Utilizing Advanced Statistics to Determine Anomalistic Conditions in Pebble-Bed Reactors

Pebble-bed reactors (PBRs) utilize hundreds of thousands of fuel pebbles, which continuously circulate through the core, in lieu of traditional fuel assemblies to generate fissions and produce power. The use of unmarked fuel pebbles presents a challenge for international safeguards verification that nuclear material is not being diverted. To ensure pebble diversion is not taking place, new methods for accounting for and monitoring the pebbles should be examined to determine an appropriate methodology for performing bulk accountancy with pebbles. Here, this work examines the use of statistical methods for determining if the reactor is within a declared range of operation by examining the statistical distribution of pebble burnup as they are discharged from the core. Using this methodology, we created a model that detects diversion over 95% of the time, over multiple diversion pathways, if the reactor core maintains a constant power density during the diversion process and only falsely labels a diversion case nominal 2% of the time. For a diversion scenario where the reactor is maintained at a constant power, the statistical analysis can correctly identify if diversion is occurring over 80% of the time; however, nearly 20% of specific diversion pathways are mislabeled nominal. These results provide a basis and framework for exploring the further use of statistical methods to determine where these methods could be most useful and where additional methods, such as machine learning, could be used to capture if diversion is occurring in pebble-bed reactors.

A machine learning approach to determine the elastic properties of printed fiber-reinforced polymers

This work focuses on the simultaneous determination of the elastic constants and the fiber orientation state for a short fiber-reinforced polymer composite by performing a minimum of experimental tests. Here we introduce a methodology that enables the inverse determination of fiber orientation state and the in-situ polymer properties by performing tensile tests at the composite coupon level. We demonstrate the approach for the extrusion deposition additive manufacturing (EDAM) process to illustrate one application of the methodology, but the development is such that it can be applied to short fiber-reinforced polymer (SFRP) systems processed via other methods. Currently, developing composites additive manufacturing digital twins require extensive material characterization. In particular, the mechanical characterization of the orthotropic elastic properties of a composite involves extensive sample preparation and testing, therefore the elasticity tensor is generally populated using a micromechanics model. This, however, requires measuring the fiber orientation state in addition to knowing the constituent material properties. Experimentally measuring the fiber orientation state can be tedious and time consuming. Further, optical methods are limited to resolving the orientation of cylindrical fibers or cluster of non-cylindrical fibers, and computed tomography (CT) methods scan regions of volume that are much smaller than a full printed bead. Therefore, we propose a methodology, accelerated by machine learning, to identify the anisotropic mechanical properties and fiber orientation state at the same time. Early results show that inference of the fiber orientation and composite properties is possible with as few as three tensile tests. Our results show that a combination of the choice of the micromechanics model and reliable set of experiments can yield the nine elastic constants, as well as, the fiber orientation state.

Identifying sequence perturbations to an intrinsically disordered protein that determine its phase-separation behavior

Phase separation of intrinsically disordered proteins (IDPs) commonly underlies the formation of membraneless organelles, which compartmentalize molecules intracellularly in the absence of a lipid membrane. Identifying the protein sequence features responsible for IDP phase separation is critical for understanding physiological roles and pathological consequences of biomolecular condensation, as well as for harnessing phase separation for applications in bioinspired materials design. To expand our knowledge of sequence determinants of IDP phase separation, we characterized variants of the intrinsically disordered RGG domain from LAF-1, a model protein involved in phase separation and a key component of P granules. Based on a predictive coarse-grained IDP model, we identified a region of the RGG domain that has high contact probability and is highly conserved between species; deletion of this region significantly disrupts phase separation in vitro and in vivo. We determined the effects of charge patterning on phase behavior through sequence shuffling. We designed sequences with significantly increased phase separation propensity by shuffling the wild-type sequence, which contains well-mixed charged residues, to increase charge segregation. This result indicates the natural sequence is under negative selection to moderate this mode of interaction. We measured the contributions of tyrosine and arginine residues to phase separation experimentally through mutagenesis studies and computationally through direct interrogation of different modes of interaction using all-atom simulations. Finally, we show that despite these sequence perturbations, the RGG-derived condensates remain liquid-like. Together, these studies advance our fundamental understanding of key biophysical principles and sequence features important to phase separation.

