Irisin attenuates lipopolysaccharide-induced acute lung injury by downregulating inflammatory cytokine expression through miR-199a-mediated Rad23b overexpression

Highlights: • Acute lung injury (ALI) is a leading cause of mortality and therapies for ALI are yet to be thoroughly investigated. • Recent evidence has shown that irisin attenuates lipopolysaccharide-induced acute lung injury. • Irisin can alleviate ALI by inhibiting miR-199a and upregulating Rad23b expression. Acute lung injury (ALI) is a leading cause of mortality as a result of inflammatory cytokine overexpression and increased rates of apoptosis. Therapies for ALI are yet to be thoroughly investigated. Recent evidence has shown that irisin exerts protective effects against many types of pathologies. The present study aimed to determine the function of irisin in an ALI mouse model induced by lipopolysaccharide (LPS) and the corresponding underlying mechanisms at the tissue, cellular, and molecular levels.

In Situ Attenuated Total Reflectance Infrared Spectroelectrochemistry (ATR-IR-SEC) for the Characterization of Molecular Redox Processes on Surface-Proximal Doped Silicon ATR Crystal Working Electrodes

In situ mid-infrared spectroscopy is a powerful technique for understanding the mechanism of CO 2 reduction (CO 2 R) catalysts because it enables the direct detection of catalytic intermediates and products. Moreover, spectroelectrochemistry (SEC), the coupling of spectroscopy with electrochemistry, allows spectroscopic changes to be correlated with applied potentials to reveal potential-dependent intermediates that are often relevant to photoelectrochemical reactions. Hybrid photoelectrodes, comprised of a narrow bandgap semiconductor, like silicon (Si), with a covalently-linked molecular catalyst, are a promising platform for sunlight-driven catalysis, but characterization of the catalytic mechanism(s) is challenging under photoelectrochemical conditions, particularly when the catalyst is present in monolayer or less concentrations. Here, we have developed a new strategy to use multiple-reflection attenuated total reflectance IR spectroscopy (ATR-IR) coupled with electrochemistry to characterize catalysts directly integrated with a semiconductor surface under applied potential. We show that by surface-proximal n-type or p-type doping of the top ~100-200 nm of the crystal surface, Si ATR crystals can be used simultaneously as the internal reflection element and semiconductor working electrode for ATR-IR-SEC measurements. The surface-proximal doping strategy yields a quasi-equipotential surface with excellent infrared transparency that would have been compromised by free carrier absorption if the crystal was uniformly doped. This approach permits the catalytically-active functionalized surface to be directly probed without modification and overcomes signal-to-noise limitations of other strategies that use separately deposited working electrodes on Si ATR crystals. Proof-of-concept ATR-IR-SEC spectra were collected during the reduction and oxidation of monolayers of Re- and Ru-based transition metal carbonyl complexes, respectively, verifying the viability of the technique to probe redox processes associated with CO 2 R catalysts on Si electrode surfaces with high sensitivity.

Engineered Endosymbionts that Modulate Primary Macrophage Function and Attenuate Tumor Growth by Shifting the Tumor Microenvironment

Modulating gene expression in macrophages can be used to improve tissue regeneration and redirect tumor microenvironments (TMEs) toward positive therapeutic outcomes. We have developed Bacillus subtilis as an engineered endosymbiont (EES) capable of residing inside the eukaryotic host cell cytoplasm and controlling the fate of macrophages. Secretion of mammalian transcription factors (TFs) from B. subtilis that expresses listeriolysin O (LLO; allowing the EES to escape destruction by the macrophage) modulated expression of surface markers, cytokines, and chemokines, indicating functional changes in a macrophage/monocyte cell line. The engineered B. subtilis LLO TF strains were evaluated in murine bone marrow-derived macrophages (BMDMs) by flow cytometry, chemokine/cytokine profiling, metabolic assays, and RNA-Seq delivery of TFs by the EES shifted BMDM gene expression, production of cytokine and chemokines, and metabolic patterns, indicating that the TF strains could guide primary macrophage function. Thereafter, the ability of the TF strains to alter the TME was characterized in vivo in an orthotopic murine model of triple-negative breast cancer to assess therapeutic effects. The TF strains altered the TME by shifting immune cell composition and attenuating tumor growth. Additionally, multiple doses of the TF strains were well-tolerated by the mice. The use of B. subtilis LLO TF strains as EES showed promise as a unique cancer immunotherapy by directing the immune function intracellularly. The uses of EES could be expanded to modulate other mammalian cells over a range of biomedical applications.

