Sexual dimorphism and the multi-omic response to exercise training in rat subcutaneous white adipose tissue

Subcutaneous white adipose tissue (scWAT) is a dynamic storage and secretory organ that regulates systemic homeostasis, yet the impact of endurance exercise training (ExT) and sex on its molecular landscape is not fully established. Utilizing an integrative multi-omics approach, and leveraging data generated by the Molecular Transducers of Physical Activity Consortium (MoTrPAC), we show profound sexual dimorphism in the scWAT of sedentary rats and in the dynamic response of this tissue to ExT. Specifically, the scWAT of sedentary females displays -omic signatures related to insulin signaling and adipogenesis, whereas the scWAT of sedentary males is enriched in terms related to aerobic metabolism. These sex-specific -omic signatures are preserved or amplified with ExT. Integration of multi-omic analyses with phenotypic measures identifies molecular hubs predicted to drive sexually distinct responses to training. Overall, this study underscores the powerful impact of sex on adipose tissue biology and provides a rich resource to investigate the scWAT response to ExT.

A model for K-shell x-ray yield from magnetic implosions at Sandia's Z machine

A zero-dimensional magnetic implosion model with a coupled equivalent circuit for the description of an imploding nested wire array or gas puff is presented. Circuit model results have been compared with data from imploding stainless steel wire arrays, and good agreement has been found. The total energy coupled to the load, Ej×B, has been applied to a simple semi-analytic K-shell yield model, and excellent agreement with previously reported K-shell yields across all wire array and gas puff platforms is seen. Trade space studies in implosion radius and mass have found that most platforms operate near the predicted maximum yield. In some cases, the K-shell yield may be increased by increasing the mass or radius of the imploding array or gas puff.

Platform for 100 s Mbar equation of state measurements on the National Ignition Facility

Equation of state (EOS) measurements in the 100 s Mbar range are needed to underwrite models employed in the simulation of high energy density plasmas. To this end, a platform has been developed for fielding on the National Ignition Facility, capable of producing high-quality impedance match EOS data, wherein a planar, high-pressure, steady shock is driven into a sample package, and sample and reference standard shock velocities are measured. This platform, dubbed planar high pressure, or PHP, was fielded with an initial proof-of-concept shot in January 2023. The first PHP shot, aiming to study gold, demonstrated a pressure close to 400 Mbar, two orders of magnitude higher than previously reported gold EOS data.

The role of thermal conduction on the size scaling of laser direct-drive implosions

Ablation pressure is a key parameter controlling laser drive performance in direct-drive inertial confinement fusion implosions. In this work, we develop a theoretical framework to clarify the role of thermal conduction in determining how ablation pressure scales with laser intensity, wavelength, and target size. We focus on a steady-state, spherically expanding plasma produced when an over-dense pellet is irradiated by a high-intensity laser. Scaling laws for ablation pressure and the mass ablation rate are derived with respect to laser intensity, wavelength, and target size. As a result, the theoretical predictions are found to be in good agreement with one-dimensional hydrodynamics simulations in spherical geometry.

Physiological Adaptations to Progressive Endurance Exercise Training in Adult and Aged Rats: Insights from the Molecular Transducers of Physical Activity Consortium (MoTrPAC)

While regular physical activity is a cornerstone of health, wellness, and vitality, the impact of endurance exercise training on molecular signaling within and across tissues remains to be delineated. The Molecular Transducers of Physical Activity Consortium (MoTrPAC) was established to characterize molecular networks underlying the adaptive response to exercise. Here, we describe the endurance exercise training studies undertaken by the Preclinical Animal Sites Studies component of MoTrPAC, in which we sought to develop and implement a standardized endurance exercise protocol in a large cohort of rats. To this end, Adult (6-mo) and Aged (18-mo) female (n = 151) and male (n = 143) Fischer 344 rats were subjected to progressive treadmill training (5 d/wk, ~70%–75% VO 2 max) for 1, 2, 4, or 8 wk; sedentary rats were studied as the control group. A total of 18 solid tissues, as well as blood, plasma, and feces, were collected to establish a publicly accessible biorepository and for extensive omics-based analyses by MoTrPAC. Treadmill training was highly effective, with robust improvements in skeletal muscle citrate synthase activity in as little as 1–2 wk and improvements in maximum run speed and maximal oxygen uptake by 4–8 wk. For body mass and composition, notable age- and sex-dependent responses were observed. This work in mature, treadmill-trained rats represents the most comprehensive and publicly accessible tissue biorepository, to date, and provides an unprecedented resource for studying temporal-, sex-, and age-specific responses to endurance exercise training in a preclinical rat model.

Enabling Interoperable SCADA Communications for PV Inverters through Embedded Controllers

The percentage integration of photovoltaic (PV) inverters in the field has increased significantly in the past 5 years. Regardless of the size of the PV plants and the inverters (residential vs. commercial), it is becoming crucial that these devices have the capability to communicate with peers (other smart devices) and with components that are at a hierarchy above the inverters (e.g., supervisory control and data acquisition (SCADA) systems, distributed controllers, and data managers). This project aims to develop a standard SCADA software code for inverters’ embedded controllers that will enable interoperability with other components in the system. To achieve this, the code will be developed using two different protocols: Distributed Network Protocol 3 and International Electrotechnical Commission 61850. The developed code is aimed to be deployed in simple embedded controllers. It will be tested in the National Renewable Energy Laboratory’s (NREL’s) Energy Systems Integration Facility. The tested code will then be made available through Triangle MicroWorks’s (TMW’s) software platform. The primary objectives of this project include training the NREL team with TMW’s embedded controller libraries, developing an interoperable communication code for embedded controllers, successfully testing and deploying the code, and demonstrating the newly developed code in a conference.
