ORBITaL-Net: A labeled training library for large-scale building feature extraction

Over the course of several years, nearly 1.5 million building outlines have been created from approximately 128,000 training tiles covering roughly 7,000 km 2 of very high-resolution multispectral overhead imagery, primarily dated between 2010 and 2020. This dataset, dubbed the Oak Ridge Building Image and TrAining Label Net (ORBITaL-Net), is designed for machine learning applications and is global in scope, with samples drawn from 72 countries across North America, South America, Africa, Europe, and Asia. ORBITaL-Net captures a great diversity in geographic setting, structural characteristics, land use (urban and rural), terrain, and imagery conditions. While the labeled building outlines are themselves valuable, the dataset’s true strength lies in the pairing of these labels with corresponding reference imagery, which is being released for open source use. Similar to SpaceNet and Replicable AI For Microplanning (ramp), this building outline dataset will allow the larger computer vision community from academia, government, and industry the opportunity to develop robust, scalable, and generalizable geospatial machine learning techniques. Unlike SpaceNet and ramp, which offer high resolution labels and imagery primarily for large urban cities, ORBITaL-Net is not focused on training samples from heavily populated areas but instead aims to capture the innate variability of conditions present in both the physical environment and imagery collections.

Liquid state properties and solidification features of the pseudo binary BaS-La 2 S 3

The high temperature thermodynamic properties of chalcogenides materials based on BaS remain elusive. Herein the pseudo binary BaS-La 2 S 3 is investigated above 1573 K. The liquid properties of BaS-La 2 S 3 are measured by means of high resolution in-situ visualization coupled with thermal arrest measurements in a thermal imaging furnace. This enables to report the first observation of such melts in a container-less setting. The melting points of BaS and La 2 S 3 are revisited at 2454 K and 2004 K respectively. La 2 S 3 demonstrates a high stability in its liquid state, in strike difference with the sublimation observed for BaS. BaS is however partially stabilized with the addition of few percents of La 2 S 3 . The remarkable chemical and thermal stability of La 2 S 3 -rich samples contrasts with the partial decomposition and high vapor pressure observed for BaS-rich samples. Observations and analysis of the solidified samples suggest three different solid solutions. Solid and liquid densities are investigated along the different compositions, supporting a first estimate of the volumetric thermal expansion coefficient for La 2 S 3 .

Cacao pod transcriptome profiling of seven genotypes identifies features associated with post-penetration resistance to Phytophthora palmivora

Abstract The oomycete Phytophthora palmivora infects the fruit of cacao trees ( Theobroma cacao ) causing black pod rot and reducing yields. Cacao genotypes vary in their resistance levels to P. palmivora , yet our understanding of how cacao fruit respond to the pathogen at the molecular level during disease establishment is limited. To address this issue, disease development and RNA-Seq studies were conducted on pods of seven cacao genotypes (ICS1, WFT, Gu133, Spa9, CCN51, Sca6 and Pound7) to better understand their reactions to the post-penetration stage of P. palmivora infection. The pod tissue- P. palmivora pathogen assay resulted in the genotypes being classified as susceptible (ICS1, WFT, Gu133 and Spa9) or resistant (CCN51, Sca6 and Pound7). The number of differentially expressed genes (DEGs) ranged from 1625 to 6957 depending on genotype. A custom gene correlation approach identified 34 correlation groups. De novo motif analysis was conducted on upstream promoter sequences of differentially expressed genes, identifying 76 novel motifs, 31 of which were over-represented in the upstream sequences of correlation groups and associated with gene ontology terms related to oxidative stress response, defense against fungal pathogens, general metabolism and cell function. Genes in one correlation group (Group 6) were strongly induced in all genotypes and enriched in genes annotated with defense-responsive terms. Expression pattern profiling revealed that genes in Group 6 were induced to higher levels in the resistant genotypes. An additional analysis allowed the identification of 17 candidate cis -regulatory modules likely to be involved in cacao defense against P. palmivora . This study is a comprehensive exploration of the cacao pod transcriptional response to P. palmivora spread after infection. We identified cacao genes, promoter motifs, and promoter motif combinations associated with post-penetration resistance to P. palmivora in cacao pods and provide this information as a resource to support future and ongoing efforts to breed P. palmivora -resistant cacao.

Incorporating long-range dependence and fractal features in turbulence spectra

We introduce an advanced turbulence spectrum model developed from mathematical foundations from a covariance function class and empirically validated using extensive field data. This model captures the complex dynamics of long-range dependence, and fractal characteristics prevalent in riverine and atmospheric boundary layer (ABL) flows that are ignored by classical spectrum models, such as IEC (International Electrotechnical Commission) von Kármán and Kaimal model. The model delineates scaling behaviors across distinct frequency bands and offers substantial flexibility through five well-defined parameters each characterizing a distinct physical aspect of the velocity time series. A detailed procedure for obtaining each parameter from time series data is outlined. The comprehensive validations with field data from tidal currents and ABL flows substantiate the model’s fidelity in accurately replicating observed phenomena. This validation establishes the reliability of the proposed model and, when incorporated into stochastic full-field simulators such as TurbSim, demonstrates its potential to advance the predictive modeling and analysis of turbulent flows in environmental science and engineering contexts.

