Development of dual‐inducible duet‐expression vectors for tunable gene expression control and CRISPR interference‐based gene repression in Pseudomonas putida KT2440

Summary The development of P. putida as an industrial host requires a sophisticated molecular toolbox for strain improvement, including vectors for gene expression and repression. To augment existing expression plasmids for metabolic engineering, we developed a series of dual‐inducible duet‐expression vectors for P. putida KT2440. A number of inducible promoters (P lac , P tac , P tetR/tetA and P bad ) were used in different combinations to differentially regulate the expression of individual genes. Protein expression was evaluated by measuring the fluorescence of reporter proteins (GFP and RFP). Our experiments demonstrated the use of compatible plasmids, a useful approach to coexpress multiple genes in P. putida KT2440. These duet vectors were modified to generate a fully inducible CRISPR interference system using two catalytically inactive Cas9 variants from S. pasteurianus (dCas9) and S. pyogenes (spdCas9). The utility of developed CRISPRi system(s) was demonstrated by repressing the expression of nine conditionally essential genes, resulting in growth impairment and prolonged lag phase for P. putida KT2440 growth on glucose. Furthermore, the system was shown to be tightly regulated, tunable and to provide a simple way to identify essential genes with an observable phenotype.

The genotype-phenotype landscape of an allosteric protein

Allostery is a fundamental biophysical mechanism that underlies cellular sensing, signaling, and metabolism. Yet a quantitative understanding of allosteric genotype-phenotype relationships remains elusive. Here, we report the large-scale measurement of the genotype-phenotype landscape for an allosteric protein: the lac repressor from Escherichia coli , LacI. Using a method that combines long-read and short-read DNA sequencing, we quantitatively measure the dose-response curves for nearly 10 5 variants of the LacI genetic sensor. The resulting data provide a quantitative map of the effect of amino acid substitutions on LacI allostery and reveal systematic sequence-structure-function relationships. We find that in many cases, allosteric phenotypes can be quantitatively predicted with additive or neural-network models, but unpredictable changes also occur. For example, we were surprised to discover a new band-stop phenotype that challenges conventional models of allostery and that emerges from combinations of nearly silent amino acid substitutions.

System-Independent X-Ray Characterization

We have developed a method for image reconstruction and analysis of dual-energy X-ray scans that has been demonstrated to yield a physically intuitive characterization of materials (by electron density and effective atomic number) in a system-independent way. Our methodology stands in contrast to common practice of reconstructing images to yield linear attenuation coefficients, which depend strongly on system spectral response. At the core of our method, a technique called dual-energy decomposition is used to generate spectral energy-independent characterizations of the attenuation properties of a material. Inputs to this decomposition include system spectral response estimates and dual-energy X-ray projection data. X-ray cross-section tables are used to convert decomposition results to physical feature estimates (electron density, effective atomic number). Our method currently works best for materials with atomic numbers below 22 (titanium). In prior work for DHS, we demonstrated in a controlled, no-clutter environment that the decomposition technique could be applied to scans of a well-characterized materials on multiple systems with different X-ray spectral responses. In cases where LAC values showed a standard deviation of as much as 20% of mean value, application of our method yielded results with standard deviations as low as 1–3% of mean value.

Management, Efficiency and Treatment of Silica, Chlorides, and other Pollutants (METSiCloPs) of Cooling Tower Water [Slides]

Site-wide water balance suggests that CTs across eight LANL facilities use about 58% of the total potable water bought from LAC (271 MGY). It is vital to make a conscious and calculated effort to reduce the dependence on external water supplies, and potable water. We need to strive to increase CoCs at CTs and improve water efficiency to reduce water consumption.

First-principles Prediction of X-Ray Polarization from Magnetic Reconnection in High-frequency BL Lacertae Objects

Relativistic magnetic reconnection is a potential particle acceleration mechanism for high-frequency BL Lac objects (HBLs). The Imaging X-ray Polarimetry Explorer (IXPE) scheduled to launch in 2021 has the capability to probe the evolution of magnetic field in HBLs, examining the magnetic reconnection scenario for the HBL flares. In this paper, we make the first attempt to self-consistently predict HBL X-ray polarization signatures arising from relativistic magnetic reconnection via combined particle-in-cell and polarized radiation transfer simulations. Here, we find that although the intrinsic optical and X-ray polarization degrees are similar on average, the X-ray polarization is much more variable in both the polarization degree and angle (PD and PA). Given the sensitivity of the IXPE, it may obtain one to a few polarization data points for one flaring event of nearby bright HBLs Mrk 421 and 501. However, it may not fully resolve the highly variable X-ray polarization. Due to temporal depolarization, where the integration of photons with variable polarization states over a finite period of time can lower the detected PD, the measured X-ray PD can be considerably lower than the optical counterpart or even undetectable. The lower X-ray PD than the optical thus can be a characteristic signature of relativistic magnetic reconnection. For very bright flares where the X-ray polarization is well resolved, relativistic magnetic reconnection predicts smooth X-ray PA swings, which originate from large plasmoid mergers in the reconnection region.

