Determining a carbohydrate profile for Hansenula polymorpha

The determination of the levels of carbohydrates in the yeast Hansenula polymorpha required the development of new analytical procedures. Existing fractionation and analytical methods were adapted to deal with the problems involved with the lysis of whole cells. Using these new procedures, the complete carbohydrate profiles of H. polymorpha and selected mutant strains were determined and shown to correlate favourably with previously published results.

Development of an expert system prototype for determining software functional requirements for command management activities at NASA Goddard

The development of an expert system prototype for software functional requirement determination for NASA Goddard's Command Management System, as part of its process of transforming general requests into specific near-earth satellite commands, is described. The present knowledge base was formulated through interactions with domain experts, and was then linked to the existing Knowledge Engineering Systems (KES) expert system application generator. Steps in the knowledge-base development include problem-oriented attribute hierarchy development, knowledge management approach determination, and knowledge base encoding. The KES Parser and Inspector, in addition to backcasting and analogical mapping, were used to validate the expert system-derived requirements for one of the major functions of a spacecraft, the solar Maximum Mission. Knowledge refinement, evaluation, and implementation procedures of the expert system were then accomplished.

Nondestructive determination of mechanical properties

The sensitivity of the velocity of ultrasonic waves to the elasticity of the materials (acousto-elastic constant - AEC) as controlled by the microstructure of the materials was examined experimentally. Rolled-plate specimens of five different Al alloys were subjected to tensile tests while being monitored for the velocity change of a 10 MHz longitudinal wave as a function of the applied tensile stress. The acoustic propagation velocity was measured perpendicularly to the stress. Determinations were made of the yield strength and the hardnesses of the specimens. Samples of the plate materials examined microscopically to characterize the solid phases. The AECs varied linearly with the percentage of solid solution phase of the alloys. The trend for strain-hardened samples was, however, opposite from that in the precipitation-hardened alloys. The results underscore the necessity of considering the microstructural dependencies of the AECs when using ultrasonic NDE methods, and also opened the potential of determining some mechanical properties with NDE techniques.

Modeling, estimation and identification methods for static shape determination of flexible structures

This paper outlines methods for modeling, identification and estimation for static determination of flexible structures. The shape estimation schemes are based on structural models specified by (possibly interconnected) elliptic partial differential equations. The identification techniques provide approximate knowledge of parameters in elliptic systems. The techniques are based on the method of maximum-likelihood that finds parameter values such that the likelihood functional associated with the system model is maximized. The estimation methods are obtained by means of a function-space approach that seeks to obtain the conditional mean of the state given the data and a white noise characterization of model errors. The solutions are obtained in a batch-processing mode in which all the data is processed simultaneously. After methods for computing the optimal estimates are developed, an analysis of the second-order statistics of the estimates and of the related estimation error is conducted. In addition to outlining the above theoretical results, the paper presents typical flexible structure simulations illustrating performance of the shape determination methods.

Automated orbit determination using tracking and data relay satellite (TDRS) data

A prototype for an onboard satellite orbit determination system has been developed at the Goddard Space Flight Center (GSFC) to aid in the research and development of onboard navigation. Two versions of the system, one for laboratory investigation and one for ground demonstration, are being used to gain insight into the feasibility and practicality of microprocessor-based automated orbit determination. This paper describes the mathematical modeling and the software structure of each system and presents the results of laboratory evaluation in terms of computational accuracy and real-time processing performance.

Comments on determination of bandgap narrowing from activation plots

A determination is made of the temperature-dependence of emitter saturation current in bipolar devices which allows the derivation of a value for bandgap narrowing that is in better agreement with other determinations than previous results based on ohmic contact measurements of temperature dependence. The new values were obtained by varying the surface recombination velocity at the emitter surface. This improves accuracy by varying the minority carrier surface recombination velocity at the emitter contacts of otherwise indistinguishable emitters.

Preliminary design of cracked-lap shear specimen thickness for determination of interlaminar fracture toughness

This paper outlines a technique for preliminary sizing of the adherends for cracked-lap-shear specimen to assure delamination instead of adherend failure. The cracked-lap-shear specimen is a popular specimen used for determination of mixed mode interlaminar fracture toughness of composites. Thin adherends, on the order of three plies thick, have been used successfully in the past for determination of fracture toughness of rather brittle matrix systems. However, the tougher matrices now being considered for composite applications require thicker adherends in order to get composite delamination rather than adherend failure. The optimum thickness is obtained on the basis that the critical value of the strain energy release rate is reached at the delamination front before the ultimate strength is reached in the adherend. By proper design the specimen would delaminate and not use excessive material or load. A simple analysis for the optimum adherend thickness and experimental verification are included.

Magellan spacecraft - Attitude determination, updates and gyro parameter calibration

A detailed mathematical algorithm for onboard autonomous attitude determination and parameter calibration in the Magellan Venus-orbiting spacecraft has been developed, and its performance has been estimated. This algorithm uses a contamination reduction attitude profile and retractive star scan maneuvers to minimize the interparameter error contamination and to maximize the parameter observability. The details of the underlying theory and strategy are described, and an extensive performance analysis with worst-case parameter errors is reported which indicates that the in-flight attitude determination unit/parameter calibration (ADU/PC) algorithm will be able to meet all the different mission phase requirements with comfortable margins. The problem of nonexact mapping retrace due to uncertain performance of control authority is addressed, and its lack of impact on the system performance is demonstrated.

