TCF Base Technology-Specific Final Report: Engineering Enzymes for Crystalline PET Substrate

The primary objective of this project was to develop a new polyethylene terephthalate (PET) hydrolase enzyme to depolymerize relevant PET substrates for Birch Biosciences, using high-throughput protein expression, purification, and assaying systems and machine learning-guided enzyme design. As a secondary project objective, we also aimed to develop a more energy-efficient ethylene glycol (EG) recovery strategy relative to distillation.

Each year, the R&D 100 Awards recognize the top 100 innovations from a pool of international entries. Technologies developed at Lawrence Livermore earned four 2025 R&D 100 Awards, raising the Laboratory’s total to 186. A series of articles beginning on p. 4 describes each winning innovation: monolithic telescopes, the flexible imaging diffraction diagnostic for laser experiments, the in-air drop encapsulation apparatus, and the metaoptics-enabled large-scale 3D nanolithography platform.

SPRUCE Photosynthesis and Respiration of Picea mariana and Larix laricina in SPRUCE Experimental Plots, 2019

This dataset contains physiological, morphological, and chemical measurements of the two dominant coniferous species, Picea mariana and Larix laricina, in August 2019 (2019-08-20 to 2019-08-22) at the SPRUCE (Spruce and Peatland Responses under Changing Environments) experiment site in the Marcell Experimental Forest in northern Minnesota, USA. These observations help to assess the effects of whole ecosystem scale warming and elevated carbon dioxide (CO2) concentrations on peatland ecosystems. Measurements include light-saturated photosynthesis and foliar dark respiration measurements under standard conditions and growth conditions involving varying temperatures and atmospheric CO2 concentrations, as well as leaf morphology measurements (leaf mass per unit leaf area) and nitrogen content based on mass and leaf area. Net photosynthesis and dark respiration measurements were taken using portable photosynthesis systems (LI6400XT, LI6800, LI-COR Biosciences, USA). This dataset contains one data file in comma-separate values (*.csv) format. Additional metadata are provided: a data dictionary and a file-level metadata file in comma-separate values (.csv) format and a user guide in PDF (*.pdf) format.

New Enzymes for Plastics Recycling from Biosensor-Guided Evolution and Machine Learning: Cooperative Research and Development Final Report, CRADA Number CRD-23-24335

The Agile BioFoundry (ABF) is a consortium of national laboratories dedicated to accelerating biomanufacturing and enabling the bioeconomy. The ABF operates a flexible biotechnology platform that can adjust to the needs of numerous government, academic, and industrial partners, thus enabling them to rapidly develop and optimize the production of a wide range of bioproducts. In this project, the ABF partnered with Birch Biosciences, Inc. to use ABF technology to assist in the development of high-performance enzymes that enable circular, sustainable recycling of poly(ethylene terephthalate) (PET) plastics. A primary goal of this project is to enable cost-effective, high yield, sustainable recycling of PET plastic packaging products.

SPACE SCIENCE-EARTH, SUN, AND STARS

Space science is science carried out in space. There is much that is exciting, but nothing mysterious about it. Space science calls upon physics, chemistry, astronomy, biosciences, and other scientific disciplines to attack some of the most important and challenging scientific problems of today. Among these problems are the investigation of the Earth and Sun, Moon and planets, stars and galaxies, and life in space from the vantage point provided by the orbiting satellite or the dep space probe.