The global Sahel monsoon ocean-pressure index reconciles its regional and large-scale features

Monitoring Sahelian rainfall variability is increasingly critical as climate extremes intensify across the region. Here, we develop the Sahelian Monsoon Ocean-Pressure Index (SMOPI), a novel global synthetic indicator constructed from five dynamically coherent sea-level pressure regions statistically linked to June-September Sahel monsoon rainfall. SMOPI captures intra-seasonal and interannual variability, and crucially, reflects the influence of both regional processes and large-scale teleconnections on monsoon dynamics. It aligns with the dominant rainfall variability mode in reanalyses and 29 CMIP6 models. Strong/positive SMOPI phases coincide with wet years and are associated with enhanced convergence, favorable jet configurations, and robust Pacific, Atlantic, and Indian Ocean teleconnections. Conversely, weak/negative SMOPI phases correspond to drought conditions and divergent moisture fluxes. SMOPI exposes model failures in reproducing historical droughts and offers new physical insights into rainfall-driving mechanisms. It stands out as a scalable, potentially transferable diagnostic tool for monitoring/forecasting and evaluating Sahelian monsoon rainfall under global warming.

Structural features of Dnase1L3 responsible for serum antigen clearance

Autoimmunity develops when extracellular DNA released from dying cells is not cleared from serum. While serum DNA is primarily digested by Dnase1 and Dnase1L3, Dnase1 cannot rescue autoimmunity arising from Dnase1L3 deficiencies. Dnase1L3 uniquely degrades antigenic forms of cell-free DNA, including DNA complexed with lipids and proteins. The distinct activity of Dnase1L3 relies on its unique C-terminal Domain (CTD), but the mechanism is unknown. We used multiple biophysical techniques and functional assays to study the interplay between the core catalytic domain and the CTD. While the core domain resembles Dnase1, there are key structural differences between the two enzymes. First, Dnase1L3 is not inhibited by actin due to multiple differences in the actin recognition site. Second, the CTD augments the ability of the core to bind DNA, thereby facilitating the degradation of complexed DNA. Together, these structural insights will inform the development of Dnase1L3-based therapies for autoimmunity.

Sensitivity of mesoscale modeling to urban morphological feature inputs and implications for characterizing urban sustainability

We examine the differences in meteorological output from the Weather Research and Forecasting (WRF) model run at 270 m horizontal resolution using 10 m, 100 m and 1 km resolution 3D neighborhood morphological inputs and with no morphological inputs. We find that the spatial variability in temperature, humidity, and other meteorological variables across the city can vary with the resolution and the coverage of the 3D urban morphological input, and that larger differences occur between simulations run without 3D morphological input and those run with some type of 3D morphology. We also find that the inclusion of input-building-defined roughness length calculations would improve simulation results further. We show that these inputs produce different patterns of heat wave spatial heterogeneity across the city of Washington, DC. These findings suggest that understanding neighborhood level urban sustainability under extreme heat waves, especially for vulnerable neighborhoods, requires attention to the representation of surface terrain in numerical weather models.

Simultaneous removal of C 2 H 2 and C 2 H 6 for C 2 H 4 purification by robust MOFs featuring a high density of heteroatoms

Simultaneous removal of C 2 H 6 and C 2 H 2 from C 2 H 4 streams is of great importance in the petrochemical industry but remains a challenging task. To address this challenge, we have selected three isoreticular MOFs with high stability, low cost, and desirable scale-up ability, namely, MOF-303, MIL-160, and CAU-23 and assessed their potential in simultaneous removal of acetylene and ethane for ethylene purification. Each MOF exhibits desirable C 2 H 2 and C 2 H 6 uptake capacity (>5.5 mmol g –1 and >4 mmol g –1 , respectively), as well as good C 2 H 2 /C 2 H 4 selectivity (>2) and C 2 H 6 /C 2 H 4 selectivity (>1.5). Notably, MOF-303 takes up 4.96 mmol g –1 C 2 H 6 at 298 K and 1 bar, the highest value among the three MOFs, with C 2 H 6 /C 2 H 4 selectivity in the range of 1.55–2.47. MIL-160 possesses a very high C 2 H 2 uptake (9.1 mmol g –1 ) and C 2 H 2 /C 2 H 4 selectivity, 10.6 (1 : 1, v/v), at 298 K, much higher than those of all other MOFs tested to date for simultaneous removal of C 2 H 6 and C 2 H 2 from C 2 H 4 . The results from breakthrough experiments confirm that all three MOFs demonstrate excellent performance for C 2 H 4 purification in a ternary mixture of C 2 H 6 /C 2 H 4 /C 2 H 2 (1 : 1 : 1, v/v/v). Here, for MOF-303, MIL-160, and CAU-23, polymer-grade C 2 H 4 up to 0.164, 0.21, and 0.181 mmol g –1 can be obtained from the equimolar ternary mixture in a single separation step from the breakthrough experiment. Additionally, DFT calculations have been performed to further investigate the mechanism of adsorption/separation for C 2 H 6 , C 2 H 4 , and C 2 H 2 .
