Expression of a Bacterial Trehalose 6-Phosphate Synthase Gene otsA in Camelina sativa Seeds Promotes the Channelling of Carbon Towards Oil Accumulation

Improving seed oil yield is essential for developing Camelina sativa as a sustainable biofuel crop. Fatty acid synthesis depends on the production of acetyl-CoA from photosynthetically derived sugars. Trehalose 6-phosphate (T6P), a proxy for sucrose availability, can link sugar status to plant growth and development. Synthesised by trehalose 6-phosphate synthase (TPS) from UDP-glucose and glucose-6-phosphate, T6P plays a regulatory role in metabolism. Our previous studies on Arabidopsis transgenic lines constitutively expressing the E. coli otsA (encoding TPS) showed increased T6P levels and seed triacylglycerol, along with stunted growth. In the present study we express otsA in camelina under the control of a seed-specific Phaseolin promoter. Seeds of the resulting transgenic lines accumulated high levels of T6P, and a 15%–20% increase in total fatty acids and triacylglycerol compared to wild-type. Molecular analysis showed the transgenic seeds had reduced SnRK1 activity, elevated WRI1 protein levels, and increased the levels of WRI1 and its target genes, along with enhanced rates of fatty acid synthesis that increased seed weights relative to wild type. Notably, the increase in oil did not affect seed protein levels but did reduce the soluble metabolite fraction. Crucially, seed-specific expression of otsA mitigated the growth defects associated with constitutive otsA expression, and the transgenic lines showed normal seed development and germination. These findings demonstrate that targeted T6P modulation via seed-specific otsA expression is an effective metabolic engineering strategy to boost oil production in camelina and potentially in other oilseed crops and bioenergy crops such as energycane, sorghum and miscanthus.

Electrolyte Takeover Strategy for Performance Recovery in Polysulfide-Permanganate Flow Batteries

The abundance of active material precursors for a polysulfide-permanganate flow battery makes it a compelling chemistry for large-scale, and potentially long-duration (>10 h), grid electricity storage. Precipitation, arising from either reactant crossover or electrolyte side reactions, decrease cell efficiencies during charge/discharge cycling. Regardless of the abundance and low cost of active materials, a system without high cyclability cannot meet grid electricity storage economic targets for applications that cycle regularly. Precipitated species can be removed, and reactor efficiency performance restored, by using an electrolyte takeover process, or ETP. Two ETP methods are investigated. One ETP uses the negative electrolyte, an alkaline polysulfide (pS) solution, as takeover solution, and another uses dilute acidic peroxide (DAP) as the takeover solution. Both ETPs maintain functional cell operation within an acceptable performance range over >1000 h and >200 cycles, a duration over which cells that do not undergo ETPs clog and fail. The DAP ETP proves especially effective and limits irrecoverable voltage efficiency fade below 0.02%/cycle. These ETPs, either individually, or in combination, can enable the requisite cyclability for practical polysulfide-permanganate flow battery systems.

Procedure for locating oil and gas wells in the Appalachian Basin

Locating undocumented (or poorly documented) oil and gas wells for environmental assessment is often difficult. Remnant features that confirm the presence of a well (intact casing/wellhead, well bore, etc.) are typically less than a meter in size and often are obscured from direct observation on the ground or from the air (by dense vegetation, for example). To efficiently find such features, it is useful to first systematically compile publicly available digital data at progressively smaller scales prior to embarking on field campaigns. Further, the information presented here describes the procedure developed and used by the U.S. Department of Energy's National Energy Technology Laboratory to locate potential oil and gas well sites for follow-up field verification and characterization. Digital data are first compiled from national and state resources such as well location/production databases, historical topographic maps, historical aerial photographs, and LiDAR data. Although each data set is likely to be incomplete or inaccurate to some extent, combining the data resources using geographic information system technology can generate potential well site targets with a higher degree of confidence, which improves the efficiency of fieldwork activities. This workflow was developed in the Appalachian Basin region, and although certain aspects may be unique, the general process would be applicable to locating undocumented wells in other regions.

Direct selection of functional fluorescent-protein antibody fusions by yeast display

Antibodies are important reagents for research, diagnostics, and therapeutics. Many examples of chimeric proteins combining the specific target recognition of antibodies with complementing functionalities such as fluorescence, toxicity or enzymatic activity have been described. However, antibodies selected solely on the basis of their binding specificities are not necessarily ideal candidates for the construction of chimeras. Here, we describe a high throughput method based on yeast display to directly select antibodies most suitable for conversion to fluorescent chimera. A library of scFv binders was converted to a fluorescent chimeric form, by cloning thermal green protein into the linker between VH and VL, and directly selecting for both binding and fluorescent functionality. This allowed us to directly identify antibodies functional in the single chain TGP format, that manifest higher protein expression, easier protein purification, and one-step binding assays.

