Experimental and Computational Insights into the Structural Dynamics of the Fc Fragment of IgG1 Subtype from Biosimilar VEGF‐Trap

The constant fragment (Fc) of the immunoglobulin G1 (IgG1) subtype is increasingly recognized as a crucial scaffold in the development of advanced therapeutics due to its enhanced specificity, efficacy, and extended half‐life. A prime example is VEGF‐Trap (Aflibercept), a recombinant fusion protein that merges the Fc region of the IgG1 subtype with the binding domains of vascular endothelial growth factor receptors (VEGFR)‐1 and VEGFR‐2. The Fc region's role in N‐glycosylation is particularly important, as it significantly influences protein stability. Herein, the first near‐physiological temperature structures of the N‐glycan‐bound Fc fragment of IgG1 subtype from a biosimilar VEGF‐Trap are presented, determined using the SPring‐8 Angstrom Compact free electron LAser (SACLA) and the Turkish Light Source (Turkish DeLight). Comparative analysis with cryogenic structures, including existing data, reveals alternate conformations within the glycan‐binding pocket. Furthermore, molecular dynamics simulations indicate the presence of a high degree of structural plasticity, explaining how the protein adapts its structure through conformational changes. The observed structural fluctuations/conformational changes demonstrate the effect of N‐glycans on protein stability. These findings offer new insights into the molecular basis of Fc‐mediated functions and provide valuable information for the design of next‐generation therapeutics.

Collider phenomenology of a gluino continuum

Continuum supersymmetry is a class of models in which the supersymmetric partners together with part of the standard model come from a conformal sector, broken in the IR near the TeV scale. Such models not only open new doors for addressing the problems of the standard model, but also have unique signatures at hadron colliders, which might explain why we have not yet seen any superpartners at the LHC. Here we use gauge-gravity duality to model the conformal sector, generate collider simulations, and finally analyze continuum gluino signatures at the LHC. Due to the increase in the number of jets produced the bounds are weaker than for the minimal supersymmetric standard model with the same gluino mass threshold.

Light dilaton in rare meson decays and extraction of its CP property

Abstract The dilatonϕis a pseudo-Nambu-Goldstone boson associated with the spontaneous breaking of scale invariance in a nearly conformal theory, and couples to the trace of the stress-energy tensor. We analyze experimental constraints on a light dilaton with mass in the MeV–GeV range from rare meson decays. New model-independent inclusive bounds for theb→sϕtransition largely exclude the parameter space of a light dilaton that could explain the muong− 2 anomaly. Despite similarities between a dilaton and a Higgs-portal scalar, the dilaton-photon coupling is enhanced compared to the Higgs-portal scalar due to contributions from loops of the conformal sector. Consequently, the shortened lifetime of the dilaton relaxes bounds fromK→π+ invisible searches at the NA62 experiment and constraints from the Big Bang Nucleosynthesis. We utilize this fact to search for the dilaton signature at a lepton collider such as the ongoing Belle II experiment. Further, we demonstrate how to extract the CP property of the dilaton using the variation of the differential cross-section ofe + e − →e + e − ϕwith the azimuthal angle between the outgoing leptons.

Quantum Critical Higgs: from AdS 5 to colliders

We examine distinctive signatures of Quantum Critical Higgs models at the LHC and future higher energy colliders. In these models the Higgs boson is part of a conformal sector that is softly broken at a threshold scale, and generically the scaling dimension of the Higgs is larger than in the Standard Model. In particular we examine the gg → H → ZZ, gg → H → γγ, and gg → Z → H Z channels to see how the cross sections deviate from the Standard Model in the high invariant mass region. In order to perform the calculations we use 5D duals of Quantum Critical Higgs models using the AdS/CFT correspondence, with a soft wall to break the conformal symmetry.

Integrable field theories and their CCFT duals

We compute the Mellin transforms of various two-dimensional integrable S-matrices, providing the first explicit, non-perturbative realizations of celestial CFT. In two dimensions, the Mellin transform is simply the Fourier transform in rapidity space, and the “celestial correlator” has no position dependence. The simplified setting allows us to study the analytic properties of CCFT correlators exactly as a function of the conformal dimensions. We find that the correlators exist as real distributions of the conformal weights, with asymptotics controlled by the mass spectrum and three-point couplings of the model. Coupling these models to a flat space limit of JT gravity preserves integrability and dresses the amplitudes by a rapidly varying gravitational phase. We find that the coupling to gravity smooths out certain singular aspects of the Mellin-transformed correlators.