Observational determination of the masses of active galactic nuclei

The masses of 24 active galactic nuclei are determined on the basis of published observations, using data on both the X-ray variability and the width of the narrow forbidden lines of O III. The results are presented in tables and graphs, and it is found that the mass estimates based on the X-ray variability are in good agreement with the dynamic masses determined from the O III linewidth when reasonable values are selected for the radius of the region of X-ray origin and when a standard photoionization model is adopted for the line-emitting gas. The O III dynamic masses of Seyfert-2 nuclei are shown to be systematically lower than those predicted by the (delta t)/M(O III) correlation for Seyfert-1 nuclei.

Tracking and Data Relay Satellite (TDRS) tracking and orbit determination

Analysis results were derived using the Goddard Trajectory Determination System to determine the orbit of TDRS-East. A batch weighted least-squares algorithm is used to fit the Bilateration Ranging Transponder system tracking measurements. Six state parameters (position and velocity components) and the solar radiation pressure coefficient are estimated. The solutions are obtained by fitting the data over 34 hr arcs that overlap by 10 hr. Ephemerides are generated over the 34 hr arcs and then compared over the overlapping regions. The position differences indicate the consistency of the solutions and give a measure of their accuracy. The effect of the modeling of perturbative forces (solar radiation pressure, nonspherical geopotential field) on these results are studied. Error analysis is performed using the ORAN program to estimate the effect of force-model errors and measurement-related errors on the TDRS-East ephemeris.

Ultrasonic determination of recrystallization

Ultrasonic attenuation was measured for cold worked Nickel 200 samples annealed at increasing temperatures. Localized dislocation density variations, crystalline order and colume percent of recrystallized phase were determined over the anneal temperature range using transmission electron microscopy, X-ray diffraction, and metallurgy. The exponent of the frequency dependence of the attenuation was found to be a key variable relating ultrasonic attenuation to the thermal kinetics of the recrystallization process. Identification of this key variable allows for the ultrasonic determination of onset, degree, and completion of recrystallization.

Determining the influence and effects of manufacturing variables on sulfur dioxide cells

A survey of the Li/SO2 manufacturing community was conducted to determine where variability exists in processing. The upper and lower limits of these processing variables might, by themselves or by interacting with other variables, influence safety, performance, and reliability. A number of important variables were identified and a comprehensive design experiment is being proposed to make the proper determinations.

Fluorometric method for the determination of gas-phase hydrogen peroxide

The fluorometric gas-phase hydrogen peroxide procedure is based on the technique used by Lazrus et. al. for the determination of H2O2 in the liquid phase. The analytical method utilizes the reaction of H2O2 with horseradish peroxidase and p-hydroxphenylacetic acid (POPHA) to form the fluorescent dimer of POPHA. The analytical reaction responds stoichiometrically to both H2O2 and some organic hydroperoxides. To discriminate H2O2 from organic hydroperoxides, catalase is used to preferentially destroy H2O2. Using a dual-channel flow system the H2O2 concentration is determined by difference.

Comparison of Hirs' equation of Moody's equation for determining rotordynamic coefficients of annular pressure seals

The rotordynamic coefficients of an incompressible-flow annular pressure seal were determined using a bulk-flow model in conjunction with two different friction factor relationships. The first, Hirs' equation, assumes the friction factor is a function of Reynolds number only. The second, Moody's equation, approximates Moody's diagram and assumes the friction factor is a function of both Reynolds number and relative roughness. For each value of relative roughness, Hirs' constants were determined so that both equations gave the same magnitude and slope of the friction factor. For smooth seals, both relationships give the same results. For rough seals (e/2 H sub 0 = 0.05) Moody's equation predicts 44% greater direct stiffness, 35% greater cross-coupled stiffness, 19% smaller cross-coupled damping, 59% smaller cross-coupled inertia, and nominally the same direct damping and direct inertia.

Determination of surface shear stress with the naphthalene sublimation technique

Aeolian entrainment and transport are functions of surface shear stress and particle characteristics. Measuring surface shear stress is difficult, however, where logarithmic wind profiles are not found, such as regions around large roughness elements. An outline of a method whereby shear stress can be mapped on the surface around an object is presented. The technique involves the sublimation of naphthalene (C10H8) which is a function of surface shear stress and surface temperature. This technique is based on the assumption that the transfer of momentum, heat and mass are analogous (Reynolds analogy). If the Reynolds analogy can be shown to be correct for a given situation, then knowledge of the diffusion of one property allows the determination of the others. The analytical framework and data acquisition for the method are described. The technique was tested in the Planetary Geology Wind Tunnel. Results show that the naphthalene sublimation technique is a reasonably accurate method for determining shear stress, particularly around objects where numerous point values are needed.

Determination of diffusion coefficients in polypyrrole thin films using a current pulse relaxation method

The current pulse E sub oc relaxation method and its application to the determination of diffusion coefficients in electrochemically synthesized polypyrrole thin films is described. Diffusion coefficients for such films in Et4NBF4 and MeCN are determined for a series of submicron film thicknesses. Measurement of the double-layer capacitance, C sub dl, and the resistance, R sub u, of polypyrrole thin films as a function of potential obtained with the galvanostatic pulse method is reported. Measurements of the electrolyte concentration in reduced polypyrrole films are also presented to aid in the interpretation of the data.
