Diagnostic Testing for COVID-19 Bridging Study for QIAamp Viral RNA Extraction vs Beckman RNAdvance vs Thermofisher MagMAX

This report describes testing that was performed by LANL’s Biological Agent Testing Lab (BATL) to validate modifications to the CDC EUA 2019-Novel Coronavirus (2019- nCoV) Real-Time RT-PCR Diagnostic Panel (EUA-CDC-nCoV-IFU). BATL intends to implement the modifications to increase thoughput for daily testing . BATL validated the viral RNA extraction process, using the orignial component, QIAamp Viral RNA Mini Kit (Cat # 52906) and the new components, Beckman Coulter magnetic 96-well plate RNAdvance Viral kit (Cat # C63510), Thermofisher MagMAX Viral/Pathogen Nucleic Acid Isolation Kit (Cat # A48310). Equivalency was demonstrated between the original component and the Beckman Coulter magnetic 96-well plate RNAdvance Viral kit (Cat # C63510). Equivalency was also demonstrated between the original component and the Thermofisher MagMAX Viral/Pathogen Nucleic Acid Isolation Kit (Cat # A48310). Subsequently, substitution of the original component with either of these kits for viral RNA extraction increased BATL’s extraction capability from 100 samples per day to 279 samples per day.

Bridging Multiscale Processes in Earth System Models with Physics-Guided Hierarchical Machine Learning

Focal Area(s): The focal area of this whitepaper is, “Predictive modeling through the use of AI techniques and AI-derived model components; the use of AI and other tools to design a prediction system comprising of a hierarchy of models (e.g., AI driven model/component/parameterization selection).” The ideas and frameworks described herein are deemed site agnostic. As a use case, this group will initially focus on the coupling between land processes, surface/subsurface hydrological processes, coastal processes, and human activities in the U.S. Gulf states.

Enabling Predictive Scale-Bridging Simulations through Active Learning (Institutional Computing Annual Report (Project w20_alscalebridging)) [Slides]

The goal of this project was to develop, demonstrate, and provide a new capability to achieve greater physical fidelity in large-scale simulations, rather than the usual brute-force increases in the number of mesh elements or particles. This was done by using machine learning (ML) techniques to develop emulators for subscale physics that can be used in coarse-scale continuum simulations, trained on fine-scale molecular dynamics (MD) simulations that are launched on-the-fly via active learning. raditional inertial confinement fusion (ICF) simulations rely on numerical diffusion to simulate molecular effects such as non-local transport and mixing without truly accounting for molecular interactions; our approach directly accounts for this physics.

Bridging Equipment Reliability Data and Robust Decisions in a Plant Operation Context

In order to reduce operation and maintenance (O&M) costs, nuclear power plants (NPPs) are moving from corrective and periodic maintenance to predictive maintenance strategies. Such transition requires changes on the data that needs to be retrieved and on the type of decision processes to be employed. Advanced monitoring and data analysis technologies are essential to support predictive strategies. They can in fact provide precise information about health of a component, track its degradation trends, and provide information of its expected failure time. With such information, maintenance operations for a component can be performed right before its expected failure time. This dynamic context of O&M operations requires new methods to analyze data, propagate component health information from the component to the system level, and optimize plant resources. In this respect, the risk informed asset management (RIAM) project has been tasked to develop and test this new class of methods into a risk analytics toolset. This toolset consists of data analytics tools coupled with reliability methods designed to manage plant assets and performances in a predictive maintenance context. This report shows the latest improvements on such development and the initial testing of our methods on the three main research areas that the RIAM project is focusing on. These areas are the following: equipment reliability data analytics, system reliability modeling, and plant resources optimization methods. We show how the methods developed in these areas can support predictive maintenance strategies by: 1) analyzing equipment reliability data (either in numeric and textual form), 2) assessing component and system health through an innovative margin-based reliability approach, and 3) identifying the most critical components and set optimal maintenance schedule based on plant economic and operational constraints.
