Benchmarking and parameter sensitivity of physiological and vegetation dynamics using the Functionally Assembled Terrestrial Ecosystem Simulator (FATES) at Barro Colorado Island, Panama

Plant functional traits determine vegetation responses to environmental variation, but variation in trait values is large, even within a single site. Likewise, uncertainty in how these traits map to Earth system feedbacks is large. We use a vegetation demographic model (VDM), the Functionally Assembled Terrestrial Ecosystem Simulator (FATES), to explore parameter sensitivity of model predictions, and comparison to observations, at a tropical forest site: Barro Colorado Island in Panama. We define a single 12-dimensional distribution of plant trait variation, derived primarily from observations in Panama, and define plant functional types (PFTs) as random draws from this distribution. We compare several model ensembles, where individual ensemble members vary only in the plant traits that define PFTs, and separate ensembles differ from each other based on either model structural assumptions or non-trait, ecosystem-level parameters, which include (a) the number of competing PFTs present in any simulation and (b) parameters that govern disturbance and height-based light competition. While single-PFT simulations are roughly consistent with observations of productivity at Barro Colorado Island, increasing the number of competing PFTs strongly shifts model predictions towards higher productivity and biomass forests. Different ecosystem variables show greater sensitivity than others to the number of competing PFTs, with the predictions that are most dominated by large trees, such as biomass, being the most sensitive. Changing disturbance and height-sorting parameters, i.e., the rules of competitive trait filtering, shifts regimes of dominance or coexistence between early- and late-successional PFTs in the model. Increases to the extent or severity of disturbance, or to the degree of determinism in height-based light competition, all act to shift the community towards early-successional PFTs. In turn, these shifts in competitive outcomes alter predictions of ecosystem states and fluxes, with more early-successional-dominated forests having lower biomass. It is thus crucial to differentiate between plant traits, which are under competitive pressure in VDMs, from those model parameters that are not and to better understand the relationships between these two types of model parameters to quantify sources of uncertainty in VDMs.

Evaluation of a quasi-steady-state approximation of the cloud droplet growth equation (QDGE) scheme for aerosol activation in global models using multiple aircraft data over both continental and marine environments

This research introduces a numerically efficient aerosol activation scheme and evaluates it by using stratus and stratocumulus cloud data sampled during multiple aircraft campaigns in Canada, Chile, Brazil, and China. The scheme employs a quasi-steady-state approximation of the cloud droplet growth equation (QDGE) to efficiently simulate aerosol activation, the vertical profile of supersaturation, and the activated cloud droplet number concentration (CDNC) near the cloud base. The calculated maximum supersaturation values using the QDGE scheme were compared with multiple parcel model simulations under various aerosol and environmental conditions. The differences are all below 0.18 %, indicating good performance and accuracy of the QDGE scheme. We evaluated the QDGE scheme by specifying observed environmental thermodynamic variables and aerosol information from 31 cloud cases as input and comparing the simulated CDNC with cloud observations. The average of mean relative error ($\overline{MRE}$) of the simulated CDNC for cloud cases in each campaign ranges from 17.30 % in Brazil to 25.90 % in China, indicating that the QDGE scheme successfully reproduces observed variations in CDNC over a wide range of different meteorological conditions and aerosol regimes. Additionally, we carried out an error analysis by calculating the maximum information coefficient (MIC) between the MRE and input variables for the individual campaigns and all cloud cases. MIC values were then sorted by aerosol properties, pollution level, environmental humidity, and dynamic condition according to their relative importance to MRE. Based on the error analysis, we found that the magnitude of MRE is more relevant to the specification of input aerosol pollution level in marine regions and aerosol hygroscopicity in continental regions than to other variables in the simulation.

Pythia8 Quark and Gluon Jets (float8 e4m3FN)

A float8 (e4m3FN) quantized version of the quark and gluon jet dataset originally published by Komiske, Metodiev, and Thaler (Zenodo record 3164691). Only the 20-file subset without charm and bottom quark jets is included here. All simulation parameters and jet selection criteria are identical to the original: Pythia 8.226, √s = 14 TeV Quarks from WeakBosonAndParton:qg2gmZq, gluons from WeakBosonAndParton:qqbar2gmZg with the Z decaying to neutrinos FastJet 3.3.0, anti-k_t jets with R = 0.4 p_T^jet ∈ [500, 550] GeV, |y^jet| < 1.7 There are 20 files, each in compressed NumPy format (QG_jets_fp8e4m3fn_0.npz through QG_jets_fp8e4m3fn_19.npz). Each file contains two arrays: X: (100000, M, 4) — 50k quark and 50k gluon jets, randomly sorted, padded to max multiplicity M, with particle features (pt, rapidity, azimuthal angle, pdgid) y: (100000,) — jet labels, gluon = 0, quark = 1 Since NumPy has no native fp8 dtype, X is stored as float32, but the values have been quantized through TensorFlow's float8_e4m3fn type and carry only fp8 precision. The quantization procedure is as follows: a global per-channel scale factor is computed from the absolute maximum value across all 20 chunks (with FP8_MAX = 448.0, the maximum representable value of e4m3FN). Each chunk is then scaled into the fp8 dynamic range, round-tripped through tf.experimental.float8_e4m3fn, and scaled back. This global scaling ensures a consistent quantization grid across the full dataset. The y labels are unchanged. Users should be aware that e4m3FN has limited dynamic range and precision. We recommend verifying this format is appropriate for your application; for a less aggressive reduction see the float16 and float32 versions linked below. If you use this dataset, please cite the original Zenodo record and its associated paper: Komiske, Metodiev, Thaler, Energy Flow Networks: Deep Sets for Particle Jets, JHEP 01 (2019) 121, arXiv:1810.05165

