Implementation of a Web Interface to Display Real-Time Statistics of a Dilution Refrigerator

The purpose of the project is to communicate with a Bluefors control unit and extract data. The Bluefors control unit is a piece of experimental equipment that measures various different data points in a dilution refrigerator and stores them. The goal was to access the control unit using the websockets library in Python. After accessing the control unit, the goal was to export the data to a web page in HTML/CSS. The tools that were used changed throughout the project. Python was used to communicate with the Bluefors unit via JSON requests and JSON objects, then the Python hosted its own websocket server to act as a proxy server to allow for the web page to access the data. This was done to bypass SSL verification. Then, the web page, which was created using JavaScript, React, Next.js, and NodeJS, accesses the proxy server to import the data from the dilution refrigerator and display the values in real time on the page.

On display: Replica instruments used to discover electrons, neutrons and more

Believe it or not, the electron was discovered in 1897 using only a hand-blown glass tube, a vacuum pump and some electrodes. Historical instrument replicas now on display in the lobby of the J.R. Oppenheimer Center give visitors a chance to remember the elegance — and relative simplicity — of the instruments scientists built and used to make pivotal discoveries.

NEAr Transporter (NEAT) Domains: Unique Surface Displayed Heme Chaperones That Enable Gram-Positive Bacteria to Capture Heme-Iron From Hemoglobin

Iron is an important micronutrient that is required by bacteria to proliferate and to cause disease. Many bacterial pathogens forage iron from human hemoglobin (Hb) during infections, which contains this metal within heme (iron–protoporphyrin IX). Several clinically important pathogenic species within the Firmicutes phylum scavenge heme using surface-displayed or secreted NEAr Transporter (NEAT) domains. In this review, we discuss how these versatile proteins function in the Staphylococcus aureus Iron-regulated surface determinant system that scavenges heme-iron from Hb. S. aureus NEAT domains function as either Hb receptors or as heme-binding chaperones. In vitro studies have shown that heme-binding NEAT domains can rapidly exchange heme amongst one another via transiently forming transfer complexes, leading to the interesting hypothesis that they may form a protein-wire within the peptidoglycan layer through which heme flows from the microbial surface to the membrane. In Hb receptors, recent studies have revealed how dedicated heme- and Hb-binding NEAT domains function synergistically to extract Hb’s heme molecules, and how receptor binding to the Hb-haptoglobin complex may block its clearance by macrophages, prolonging microbial access to Hb’s iron. The functions of NEAT domains in other Gram-positive bacteria are also reviewed.

Mercury-Tolerant Ensifer medicae Strains Display High Mercuric Reductase Activity and a Protective Effect on Nitrogen Fixation in Medicago truncatula Nodules Under Mercury Stress

Mercury (Hg) is extremely toxic for all living organisms. Hg-tolerant symbiotic rhizobia have the potential to increase legume tolerance, and to our knowledge, the mechanisms underlying Hg tolerance in rhizobia have not been investigated to date. Rhizobial strains of Ensifer medicae, Rhizobium leguminosarum bv. trifolii and Bradyrhizobium canariense previously isolated from severely Hg-contaminated soils showed different levels of Hg tolerance. The ability of the strains to reduce mercury Hg 2+ to Hg o , a volatile and less toxic form of mercury, was assessed using a Hg volatilization assay. In general, tolerant strains displayed high mercuric reductase activity, which appeared to be inducible in some strains when grown at a sub-lethal HgCl 2 concentration. A strong correlation between Hg tolerance and mercuric reductase activity was observed for E. medicae strains, whereas this was not the case for the B. canariense strains, suggesting that additional Hg tolerance mechanisms could be playing a role in B. canariense. Transcript abundance from merA, the gene that encodes mercuric reductase, was quantified in tolerant and sensitive E. medicae and R. leguminosarum strains. Tolerant strains presented higher merA expression than sensitive ones, and an increase in transcript abundance was observed for some strains when bacteria were grown in the presence of a sub-lethal HgCl 2 concentration. These results suggest a regulation of mercuric reductase in rhizobia. Expression of merA genes and mercuric reductase activity were confirmed in Medicago truncatula nodules formed by a sensitive or a tolerant E. medicae strain. Transcript accumulation in nodules formed by the tolerant strain increased when Hg stress was applied, while a significant decrease in expression occurred upon stress application in nodules formed by the Hg-sensitive strain. The effect of Hg stress on nitrogen fixation was evaluated, and in our experimental conditions, nitrogenase activity was not affected in nodules formed by the tolerant strain, while a significant decrease in activity was observed in nodules elicited by the Hg-sensitive bacteria. Our results suggest that the combination of tolerant legumes with tolerant rhizobia constitutes a potentially powerful tool in the bioremediation of Hg-contaminated soils.

