Genome-Scale Transcription-Translation Mapping Reveals Features of Zymomonas mobilis Transcription Units and Promoters

Efforts to rationally engineer synthetic pathways in Zymomonas mobilis are impeded by a lack of knowledge and tools for predictable and quantitative programming of gene regulation at the transcriptional, posttranscriptional, and posttranslational levels. With the detailed functional characterization of the Z. mobilis genome presented in this work, we provide crucial knowledge for the development of synthetic genetic parts tailored to Z. mobilis . This information is vital as researchers continue to develop Z. mobilis for synthetic biology applications. Our methods and statistical analyses also provide ways to rapidly advance the understanding of poorly characterized bacteria via empirical data that enable the experimental validation of sequence-based prediction for genome characterization and annotation.

Genomic Features and Pervasive Negative Selection in Rhodanobacter Strains Isolated from Nitrate and Heavy Metal Contaminated Aquifer

Despite the dominance of Rhodanobacter species in the subsurface of the contaminated Oak Ridge Reservation (ORR) site, very little is known about the mechanisms underlying their adaptions to the various stressors present at ORR. Recently, multiple Rhodanobacter strains have been isolated from the ORR groundwater samples from several wells with varying geochemical properties.

WHONDRS Surface Water and Sediment Non-Purgeable Organic Carbon and FTICR-MS across Glacial Features in Svalbard 2021 (v3)

This dataset supports a broader study examining land-atmosphere gas exchange associated with permafrost and glaciers in Svalbard. The dataset provides geochemistry and organic matter characterization data generated from surface water and sediment. Samples were collected from 8 spatially diverse sites across central Svalbard and also from a more comprehensive temporal and spatial study within one glacial catchment area throughout one summer melt season. Related data were collected and will be published separately in collaboration with Yde and Kleber. This data package was originally published in September 2022. It was updated in March 2023 (v2; new and modified files), and July 2025 (v3; modified files). See the change history section in the readme for details. For details on how to navigate data packages generated by this project, see https://data.ess-dive.lbl.gov/portals/PNNLRiverCorridorSFA/About. This dataset is comprised of one main data folder containing (1) file-level metadata; (2) data dictionary; (3) field metadata; (4) dissolved organic carbon (DOC; measure as non-purgeable organic carbon; NPOC); (5) surface water sampling protocol; (6) sediment extraction protocol; (7) readme; (8) methods codes; (9) international generic sample number (IGSN) mapping file; and (10) folder of high resolution characterization of organic matter via 7 and 12 Tesla (7T and 12T) Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) generated through the Environmental Molecular Sciences Laboratory (EMSL; https://www.pnnl.gov/environmental-molecular-sciences-laboratory). The FTICR folder contains two subfolders containing the 7T and 12T .xml data files and another subfolder containing instructions for using Formularity (https://omics.pnl.gov/software/formularity) and an R script to process the data based on the user's specific needs. All files are .csv, .pdf, .R, .ref, or .xml.

Design Considerations and Operational Features of the Collimators for the Fermilab Main Injector and Recycler

The Fermilab Main Injector system delivers 700 kW of 120 GeV Proton beam for neutrino experiments. Since 2013 this has been achieved using slip stacking accumulation in the Recycler with up to 12 batches from the Fermilab Booster per Main Injector Ramp Cycle. To control activation from beam loss, collimation systems in the Booster to Recycler transfer line, in the Recycler and in the Main Injector are employed. Residual radiation measurements around the ring with detailed studies at the collimators are required to maintain adequate loss control. We will review design considerations, operational parameters and activation results for more than ten years of operation. Simulations with MARS15 are used to explore the activation rates and the isotopic composition of the resulting activation.

Development of Anti- Yersinia pestis Human Antibodies with Features Required for Diagnostic and Therapeutic Applications

Yersinia pestis is a category A infective agent that causes bubonic, septicemic, and pneumonic plague. Notably, the acquisition of antimicrobial or multidrug resistance through natural or purposed means qualifies Y. pestis as a potential biothreat agent. Therefore, high-quality antibodies designed for accurate and sensitive Y. pestis diagnostics, and therapeutics potentiating or replacing traditional antibiotics are of utmost need for national security and public health preparedness. Here, we describe a set of human monoclonal immunoglobulins (IgG1s) targeting Y. pestis fraction 1 (F1) antigen, previously derived from in vitro evolution of a phage-display library of single-chain antibodies (scFv). We extensively characterized these antibodies and their effect on bacterial and mammalian cells via: ELISA, flow cytometry, mass spectrometry, spectroscopy, and various metabolic assays. Two of our anti-F1 IgG (αF1Ig 2 and αF1Ig 8) stood out for high production yield, specificity, and stability. These two antibodies were additionally attractive in that they displayed picomolar affinity, did not compete when binding Y. pestis, and retained immunoreactivity upon chemical derivatization. Most importantly, these antibodies detected < 1,000 Y. pestis cells in sandwich ELISA, did not harm respiratory epithelial cells, induced Y. pestis agglutination at low concentration (350 nM), and caused apparent reduction in cell growth when radiolabeled at a nonagglutinating concentration (34 nM). These antibodies are amenable to the development of accurate and sensitive diagnostics and immuno/radioimmunotherapeutics.

Housedon-Hill - A ChemCam/RMI mega mosaic to investigate distant features

The ChemCam Remote Microscopic Imager (RMI) onboard Curiosity was originally designed to document the tiny areas analyzed by ChemCam’s laser-induced breakdown spectroscopy (LIBS) technique on rocks at few meters distance around the rover. The RMI produces 1024 x 1024 pixel black and white images directly from ChemCam’s telescope focal plane. Early in the mission, it was recognized that, thanks to its powerful optics, RMI could also play a role as a long-distance reconnaissance tool, complementing other cameras on the remote sensing mast, and taking advantage of its very long 700 mm focal length. Imaging areas several kilometers from the rover provides pictures that are very complementary to orbital observations and gives a more human-like, ground-based perspective. Between sols 2878 and 2921, Curiosity stayed parked at the same place to perform various rock sampling analyses. This opportunity was used to target very distant areas of interest, daily building an RMI mosaic of 216 overlapping images covering an area ranging from the bottom layers of Mount Sharp to the edge of Vera Rubin Ridge. This product is so far the largest RMI mosaic acquired during the entire mission.

Sights of summer from Lab’s earliest days digitization preserves images from 1940s, features early staff enjoying Los Alamos

Working at the Lab during World War II was serious business, but even these world-changing scientists and staff found time to play during the summer months. The National Security Research Center (NSRC), which is the Lab’s classified library and also houses unclassified artifacts from Los Alamos history, recently rescanned candid footage from two of contributors to the Manhattan Project.