Accelerating crystal structure determination with iterative AlphaFold prediction

Experimental structure determination can be accelerated with artificial intelligence (AI)-based structure-prediction methods such as AlphaFold . Here, an automatic procedure requiring only sequence information and crystallographic data is presented that uses AlphaFold predictions to produce an electron-density map and a structural model. Iterating through cycles of structure prediction is a key element of this procedure: a predicted model rebuilt in one cycle is used as a template for prediction in the next cycle. This procedure was applied to X-ray data for 215 structures released by the Protein Data Bank in a recent six-month period. In 87% of cases our procedure yielded a model with at least 50% of C α atoms matching those in the deposited models within 2 Å. Predictions from the iterative template-guided prediction procedure were more accurate than those obtained without templates. It is concluded that AlphaFold predictions obtained based on sequence information alone are usually accurate enough to solve the crystallographic phase problem with molecular replacement, and a general strategy for macromolecular structure determination that includes AI-based prediction both as a starting point and as a method of model optimization is suggested.

Differential early–life survival underlies the adaptive significance of temperature–dependent sex determination in a long–lived reptile

1. Many ectotherms rely on temperature cues experienced during development to determine offspring sex. The first descriptions of temperature-dependent sex determination (TSD) were made over 50 years ago, yet an understanding of its adaptive significance remains elusive, especially in long-lived taxa. 2. One novel hypothesis predicts that TSD should be evolutionarily favoured when two criteria are met—(a) incubation temperature influences annual juvenile survival and (b) sexes mature at different ages. Under these conditions, a sex-dependent effect of incubation temperature on offspring fitness arises through differences in age at sexual maturity, with the sex that matures later benefiting disproportionately from temperatures that promote juvenile survival. 3. The American alligator (Alligator mississippiensis) serves as an insightful model in which to test this hypothesis, as males begin reproducing nearly a decade after females. Here, through a combination of artificial incubation experiments and mark-recapture approaches, we test the specific predictions of the survival-to-maturity hypothesis for the adaptive value of TSD by disentangling the effects of incubation temperature and sex on annual survival of alligator hatchlings across two geographically distinct sites. 4. Hatchlings incubated at male-promoting temperatures (MPTs) consistently exhibited higher survival compared to those incubated at female-promoting temperatures. This pattern appears independent of hatchling sex, as females produced from hormone manipulation at MPT exhibit similar survival to their male counterparts. 5. Additional experiments show that incubation temperature may affect early-life survival primarily by affecting the efficiency with which maternally transferred energy resources are used during development. 6. Results from this study provide the first explicit empirical support for the adaptive value of TSD in a crocodilian and point to developmental energetics as a potential unifying mechanism underlying persistent survival consequences of incubation temperature.

Genome-wide DNA methylation patterns harbour signatures of hatchling sex and past incubation temperature in a species with environmental sex determination

Conservation of thermally sensitive species depends on monitoring organismal and population-level responses to environmental change in real time. Epigenetic processes are increasingly recognized as key integrators of environmental conditions into developmentally plastic responses, and attendant epigenomic data sets hold potential for revealing cryptic phenotypes relevant to conservation efforts. Here, we demonstrate the utility of genome-wide DNA methylation (DNAm) patterns in the face of climate change for a group of especially vulnerable species, those with temperature-dependent sex determination (TSD). Due to their reliance on thermal cues during development to determine sexual fate, contemporary shifts in temperature are predicted to skew offspring sex ratios and ultimately destabilize sensitive populations. Using reduced-representation bisulphite sequencing, we profiled the DNA methylome in blood cells of hatchling American alligators (Alligator mississippiensis), a TSD species lacking reliable markers of sexual dimorphism in early life stages. We identified 120 sex-associated differentially methylated cytosines (DMCs; FDR < 0.1) in hatchlings incubated under a range of temperatures, as well as 707 unique temperature-associated DMCs. We further developed DNAm-based models capable of predicting hatchling sex with 100% accuracy (in 20 training samples and four test samples) and past incubation temperature with a mean absolute error of 1.2°C (in four test samples) based on the methylation status of 20 and 24 loci, respectively. Though largely independent of epigenomic patterning occurring in the embryonic gonad during TSD, DNAm patterns in blood cells may serve as nonlethal markers of hatchling sex and past incubation conditions in conservation applications. These findings also raise intriguing questions regarding tissue-specific epigenomic patterning in the context of developmental plasticity.

