Competitive binding of STATs to receptor phospho-Tyr motifs accounts for altered cytokine responses

Cytokines elicit pleiotropic and non-redundant activities despite strong overlap in their usage of receptors, JAKs and STATs molecules. We use IL-6 and IL-27 to ask how two cytokines activating the same signaling pathway have different biological roles. We found that IL-27 induces more sustained STAT1 phosphorylation than IL-6, with the two cytokines inducing comparable levels of STAT3 phosphorylation. Mathematical and statistical modeling of IL-6 and IL-27 signaling identified STAT3 binding to GP130, and STAT1 binding to IL-27Rα, as the main dynamical processes contributing to sustained pSTAT1 levels by IL-27. Mutation of Tyr613 on IL-27Rα decreased IL-27-induced STAT1 phosphorylation by 80% but had limited effect on STAT3 phosphorgylation. Strong receptor/STAT coupling by IL-27 initiated a unique gene expression program, which required sustained STAT1 phosphorylation and IRF1 expression and was enriched in classical Interferon Stimulated Genes. Interestingly, the STAT/receptor coupling exhibited by IL-6/IL-27 was altered in patients with systemic lupus erythematosus (SLE). IL-6/IL-27 induced a more potent STAT1 activation in SLE patients than in healthy controls, which correlated with higher STAT1 expression in these patients. Partial inhibition of JAK activation by sub-saturating doses of Tofacitinib specifically lowered the levels of STAT1 activation by IL-6. Our data show that receptor and STATs concentrations critically contribute to shape cytokine responses and generate functional pleiotropy in health and disease.

THEORETICAL STUDY OF ATOMIC AND MOLECULAR GASES AND THEIR REACTIONS IN THE UPPER ATMOSPHERE. SUMMARY OF WORK DONE IN THE PERIOD NOV. 1, 1960 TO APR. 30, 1961

In the previous report it was stated that the major mathematical problems of computations were nearly solved and an attempt is being made to start application of "Tables of Integrals" to the computation of the different states of atoms and molecules. In this respect I like to report that the computation of a number of states of atoms and molecules has been carried out. The results are so far confined only to the equilibrium distance of molecules. We are extending the calculation to other distances so as to compute potential energy curves of different states of molecules. We are also developing Thomas Fermi Statistical model of the atom to verify our results of more accurate calculation. The results so far have been very encouraging and I hope to report some of these results in the next paper.

Computer discrimination procedures applicable to aerial and ERTS multispectral data

Two statistical models are compared in the classification of crops recorded on color aerial photographs. A theory of error ellipses is applied to the pattern recognition problem. An elliptical boundary condition classification model (EBC), useful for recognition of candidate patterns, evolves out of error ellipse theory. The EBC model is compared with the minimum distance to the mean (MDM) classification model in terms of pattern recognition ability. The pattern recognition results of both models are interpreted graphically using scatter diagrams to represent measurement space. Measurement space, for this report, is determined by optical density measurements collected from Kodak Ektachrome Infrared Aero Film 8443 (EIR). The EBC model is shown to be a significant improvement over the MDM model.

Photographic astrometry and overlap reduction techniques

Rigorous adjustment algorithms are discussed which extract hidden information contained in overlapping photographs of star fields. In particular, the development of adequate physical and statistical models is emphasized.

Comments on the possibility for determining detailed atmospheric structure from transhorizon radio propagation measurements

A deterministic approach is used to examine the atmospheric structure at any instant in time, with an assumption as to the degree of anisotropy. A statistical model is also used to relate the experimental data to atmospheric structure. With this model, the three dimensional spectrum of spatial wave numbers of the refractivity fluctuations are determined, along with correlations which exist within the spectrum.

Statistical distribution of spacecraft maximum structural response.

Under nonstationary random excitations resulting from booster engine shutdown, a direct statistical analysis of spacecraft maximum response is performed, and the spacecraft structural reliability is obtained. It is found that the Gumbel (1958) Type I asymptotic distribution of maximum values provides a reasonably good statistical model for spacecraft maximum responses. This approach makes it possible to perform the reliability-based optimum design of spacecraft structures.

Analysis of stress-rupture data from S-glass composites.

Both composite and free strands have been tested to study matrix effectiveness. Glass from different lots were used, and processing parameters were varied to demonstrate the effects of material variability. The statistical patterns that emerge from these experiments show that lifetime distributions are characteristically skewed. As a class, the lifetime distributions are well approximated by the reduced form of the Weibull distribution. The strength retention data display little early degradation. The retained-strength distributions can also be classed as Weibull distributions. A statistical model applied to the data gives predictions of progressive failure, strength retention path, and expected life for two basic types of internal fracture processes.
