Hydrodynamics of countercurrent flow in an additive-manufactured column with triply periodic minimal surfaces for carbon dioxide capture

Computational investigation of the countercurrent flows in an additively manufactured (AM) column packed with triply periodic minimal surface (TPMS) is challenging. Multiphase flow simulations using the volume of fluid (VOF) method can explain the column’s hydrodynamic characteristics, including interfacial area, liquid holdup, pressure drop, flow regime, etc. The interfacial area is a key factor dictating the mass transfer via the absorption process, and thereby the overall efficiency of the column. The impacts of a solvent's physical properties, liquid and gas loads, and contact angle on the hydrodynamics are extensively explored. The solvents include traditional solvent (monoethanolamine [MEA]) used in industry as well as a water-lean solvent (EEMPA) as prospective solvents for carbon capture. As expected, interfacial area and liquid holdup increase with increasing liquid loads (q L ). The EEMPA exhibits higher values for the interfacial area and liquid holdup compared to those for the MEA. The dry and wet pressure drops per unit length in the column at different gas loads (F G ) are calculated and further compared with the empirical correlations for traditional packings. At low gas loads, i.e., in the preloading regime, the predicted values of the wet and dry pressure drops match well with the corresponding values calculated from available correlations. The gas load has a marginal impact on the interfacial area at its lower value, but the higher value of F G (>2.18 $\sqrt{Pa}$) causes flooding in the column. The effect of varying contact angles on the interfacial area and liquid holdup in the preloading regime is also studied. Both interfacial area and liquid holdup decrease with the increase in the value of the contact angle. Altogether, the effect of the contact angle on interfacial area is more pronounced than other parameters influencing the interfacial area.

Integrated lignocellulosic biorefinery for efficient production of furans and photothermal materials

Integrated lignocellulosic biorefineries offer a great potential to valorize all the components in lignocellulose into products, including fuels, chemicals, and materials. However, because of lignocellulose recalcitrance, conversion of bioresources remains a techno-economic challenge for many lignocellulosic biorefineries. In this work, we have proposed a sustainable and profitable biorefinery strategy for lignocellulose fractionation and conversion. In this design, a biphasic solvent consisting of a molten salt hydrate LiCl·4H 2 O and γ-valerolactone (GVL) was initially used for separating hemicellulose from lignocellulose. More interestingly, 100 wt% of biorefinery products from lignin were directly converted to functional photothermal materials by coordinating with Fe 3+ for solar-thermal-electricity conversion. Attributed to this rational design, we report the techno-economic analysis predicts a revenue of 439.3 USD by processing 100 kg of lignocellulosic biomass using the above developed method.

Constructing uniform oxygen defect engineering on primary particle level for high-stability lithium-rich cathode materials

Lithium-rich layered cathode materials are considered to be research focus of cathode candidates for next generation lithium-ion batteries due to their high specific capacity and low cost. However, lattice deoxidation associated with elemental migration and internal local shrinkage usually results in deteriorated cyclic performance and notorious voltage attenuation, severely limiting its application. In this paper, we have successfully injected uniform oxygen defects into surface region of Li 1.2 Ni 0.13 Co 0.13 Mn 0.54 O 2 primary particles under the high-pressure and weak carbonate environment. Various experimental investigations indicate that the injected robust oxygen defects can not only mitigate detrimental interfacial reactions but also suppress unfavorable lattice variation and particle breakage. More importantly, theoretical calculations unravel the critical roles of oxygen defects in regulating energy band structure for strengthened anionic reversibility. Owing to stabilization effects of unique oxygen defect engineering, the modified Li 1.2 Ni 0.13 Co 0.13 Mn 0.54 O 2 cathode has harvested dramatically enhanced electrochemical performance including a high initial coulombic efficiency of 96.6%, an outstanding capacity retention of 91.96% (1C, 200 cycles) and suppressed voltage decay of only 1.62 mV per cycle. Therefore, this facile and effective defect engineering strategy could establish new guidance for promoting practical application of Li-rich Mn-based cathode material.

