Aerial photography for sensing plant anomalies

Changes in the red tonal response of Kodak Ektrachrome Infrared Aero 8443 film (EIR) are often incorrectly attributed solely to variations in infrared light reflectance of plant leaves, when the primary influence is a difference in visible light reflectance induced by varying chlorophyll contents. Comparisons are made among aerial photographic images of high- and low-chlorophyll foliage. New growth, foot rot, and boron and chloride nutrient toxicites produce low-chlorophyll foliage, and EIR transparency images of light red or white compared with dark-red images of high-chlorophyll foliage. Deposits of the sooty mold fungus that subsists on the honeydew produced by brown soft scale insects, obscure the citrus leaves' green color. Infected trees appear as black images on EIR film transparencies compared with red images of healthy trees.

The development of spectro-signature indicators of root disease impacts on forest stands

A field research program was begun in 1969 and intensified in 1970 on the physiology and biophysical responses of second-growth Douglas fir infected with root rot fungus. A double tramway system was suspended between three 100-foot instrument towers to carry sensors for measuring the energy response from above both healthy and infected trees. Processing and analysis was completed of airborne multispectral scanner imagery collected over the Wind River research area in 1969. Likelihood ratio processing of three-channel infrared data and Euclidean distance analysis of ten-channel spectrometer data did not identify incipient root rot infection outside the training sets. In all cases infected fir was misclassified as healthy fir. It was concluded from careful examination of physiological data that Poria root rot infection has little effect on water metabolism and energy exchange. What was identified was a low-grade stress that affects respiration and metabolism over long periods of time. This led to minor changes in the external physical symptoms of Poria-infected trees which was revealed only in the shortwave reflectance data.

Analysis of commercial equipment and instrumentation for Spacelab payloads. Volume 3: Design analysis and trade studies

A detailed analysis is presented of each selected equipment item, and suitability and cost analyses were documented by equipment item. Tradeoffs of alternative specification requirements are presented which include possible relaxation of vibration, material control, fungus and corrosion requirements for experiment equipment. An additional tradeoff was performed to determine whether it is cost effective to modify experiment equipment to be compatible with a 28-volt dc power source rather than the conventional 110-volt ac source. Programmatic analysis data are given which were used as the basis for the extension of results from the analyses of specific equipment items to the entire spacelab experiment program.

Environmental testing of flat plate solar cell modules

Commercially available flat-plate solar cell modules have been subjected to a variety of environmental tests designed to simulate service conditions. Among the tests are those simulating heat and rain, wind-driven rains, humidity and freezing, humidity and heat, humidity with a voltage bias, salt fog, hail impact, and fungus infestation. Tests for optical surface soiling and the combined effects of temperature, humidity and UV irradiation are under development. A correlation has been demonstrated between degradation caused by the qualification tests and such observed field effects as power loss.

Neurospora circadian rhythms in space - A reexamination of the endogenous-exogenous question

To test the functioning of circadian rhythms removed from periodicities of the earth's 24-hour rotation, the conidiation rhythm of the fungus Neurospora crassa was monitored in constant darkness during spaceflight. The free-running period of the rhythm was the same in space as on the earth, but there was a marked reduction in the clarity of the rhythm, and apparent arrhythmicity in some tubes. At the current stage of analysis of the results there is insufficient evidence to determine whether the effect seen in space was related to removal from 24-hour periodicities and whether the circadian timekeeping mechanism, or merely its expression, was affected.

Spacelab 1 and the Life Sciences Flight Experiments Program

The Life Sciences Flight Experiments Program (LSFEP) was established by NASA in 1978 to plan and direct efforts necessary to conduct a continuing program of in-flight life science investigations throughout the Space Shuttle era. The Spacelab 1 (SL-1) mission, conducted from November 28 to December 8, 1983, was to verify Spacelab performance through a variety of scientific experiments including life science. A description is given of the seven NASA life sciences experiments, which consisted of four human experiments, a fungus experiment, a plant experiment, and radiation experiments. Ten life sciences experiments from the European Space Agency were also flown. The experiments include studies of the circadian rhythms in Neurospora crassa, the nutation of Helianthus annus, the vestibular function during weightlessness, the influence of space flight on erythrokinetics in man, and the adaptation of vestibulo-spinal reflex mechanisms during space flight.

Preliminary characterization of persisting circadian rhythms during space flight

In order to evaluate the function of the circadian timing system in space, the circadian rhythm of conidiation of the fungus Neurospora crassa was monitored in constant darkness on the STS 9 flight of the Space Shuttle Columbia. During the first 7 days of spaceflight many tubes showed a marked reduction in the apparent amplitude of the conidiation rhythm, and some cultures appeared arrhythmic. There was more variability in the growth rate and circadian rhythms of individual cultures in space than is usually seen on earth. The results of this experiment indicate that while the circadian rhythm of Neurospora conidiation can persist outside of the earth's environment, either the timekeeping process or its expression is altered in space.

Rust Inhibitor And Fungicide For Cooling Systems

Mixture of benzotriazole, benzoic acid, and fungicide prevents growth of rust and fungus. Water-based cooling mixture made from readily available materials prevents formation of metallic oxides and growth of fungi in metallic pipes. Coolant remains clear and does not develop thick sludge tending to collect in low points in cooling systems with many commercial rust inhibitors. Coolant compatible with iron, copper, aluminum, and stainless steel. Cannot be used with cadmium or cadmium-plated pipes.

