CRISPR/Cas9 Directed Reprogramming of iPSC for Accelerated Motor Neuron Differentiation Leads to Dysregulation of Neuronal Fate Patterning and Function

Neurodegeneration causes a significant disease burden and there are few therapeutic interventions available for reversing or slowing the disease progression. Induced pluripotent stem cells (iPSCs) hold significant potential since they are sourced from adult tissue and have the capacity to be differentiated into numerous cell lineages, including motor neurons. This differentiation process traditionally relies on cell lineage patterning factors to be supplied in the differentiation media. Genetic engineering of iPSC with the introduction of recombinant master regulators of motor neuron (MN) differentiation has the potential to shorten and streamline cell developmental programs. We have established stable iPSC cell lines with transient induction of exogenous LHX3 and ISL1 from the Tet-activator regulatory region and have demonstrated that induction of the transgenes is not sufficient for the development of mature MNs in the absence of neuron patterning factors. Comparative global transcriptome analysis of MN development from native and Lhx-ISL1 modified iPSC cultures demonstrated that the genetic manipulation helped to streamline the neuronal patterning process. However, leaky gene expression of the exogenous MN master regulators in iPSC resulted in the premature activation of genetic pathways characteristic of the mature MN function. Dysregulation of metabolic and regulatory pathways within the developmental process affected the MN electrophysiological responses.

High Precision Piezoelectric Linear Motors for Operations at Cryogenic Temperatures and Vacuum

The Jet Propulsion Laboratory evaluated the use of an electromechanical device for optically positioning a mirror system during the pre-project phase of the Pluto-Fast-Flyby (PFF) mission. The device under consideration was a piezoelectric driven linear motor functionally dependent upon a time varying electric field which induces displacements ranging from submicrons to millimeters with positioning accuracy within nanometers. Using a control package, the mirror system provides image motion compensation and mosaicking capabilities. While this device offers unique advantages, there were concerns pertaining to its operational capabilities for the PFF mission. The issues include irradiation effects and thermal concerns. A literature study indicated that irradiation effects will not significantly impact the linear motor's operational characteristics. On the other hand, thermal concerns necessitated an in depth study.

Actuation Using Piezoelectric Materials: Application in Augmenters, Energy Harvesters, and Motors

Piezoelectric actuators are used in many manipulation, movement, and mobility applications as well as transducers and sensors. When used at the resonance frequencies of the piezoelectric stack, the actuator performs at its maximum actuation capability. In this Space Grant internship, three applications of piezoelectric actuators were investigated including hammering augmenters of rotary drills, energy harvesters, and piezo-motors. The augmenter shows improved drill performance over rotation only. The energy harvesters rely on moving fluid to convert mechanical energy into electrical power. Specific designs allow the harvesters more freedom to move, which creates more power. The motor uses the linear movement of the actuator with a horn applied to the side of a rotor to create rotational motion. Friction inhibits this motion and is to be minimized for best performance. Tests and measurements were made during this internship to determine the requirements for optimal performance of the studied mechanisms and devices.

Pot-ability Assessment of Litz Wires for High Power Density Electric Motor

An alternative process technique, namely vacuum-assisted axial injection potting (VaAIP), has been developed to pot the Litz wires in the stator winding of high power density electric motors for the future electrified aircrafts. Initial trials of the process showed significant improvement in potting quality with less voids, thus potential improvement in thermal management of the motors. As an initial effort of pot-ability assessment, microstructures, 2-D and 3-D, of the Litz wires including dimensions and distribution of conductor filament, coating, and open spaces; packing patterns; shape/configuration changes of each bundles or the overall cross-sections per degree of twist were determined and quantified successfully. The microstructure analyses were performed not only for effective potting process development but also for more realistic electro-thermal modeling solutions. This paper will present results of the microstructure analyses, potentials of the VaAIP process from the trials, and future plans for scale-up and implementation of the process into a full-scale prototype stator winding.

Designed Experiment Results for Constituent Effects on NBR Based Solid Rocket Motor Insulation Performance

Solid rocket motor internal insulations are facing serious issues with obsolescence of constituents and the need for improved performance. The design community is at a unique point with a large number of rocket motors requiring reformulation of insulation or design upgrades at the same time. Unfortunately, performance data related to component choice is not generally available to aid development. A study of insulation component materials, conducted at Marshall Space Flight Center over the last four years, used designed experiments to understand the contribution of each material to the performance of a formulation. This report covers work with Nitrile Butadiene Rubber (NBR) based formulations. In-house compounding provided modified formulations allowing for isolation of both individual and synergistic effects. All formulas were tested for mechanical, thermal and producibility properties to evaluate performance. This presentation will describe the methodology for, and results from, identifying the individual and combined effects of components used in our formulations. Specific properties investigated include, rheology, cure characteristics, density, tensile strength, along with erosion/ablation under various heat loads and flame chemistries. Scale up of several formulas from lab to production size batches was accomplished. We will discuss troubleshooting problems of scale and the new set of designed experiments with Ethylene Propylene Diene Monomer (EPDM) based formulations.

Modeling and Mitigation of dv/dt and Transmission Line Effect for Motor Drive System in Electric Aircraft Propulsion

Due to constraints in space and structure as well as other practical limitations in aircraft system, the motor controller or electric motor are often installed far away from each other and interconnected via long cables. This amplifies the transmission line effect excited by the dv/dt PWM voltage and induces high voltage stress on the machine winding. This paper investigates the state-of-art modeling approaches for such systems, identifies several limitations in matching the tested voltage reflection waveform in terms of peak voltage, oscillation frequency and damping speed, and offers several key considerations not well covered in literature or in engineering practice. It then proposes a practical modeling methodology which, as validated by experimental results, truly captures these critical reflection voltage specs. To mitigate the high peak voltage and strong oscillation, dv/dt filter is applied and a parametric design optimization based on widely adopted LC-R topology is illustrated, which not only alleviates the voltage stress but also minimized the power loss of the passive damping resistor.

Residual Thrust in a Solid Rocket Motor: A Model Applied to the Mars Ascent Vehicle

A model for solid rocket motor residual thrust is built from the concept that hot nozzle phenolic or carbon materials provide a radiating source of heat flux to the insulation, leading to continued off-gassing by pyrolysis. This model is 1. Fitted to flight datasets and scaled to the Mars Ascent Vehicle second stage motor (MAV SRM2), and 2. Derived with MAV SRM2 design and material estimates. The two methods show a potential range of modeling uncertainty for use by mission analysts. Further loss methods that could reduce the maximum residual impulse are identified but left to further work to evaluate.

Extended Investigation into Fault-Tolerant Integrated Motor Drive for a Quadrotor Urban Air Mobility (UAM) Aircraft

Electric propulsor machines and their associated power electronics have been identified in past studies as low-reliability components in emerging Urban Air Mobility (UAM) Vertical Takeoff and Landing (VTOL) vehicles that raise their predicted catastrophic failure rates. This research effort extends previously presented work investigating the use of fault-tolerant (FT) motor drives in quadrotor aircraft as a promising approach to significantly increase their mean-time-to-failure (MTTF). In particular, fault-tolerant modular motor drives (FT-MMDs) are identified as strong candidates for closing the electric propulsor reliability gap. These FT-MMDs divide a machine drive into multiple redundant modules, each consisting of three phase stator windings and power electronics units, enabling continued machine operation after a module failure. Achieving the highest possible isolation between modules (physical, magnetic, thermal, electrical) is critically important in FT-MMDs to prevent the propagation of any failure between modules. Key additional requirements for achieving the highest possible reliability characteristics with FT-MMDs are high repair rates and aggressive suppression of all single-point failures.