Stabilizing high-Ni cathodes with gradient surface Ti-enrichment

High-Ni cathodes are being intensely pursued worldwide for electric vehicles and other energy-dense applications due to their high capacity and low cost. However, structural instabilities during electrochemical cycling and when subjected to thermal treatment have been the major issues hindering their practical deployment. We here report a rational design of coating-integrated-into-synthesis protocol for fabricating surface Ti-enriched LiNi 0.8 Mn 0.1 Co 0.1 O 2 (NMC811#Ti) material. The coating to intermediates is crucial to obtain high structural ordering, both in the bulk and surface of high-Ni cathodes, and the Ti substitute has a unique tri-valence (Ti 3+ ) in a gradient surface distribution. Further, the simulations of projected density of states in the atomistic understanding further certify significantly enhanced stability of lattice oxygen for the NMC811 through such a Ti 3+ -based structure reinforcement. Consequently, the NMC811#Ti cathode delivers a high capacity up to 200mAhg -1 at 0.1 C, along with superior stabilities during air-storage and thermal treatment (up to 297°C at the fully charged state under differential scanning calorimetric measurements). The corresponding NMC811#Ti||graphite full cell exhibits a desired 83.6% capacity retention after 1000 cycles at 0.5 C in a voltage range of 2.8–4.3V. This work demonstrates a delicate surface reinforcement to stabilize high-Ni cathodes for long-life and safe lithium-ion batteries.

Countercurrent flow characteristics of next generation solvent in novel 3D printed columns for carbon capture

Solvent based absorption for carbon dioxide capture in a packed column is being considered as an efficient technology for the decarbonization process of point source. Significant efforts are underway to improve the design of packings with the cutting edge 3D printing technology for efficient carbon capture. Accordingly, multiphase flow studies were conducted to assess the performance of novel 3D printed columns with various triply periodic minimal surface (TPMS) designs. The effects of solvent properties, liquid and gas loads on the performance of TPMS columns are extensively explored. Hydrodynamics of the potential water-lean solvents (EEMPA) as well as aqueous monoethanolamine (MEA) solvent for carbon capture are evaluated and compared. The interfacial area and liquid holdup increase with increasing liquid loads (q L ) for TPMS columns. Schwarz column consistently shows the highest liquid holdup value. The EEMPA exhibits higher values for the interfacial area (10–15 %) and liquid holdup (~4 %) in comparison to the MEA. TPMS columns exhibit the intermediate value of dry pressure drop between the random and the structure packed columns. Among selected TPMS columns, the gyroid packing shows the lowest pressure drop. The gas load has marginal impact on the interfacial area at lower value while a higher gas load leads to column’s flooding. Prior to the flooding, the interfacial area in TMPS packings rises with increased gas load at a fixed liquid load except Schwarz packing where interfacial area is incentive to the gas load. Additionally, the liquid holdup and wet pressure drop rise as gas load increases in TPMS columns. Overall, Fisher Koch packing is more susceptible to flood as compared to other TPMS packings. Flow regimes: loading and flooding are also delineated in the TPMS packings.

helixCAM: A platform for programmable cellular assembly in bacteria and human cells

Interactions between cells are indispensable for signaling and creating structure. The ability to direct precise cell-cell interactions would be powerful for engineering tissues, understanding signaling pathways, and directing immune cell targeting. In humans, intercellular interactions are mediated by cell adhesion molecules (CAMs). However, endogenous CAMs are natively expressed by many cells and tend to have cross-reactivity, making them unsuitable for programming specific interactions. Here, we showcase “helixCAM,” a platform for engineering synthetic CAMs by presenting coiled-coil peptides on the cell surface. helixCAMs were able to create specific cell-cell interactions and direct patterned aggregate formation in bacteria and human cells. Based on coiled-coil interaction principles, we built a set of rationally designed helixCAM libraries, which led to the discovery of additional high-performance helixCAM pairs. Finally, we applied this helixCAM toolkit for various multicellular engineering applications, such as spherical layering, adherent cell targeting, and surface patterning.

