Optogenetics and biosensors set the stage for metabolic cybergenetics

We report cybergenetic systems use computer interfaces to enable feed-back controls over biological processes in real time. The complex and dynamic nature of cellular metabolism makes cybergenetics attractive for controlling engineered metabolic pathways in microbial fermentations. Cybergenetics would not only create new avenues of research into cellular metabolism, it would also enable unprecedented strategies for pathway optimization and bioreactor operation and automation. Implementation of metabolic cybergenetics, however, will require new capabilities from actuators, biosensors, and control algorithms. The recent application of optogenetics in metabolic engineering, the expanding role of genetically encoded biosensors in strain development, and continued progress in control algorithms for biological processes suggest that this technology will become available in the not so distant future.

Context and mitigation of lost circulation during geothermal drilling in diverse geologic settings

Lost circulation is one of the most common and expensive problems facing geothermal energy development, representing up to 30% of drilling costs. We examined drilling records from four geothermal fields—McGinness Hills in central Nevada, Don A. Campbell and Steamboat Hills in western Nevada, and Puna Geothermal Venture on the Big Island of Hawai'i— to identify geologies most prone to lost circulation, as well as common mitigation strategies. Depths of lost circulation events varied, but their frequency often increased in the production interval. Lost circulation commonly occurred near fault intersections, and heavily faulted fields like McGinness Hills and Don A. Campbell showed secondary mineralization within approximately 100 m (328 ft) or less of where circulation was lost. Lost circulation mitigation strategies included using locally available materials (e.g., cotton seed hulls) as well as more expensive proprietary lost circulation materials, cement plugs above the reservoir, and drilling blind with aerated, polymer-based mud in the production zone. Addressing lost circulation using a well thought out decision-making approach and materials above the reservoir will save time and cost, and provide needed well integrity. Mitigation often requires a series of steps, typically applied from perceived least expensive to most, and are dependent on the severity and location in the well where circulation was lost and availability of materials. Placing cement plugs can cure lost circulation events, however these plugs are often expensive, time-consuming, and may not be successful.

Analysis of backwash settings to maximize net water production in an engineering-scale ultrafiltration system for water reuse

Ultrafiltration (UF) has been widely utilized as water pretreatment for different applications especially in water reuse. The UF system operation is characterized by a filtration phase, where particles accumulate on the membrane surface resulting in an increase in the transmembrane pressure (TMP) and a cleaning phase, where foulants are removed through cleaning cycles including physical backwash and chemical-enhanced backwash (CEB). In this study, data from an engineering-scale UF system treating reclaimed wastewater were used to assess the impact of backwashing on the filtration process. TMP backwash trigger, backwash duration, and CEB frequency were purposely varied for a cycle-by-cycle investigation on the net water production, water recovery, initial operating TMP, and filtration cycle duration. As the TMP backwash trigger was varied between 62 and 145kPa, the maximum net water production (63 m 3 /d) was achieved at 103kPa and water recovery remained relatively constant at approximately 92%. Backwash durations of 45, 65, and 85s were performed where both net water production and water recovery yielded similar results (~63 m 3 /d and ~ 91%) compared with 103kPa TMP backwash trigger. The CEB frequency was also lowered from one every three backwashes (1/3) to 1/6 and 1/12 and resulted in decreased net water production and water recovery while the initial TMP increased. Interestingly, the total number of CEBs remained approximately constant regardless of their frequency. In conclusion, results suggest that CEB is an important fouling control process to maximize water production.

The electronic set-up for the scintillation light detection system of ICARUS-SBN at Fermilab

The ICARUS-T600 Liquid Argon (LAr) Time Projection Chamber (TPC) is presently used as the far detector of the Short Baseline Neutrino (SBN) program at Fermilab (USA) to search for a possible sterile neutrino signal with the Booster Neutrino Beam (BNB). We discuss a light detection system, based on 360 Hamamatsu R5912-MOD Photo-Multiplier Tubes (PMTs), which has been realized to detect vacuum ultraviolet (VUV) photons produced after the passage of ionizing particles in LAr. High performance electronics allows recording of the PMT signals and provides a fast discrimination for the exploitation of the scintillation light for trigger purposes. This system is fundamental for the TPC operation, contributing to the trigger system and for the 3D reconstruction of events.
