and the rest of the accelerator chain. The first section of the Linac is a Drift-Tube Linac (DTL), which in its current state, suffers from a lack of instrumentation along its length. As a result, operational staff do not have access to the diagnostic information needed to tune the critical components of this accelerator, such as the quadrupole magnets within the drift tubes. This work presents an effort to utilize both fixed and translating scintillation detectors to investigate beam loss along the first two cavities of the DTL.
