Preliminary Design and Testing of Candidate Coating Materials Applied to Inconel Bubblers for Improved Corrosion Resistance

The US Department of Energy (DOE) Office of Environmental Management Technology Development (EM-TD) has requested Savannah River National Laboratory (SRNL) evaluate ceramic coating materials that can extend the service life of Hanford melter bubblers. The proposed Task will focus on the development of ceramic coating materials for Inconel bubblers to reduce corrosion and increase the service life of the bubblers.

Nanopore Activity Assays for Detection of Biomarker Protease Activity: Design and Testing of Substrates for Both Nanopore Sequencing and PCR-Based Detection Methods

The work performed in this project has demonstrated the ability to construct proteolytic enzyme substrates that are PCR and sequencing-readable reporter molecules. Specifically, the goal was to detect those reporter molecules via PCR and Oxford Nanopore Technologies MinION sequencing methods following exposure to the biomarker protease thrombin. The assay development focused on binding the constructed peptide-oligonucleotide chimera to immobilized streptavidin. The action of thrombin on the peptide portion of the molecule released the oligonucleotide for detection. Detection of protease activity was demonstrated in a concentration-dependent manner using MALDI-MS, RT-PCR and DNA sequencing. Additional steps to remove background release of reporter molecules during the assay was used to improve the difference in detected oligonucleotide reporter following protease activity. Additional steps in assay development will be to (1) test the assay in an appropriate matrix, (2) investigate detection using additional DNA sequencing platforms and (3) demonstrate multiplexed detection of multiple protease markers in a single reaction.

Design and test of a special magnet for the Mu2e experiment

The Mu2e experiment (Muon-to-Electron Conversion Experiment) is looking for evidence that a muon can change into an electron without the emission of neutrinos. The experiment will search for the charged-lepton flavor violating (CLFV) neutrino-less conversion of a negative muon into an electron in the field of a nucleus. The conversion process results in a monochromatic electron with an energy of 104.97 MeV, slightly below the muon rest mass. The goal of the experiment is to improve the previous upper limit by four orders of magnitude and reach a SES (single event sensitivity) of 3 × 10 -17 on the conversion rate, a 90% CL of 8 × 10 -17 , and a 5σ discovery reach at 2 × 10 -16 . The experiment will use an intense pulsed negative muon beam, which is essential to reducing backgrounds. The other essential element is a sophisticated magnetic system composed of three consecutive solenoids (figure 1) that form the muon beam.

Methods for rule-based genome design

Methods and systems for designing, testing, and validating genome designs based on rules or constraints or conditions or parameters or features and scoring are described herein. A computer-implemented method includes receiving data for a known genome and a list of alleles, identifying and removing occurrences of each allele in the known genome, determining a plurality of allele choices with which to replace occurrences in the known genome, generating a plurality of alternative gene sequences for a genome design based on the known genome, wherein each alternative gene sequence comprises a different allele choice, applying a plurality of rules or constraints or conditions or parameters or features to each alternative gene sequence by assigning a score for each rule or constraint or condition or parameter or feature in each alternative gene sequence, resulting in scores for the applied plurality of rules or constraints or conditions or parameters or features, scoring each alternative gene sequence based on a weighted combination of the scores for the plurality of rules or constraints or conditions or parameters or features, and selecting at least one alternative gene sequence as the genome design based on the scoring.