Effects of natural environment on first generation solid rocket booster thermal protection system materials

The effort to demonstrate, by real-time exposure, the effects of the natural environment at Kennedy Space Center, Florida, upon the Thermal Protection System (TPS) of the Solid Rocket Booster (SRB) is summarized, and that the overall SRB TPS configuration is verified to meet all requirements for resistance to the conditions associated with outdoor weathering, including: solar radiation; temperature; humidity; precipitation; wind; sand/dust abrasion; static electricity; salt spray; fungus; and atmospheric oxidants. The evaluation criterion for this project was based upon flatwise tensile properties, visual inspection, color change, and thermal performance. Based upon the evaluation of the changes in these properties, it is concluded that properly applied and topcoat-protected TPS can satisfactorily withstand the conditions of the natural environment at KSC for exposures up to six months.

Preliminary crystallographic examination of a novel fungal lysozyme from Chalaropsis

The lysozyme from the fungus of the Chalaropsis species has been crystallized. This lysozyme displays no sequence homology with avian, phage, or mammalian lysozymes, however, preliminary studies indicate significant sequence homology with the bacterial lysozyme from Streptomyces. Both enzymes are unusual in possessing beta-1,4-N-acetylmuramidase and beta-1,4-N,6-O-diacetylmuramidase activity. The crystals grow from solutions of ammonium sulfate during growth periods from several months to a year. The space group is P2(1)2(1)2(1) with a = 34.0 A, b = 42.6 A, c = 122.1 A. Preliminary data indicate that there is 1 molecule/asymmetric unit.

Identification of a far-red reflectance response to ectomycorrhizae in slash pine

Spectral reflectances were measured for the canopies of Pinus elliottii seedlings that were inoculated versus noninoculated with the ectomycorrhizal fungus Pisolithus tinctorius. Reflectances of the less vigorous, noninoculated seedlings increased substantially at visible and near-IR wavelengths compared to the more vigorous, inoculated seedlings. An algorithm was developed to indicate the spectral regions in which reflectance was most sensitive to ectomycorrhizae. The reflectance curve of the more vigorous seedlings was used as a standard of comparison. The spectral regions defined by this algorithm are denoted as regions of maximum reflectance sensitivity. Far-red and near-IR photography filtered to the region of maximum sensitivity (700 nm) were used to enable visualization of a 'blue shift' reflectance response to decreased vigor in the noninoculated seedlings in response to these differences of mycorrhizal development.

PCR Based Microbial Monitor for Analysis of Recycled Water Aboard the ISSA: Issues and Prospects

The monitoring of spacecraft life support systems for the presence of health threatening microorganisms is paramount for crew well being and successful completion of missions. Development of technology to monitor spacecraft recycled water based on detection and identification of the genetic material of contaminating microorganisms and viruses would be a substantial improvement over current NASA plans to monitor recycled water samples that call for the use of conventional microbiology techniques which are slow, insensitive, and labor intensive. The union of the molecular biology techniques of DNA probe hybridization and polymerase chain reaction (PCR) offers a powerful method for the detection, identification, and quantification of microorganisms and viruses. This technology is theoretically capable of assaying samples in as little as two hours with specificity and sensitivity unmatched by any other method. A major advance in probe-hybridization/PCR has come about in a technology called TaqMan(TM), which was invented by Perkin Elmer. Instrumentation using TaqMan concepts is evolving towards devices that could meet NASA's needs of size, low power use, and simplicity of operation. The chemistry and molecular biology needed to utilize these probe-hybridization/PCR instruments must evolve in parallel with the hardware. The following issues of chemistry and biology must be addressed in developing a monitor: Early in the development of a PCR-based microbial monitor it will be necessary to decide how many and which organisms does the system need the capacity to detect. We propose a set of 17 different tests that would detect groups of bacteria and fungus, as well as specific eukaryotic parasites and viruses; In order to use the great sensitivity of PCR it will be necessary to concentrate water samples using filtration. If a lower limit of detection of 1 microorganism per 100 ml is required then the microbes in a 100 ml sample must be concentrated into a volume that can be added to a PCR assay; There are not likely to be contaminants in ISSA recycled water that would inhibit PCR resulting in false-negative results; The TaqMan PCR product detection system is the most promising method for developing a rapid, highly automated gene-based microbial monitoring system. The method is inherently quantitative. NASA and other government agencies have invested in other technologies that, although potentially could lead to revolutionary advances, are not likely to mature in the next 5 years into working systems; PCR-based methods cannot distinguish between DNA or RNA of a viable microorganism and that of a non-viable organism. This may or may not be an important issue with reclaimed water on the ISSA. The recycling system probably damages the capacity of the genetic material of any bacteria or viruses killed during processing to serve as a template in a PCR desinged to amplify a large segment of DNA (less than 650 base pairs). If necessary, vital dye staining could be used in addition to PCR, to enumerate the viable cells in a water sample; The quality control methods have been developed to insure that PCR's are working properly, and that reactions are not contaminated with PCR carryover products which could lead to the generation of false-positive results; and The sequences of the small rRNA subunit gene for a large number of microorganisms are known, and they consititue the best database for rational development of the oligonucleotide reagents that give PCR its great specificity. From those gene sequences, sets of oligonucleotide primers for PCR and Taqman detection that could be used in a NASA microbial monitor were constructed using computer based methods. In addition to space utilization, a microbial monitior will have tremendous terrestrial applications. Analysis of patient samples for microbial pathogens, testing industrial effluent for biofouling bacteria, and detection biological warfare agents on the battlefield are but a few of the diverse potential uses for this technology. Once fully developed, gene-based microbial monitors will become the fundamental tool in every lab that tests for microbial contaminants, and serve as a powerful weapon in mankind's war with the germ world.