Tailoring Microbial Fitness Through Computational Steering and CRISPRi-Driven Robustness Regulation

The widespread application of genetically modified microorganisms (GMMs) across diverse sectors underscores the pressing need for robust strategies to mitigate the risks associated with their potential uncontrolled escape. This study merges computational modeling with CRISPR interference (CRISPRi) to refine GMM metabolic robustness. Utilizing ensemble modeling, we achieved high-throughput in silico screening for enzymatic targets susceptible to expression alterations. Translating these insights, we developed functional CRISPRi, boosting fitness control via multiplexed gene knockdown. Our method, enhanced by an insulator-improved gRNA structure and an off-switch circuit controlling a compact Cas12m, resulted in rationally engineered strains with escape frequencies below National Institutes of Health standards. The effectiveness of this approach was confirmed under various conditions, showcasing its ability for secure GMM management. This research underscores the resilience of microbial metabolism, strategically modifying key nodes to halt growth without provoking significant resistance, thereby enabling more reliable and precise GMM control. A record of this paper's transparent peer review process is included in the supplemental information.

Investigation of countercurrent flow profile and liquid holdup in random packed column with local CFD data

Liquid holdup and mass transfer area are critical parameters used in packed column design and CO 2 capture efficiency prediction. Compared to experimental methods, 3D computational fluid dynamics (CFD) can provide high-resolution data for studying countercurrent flow hydrodynamics in a packed column. In this paper, a framework was established for modeling the liquid–gas countercurrent flow in a random packed column with pall rings. Besides the column-averaged information, the radial pall ring distribution, velocity, and liquid holdup profiles are obtained in the simulations. The radial two-phase velocity and liquid holdup profiles show significant fluctuations caused by the non-uniform pall ring distribution. The entrance effect and wall influence are also observed in the packed column. By using local countercurrent flow information, the packing specific area $a_p$ and liquid velocity $u_L$ range are significantly expanded for liquid holdup hL prediction. This enables us to develop the liquid holdup correlation with one packing size and six CFD runs only. The proposed power law of the liquid holdup correlation $h_L~u_L^{0.44}$ indicates the random packed column falls in a viscous to turbulent transition regime with a Reynolds Number range of [6.7–40.2]. The derived liquid holdup correlation is in good agreement with existing correlations that are developed using the column-averaged experimental data.

Differentiation of static and dynamic interfacial area in the structured packed column

Effective mass-transfer area plays a key role for post-combustion carbon capture. The static area, an inactive part relative to mass transfer, needs to be differentiated in the interfacial area. In the present study, CFD method is employed to investigate static and dynamic interfacial areas in Mellapak 500.Y structured packing. Here, a wide range of physical properties and loading conditions are included to understand their effects on both areas. The result shows that the relationship between interfacial area and interfacial velocity at the local scale can be described by gamma distribution with good accuracy. The influences of various physical properties derived from simulation are compared with existing experiment-based correlations for both static and interfacial areas. The result shows the dominant influence of viscosity on the static area, and the most importance of the influence of contact angle on the interfacial area. This study also highlights the importance of viscosity, which is usually ignored.

VRC34-Antibody Lineage Development Reveals How a Required Rare Mutation Shapes the Maturation of a Broad HIV-Neutralizing Lineage

Rare mutations have been proposed to restrict the development of broadly neutralizing antibodies against HIV-1, but this has not been explicitly demonstrated. We hypothesized that such rare mutations might be identified by comparing broadly neutralizing and non-broadly neutralizing branches of an antibody-developmental tree. Because sequences of antibodies isolated from the fusion peptide (FP)-targeting VRC34-antibody lineage suggested it might be suitable for such rare mutation analysis, we carried out next-generation sequencing (NGS) on B cell transcripts from donor N123, the source of the VRC34 lineage, and functionally and structurally characterized inferred intermediates along broadly neutralizing and poorly neutralizing developmental branches. The broadly neutralizing VRC34.01 branch required the rare heavy-chain mutation Y33P to bind FP, whereas the early bifurcated VRC34.05 branch did not require this rare mutation and evolved less breadth. Our results demonstrate how a required rare mutation can restrict development and shape the maturation of a broad HIV-1-neutralizing antibody lineage.