A cryogenic white light absorption spectroscopy setup with in situ gamma irradiation and thermo-optical annealing for optical fiber radiation-induced attenuation characterization

Silica optical fiber sensors offer a fast, distributed measurement solution in various cryogenic and radiation environments, such as magnets for fusion power. Under these conditions, light-absorbing point defects limit lifetime via radiation-induced attenuation (RIA). To support RIA kinetics prediction, we present an in situ, broadband absorption spectroscopy setup combining gamma irradiation with liquid nitrogen cooling. The apparatus enables continuous monitoring of narrowband RIA levels and the use of secondary optical annealing light sources, alongside broadband spectrum measurements to characterize RIA defects through spectrum decomposition. Initial results confirm the inevitable photobleaching effect of the probe light source, which must be accounted for. In addition, we report RIA kinetics during cycles of gamma irradiation at 77 K and isochronal thermal annealing steps from liquid nitrogen to room temperature, showcasing the setup’s ability to replicate real fiber operation scenarios. These instruments form an ideal platform to further study the kinetics of RIA coupled with thermal and optical annealing.

ENPP1’s regulation of extracellular cGAMP is a ubiquitous mechanism of attenuating STING signaling

The metazoan innate immune second messenger 2'3'-cGAMP is present both inside and outside cells. However, only extracellular cGAMP can be negatively regulated by the extracellular hydrolase ENPP1. Here, we determine whether ENPP1’s regulation of extracellular cGAMP is a ubiquitous mechanism of attenuating stimulator of interferon genes (STING) signaling. We identified ENPP1 H362A , a point mutation that cannot degrade the 2'-5' linkage in cGAMP while maintaining otherwise normal function. The selectivity of this histidine is conserved down to bacterial nucleotide pyrophosphatase/phosphodiesterase (NPP), allowing structural analysis and suggesting an unexplored ancient history of 2'-5' cyclic dinucleotides. Enpp1 H362A mice demonstrated that extracellular cGAMP is not responsible for the devastating phenotype in ENPP1-null humans and mice but is responsible for antiviral immunity and systemic inflammation. Our data define extracellular cGAMP as a pivotal STING activator, identify an evolutionarily critical role for ENPP1 in regulating inflammation, and suggest a therapeutic strategy for viral and inflammatory conditions by manipulating ENPP1 activity.

Lifetime measurements of 0 + states in Er 168 with the Doppler-shift attenuation method

Here, the lowest-lying shape oscillations of deformed nuclei have been described as quadrupole in nature (λ = 2), resulting in two types of vibrations or oscillations: β vibrations with oscillations along the symmetry axis (K π = 0 + ) and γ vibrations breaking axial symmetry with a projection of K π = 2 + on the symmetry axis. The γ vibration seems to be well characterized as the first K π = $2^+_1$ (or $2^+_γ$) band in deformed nuclei and exhibits a systematic behavior across the region. The nature of the K π = 0 + excitations, however, has remained poorly understood and has been open to debate for some decades. The goal of this work is to understand the nature of 0 + states observed in 168 Er through measurements of the lifetimes of these states and to determine if they are consistent with oscillations built on a deformed ground state, the minima of other coexisting shapes, single-particle states, or a mixture of effects. Lifetimes of excited states in the 168 Er nucleus were measured with the Doppler Shift Attenuation Method (DSAM) and the inelastic neutron scattering reaction, (n, n'γ), at the University of Kentucky Accelerator Laboratory. Numerous 0 + states had been observed by the (p, t) reaction. We confirm the 0 + states at 1217.2, 1421.5, 1833.6, 2364.9, 2392.1, and 2643.0 keV in 168 Er. We could not, however, support the previous assignments of 0 + levels at 2114.1, 2200.6, 2572.5, and 2617.4 keV. We report measured lifetimes for six confirmed 0 + excitations and additional members of 0 + bands. The results for 168 Er show that it is the third excited K π = 0 + ($0^+_4$) excitation that carries the collective strength and, therefore, the potential to be an oscillation on the ground state. This result is similar to the case in 166 Er, where it was also the $0^+_4$ state that exhibited greater collectivity than the first excited K π = 0 + band. The Delaroche et al. prediction for a collective K π = 0 + band is at E T =1.818 MeV which corresponds the third excited K π = 0 + band.