AdS Higgs mechanism from double trace deformed CFT

Explicit breaking of a global symmetry in a conformal field theory is holographically dual to giving mass to a gauge field living in AdS via the Higgs mechanism. We show that if this breaking is induced via a double trace deformation the Higgs mechanism is induced via a scalar loop diagram. The mass can be calculated analytically in both bulk and field theory and we find perfect agreement. While representing familiar physics, the mechanism is identical to how the graviton picks up a mass in the holographic dual of a conformal field theory coupled to a bath.

Neutrino masses from low scale partial compositeness

We consider a class of models in which the neutrinos acquire Majorana masses through mixing with singlet neutrinos that emerge as composite states of a strongly coupled hidden sector. In this framework, the light neutrinos are partially composite particles that obtain their masses through the inverse seesaw mechanism. We focus on the scenario in which the strong dynamics is approximately conformal in the ultraviolet, and the compositeness scale lies at or below the weak scale. The small parameters in the Lagrangian necessary to realize the observed neutrino masses can naturally arise as a consequence of the scaling dimensions of operators in the conformal field theory. We show that this class of models has interesting implications for a wide variety of experiments, including colliders and beam dumps, searches for lepton flavor violation and neutrinoless double beta decay, and cosmological observations. At colliders and beam dumps, this scenario can give rise to striking signals involving multiple displaced vertices. The exchange of hidden sector states can lead to observable rates for flavor violating processes such as μ → eγ and μ → e conversion. If the compositeness scale lies at or below a hundred MeV, the rate for neutrinoless double beta decay is suppressed by form factors and may be reduced by an order of magnitude or more. The late decays of relic singlet neutrinos can give rise to spectral distortions in the cosmic microwave background that are large enough to be observed in future experiments.

Universal entropy and hawking radiation of near-extremal AdS4 black holes

We compute the Bekenstein-Hawking entropy of near-extremal asymptotically AdS 4 electrically charged rotating black holes using three different methods: (i) from the gravity solution, (ii) from the near-horizon Kerr/CFT correspondence and (iii) from the boundary conformal field theory. The results from these three different approaches match exactly, giving us a unique and universal expression for the entropy and the microstate counting of near-extremal AdS black holes via the AdS/CFT correspondence. In the second method, we extend the Kerr/CFT correspondence to the near-extremal case to compute the left and right central charges. We also use hidden conformal symmetry of the near-horizon geometry to compute the Frolov-Thorne temperatures. From the results of the near-extremal AdS 4 black hole entropy, we provide a microscopic foundation for Hawking radiation.

Line operators in Chern-Simons-Matter theories and Bosonization in Three Dimensions II: Perturbative analysis and all-loop resummation

We study mesonic line operators in Chern-Simons theories with bosonic or fermionic matter in the fundamental representation. In this paper, we elaborate on the classification and properties of these operators using all loop resummation of large N perturbation theory. We show that these theories possess two conformal line operators in the fundamental representation. One is a stable renormalization group fixed point, while the other is unstable. They satisfy first-order chiral evolution equations, in which a smooth variation of the path is given by a factorized product of two mesonic line operators. The boundary operators on which the lines can end are classified by their conformal dimension and transverse spin, which we compute explicitly at finite ’t Hooft coupling. We match the operators in the bosonic and fermionic theories. Finally, we extend our findings to the mass deformed theories and discover that the duality still holds true.

Moonshine, superconformal symmetry, and quantum error correction

Special conformal field theories can have symmetry groups which are interesting sporadic finite simple groups. Famous examples include the Monster symmetry group of a c = 24 twodimensional conformal field theory (CFT) constructed by Frenkel, Lepowsky and Meurman, and the Conway symmetry group of a c = 12 CFT explored in detail by Duncan and Mack-Crane. The Mathieu moonshine connection between the K3 elliptic genus and the Mathieu group M24 has led to the study of K3 sigma models with large symmetry groups. A particular K3 CFT with a maximal symmetry group preserving (4, 4) superconformal symmetry was studied in beautiful work by Gaberdiel, Taormina, Volpato, and Wendland [41]. The present paper shows that in both the GTVW and c = 12 theories the construction of superconformal generators can be understood via the theory of quantum error correcting codes. The automorphism groups of these codes lift to symmetry groups in the CFT preserving the superconformal generators. In the case of the N = 1 supercurrent of the GTVW model our result, combined with a result of T. Johnson-Freyd implies the symmetry group is the maximal subgroup of M24 known as the sextet group. (The sextet group is also known as the holomorph of the hexacode.) Building on [41] the Ramond-Ramond sector of the GTVW model is related to the Miracle Octad Generator which in turn leads to a role for the Golay code as a group of symmetries of RR states. Moreover, (4, 1) superconformal symmetry suffices to define and decompose the elliptic genus of a K3 sigma model into characters of the N = 4 superconformal algebra. The symmetry group preserving (4, 1) is larger than that preserving (4, 4).