Using optimal estimation to retrieve winds from velocity-azimuth display (VAD) scans by a Doppler lidar

Abstract. Low-powered commercially available coherent Doppler lidar (CDL) wind profilers provide continuous measurement of vertical profiles of wind in the lower troposphere, usually close to or up to the top of the planetary boundary layer. The vertical extent of these wind profiles is limited by the availability of scatterers and thus varies substantially throughout the day and from one day to the next. This makes it challenging to develop continuous products that rely on CDL-observed wind profiles. In order to overcome this problem, we have developed a new method for wind profile retrievals from CDL that combines the traditional velocity-azimuth display (VAD) technique with optimal estimation (OE) to provide continuous wind profiles up to 3 km. The new method exploits the level-to-level covariance present in the wind profile to fill in the gaps where the signal-to-noise ratio of the CDL return is too low to provide reliable results using the traditional VAD method. Another advantage of the new method is that it provides the full error covariance matrix of the solution and profiles of information content, which more easily facilitates the assimilation of the observed wind profiles into numerical weather prediction models. This method was tested using yearlong CDL measurements at the Atmospheric Radiation Measurement (ARM) Southern Great Plains (SGP) Central Facility in 2019. Comparison with the ARM operational CDL wind profile product and collocated radiosonde wind measurements shows excellent agreement (R2>0.99) with no degradation in results where the traditional VAD provided a valid solution. In the region where traditional VAD does not provide results, the OE wind speed and wind vector have uncertainties of 3.44 and 4.33 m s−1, respectively. As a result, the new method provides additional information over the standard technique and increases the effective range of existing CDL systems without the need for additional hardware.

System and method for liquid crystal display system incorporating wire grid polarizers for large scale and large volume stereolithography

The present disclosure relates to a liquid crystal display (LCD) system. The system in one example has a light source for generating unpolarized light, and an LCD screen arranged in a path of transmittance of the unpolarized light. First and second wire grid polarizers are arranged adjacent to the LCD screen and each have a plurality of nano-scale wires, with the first and second wire grid polarizers have differing polarizations. A pitch of each of the nano-scale wires is no larger than one-third a wavelength of the unpolarized light from the light source. The wire grid polarizers create, in connection with operation of the LCD screen, a 2D light mask suitable for initiating the polymerization of an optically curable material.

Performance Assessment of Low-GWP Refrigerated Reach-In Display Cases

This project is part of an effort by Commonwealth Edison Company (ComEd) to evaluate the energy and peak demand savings potential of emerging technologies in the Chicago area. This document focuses on the assessment of energy-efficient, medium-temperature, self-contained refrigerated display cases utilizing environmentally friendly refrigerants. The results of this evaluation will be considered by ComEd and CLEAResult to develop a new energy efficiency rebate measure for ComEd's incentive programs. This rebate measure will become an addition to the Technical Reference Manual. In 2016, the United Nations passed the Kigali Montreal Protocol Amendment, which placed restrictions on certain types of refrigerants. In compliance with this amendment, the U.S. Environmental Protection Agency (EPA) placed a ban on the manufacture of refrigeration systems using hydrofluorocarbons including R134a starting in January 2020. Although the ban has halted manufacture, the EPA continues to allow the use of these refrigerants. Therefore, it is critical to provide incentives for replacing these refrigerants with other environmentally friendly and energy-efficient alternatives.

Use of wire grid polarizers with liquid crystal display for large-volume stereolithography

Spatial light modulators that use liquid crystal devices (LCDs) to pattern ultraviolet light for vat polymerization suffer from a few limitations: limited range of wavelengths that can be patterned, low damage threshold for incoming light, and low efficiency of light transmission. A monochrome LCD can more efficiently transmit useful wavelengths of light for stereolithography (365–405 nm) than a color LCD, but it still contains two absorptive film polarizers that suffer from the aforementioned limitations. Wire grid polarizers reflect rather than absorb the blocked polarizations of light, resulting in a higher intensity damage threshold. They can also polarize lower wavelength light that is limited only by the wire pitch. In this work, this research investigates the effects of replacing an LCD’s film polarizers with wire grid polarizers to enable polarization and patterning of high intensity UV light, resulting in shorter curing times and the ability to print a wider range of stereolithography feedstocks.