Cooperative Research and Development Agreement between National Energy Technology Laboratory and Pyrochem Catalyst Company [Abstract]

A catalytic converter is a key enabling technology for more advanced engine technologies to meet the more stringent fuel economy and emission standards of the future, particularly the requirement to treat emissions formed during a cold start (which account for between 60-80% of total emissions). As internal combustion engine technology is expected to remain a dominant force in the market, new and innovative catalyst technologies are needed with low temperature activity to reduce exhaust pollutants by 90% at temperatures as low as 150°C, with a performance lifetime that exceeds 150,000 miles of driving. Considering current catalyst technology only starts to convert pollutants at 200°C, the challenge is significant. Pyrochem Catalyst Company (PCC) has developed a promising PyroCatTM pyrochlore catalyst technology (that incorporates an NETL technology licensed by PCC), which has shown activity better than current state of the art and approaching the 150°C target. NETL and PCC will collaborate to transition PyroCatTM from lab-scale testing to commercialization through the generation of a commercially representative monolith catalyst that can be sent to automotive companies and catalyst manufacturers to validate in their reactor systems.

Modification of Fractured Rock Permeability Field Using Polymer Foam to Improve Geothermal System Efficiency

The project aims at developing novel methods for blocking high-permeability pathways in geothermal reservoirs to improve heat exchange efficiency. While the project was at DRI, and now when it has been transferred to UNR, the goal of blocking high-permeability pathways is achieved through targeted injection of heat-sensitive polymer foams, particularly foamed epoxy resins, whose activation and curing are triggered by the ambient thermal field.

Comet nucleus sample return

A comet nucleus sample return mission has been proposed for implementation near the end of this century. The objective of the mission is to collect a sample of undisturbed material from beneath the surface of an active comet and return it to earth in a minimally altered state. Potential targets include the short-period comets Encke, Tempel 2 and Wild 2. This article defines such a mission and describes its requirements with regard to science, sampling, thermal protection and performance. It has been determined that, with present launch capabilities, Solar Electric Propulsion (SEP) is an enabling technology for this mission.

Ionic binding properties of carrier ampholytes

Radioactive ampholytes were synthesized with specific activity of 638 microCi/g. These were used in studies of ampholyte binding to target proteins under nonionic conditions. These radioactive ampholytes bound to target proteins but were dissociable in sodium chloride solutions with dissociation occurring in a concentration dependent way. The ampholytes could be dissociated from target molecules using excess unlabelled ampholytes synthesized in the laboratory as well as commercial ampholytes. Radioactive ampholytes were bound to target proteins with different isoelectric points, and the bound ampholytes were eluted and analyzed by recycling isoelectric focusing. The results showed that acidic proteins bound basic ampholytes and basic proteins bound acidic ampholytes.

Video-sensor/electronic-feedback control system being developed to provide automatic control of final stage of approach of active vehicle to passive vehicle or object. No communication between vehicles necessary. Docking target, mounted on post on passive vehicle, has three reflective dots, positions of which measured in image to determine relative position and orientation of target and vehicle. Control channels process video-sensor feedback into motion-control signals (thrustor commands in spacecraft or motor commands in robot). Conceived for use aboard chasing spacecraft to control its maneuvers in docking with chased spacecraft, concept also useful in controlling coupling of aircraft in flight or vessels on water or in controlling approach of robot to object to be manipulated.

COMPTEL solar flare observations

COMPTEL as part of a solar target of opportunity campaign observed the sun during the period of high solar activity from 7-15 Jun. 1991. Major flares were observed on 9 and 11 Jun. Although both flares were large GOES events (greater than or = X10), they were not extraordinary in terms of gamma-ray emission. Only the decay phase of the 15 Jun. flare was observed by COMPTEL. We report the preliminary analysis of data from these flares, including the first spectroscopic measurement of solar flare neutrons. The deuterium formation line at 2.223 MeV was present in both events and for at least the 9 Jun. event, was comparable to the flux in the nuclear line region of 4-8 MeV, consistent with Solar-Maximum Mission (SSM) Observations. A clear neutron signal was present in the flare of 9 Jun. with the spectrum extending up to 80 MeV and consistent in time with the emission of gamma-rays, confirming the utility of COMPTEL in measuring the solar neutron flux at low energies. The neutron flux below 100 MeV appears to be lower than that of the 3 Jun. 1982 flare by more than an order of magnitude. The neutron signal of the 11 Jun. event is under study. Severe dead time effects resulting from the intense thermal x-rays require significant corrections to the measured flux which increase the magnitude of the associated systematic uncertainties.

Continued Development of an Ultra-Narrow Bandpass Filter for Solar Research

The objective of work under this task was to develop ultranarrow optical bandpass filters and related technology necessary for construction of a compact solar telescope capable of operating unattended in space. The scientific problems to which such a telescope could be applied include solar seismology, solar activity monitoring, solar irradiance variations, solar magnetic field evolution, and the location of targets for narrow-field specialized telescopes. We have demonstrated a Y-cut lithium-niobate Fabry-Perot etalon. This filter will be used on the Flare Genesis Experiment. We also obtained solar images with a Z-cut etalon. The technical report on etalon filters is attached to this final report. We believe that work under this grant will lead to the commercial availability of a universal optical filter with approximately 0.1 A bandwidth. Progress was made toward making a suitable 1-2 A tunable blocker filter, but it now appears that the best approach is to make a double-cavity etalon that will not require such a narrow blocker. Broader band blockers are commercially available.