Global symmetry and integral constraint on superconformal lines in four dimensions

We study properties of point-like impurities preserving flavor symmetry and supersymmetry in four-dimensional 𝒩 = 2 field theories. At large distances, such impurities are described by half-BPS superconformal line defects. By working in the AdS 2 × S 2 conformal frame, we develop a novel and simpler way of deriving the superconformal Ward identities relating the various two-point functions of flavor current multiplet operators in the presence of the defect. We use these relations to simplify a certain integrated two-point function of flavor current multiplet operators that, in Lagrangian theories, can be computed using supersymmetric localization. The simplification gives an integral constraint on the two-point function of the flavor current multiplet superconformal primary with trivial integration measure in the AdS 2 × S 2 conformal frame. We provide several consistency checks on our Ward identities.

Janus on the brane

We present a non-supersymmetric deformation of probe branes describing conformal defects of codimension two in AdS/CFT. The worldvolume of the probe branes is deformed from AdS p × S 1 embedded in an AdS p+2 × M D–p–2 background to an embedding of Janus form, which uses an AdS p–1 slicing of AdS p and in which the brane bends along the slicing coordinate. In field theory terms this realizes conformal interfaces on codimension-two defects. We discuss these "Janus on the brane" solutions for AdS 3 × S 1 D3-branes in the AdS 5 × S 5 solution of Type IIB, realizing interfaces on surface defects in N = 4 SYM, and show that similar solutions exist for probe branes in AdS p+2 × S 9–p vacua of M-theory and in the AdS 6 × S 4 solution of massive Type IIA.

(2, 2) Scattering and the celestial torus

Analytic continuation from Minkowski space to (2, 2) split signature spacetime has proven to be a powerful tool for the study of scattering amplitudes. Here we show that, under this continuation, null infinity becomes the product of a null interval with a celestial torus (replacing the celestial sphere) and has only one connected component. Spacelike and timelike infinity are time-periodic quotients of AdS 3 . These three components of infinity combine to an S 3 represented as a toric fibration over the interval. Privileged scattering states of scalars organize into SL(2, $\mathbb{R}$) L ×SL(2, $\mathbb{R}$) R conformal primary wave functions and their descendants with real integral or half-integral conformal weights, giving the normally continuous scattering problem a discrete character.

Celestial amplitudes from UV to IR

Celestial amplitudes represent 4D scattering of particles in boost, rather than the usual energy-momentum, eigenstates and hence are sensitive to both UV and IR physics. We show that known UV and IR properties of quantum gravity translate into powerful constraints on the analytic structure of celestial amplitudes. For example the soft UV behavior of quantum gravity is shown to imply that the exact four-particle scattering amplitude is meromorphic in the complex boost weight plane with poles confined to even integers on the negative real axis. Would-be poles on the positive real axis from UV asymptotics are shown to be erased by a flat space analog of the AdS resolution of the bulk point singularity. The residues of the poles on the negative axis are identified with operator coefficients in the IR effective action. Far along the real positive axis, the scattering is argued to grow exponentially according to the black hole area law. Exclusive amplitudes are shown to simply factorize into conformally hard and conformally soft factors. The soft factor contains all IR divergences and is given by a celestial current algebra correlator of Goldstone bosons from spontaneously broken asymptotic symmetries. The hard factor describes the scattering of hard particles together with the boost-eigenstate clouds of soft photons or gravitons required by asymptotic symmetries. These provide an IR safe S-matrix for the scattering of hard particles.