Multiwavelength Observations of the Blazar VER J0521+211 during an Elevated TeV Gamma-Ray State

We report on a long-lasting, elevated gamma-ray flux state from VER J0521+211 observed by VERITAS, MAGIC, and Fermi-LAT in 2013 and 2014. The peak integral flux above 200 GeV measured with the nightly binned light curve is (8.8 ± 0.4) × 10 –7 photons m –2 s –1 , or ~37% of the Crab Nebula flux. Multiwavelength observations from X-ray, UV, and optical instruments are also presented. A moderate correlation between the X-ray and TeV gamma-ray fluxes was observed, and the X-ray spectrum appeared harder when the flux was higher. Using the gamma-ray spectrum and four models of the extragalactic background light (EBL), a conservative 95% confidence upper limit on the redshift of the source was found to be z ≤ 0.31. Unlike the gamma-ray and X-ray bands, the optical flux did not increase significantly during the studied period compared to the archival low-state flux. The spectral variability from optical to X-ray bands suggests that the synchrotron peak of the spectral energy distribution (SED) may become broader during flaring states, which can be adequately described with a one-zone synchrotron self-Compton model varying the high-energy end of the underlying particle spectrum. The synchrotron peak frequency of the SED and the radio morphology of the jet from the MOJAVE program are consistent with the source being an intermediate-frequency-peaked BL Lac object.

The Peculiar Variable X-Ray Spectrum of the Active Galactic Nucleus PKS 2005–489

PKS 2005-489 is a well-known, bright southern BL Lac object that has been detected up to TeV energies. In a low-flux state it exhibits the expected multiwavelength double-peaked spectrum in the radio- γ -ray band. The high-flux state shows extreme flux variations in the X-ray band with a hardening as well as a peculiar curved feature in the spectrum. Thus far, PKS 2005-489 is the only source to exhibit such a feature. To study the X-ray variability further, we obtained the first hard X-ray spectrum of the source with the Nuclear Spectroscopic Telescope Array. We compare quasi-simultaneous radio, optical, UV, soft and hard X-ray, and γ -ray data of PKS 2005-489 to archival data in order to study its broadband behavior. We find a very consistent quiet state in the spectral energy distribution, with little variation in spectral shape or flux between the 2012 and 2020 data. A possible explanation for the peculiar X-ray spectrum in the flaring state is an additional component in the jet, possibly accelerated via magnetic reconnection, that is not co-spatial to the low-flux state emission region.

Distortion of a Relativistic Jet Echoing a Magnetic Flux Eruption

Magnetized accretion onto spinning black holes (BHs) can accumulate a large magnetic flux across the event horizon and launch a pair of relativistic jets via the Blandford–Znajek mechanism. In the magnetically saturated (arrested) state, excess magnetic flux is ejected from the BH in episodic magnetic flux eruptions, which result in a significant yet temporary reduction of jet power. We analyze results of a high-resolution 3D general-relativistic magnetohydrodynamic numerical simulation of geometrically thick magnetically saturated accretion onto a high-spin Kerr BH for a single cycle of magnetic flux eruption and accumulation. We show that following an eruption, a weakened jet develops a strong helical distortion with a distinct structure of magnetic fields—the poloidal field along the jet core is unaffected by the eruption; while toroidal field lines, ejected from the BH during the eruption and later readvected onto it, form poloidal “bypasses” along the inner jet sheath. Such a distortion may appear in sources fed by geometrically thick accretion flows as an asymmetric superluminal knot, strongly interacting with the jet sheath along an oblique working surface. The jet section repowered by magnetic flux reaccumulated on the BH is tilted by a few degrees, implying significant variations in radiation boost towards observers of BL Lac blazars. The intrinsic structure of the jet spine is consistent with axisymmetric semi-analytical models.

