Investigation into Stimulated Microgravity Techniques used to Study Biofilm Growth

Bacterial growth in liquid media in microgravity conditions is not well understood. Trends such as a shortened lag phase, longer log phase, slower growth rate, and a higher final population concentration have been noted but the underlying cause remains unclear. At the single cell level, it is predicted that bacteria are less gravity-sensitive than larger species. The effects on their immediate environment, including the lack of cell settlement and slower mass transfer of nutrients due to lack of density driven convection, could help explain the trends. Ground-based spaceflight analogs, or simulated microgravity devices, are often employed to achieve different attributes of weightlessness to study effects on bacterial growth. Though these technologies could isolate gravity’s role in various biological processes, they cannot replicate all its effects and underlying mechanisms. Hence, interpretation of results could be misleading, even if similar to spaceflight. In this study two common simulated microgravity devices were investigated to determine whether they could simulate relevant microgravity conditions for bacterial growth. A bioreactor, the high aspect ratio vessel (HARV), was used with dyes of different density mounted on a random positioning machine (RP machine) or a rotating wall vessel (RWV). The RP machine displayed higher mixing rates than the RWV. The RWV was further tested at different rotations per minute (RPM). The range to minimize effects of density driven convection (low speeds) or centrifugal forces (high speeds) was between a range of 15-20 RPM. These results will help inform the selection of simulated microgravity device as well as interpretation of subsequent biofilm growth results.

High-Temperature Stability of Aqueous Foams as Potential Waterless Hydrofracking Fluid for Geothermal Reservoir Stimulation

Hydraulic fracturing is the process of fracturing rock formations with high-pressure water-based fluids. In Enhanced Geothermal Systems (EGS) hydraulic fracturing is carried out by injecting high-pressure fluids into the Hot Dry Rocks (HDR) under carefully controlled conditions. The fluid used for fracturing is an important component for EGS, not only concerning the technical approch but also environmental impact. Recent research has been carried out to develop waterless fracturing technologies for EGS, including foam-based hydrofracking, where foams are mixtures of gas and liquid fluids. Foam fracturing fluids have potential benefits over water-based fluids because of less water consumption, less damage in water sensitive formations, and less liquid to recover and handle after fracturing process. One challenge for implementing foam fracturing in EGS is to achieve stable foams at high temperatures, as the foam stability tends to decay with increase in temperature. This research is focused on investigating the stability of foams at high temperatures. Preliminary results show that 1) N2 foams are more stable compared to CO2 foams, 2) foams containing only surfactants were not stable at high temperatures, and 3) addition of different stabilizing agents increased the thermal stability of all the foams. It was observed that different stabilizing agents interacted differently with anionic, nonionic, and cationic surfactants. It was further shown that the crosslinking agent with a base solution of pH 6 and graphene oxide dispersion was the most effective stabilizing agent for anionic surfactant AOS, while bentonite clay effectively enhanced thermal stability for another anionic surfactant SDS. SiO2 nanoparticles showed limited improvement in thermal stability at high temperatures compared to other stabilizing agents.

Solvent depression of transition temperature to selectively stimulate actuation of shape memory polymer foams

An embodiment of the invention is a shape memory polymer (SMP) foam designed to recover its original shape through exposure to a solvent. Thermo-responsive SMPs are polymers designed to maintain a programmed secondary shape until heated above their transition temperature, upon which the polymer recovers its original, or primary, shape. The thermo-responsive SMP foam is programmed to its secondary shape prior to use, typically compression of the foam to a small volume, and remains in this programmed shape until exposed to a selected solvent such as dimethyl sulfoxide or ethyl alcohol. Upon exposure to the solvent, the transition temperature of the SMP foam decreases below the temperature of the environment and the SMP foam actuates to its primary shape. The SMP foam is tailored to actuate upon exposure to specific solvents while minimizing or preventing actuation when exposed to water or other solvents. This selective solvent actuation can be used to increase working time of a SMP foam device, that is, the time allowed for use of a device without undesired actuation, while maintaining functional SMP actuation. Solvent actuated SMP foams can be used in various applications including, but not limited to, treatment of aneurysms and arterio-venous malformations, tissue engineering, and wound healing.

Peptides having reduced toxicity that stimulate cholesterol efflux

The present invention provides a family of non-naturally occurring polypeptides having cholesterol efflux activity that parallels that of full-length apolipoproteins (e.g., Apo AI and Apo E), and having high selectivity for ABCA1 that parallels that of full-length apolipoproteins. Further, the peptides of the invention have little or no toxicity when administered at therapeutic and higher doses. The invention also provides compositions comprising such polypeptides, methods of identifying, screening and synthesizing such polypeptides, and methods of treating, preventing or diagnosing diseases and disorders associated with dyslipidemia, hypercholesterolemia, or inflammation; or diseases involving abnormal glucose metabolism, e.g., diabetes, metabolic syndrome; or Alzheimer's Disease or frontotemporal dementia.

Cylindrical microelectrode array for neural stimulation and recording

A cylindrical microelectrode array having an elongated cylindrical core, and a multilayer structure conformally folded around and affixed to the cylindrical core so as to extend between opposite ends of the core. The multilayer structure has integrated sections including an electrode section with electrodes exposed through electrically insulating layers, a connector section with conductive bond pads for interfacing with external electronics, and a cable section with conductive traces encapsulated in electrically insulating layers and which connect between the electrodes and their corresponding bond pads. The array may be fabricated using a planar multilayer structure having the electrode, connector, and cable sections, and conformally folding the multilayer structure around and affixing to the cylindrical core. The cable section in particular may be conformally coiled around and affixed to the cylindrical core so that the electrical conduits helically extend between the connector and electrode sections.
