Effects of Polyhydroxybutyrate Production on Cell Division

Synthetic biological engineering can be utilized to aide the advancement of improved long-term space flight. The potential to use synthetic biology as a platform to biomanufacture desired equipment on demand using the three dimensional (3D) printer on the International Space Station (ISS) gives long-term NASA missions the flexibility to produce materials as needed on site. Polyhydroxybutyrates (PHBs) are biodegradable, have properties similar to plastics, and can be produced in Escherichia coli using genetic engineering. Using PHBs during space flight could assist mission success by providing a valuable source of biomaterials that can have many potential applications, particularly through 3D printing. It is well documented that during PHB production E. coli cells can become significantly elongated. The elongation of cells reduces the ability of the cells to divide and thus to produce PHB. I aim to better understand cell division during PHB production, through the design, building, and testing of synthetic biological circuits, and identify how to potentially increase yields of PHB with FtsZ overexpression, the gene responsible for cell division. Ultimately, an increase in the yield will allow more products to be created using the 3D printer on the ISS and beyond, thus aiding astronauts in their missions.

Code TS: Entry Systems and Technology Division

The TS division (Entry Systems and Technology Division) includes people who 1) Help design spacecraft for different exploration missions; 2) Figure out how hot the environments around a spacecraft will get; 3) Invent new materials that can protect the spacecraft; 4) Figure out how those materials will behave on a spacecraft and how thick they need to be; 5) Plan and perform tests on those materials and spacecraft designs to prove they will fly successfully; and 6) Help get those spacecraft ready to launch. This presentation will describe a little bit about all 6 areas.

NASA Earth Science Division’s Commitment to Increasing Safety in the Field

NASA's Earth Science Division (ESD) has led and supported field campaign research over many decades focused on advancing fundamental research, testing new instrument technologies, and promoting career development. ESD field campaign research is conducted over a wide range of projects that vary in size, science focus area, location, platform type, and people. ESD leadership has created a task team to address campaign physical and mental safety, with the goal of providing all participants in NASA field campaign research with an environment that promotes research, safety, inclusivity, and a positive experience. Building on resources that have been developed both within and outside NASA, we report on the task team’s accomplishments and near-term plans, including the recent establishment of a best practices document and guidelines for the development of “agreement of behaviors” document for field teams. We also describe progress in the development of an online training module for field campaign participation, the incorporation of campaign safety language in forthcoming NASA ROSES solicitations, and outreach activities. Finally, we report on recent joint interactions with the NASA Planetary Science Division’s Ethics in Fieldwork team.

Experimental and Analytical Verification of ASME Section IiII Division 5 Creep-Fatigue Design Rules

The continuous advancement of structural materials and the growing demands for more reliable and economical structural components in high-temperature reactor applications have necessitated the development of comprehensive design methodologies and design rules. Mechanical degradation of structural components at elevated temperatures subjected to cyclic deformation is controlled by the creep-fatigue damage. Over the past few decades, diligent research efforts have been dedicated to refining the development of elevated temperature design rules in the American Society of Mechanical Engineers (ASME) Boiler and Pressure Vessel Code (BPVC), Section III, Division 5 and to develop conservative design rules that can effectively guard against the risk of creep-fatigue failure. In ASME Section III, Division 5, for a design to pass the creep-fatigue acceptance criteria, creep damage and fatigue damage are evaluated separately, and these damages must not violate the bi-linear creep-fatigue interaction diagram, i.e., the so-called D-diagram. The creep-fatigue damage evaluation procedure assumes that the effects of the actual cyclic loading sequence can be bounded by assuming that the individual loading cycles are uniformly distributed throughout the component design life. In this study, creep-fatigue experiments with variable amplitudes and loading sequencies were designed and performed on Alloy 617 at high temperatures. The results were analyzed to evaluate the loading history effect on creep-fatigue damage accumulation and to verify the assumptions for the creep-fatigue evaluation design rules.

ECAR-6564 Rev 1 MARVEL Project Primary Coolant System ASME BPVC Section III Division 5 Design by Analysis

This report demonstrates that the MARVEL Reactor Primary Coolant Boundary (herein referred to as the Primary Coolant System, or PCS) is designed to meet ASME BPVC Section III Division 5 elevated temperature service design-by-analysis criteria. The analysis approach that delineates division of responsibilities to meet project objectives is discussed herein. In short, Design and Service Level A, B, and C code calculations are detailed in ECAR-6580 [9] for the majority of the PCS with complex geometry, while the Lower Downcomers, Bottom Head, and Reactor Core Barrel are analyzed in ANL-24/36. Service Level D code calculations are detailed in this document.

Preliminary design analysis workflow for Division 5 HHA-3200 requirements for graphite core components

This report presents a design analysis workflow for graphite core components and assemblies, based on the design rules of ASME Boiler Pressure and Vessel Code, Section III, Division 5, Article HHA-3000. The workflow contains three stages: developing the design of the graphite core component, modeling the component with the finite element software MOOSE, and assessing if the component passes/fails the criteria of the HHA-3000 design rules. Since the design rules use probabilistic metrics specifically established to evaluate brittle materials, we developed a python library that performs all the statistical calculations necessary for the evaluations of the HHA-3000 criteria.

The role of a cell surface inhibitor in early signal transduction associated with the regulation of cell division and differentiation

Serum stimulation of quiescent human fibroblast cultures resulted in a hyperphosphorylation of the nuclear retinoblastoma gene susceptibility product (RB). However, serum stimulation in the presence of 9 x 10(-8) M of a purified bovine sialoglycopeptide (SGP) cell surface inhibitor abrogated the hyperphosphorylation of the RB protein and the subsequent progression of cells through the mitotic cycle. The experimental results suggest that the SGP mediated its cell cycle arrest at a site in the cell cycle that was at the time of RB phosphorylation or somewhat upstream of the modification of this regulatory protein of cell division. Both cells serum-deprived and serum stimulated in the presence of the SGP displayed only a hypophosphorylated RB protein, consistent with the SGP-mediated cell cycle arrest point being near the G1/S interface.