The architecture of resilience: a genome assembly of Myrothamnus flabellifolia sheds light on desiccation tolerance and sex determination

Myrothamnus flabellifolia is a dioecious resurrection plant endemic to southern Africa that has become an important model for understanding desiccation tolerance. Despite its ecological and medicinal significance, genomic and transcriptomic resources for the species are limited. We generated a chromosome-level, haplotype-resolved reference genome assembly and annotation for M. flabellifolia and conducted transcriptomic profiling across a natural dehydration–rehydration time course in the field. Genome architecture and sex determination were characterized, and co-expression network and cis-regulatory element (CRE) enrichment analyses were used to investigate dynamic responses to desiccation. The 1.28-Gb genome exhibits unusually consistent chromatin architecture with unique chromosome organization across highly divergent haplotypes. We identified an XY sexual system with a small sex-determining region on Chromosome 8. Transcriptomic responses varied with dehydration severity, pointing to early suppression of growth, progressive activation of protective mechanisms, and subsequent return to homeostasis upon rehydration. Late embryogenesis abundant and early light-induced protein transcripts were dynamically regulated and showed enrichment of abscisic acid and stress-responsive CREs pointing toward conserved responses. Together, this study provides foundational resources for understanding the genomic architecture and reproductive biology of M. flabellifolia and offers new insights into the mechanisms of desiccation tolerance.

Towards Compact Neural Networks via End-to-End Training: A Bayesian Tensor Approach with Automatic Rank Determination

Post-training model compression can reduce the inference costs of deep neural networks, but uncompressed training still consumes enormous hardware resources and energy. To enable low-energy training on edge devices, it is highly desirable to directly train a compact neural network from scratch with a low memory cost. Low-rank tensor decomposition is an effective approach to reduce the memory and computing costs of large neural networks. However, directly training low-rank tensorized neural networks is a very challenging task because it is hard to determine a proper tensor rank a priori, and the tensor rank controls both model complexity and accuracy. Here, this paper presents a novel end-to-end framework for low-rank tensorized training. We first develop a Bayesian model that supports various low-rank tensor formats (e.g., CANDECOMP/PARAFAC, Tucker, tensor-train, and tensor-train matrix) and reduces neural network parameters with automatic rank determination during training. Then we develop a customized Bayesian solver to train large-scale tensorized neural networks. Our training methods shows orders-of-magnitude parameter reduction and little accuracy loss (or even better accuracy) in the experiments. On a very large deep learning recommendation system with over 4.2 ×10 9 model parameters, our method can reduce the parameter number to 1.6 ×10 5 automatically in the training process (i.e., by 2.6 ×10 4 times) while achieving almost the same accuracy. Sex Determination and Ovarian Development in Reptiles and Amphibians: From Genetic Pathways to Environmental Influences

Background: Reptiles and amphibians provide untapped potential for discovering how a diversity of genetic pathways and environmental conditions are incorporated into developmental processes that can lead to similar functional outcomes. These groups display a multitude of reproductive strategies, and whereas many attributes are conserved within groups and even across vertebrates, several aspects of sexual development show considerable variation. Summary: Here, in this review, we focus our attention on the development of the reptilian and amphibian ovary. First, we review and describe the events leading to ovarian development, including sex determination and ovarian maturation, through a comparative lens. We then describe how these events are influenced by environmental factors, focusing on temperature and exposure to anthropogenic chemicals. Lastly, we identify critical knowledge gaps and future research directions that will be crucial to moving forward in our understanding of ovarian development and the influences of the environment in reptiles and amphibians. Key Messages: Reptiles and amphibians provide excellent models for understanding the diversity of sex determination strategies and reproductive development. However, a greater understanding of the basic biology of these systems is necessary for deciphering the adaptive and potentially disruptive implications of embryo-by-environment interactions in a rapidly changing world.

Improving SGP4 Orbit Determination with New State Estimation Algorithm

The Simplified General Perturbations 4 Model (SGP4) is a well-known tool for performing satellite orbit determination. However, uncertainties and inaccuracies in the initial state inputs (required by SGP4) degrade the performance of the propagator. We present a new state estimation algorithm that allows for independent computation of these initial inputs using Unscented Kalman Filtering and GPS data from a satellite. The algorithm is tested on real flight data and demonstrates a notable performance improvement over the standard method of orbit determination using SGP4.

Analyzing Relationships Between Economic and Neighborhood-Related Social Determinants of Health and Intensive Care Unit Length of Stay for Critically Ill Children With Medical Complexity Presenting With Severe Sepsis

