Biological engineering of microorganisms to produce value-added chemicals is a promising route to sustainable manufacturing. However, overproduction of metabolic intermediates at high titer, rate, and yield from inexpensive substrates is challenging in non-model systems where limited information is available regarding metabolic flux and its control in production conditions. Integrated multi-omic analyses of engineered strains offers an in-depth look at metabolites and proteins directly involved in growth and production of target and non-target bioproducts. Here we applied multi-omic analyses to overproduction of the polymer precursor 3-hydroxypropionic acid (3HP) in the filamentous fungus Aspergillus pseudoterreus. A synthetic pathway consisting of aspartate decarboxylase, beta-alanine pyruvate transaminase, and 3HP dehydrogenase was designed and built for A. pseudoterreus. Strains with single- and multi-copy integration events were isolated and multi-omics analysis consisting of intracellular and extracellular metabolomics and targeted and global proteomics was used to interrogate the strains in shake-flask and bioreactor conditions. Production of a variety of co-products (organic acids and glycerol) and oxidative degradation of 3HP were identified as metabolic pathways competing with 3HP production. Intracellular accumulation of nitrogen as 2,4-diaminobutanoate was identified as an off-target nitrogen sink that may also limit flux through the engineered 3HP pathway. Elimination of the high-expression oxidative 3HP degradation pathway by deletion of a putative malonate semialdehyde dehydrogenase improved the yield of 3HP by 3.4× after 10 days in shake-flask culture. This is the first report of 3HP production in a filamentous fungus amenable to industrial scale biomanufacturing of organic acids at high titer and low pH.

The heat-transfer characteristics of a chordwise-finned, impingement-cooled vane were investigated in both a modified J-57 research engine and a four-vane cascade. The data were compared by a correlation of temperature difference ratio with coolant- to gas-flow ratio and also by two modifications of this correlation. The results indicated that the cascade vane temperature data can generally be used to represent the engine vane temperature data. A discussion of engine and cascade gas-side heat-transfer coefficients is also presented. A redesign of the vane leading edge could significantly increase the potential turbine-inlet temperature operating limit.

A uranium dioxide (UO2) fueled capsule was fabricated and irradiated in the NASA Plum Brook Reactor Facility. The capsule consisted of two bulk UO2 specimens clad with chemically vapor deposited tungsten (CVD W) 0.762 and 0.1016 cm (0.030-and 0.040-in.) thick, respectively. The second specimen with 0.1016-cm (0.040-in.) thick cladding was irradiated at temperature for 2607 hours, corresponding to an average burnup of 1.516 x 10 to the 20th power fissions/cu cm. Postirradiation examination showed distortion in the bottom end cap, failure of the weld joint, and fracture of the central vent tube. Diametral growth was 1.3 percent. No evidence of gross interaction between CVD tungsten or arc-cast tungsten cladding and the UO2 fuel was observed. Some of the fission gases passed from the fuel cavity to the gas surrounding the fuel specimen via the vent tube and possibly the end-cap weld failure. Whether the UO2 loss rates through the vent tube were within acceptable limits could not be determined in view of the end-cap weld failure.
One Thiokol Chemical Corporation TE-M-521-5 solid-propellant apogee rocket motor was successfully fired at an average simulated altitude of about 108,000 ft while spinning at 46 rpm. The general program objectives were to verify compliance of motor performance with the manufacturer's specifications. Specific primary objectives were to determine vacuum ballistic performance of the motor after prefire vibration conditioning and temperature conditioning at 40F, altitude ignition characteristics, motor structural integrity, and motor temperature-time history during and after motor operation. Additional objectives were to measure the lateral (nonaxial) thrust component during motor operation and to measure radiation heat flux in the vicinity of the nozzle exit plane.