Dihydrogen Adduct (Co–H 2 ) Complexes Displaying H‐Atom and Hydride Transfer

Abstract The prototypical reactivity profiles of transition metal dihydrogen complexes (M‐H 2 ) are well‐characterized with respect to oxidative addition (to afford dihydrides, M(H) 2 ) and as acids, heterolytically delivering H + to a base and H − to the metal. In the course of this study we explored plausible alternative pathways for H 2 activation, namely direct activation through H‐atom or hydride transfer from the σ‐H 2 adducts. To this end, we describe herein the reactivity of an isostructural pair of a neutral S = and an anionic S =0 Co‐H 2 adduct, both supported by a trisphosphine borane ligand (P 3 B ). The thermally stable metalloradical, (P 3 B )Co(H 2 ), serves as a competent precursor for hydrogen atom transfer to t Bu 3 ArO ⋅ . What is more, its anionic derivative, the dihydrogen complex [(P 3 B )Co(H 2 )] 1− , is a competent precursor for hydride transfer to BEt 3 , establishing its remarkable hydricity. The latter finding is essentially without precedent among the vast number of M‐H 2 complexes known.

Ecological niche models display nonlinear relationships with abundance and demographic performance across the latitudinal distribution of Astragalus utahensis (Fabaceae)

Abstract The potential for ecological niche models (ENMs) to accurately predict species' abundance and demographic performance throughout their geographic distributions remains a topic of substantial debate in ecology and biogeography. Few studies simultaneously examine the relationship between ENM predictions of environmental suitability and both a species' abundance and its demographic performance, particularly across its entire geographic distribution. Yet, studies of this type are essential for understanding the extent to which ENMs are a viable tool for identifying areas that may promote high abundance or performance of a species or how species might respond to future climate conditions. In this study, we used an ensemble ecological niche model to predict climatic suitability for the perennial forb Astragalus utahensis across its geographic distribution. We then examined relationships between projected climatic suitability and field‐based measures of abundance, demographic performance, and forecasted stochastic population growth (λ s ). Predicted climatic suitability showed a J‐shaped relationship with A. utahensis abundance, where low‐abundance populations were associated with low‐to‐intermediate suitability scores and abundance increased sharply in areas of high predicted climatic suitability. A similar relationship existed between climatic suitability and λ s from the center to the northern edge of the latitudinal distribution. Patterns such as these, where density or demographic performance only increases appreciably beyond some threshold of climatic suitability, support the contention that ENM‐predicted climatic suitability does not necessarily represent a reliable predictor of abundance or performance across large geographic regions.

Acute Copper Toxicity Displays a Nonmonotonic Relationship with Age Across the Medaka ( Oryzias latipes ) Life Span

The ability of an organism to cope with environmental stressors varies across the life span because of developmental stage–specific responses and age-related functional declines. In the present study, we examined the effect of age on acute copper toxicity in Japanese medaka (Oryzias latipes). We first determined the median lethal concentration (LC50) at 96 h for embryos, 7-day-old fry, and 6-month-old medaka. Embryos were exposed to 0, 15, 30, 60, 125, 250, and 500 ppb CuSO 4 through hatching. Fry were exposed to 0, 20, 50, 75, 100, 150, 250, and 500 ppb CuSO 4 for 96 h. Adult fish were exposed to 0, 100, 150, 200, 250, and 300 ppb CuSO 4 for 96 h. The 96-h LC50 was 804 ppb for embryos, 262 ppb for embryonically exposed larvae, 60.3 ppb for 7-day-old fry, and 226 ppb for adults. We then challenged cohorts of fish aged 2, 3, 5, 6, 7, 8, 9, 10, 11, 13, 14, 15, and 16 months with a 225-ppb CuSO 4 exposure to determine the acute toxicity across the life span. The fish exhibited a bimodal tolerance to copper, with tolerance peaking in 2- and 3-month-old fish and again at 10 and 11 months of age. Our data demonstrate that copper sensitivity is dynamic throughout the medaka life span and may be influenced by trade-offs with reproduction.