Objectives: Of the Social Determinants of Health (SDoH), we evaluated socioeconomic and neighborhood-related factors which may affect children with medical complexity (CMC) admitted to a Pediatric Intensive Care Unit (PICU) in Shelby County, Tennessee with severe sepsis and their association with PICU length of stay (LOS). We hypothesized that census tract-level socioeconomic and neighborhood factors were associated with prolonged PICU LOS in CMC admitted with severe sepsis in the underserved community. Methods: This single-center retrospective observational study included CMC living in Shelby County, Tennessee admitted to the ICU with severe sepsis over an 18-month period. Severe sepsis CMC patients were identified using an existing algorithm incorporated into the electronic medical record at a freestanding children's hospital. SDoH information was collected and analyzed using patient records and publicly available census-tract level data, with ICU length of stay as the primary outcome. Results: 83 encounters representing 73 patients were included in the analysis. The median PICU LOS was 9.04 days (IQR 3.99–20.35). The population was 53% male with a median age of 4.1 years (IQR 1.96–12.02). There were 57 Black/African American patients (68.7%) and 85.5% had public insurance. Based on census tract-level data, about half (49.4%) of the CMC severe sepsis population lived in census tracts classified as suffering from high social vulnerability. There were no statistically significant relationships between any socioeconomic and neighborhood level factors and PICU LOS. Conclusion: Pediatric CMC severe sepsis patients admitted to the PICU do not have prolonged lengths of ICU stay related to socioeconomic and neighborhood-level SDoH at our center. A larger sample with the use of individual-level screening would need to be evaluated for associations between social determinants of health and PICU outcomes of these patients.

A First Look at Orbit Determination for the Cassini Mission, Part 2: Saturn Tour

This paper completes the presentation of the first round of orbit determination analysis accomplished as part of Cassini navigation studies. The thrust of the analysis was to characterize operational orbit determination accuracy to first order for selected phases of the mission. The previous paper, Part 1, covered early phases from inner solar system cruise through the approach to Saturn and delivery of the Huygens probe to Titan. Part 2 resumes chronologically where Part 1 left off, covering the tour phase after delivery of the Huygens probe.

Atmospheric drag model for Cassini orbit determination during low altitude Titan flybys

On April 16, 2005, the Cassini spacecraft performed its lowest altitude flyby of Titan to date, the Titan-5 flyby, flying 1027 km above the surface of Titan. This document discusses the development of a Titan atmospheric drag model for the purpose of the orbit determination of Cassini. Results will be presented for the Titan A flyby, the Titan-5 flyby as well as the most recent low altitude Titan flyby, Titan-7. Different solutions will be compared against OD performance in terms of the flyby B-plane parameters, spacecraft thrusting activity and drag estimates. These low altitude Titan flybys were an excellent opportunity to observe the effect of Titan's atmospheric drag on the orbit determination solution and results show that the drag was successfully modeled to provide accurate flyby solutions.

Real-time Sub-cm Differential Orbit Determination of two Low-Earth Orbiters with GPS Bias Fixing

An effective technique for real-time differential orbit determination with GPS bias fixing is formulated. With this technique, only real-time GPS orbits and clocks are needed (available from the NASA Global Differential GPS System with 10-20 cm accuracy). The onboard, realtime orbital states of user satellites (few meters in accuracy) are used for orbit initialization and integration. An extended Kalman filter is constructed for the estimation of the differential orbit between the two satellites as well as a reference orbit, together with their associating dynamics parameters. Due to close proximity of the two satellites and of similar body shapes, the differential dynamics are highly common and can be tightly constrained which, in turn, strengthens the orbit estimation. Without explicit differencing of GPS data, double-differenced phase biases are formed by a transformation matrix. Integer-valued fixing of these biases are then performed which greatly strengthens the orbit estimation. A 9-day demonstration between GRACE orbits with baselines of approx.200 km indicates that approx.80% of the double-differenced phase biases can successfully be fixed and the differential orbit can be determined to approx.7 mm as compared to the results of onboard K-band ranging.

GRAIL Science Data System Orbit Determination : Approach, Strategy, and Performance

This paper details orbit determination techniques and strategies employed within each stage of the larger iterative process of preprocessing raw GRAIL data into the gravity science measurements used within gravity field solutions. Each orbit determination pass used different data, corrections to them, and/or estimation parameters. We compare performance metrics among these passes. For example, for the primary mission, the magnitude of residuals using our orbits progressed from approximately or equal to19.4 to 0.077 approximately or equal to m/s for inter-satellite range rate data and from approximately or equal to 0.4 to approximately or equal to 0.1 mm/s for Doppler data.

Filter Strategies for Mars Science Laboratory Orbit Determination

The Mars Science Laboratory (MSL) spacecraft had ambitious navigation delivery and knowledge accuracy requirements for landing inside Gale Crater. Confidence in the orbit determination (OD) solutions was increased by investigating numerous filter strategies for solving the orbit determination problem. We will discuss the strategy for the different types of variations: for example, data types, data weights, solar pressure model covariance, and estimating versus considering model parameters. This process generated a set of plausible OD solutions that were compared to the baseline OD strategy. Even implausible or unrealistic results were helpful in isolating sensitivities in the OD solutions to certain model parameterizations or data types.
