Index tests for geosynthetic clay liners containing bentonite–polymer composites

The hydraulic conductivity of geosynthetic clay liners (GCLs) containing bentonite–polymer composites (BPCs) is controlled by a pore clogging mechanism related to the swelling of sodium bentonite (NaB) granules and formation of polymer gel in the pore space. Three index testing procedures that are influenced by bentonite swelling and rheology of the polymer gel were evaluated in this study for screening BPC GCLs for hydraulic conductivity: viscosity, liquid limit by fall-cone penetration and active water content. Experiments were conducted using polyacrylamide (PAM) polymer, dry-blended mixtures of PAM and NaB and a BPC from a commercially available GCL. These materials were contacted with inorganic solutions representing leachates from a range of waste containment scenarios. For the BPC that was evaluated, hydraulic conductivities less than 10 -10 m/s were obtained when the viscosity exceeded 220 mPa s and the active water content exceeded 300%. Hydraulic conductivities less than 10 -11 m/s were achieved for viscosity exceeding 350 mPa s, fall-cone liquid limit exceeding 190 and active water content exceeding 300%. Additional testing is needed to assess the generality of these index tests and the identified thresholds when applied to other BPC GCLs and other permeant liquids.

Delta-Rice: A HDF5 Compression Plugin optimized for Digitized Detector Data

Delta-Rice is an HDF5 (The HDF Group et al., 2020) filter plugin that was developed to compress digitized detector signals recorded by the Nab experiment (Fry et al., 2019), a fundamental neutron physics experiment. This is a two-step process where incoming data is passed through a pre-processing filter and then compressed with Rice coding. A routine for determining the optimal pre-processing filter for a dataset is provided along with an example GPU deployment. When applied to data collected by the Nab data acquisition system, this method produced output files 29% their initial size, and was able to do so with an average read/write throughput in excess of 2 GB/s on a single CPU. Compared to the widely used Gzip compression routine, Delta-Rice reduces the file size by 33% more with over an order of magnitude increase in read/write throughput. Delta-Rice is available on CPU to users through the HDF5 library.

A novel, steerable, low-energy proton source for detector characterization

We report on the conversion of the Manitoba II mass spectrometer into a versatile low-energy proton beam facility. This infrastructure is adaptable to any detector-under-test (DUT), and has proven itself effective with the characterization of silicon detectors used in subatomic beyond-the-Standard-Model (BSM) searches, namely the Nab experiment. A pencil beam of mono-energetic protons can be produced in a range from 25 keV to 35 keV, achieving a beam current of ~1 x 10 -18 A. Electrostatic steering plates were constructed to direct the Gaussian-profile proton beam over a 117 mm diameter area-of-interest with full-width at half-maxima (FWHM) ranging from 0.6 mm to 1.26 mm. This work discusses the modifications and subsequent tests to confirm the beam specifications met the demands of the aforementioned detectors.

A Germline-Targeting Chimpanzee SIV Envelope Glycoprotein Elicits a New Class of V2-Apex Directed Cross-Neutralizing Antibodies

HIV-1 and its SIV precursors share a broadly neutralizing antibody (bNAb) epitope in variable loop 2 (V2) at the envelope glycoprotein (Env) trimer apex. Here, we tested the immunogenicity of germ line-targeting versions of a chimpanzee SIV (SIVcpz) Env in human V2-apex bNAb heavy-chain precursor-expressing knock-in mice and as chimeric simian-chimpanzee immunodeficiency viruses (SCIVs) in rhesus macaques (RMs). Trimer immunization of knock-in mice induced V2-directed NAbs, indicating activation of V2-apex bNAb precursor-expressing mouse B cells. SCIV infection of RMs elicited high-titer viremia, potent autologous tier 2 neutralizing antibodies, and rapid sequence escape in the canonical V2-apex epitope. Six of seven animals also developed low-titer heterologous plasma breadth that mapped to the V2-apex. Antibody cloning from two of these animals identified multiple expanded lineages with long heavy chain third complementarity determining regions that cross-neutralized as many as 7 of 19 primary HIV-1 strains, but with low potency. Negative stain electron microscopy (NSEM) of members of the two most cross-reactive lineages confirmed V2 targeting but identified an angle of approach distinct from prototypical V2-apex bNAbs, with antibody binding either requiring or inducing an occluded-open trimer. Probing with conformation-sensitive, nonneutralizing antibodies revealed that SCIV-expressed, but not wild-type SIVcpz Envs, as well as a subset of primary HIV-1 Envs, preferentially adopted a more open trimeric state. These results reveal the existence of a cryptic V2 epitope that is exposed in occluded-open SIVcpz and HIV-1 Env trimers and elicits cross-neutralizing responses of limited breadth and potency.

Community-Scale Solar Deployment in the Northwest Arctic

NANA Regional Corporation (NRC, or NANA) was formed as an Alaska Native Corporation (ANC) pursuant to the Alaska Native Claims Settlement Act of 1971. Our lands cover 39,000 square miles of the Northwest Arctic region of Alaska. We partner closely with the Northwest Arctic Borough (NAB) and our 11 remote communities on numerous projects and activities, especially around clean energy initiatives that improve quality of life and help to reduce extremely high energy costs experienced by the communities in our region. Collectively, the regional partnership has developed numerous successful solar, wind, biomass, and energy storage, distribution upgrade, and efficiency projects. To further progress, we created the Northwest Arctic Energy Steering Committee to share and replicate these benefits across the region. NANA’s mission is to provide economic opportunities for our more than 13,500 Iñupiat shareholders and to protect and enhance NANA lands.

High Penetration Solar and Battery Project in Noatak, Alaska

Through funding from the Department of Energy’s Office of Indian Energy, the Northwest Arctic Borough (NAB) and the Native Village of Noatak (NNV) formed a Tribal Energy Development Organization (TEDO) to implement the Noatak Solar and Battery Project to reduce reliance on costly imported diesel, stabilize energy costs, and strengthen local energy sovereignty.

Defining criteria for broadly neutralizing HIV antibodies

Over the course of a few years, a small percentage of individuals with HIV-1 develop broadly neutralizing antibodies (bnAbs) capable of neutralizing diverse viruses. Although hundreds of antibodies with neutralizing activity against heterologous viruses have been referred to as bnAbs, there is no universally accepted numerical definition of a bnAb. Here, we will review important elements of HIV neutralizing antibodies and proposed definitions of bnAbs, as well as introduce a web-based tool, CAByN (Choose Antibodies by Neutralization), allowing users to identify antibodies meeting their numerical definitions of a bnAb from data in the Los Alamos HIV Databases CATNAP (Compile, Analyze and Tally NAb Panels) antibody neutralization database. Biological findings from use of CAByN are also presented here, including differential neutralizing activity for certain antibodies across viral clades, and identification of antibodies with suspected incomplete neutralization.