Minimizing total energy cost and tardiness penalty for a scheduling-layout problem in a flexible job shop system: A comparison of four metaheuristic algorithms

Job scheduling and machine layout are interrelated in improving energy consumption (EC) and productivity measures such as tardiness and represent two important decisions that must be made by manufacturers. This interdependency can be explained by transportation time, which connects scheduling and layout. Scheduling and layout, however, have not been thoroughly studied in conjunction using an integrated model in the context of sustainable manufacturing. Hence, we propose an energy-aware optimization model in which scheduling is integrated with layout in a single-level framework. More specifically, a single objective function is defined to minimize the facility energy cost and the job tardiness penalty, which control EC and tardiness respectively in a flexible job shop system. In order to model machine EC more accurately, we also consider three different machine states: a processing state and two idle states. Our case studies show that the integrated model exhibits better performance in controlling manufacturing EC and job tardiness than a non-integrated model in which machine locations are uncontrollable and transportation times between machines are unchangeable. To deal with large-sized problems, we also introduce four new metaheuristics. The performances of these new metaheuristics are compared in terms of objective function values and CPU times using various case studies. The results indicate that a hybrid ant colony optimization and simulated annealing (ACO-SA) algorithm provides better performance than the other algorithms. Specifically, our case studies show that the integrated model using ACO-SA can improve the objective function value by around 5% when compared to the non-integrated model.

A two-equation soot-in-flamelet modeling approach applied under Spray A conditions

Soot production (including formation and oxidation) is studied in the transient, high-pressure and turbulent n-dodecane Spray A flames from the Engine Combustion Network (ECN) using computational fluid dynamics (CFD) simulations. A two-equation soot-in-flamelet modeling approach is applied within the framework of the Unsteady Flamelet Progress Variable (UFPV) model and results are validated against experimental data. Equations for soot mass fraction and soot number density derived in the mixture fraction space are solved in the context of detailed flamelet calculations. Source terms for the different steps in the soot chemistry are tabulated and incorporated in the flamelet manifold. For the reference condition, the modeling approach based on the tabulated flamelet manifold reduces the computational cost of a CFD calculation by approximately 40 times compared to a non-tabulated well-mixed (WM) modeling approach. The soot-in-flamelet approach is then extended to study the effect of ambient oxygen concentration, ambient mixture composition and ambient temperature on soot production. Results show that the modeling approach is able to capture the experimental trends for the soot volume fraction (SVF) with good quantitative agreement, especially in the soot ramp-up region.

Low- and intermediate-temperature oxidation of dimethyl ether up to 100 atm in a supercritical pressure jet-stirred reactor

Understanding the low- and intermediate-temperature oxidation chemistry of oxygenated fuels like dimethyl ether (DME) at high pressure is paramount to the development of advanced engines with low carbon emissions. The supercritical pressure jet-stirred reactor (SP-JSR) recently developed at Princeton provides a new platform for conducting kinetic studies at low and intermediate temperatures at extremely high pressures with a uniform temperature distribution and a short flow residence time. This paper uses the SP-JSR to investigate DME oxidation at equivalence ratios of 0.175, 1.0, and 1.72, for pressures of 10 and 100 atm, and temperatures ranging from 400 to 900 K. The results demonstrate weakened NTC behavior at 100 atm relative to 10 atm due to increased flux through QOOH + O 2 = O 2 QOOH relative to QOOH = 2 CH 2 O + OH at 100 atm. Furthermore, the intermediate-temperature oxidation window is shifted to lower temperatures at 100 atm. The experimental data are compared with several chemical kinetic models from the literature. The existing models are seen to agree quite well with the experimental data at 10 atm. However, the models fail to properly capture the NTC behavior at 100 atm. Reaction pathway analyses indicate that both the low- and intermediate-temperature chemistries are controlled by RO 2 consumption pathways. The reaction rates for several of the important reactions, such as DME + OH = CH 3 OCH 2 + H 2 O, H 2 O 2 (+M) = 2 OH (+M), and 2 HO 2 = 2 OH + O 2 are updated in this work. Furthermore, the updated model improves the predictability for all key species compared to the original model.
