Development of a Freeze-Dried CRISPR-Cas12 Sensor for Detecting Wolbachia in the Secondary Science Classroom

Training the future synthetic biology workforce requires the opportunity for students to be exposed to biotechnology concepts and activities in secondary education. Detecting Wolbachia bacteria in arthropods using polymerase chain reaction (PCR) has become a common way for secondary students to investigate and apply recombinant DNA technology in the science classroom. Despite this important activity, cutting-edge biotechnologies such as clustered regularly interspaced short palindromic repeat (CRISPR)-based diagnostics have yet to be widely implemented in the classroom. To address this gap, we present a freeze-dried CRISPR-Cas12 sensing reaction to complement traditional recombinant DNA technology education and teach synthetic biology concepts. The reactions accurately detect Wolbachia from arthropod-derived PCR samples in under 2 h and can be stored at room temperature for over a month without appreciable degradation. Here, the reactions are easy-to-use and cost less than $40 to implement for a classroom of 22 students including the cost of reusable equipment. We see these freeze-dried CRISPR-Cas12 reactions as an accessible way to incorporate synthetic biology education into the existing biology curriculum, which will expand biology educational opportunities in science, technology, engineering, and mathematics.

Sodium Hydrogen Sensor and Acoustic Flow Sensor Development

This report documents the development of diffusion-type hydrogen meters (DTHMs) as part of a steam generator leak detection system (SGLDS) of sodium-cooled fast reactors (SFR) and the demonstration of the capabilities of acoustic flowmeters, including a commercial UTFM (Flexim FLUXUS F721) and an Argonne HT acoustic flowmeter running on shear-to-longitudinal mode. It also evaluates two Argonne permanent magnet flowmeters (PMFMs).

SynthEsizing Novel H2 Sensors for Operational Resilience in Pipeline Infrastructure (SENSOR) (CRADA Final Report)

Hydrogen (H₂) is gaining attention as a versatile energy carrier with potential applications across industrial processes, power generation, and transportation. However, its practical deployment, particularly in large-scale distribution systems, faces significant infrastructure challenges. Transporting hydrogen through dedicated pipelines or blending it into existing natural gas networks can lead to serious issues, such as leakage due to the small size of hydrogen molecules and material degradation in pipelines through embrittlement. These technical risks raise safety concerns and could limit the integration of hydrogen into current energy infrastructures. Additionally, using hydrogen-enriched gas mixtures in combustion systems like turbines and engines introduces new performance and compatibility challenges that must be resolved before widespread use becomes feasible.

Role of passive remote sensors. Sensor System Panel report

Capabilities of present passive systems are described and the development of passive remote sensing systems for the more abundant tropospheric trace species is recommended. The combination of nadir-viewing spectrometers and solar occultation for tropospheric measurement of those gases having large stratospheric burdens is discussed. Development of a nadir-viewing instrument capable of obtaining continuous spectra in narrower bands is recommended. Gas filter radiometers for species specific measurements and development of a spectral survey instrument are discussed. Further development of aerosol retrieval algorithms, including polarization techniques, for obtaining aerosol thickness and size distributions is advised. Recommendations of specific investigations to be pursued are presented.

Comparative analysis of different sensor data (Landsat-TM and MOMS) for earth observation and impact on future sensor development

The missions of the German Modular Optoelectronic Multispectral Scanner (MOMS) aboard two STS flights demonstrated the feasibility of a novel concept with regard to both technical and scientific objectives. On account of the successful missions, a cooperation was agreed between the German Federal Minister for Research nad Technology and NASA for comparing MOMS observations with the more familiar operational Landsat-TM data over selected test sites, as a means of obtaining some relative measure of performance. This paper summarizes the results obtained and presents the MOMS-02, a further experimental representative of the MOMS program aiming at the realization of an operational system for the mid-nineties.