The code and detailed step-by-step instructions for generating the model output data, processing results, and analysis and plotting are provided at https://github.com/IMMM-SFA/Ferencz_et_al_2026_ER_Water. The PyArtes model is a python adaptation of the Artes model. PyArtes uses many of the same input data and optimization model architecture as Artes. Documentation for the PyArtes model is provided in the Supplement to the paper. The primary data product are simulated monthly water shortages for indoor and outdoor demand under a large ensemble of drought scenarios (>13,000). The droughts are hypothetical and are not based on historical time series data of supply sources - though historical data did help inform ranges explored for supply parameters. Demands are informed by recent 2017-2021 water supply data. Demands used for the model can be accessed at https://github.com/IMMM-SFA/Ferencz_et_al_2026_ER_Water. Simulations resolve demand for over 90 water providers in the study region. The results report 36 months of water shortage data for each indoor and outdoor demand node. The study also developed a multilayer perceptron (MLP) neural network trained on a subset of the simulated shortage ensemble to emulate worst annual water shortage for a given set of parameter multipliers -- provided the parameter values fall within the ranges sampled in the ensemble. Emulated water shortages for synthetic ensembles are in the MLP-generated shortages folder. The MLP model was used to generate larger ensembles to support Sobol analysis that would have been extremely computationally expensive to simulate. Wire Arc Additive Manufacturing of Lightweight High Pressure Die Casting Tooling

Oak Ridge National Laboratory (ORNL) and Mercury Marine partnered to develop and test methods for additively manufactured tooling for aluminum die casting applications under CRADA agreement NFE-20-08193. Tooling is the largest capital expense for high production casting projects. The lead time for tooling is often measured in months with a typical project taking 9-12 months to realize Production Part Approval Process (PPAP) ready die cast samples. This project demonstrated the technical viability of rapidly produced steel components for high pressure die casting tooling via Wire Arc Additive Manufacturing (WAAM). A 410 stainless steel tool was redesigned and optimized with conformal cooling channels and additively manufactured. The finished tool was tested and used to produce over 4000 parts, which well surpassed expectations. A secondary objective was to evaluate the durability of multi-material additively manufactured (AM) components with conformal cooling. A large multi-material tool (H13 and 410SSNiMo) was manufactured using the same methods showing potential reductions in used material and cost. However, the H13 section sustained material cracking. Further analysis showed that the potential cause was the CTE mismatch of the two materials at higher temperatures. It is also suggested that the material mix can be used if the steel processing temperature does not exceed 600 ̊C.This project has shown high potential for using the WAAM technology for creating AM parts for aluminum dies casting. However, the multi-material approach requires extended study and tests.

Deep representation learning improves prediction of LacI-mediated transcriptional repression

Significance The understanding of protein function increases with new experimental and evolutionary datasets. A major challenge is to apply machine learning to these datasets to capture essential features of protein function. Here, we analyze the experimentally determined repression function for tens of thousands of mutants of the LacI protein. This study provides a continuous, noncategorical repression value across a majority of all single mutations and for thousands of higher-order mutations. To develop a top-performing model for the prediction of repression by LacI, we compare several leading variant effect prediction algorithms. A deep representation learning paradigm, first trained across millions of proteins from all known protein families and then fine-tuned using LacI experimental data, offers the highest predictive performance of repression function.

Floating photovoltaic power plants: A review of energy yield, reliability, and operation and maintenance

Photovoltaic (PV) systems are essential for the transition to sustainable energy, reducing fossil fuel dependence and mitigating climate change. Although PV requires minimal land area — PV can meet the European Union's energy needs using only 0.26% of its land — space for deployment is often scarce in densely populated regions. Floating photovoltaics (FPV) offer an effective solution to land-use challenges by installing PV systems on floating structures in water bodies. FPV is a growing niche within PV with a cumulative installed capacity reaching 7.7 GW globally by 2023. Almost 90% of the installed FPV capacity is in Asia, with close to 50% of in China alone, while the Netherlands and France are the largest markets outside Asia. FPV shows strong potential to support climate targets, but still faces challenges like regulatory barriers, cost competitiveness compared to ground-based PV (GPV), and uncertainties about environmental impacts and system reliability. FPV systems are currently installed mainly on sheltered inland waters, such as quarry lakes, irrigation ponds and reservoirs. FPV technical standards are still being developed. Guidelines have been published by the World Bank, DNV, and Solar Power Europe, and emerging national standards from South Korea, China, and Singapore address design, components, and safety. The International Electrotechnical Commission (IEC) is working on formal standards for floats, mooring systems, and electrical connectors. However, the published best practices lack quantitative guidance for yield modelling and reliability, which this report aims to address. It provides data-driven insights, models, and parameters essential for accurate energy yield, reliability, and maintenance predictions over FPV systems' lifetimes.