The Calpain-7 protease functions together with the ESCRT-III protein IST1 within the midbody to regulate the timing and completion of abscission

The Endosomal Sorting Complexes Required for Transport (ESCRT) machinery mediates the membrane fission step that completes cytokinetic abscission and separates dividing cells. Filaments composed of ESCRT-III subunits constrict membranes of the intercellular bridge midbody to the abscission point. These filaments also bind and recruit cofactors whose activities help execute abscission and/or delay abscission timing in response to mitotic errors via the NoCut/Abscission checkpoint. We previously showed that the ESCRT-III subunit IST1 binds the cysteine protease Calpain-7 (CAPN7) and that CAPN7 is required for both efficient abscission and NoCut checkpoint maintenance. Here, we report biochemical and crystallographic studies showing that the tandem microtubule-interacting and trafficking (MIT) domains of CAPN7 bind simultaneously to two distinct IST1 MIT interaction motifs. Structure-guided point mutations in either CAPN7 MIT domain disrupted IST1 binding in vitro and in cells, and depletion/rescue experiments showed that the CAPN7-IST1 interaction is required for (1) CAPN7 recruitment to midbodies, (2) efficient abscission, and (3) NoCut checkpoint arrest. CAPN7 proteolytic activity is also required for abscission and checkpoint maintenance. Hence, IST1 recruits CAPN7 to midbodies, where its proteolytic activity is required to regulate and complete abscission.

Development of an inexpensive matrix-assisted laser desorption—time of flight mass spectrometry method for the identification of endophytes and rhizobacteria cultured from the microbiome associated with maize

Many endophytes and rhizobacteria associated with plants support the growth and health of their hosts. The vast majority of these potentially beneficial bacteria have yet to be characterized, in part because of the cost of identifying bacterial isolates. Matrix-assisted laser desorption-time of flight (MALDI-TOF) has enabled culturomic studies of host-associated microbiomes but analysis of mass spectra generated from plant-associated bacteria requires optimization. In this study, we aligned mass spectra generated from endophytes and rhizobacteria isolated from heritage and sweet varieties of Zea mays. Multiple iterations of alignment attempts identified a set of parameters that sorted 114 isolates into 60 coherent MALDI-TOF taxonomic units (MTUs). These MTUs corresponded to strains with practically identical (>99%) 16S rRNA gene sequences. Mass spectra were used to train a machine learning algorithm that classified 100% of the isolates into 60 MTUs. These MTUs provided >70% coverage of aerobic, heterotrophic bacteria readily cultured with nutrient rich media from the maize microbiome and allowed prediction of the total diversity recoverable with that particular cultivation method. Acidovorax sp., Pseudomonas sp. and Cellulosimicrobium sp. dominated the library generated from the rhizoplane. Relative to the sweet variety, the heritage variety contained a high number of MTUs. The ability to detect these differences in libraries, suggests a rapid and inexpensive method of describing the diversity of bacteria cultured from the endosphere and rhizosphere of maize.

INTERNATIONAL COOPERATION IN SPACE

In the community of nations there exists today a deep and wide abyss separating one important and powerful group of nations from another. The political and philosophical differences which constitute this abyss are very real and very fundamental, and they are not likely to disappear during this century. There are smaller canyons in the landscape as well, some more permanent than others. However, it is possible to build bridges of culture, science, and cooperative effort of many kinds across these canyons so that men of good will from one side may mingle freely with those from the other side, and for a little while, at least, be as one people. Scientists have been helping to build bridges like this for a long time. Especially prominent in this sort of activity has been the International Council of Scientific Unions (ICSU) which was the mother hen, so to speak, of the International Geophysical Year. At present, problems of international cooperation in space research are the concern of a Special Committee of the International Council of Scientific Unions, known as COSPAR (Committee on Space Research).