Measurement of the production cross section of 31 GeV/ c protons on carbon via beam attenuation in a 90-cm-long target

The production cross section of 30.92 GeV/c protons on carbon is measured by the NA61/SHINE spectrometer at the CERN Super Proton Synchrotron by means of beam attenuation in a copy (replica) of the 90-cm-long target of the T2K neutrino oscillation experiment. The employed method for direct production cross-section estimation minimizes model corrections for elastic and quasielastic interactions. The obtained production cross section is $σ_{\text{prod}}$ = 227.6 ± 0.8(stat)$_{\text{-3.2}}^{\text{+1.9}}$(sys) – 0.8(mod) mb. It is in agreement with previous NA61/SHINE results obtained with a thin carbon target, while providing improved precision with a total fractional uncertainty of less than 2%. This direct measurement will reduce the uncertainty of the T2K neutrino flux prediction.

Omega-3 fatty acids attenuate cardiovascular effects of short-term exposure to ambient air pollution

Background: Exposure to air pollution is associated with elevated cardiovascular risk. Evidence shows that omega-3 polyunsaturated fatty acids (omega-3 PUFA) may attenuate the adverse cardiovascular effects of exposure to fine particulate matter (PM 2.5 ). However, it is unclear whether habitual dietary intake of omega-3 PUFA protects against the cardiovascular effects of short-term exposure to low-level ambient air pollution in healthy participants. In the present study, sixty-two adults with low or high dietary omega-3 PUFA intake were enrolled. Blood lipids, markers of vascular inflammation, coagulation and fibrinolysis, and heart rate variability (HRV) and repolarization were repeatedly assessed in 5 sessions separated by at least 7 days. This study was carried out in the Research Triangle area of North Carolina, USA between October 2016 and September 2019. Daily PM 2.5 and maximum 8-h ozone (O 3 ) concentrations were obtained from nearby air quality monitoring stations. Linear mixed-effects models were used to assess the associations between air pollutant concentrations and cardiovascular responses stratified by the omega-3 intake levels. Results: The average concentrations of ambient PM 2.5 and O 3 were well below the U.S. National Ambient Air Quality Standards during the study period. Significant associations between exposure to PM 2.5 and changes in total cholesterol, von Willebrand factor (vWF), tissue plasminogen activator, D-dimer, and very-low frequency HRV were observed in the low omega-3 group, but not in the high group. Similarly, O 3 -associated adverse changes in cardiovascular biomarkers (total cholesterol, high-density lipoprotein, serum amyloid A, soluable intracellular adhesion molecule 1, and vWF) were mainly observed in the low omega-3 group. Lag-time-dependent biphasic changes were observed for some biomarkers. Conclusions: This study demonstrates associations between short-term exposure to PM 2.5 and O 3 , at concentrations below regulatory standard, and subclinical cardiovascular responses, and that dietary omega-3 PUFA consumption may provide protection against such cardiovascular effects in healthy adults.