Wormholes with ends of the world

We study classical wormhole solutions in 3D gravity with end-of-the-world (EOW) branes, conical defects, kinks, and punctures. These solutions compute statistical averages of an ensemble of boundary conformal field theories (BCFTs) related to universal asymptotics of OPE data extracted from the 2D conformal bootstrap. Conical defects connect BCFT bulk operators; branes join BCFT boundary intervals with identical boundary conditions; kinks (1D defects along branes) link BCFT boundary operators; and punctures (0D defects) are endpoints where conical defects terminate on branes. We provide evidence for a correspondence between the gravity theory and the ensemble. In particular, the agreement of the g-function dependence results from an underlying topological aspect of the on-shell EOW brane action, from which a BCFT analog of the Schlenker-Witten theorem also follows.

Transport across interfaces in symmetric orbifolds

We examine how conformal boundaries encode energy transport coefficients — namely transmission and reflection probabilities — of corresponding conformal interfaces in symmetric orbifold theories. These constitute a large class of irrational theories and are closely related to holographic setups. Our central goal is to compare such coefficients at the orbifold point (a field theory calculation) against their values when the orbifold is highly deformed (a gravity calculation) — an approach akin to past AdS/CFT-guided comparisons of physical quantities at strong versus weak coupling. At the orbifold point, we find that the (weighted-average) transport coefficients are simply averages of coefficients in the underlying seed theory. We then focus on the symmetric orbifold of the T 4 sigma model interface CFT dual to type IIB supergravity on the 3d Janus solution. We compare the holographic transmission coefficient, which was found by [1], to that of the orbifold point. We find that the profile of the transmission coefficient substantially increases with the coupling, in contrast to boundary entropy. We also present some related ideas about twisted-sector data encoded by boundary states.

A discontinuous piecewise polynomial generalized moving least squares scheme for robust finite element analysis on arbitrary grids

A variational approach is developed with a meshless discretization to enable accurate and robust numerical simulation of partial differential equations for meshes that are of poor quality. Traditional finite element methods use the mesh to both discretize the geometric domain and to define the finite element shape functions. The latter creates a dependence between the quality of the mesh and the properties of the finite element basis that may adversely affect the accuracy of the discretized problem. Here, we propose a new approach for defining finite element shape functions that breaks this dependence and separates mesh quality from the discretization quality, which we call discontinuous piecewise polynomial generalized moving least squares (DPP-GMLS). At the core of the approach is a meshless definition of the shape functions, which limits the purpose of the mesh to representing the geometric domain and integrating the basis functions without having any role in their approximation quality. The resulting non-conforming space can be utilized within a standard discontinuous Galerkin framework, providing a rigorous foundation for solving partial differential equations on low-quality meshes. We present a collection of numerical experiments demonstrating our approach in a wide range of settings: strongly coercive elliptic problems, linear elasticity in the compressible regime, and the stationary Stokes problem. We demonstrate convergence for all problems and stability for element pairs for problems which usually require inf-sup compatibility for conforming methods, also referring to a minor modification possible through the symmetric interior penalty Galerkin framework for stabilizing element pairs that would otherwise be traditionally unstable. Mesh robustness is particularly critical for elasticity, and we provide an example that our approach provides a greater than 5 x improvement in accuracy and allows for taking an 8 x larger stable timestep for a highly deformed mesh, compared to the continuous Galerkin finite element method.

Extended isogeometric analysis of multi-material and multi-physics problems using hierarchical B-splines

Here, this paper presents an immersed, isogeometric finite element framework to predict the response of multi-material, multi-physics problems with complex geometries using locally refined discretizations. To circumvent the need to generate conformal meshes, this work uses an extended finite element method (XFEM) to discretize the governing equations on non-conforming, embedding meshes. A flexible approach to create truncated hierarchical B-splines discretizations is presented. This approach enables the refinement of each state variable field individually to meet field-specific accuracy requirements. To obtain an immersed geometry representation that is consistent across all hierarchically refined B-spline discretizations, the geometry is immersed into a single mesh, the XFEM background mesh, which is constructed from the union of all hierarchical B-spline meshes. An extraction operator is introduced to represent the truncated hierarchical B-spline bases in terms of Lagrange shape functions on the XFEM background mesh without loss of accuracy. The truncated hierarchical B-spline bases are enriched using a generalized Heaviside enrichment strategy to accommodate small geometric features and multi-material problems. The governing equations are augmented by a formulation of the face-oriented ghost stabilization enhanced for locally refined B-spline bases. We present examples for two- and three-dimensional linear elastic and thermo-elastic problems. The numerical results validate the accuracy of our framework. The results also demonstrate the applicability of the proposed framework to large, geometrically complex problems.