Manual control in space research on perceptual-motor functions under zero gravity conditions (L-10)

Are human abilities to control vehicles and other machines the same in space as those on Earth? The L-10 Manual Control Experiment of the First Materials Processing Tests (FMPT) started from this question. Suppose a pilot has the task to align the head of a space vehicle toward a target. His actions are to look at the target, to determine the vehicle movement, and to operate the manipulator. If the activity of the nervous system were the same as on Earth, the movements, of the eye and hand would become excessive because the muscles do not have to oppose gravity. The timing and amount of movement must be arranged for appropriate actions. The sensation of motion would also be affected by the loss of gravity because the mechanism of the otolith, the major acceleration sensor, depends on gravity. The possible instability of the sensation of direction may cause mistakes in the direction of control of manipulator movement. Thus, the experimental data can be used for designing man-machine systems in space, as well as for investigation of physiological mechanisms. In this experiment, the direction of vehicle heading is expressed by a light spot on an array of light emitting diodes and the manipulator is of a finger stick type. As the light spot moves up and down, the Japanese Payload Specialist, and the subject, must move the manipulator forward and backward to keep the movement of the light spot within the neighborhood of the central point of the display. The position of the light spot is computed in such a manner that when the stick is kept at the neutral position, a motion whose acceleration is proportional to the angle of deflection is added to the movement of the light spot. The Operator Describing Function, which is an expression of human control characteristics, can be calculated from 2 minutes of raw data of the light spot position and stick deflection. The 2 minutes of operation is called a run, and 8 runs with resting periods composes a session. The on-orbit experiment will be conducted on the second, fourth, and seventh days. One session of experiment of each of these days is conducted following the L-4 experiment, which uses the same apparatus. The Payload Specialist, aided by a Mission Specialist, will take our apparatus from a rack container, set up the apparatus, attach electrodes for measurements of eye movement and muscle activity, conduct the L-4 Visual Stability Experiment, conduct one session of the manual control experiment, and then dissemble and stow the apparatus. In addition to the flight experiment, pre-flight and post-flight experiments will be conducted. The data of three sessions on orbit will reflect adaption of physiological systems to microgravity. The data of post-flight experiments, on the other hand, will reflect re-adaptation of physiological systems to the gravity condition on the ground. Control data collected with and without psychological tension will be scheduled just prior to and long before launch.

Trace-element composition of Chicxulub crater melt rock, K/T tektites and Yucatan basement

The Cretaceous/Tertiary (K/T) boundary Chicxulub impact is the best preserved large impact in the geologic record. The Chicxulub crater has been buried with no apparent erosion of its intracrater deposits, and its ejecta blanket is known and is well preserved at hundreds of localities globally. Although most of the molten material ejected from the crater has been largely altered, a few localities still preserve tektite glass. Availability of intra- and extracrater impact products as well as plausible matches to the targeted rocks allows the comparison of compositions of the different classes of impact products to those of the impacted lithologies. Determination of trace-element compositions of the K/T tektites, Chicxulub melt rock, and the targeted Yucatan silicate basement and carbonate/evaporite lithologies have been made using instrumental neutron activation analysis (INAA) and inductively coupled plasma mass spectrometry (ICP-MS). Some sample splits were studied with both techniques to ensure that inter-laboratory variation was not significant or could be corrected. The concentration of a few major and minor elements was also checked against microprobe results. Radiochemical neutron activation analysis (RNAA) was used to determine Ir abundances in some samples.

SEDAC data and applications development plan V(1)

This Data and Applications Development Plan describes how the Socioeconomic Data and Applications Center (SEDAC) at the Consortium for International Earth Science Information Network (CIESIN) is responding to SEDAC's mission to develop new applications and information products that synthesize earth science and socioeconomic data in ways that support decision making. The SEDAC data and applications development effort is a primary means by which the Earth Observing System Data and Information System (EOSDIS) program can help to ensure that the scientific investment embodied in NASA's Mission to Planet Earth (MTPE) program leads to tangible benefits to the American people. The Plan describes the major issues concerned with global climate change, identifies important decision makers and other users who would benefit from improved data and information, and assesses key opportunities for responding to the needs of these users and the scientists who work with them. It then details specific plans for developing selected integrated data products and services and for making them available to the target users on an operational basis. This report provides a summary of the activities performed in connection with NASA/GSFC Letter Contract NAS5-3263 for the period shown.

The 'CommTech' Methodology: A Demand-Driven Approach to Efficient, Productive and Measurable Technology Transfer

Market research sources were used to initially gather primary technological problems and needs data from non-aerospace companies in targeted industry sectors. The company-supplied information served as input data to activate or start-up an internal, phased match-making process. This process was based on technical-level relationship exploration followed by business-level agreement negotiations, and culminated with project management and execution. Space Act Agreements represented near-term outputs. Company product or process commercialization derived from Lewis support and measurable economic effects represented far-term outputs.