Qualitative Conditions for Reasonable Estimates of Kerma Using Total and Kerma Cross Section Attenuation of Photons

Performing a full adjoint simulation in the Integrated Tiger Series (ITS) can be time consuming to obtain statistically significant results. The ray-trace capability in ITS allows for rapid scoping calculations of the uncollided kerma from photon sources without needing to run a full adjoint simulation. However, the uncollided estimate will always underestimate the full-physics kerma since it neglects scattered radiation, and under certain conditions, the result from the capability may not provide a sufficiently accurate estimate of the full-physics kerma. To exemplify the conditions in which the capability provides reasonable estimates, two problem geometries with different materials are simulated using the full adjoint capability as well as the ray-trace capability with a total cross section treatment and a new kerma-attenuation cross-section treatment. The results are then compared to show under which conditions the estimates are accurate. Reasonable estimates are provided from the ray-trace feature when there is minimal scattering into a region of interest occurring, such as with low-Z material and when the photons travel through small amounts of material.

A Single Amino Acid Substitution in the Matrix Protein (M51R) of Vesicular Stomatitis New Jersey Virus Impairs Replication in Cultured Porcine Macrophages and Results in Significant Attenuation in Pigs

In this study, we explore the virulence of vesicular stomatitis New Jersey virus (VSNJV) in pigs and its potential relationship with the virus’s ability to modulate innate responses. For this purpose, we developed a mutant of the highly virulent strain NJ0612NME6, containing a single amino acid substitution in the matrix protein (M51R). The M51R mutant of NJ0612NME6 was unable to suppress the transcription of genes associated with the innate immune response both in primary fetal porcine kidney cells and porcine primary macrophage cultures. Impaired viral growth was observed only in porcine macrophage cultures, indicating that the M51 residue is required for efficient replication of VSNJV in these cells. Furthermore, when inoculated in pigs by intradermal scarification of the snout, M51R infection was characterized by decreased clinical signs including reduced fever and development of less and smaller secondary vesicular lesions. Pigs infected with M51R had decreased levels of viral shedding and absence of RNAemia compared to the parental virus. The ability of the mutant virus to infect pigs by direct contact remained intact, indicating that the M51R mutation resulted in a partially attenuated phenotype capable of causing primary lesions and transmitting to sentinel pigs. Collectively, our results show a positive correlation between the ability of VSNJV to counteract the innate immune response in swine macrophage cultures and the level of virulence in pigs, a natural host of this virus. More studies are encouraged to evaluate the interaction of VSNJV with macrophages and other components of the immune response in pigs.

EPCAPE-PT-LANL Measurements: Humidified Cavity Attenuated Phase Shift Spectroscopy

Coastal cities offer a unique environment for studying aerosol-cloud interactions and the effects of urban emissions on cloud properties. As part of the Eastern Pacific Cloud Aerosol Precipitation Experiment (EPCAPE), the Partitioning Thrust by Los Alamos National Laboratory (EPCAPE-PT-LANL) was conducted. Our campaign focused on measuring the optical and chemical properties of aerosols and their interactions within marine stratocumulus clouds in La Jolla, California. EPCAPE-PT-LANL enhances the primary goals of EPCAPE through innovative observations of vapor-phase transitions between aerosols and cloud droplets, the impact of black carbon on aerosol-cloud dynamics, and the effects of cloud processing on aerosol optical properties. Simultaneous real-time measurement of composition, flow, attenuation, density, and pipe-wallthickness in multiphase fluids

Apparatus (10) and methods for making simultaneous measurements of composition (water-cut), fluid flow, and sound attenuation in a multiphase fluid flowing (15) through a pipe (12) in real-time, using the same apparatus (10) are described. Additionally, the apparatus (10) provides real-time pipe wall thickness monitoring for observing pipe corrosion or internal deposition. Knowledge of wall thickness is necessary to correct for water-cut (oil-water composition) automatically by adjusting the liquid path length internal to the pipe (spool). The use of short duration frequency chirp excitation signals (24) enables the apparatus to provide information that can be used to extract multiple levels of information from the same measurement in multiphase fluids including the presence of a significant quantity of gas (˜60% gas volume fraction) in different flow regimes. Besides measuring steady flow, this device is useful for measurements during fast changing flows, such as for a rod-pumped well. Measurements up to about 1000 times a second can reliably be made.
