Associations between regional blood-brain barrier permeability, aging, and Alzheimer’s disease biomarkers in cognitively normal older adults

Background Increased blood-brain barrier permeability (BBBp) has been hypothesized as a feature of aging that may lead to the development of Alzheimer’s disease (AD). We sought to identify the brain regions most vulnerable to greater BBBp during aging and examine their regional relationship with neuroimaging biomarkers of AD. Methods We studied 31 cognitively normal older adults (OA) and 10 young adults (YA) from the Berkeley Aging Cohort Study (BACS). Both OA and YA received dynamic contrast-enhanced MRI (DCE-MRI) to quantify K trans values, as a measure of BBBp, in 37 brain regions across the cortex. The OA also received Pittsburgh compound B (PiB)-PET to create distribution volume ratios (DVR) images and flortaucipir (FTP)- PET to create partial volume corrected standardized uptake volume ratios (SUVR) images. Repeated measures ANOVA assessed the brain regions where OA showed greater BBBp than YA. In OA, K trans values were compared based on sex, Aβ positivity status, and APOE4carrier status within a composite region across the areas susceptible to aging. We used linear models and sparse canonical correlation analysis (SCCA) to examine the relationship between K trans and AD biomarkers. Results OA showed greater BBBp than YA predominately in the temporal lobe, with some involvement of parietal, occipital and frontal lobes. Within an averaged ROI of affected regions, there was no difference in K trans values based on sex or Aβ positivity, but OA who were APOE4carriers had significantly higher K trans values. There was no direct relationship between averaged K trans and global Aβ pathology, but there was a trend for an Ab status by tau interaction on K trans in this region. SCCA showed increased K trans was associated with increased PiB DVR, mainly in temporal and parietal brain regions. There was not a significant relationship between K trans and FTP SUVR. Discussion Our findings indicate that the BBB shows regional vulnerability during normal aging that overlaps considerably with the pattern of AD pathology. Greater BBBp in brain regions affected in aging is related to APOE genotype and may also be related to the pathological accumulation of Aβ.

Wastewater Recycling Using a Hygroscopic Cooling System

This project by the Energy & Environmental Research Center (EERC), Baltimore Aircoil Company (BAC), and Great River Energy (GRE) evaluated the concept of recycling wastewater at a coal-fired power plant using a hygroscopic cooling system, which is an evaporative cooling technology analogous to conventional cooling towers, except that sparingly soluble dissolved solids are precipitated and removed as waste solids instead of purging them with a liquid blowdown stream. This technology can maximize the use of plant makeup water by obtaining useful evaporative cooling from wastewater while minimizing the volume of wastes needing disposal. Experimental activities were conducted in two phases, a laboratory-based evaluation of the properties of wastewater from the host site, GRE’s Coal Creek Station near Underwood, North Dakota, and a field test of a small pilot hygroscopic cooling system at the host site power plant. Findings from the laboratory study informed the design of the pilot system and the system’s field test performance served as the basis for a techno-economic analysis (TEA) of the hygroscopic recycling concept. At the preferred operating conditions identified during the TEA, the wet-bulb approach temperature of the tower was 7.3°C (13°F) and the volume of blowdown produced by the plant was reduced to 5.4% of its incoming volume. Waste solids produced during field testing were classified as nonhazardous waste based on the measured hazardous element content and evaluation of their leaching potential. However, to qualify as a solid for landfill disposal i.e., as determined by the U.S. Environmental Protection Agency’s paint filter test, it appears that a dewatering step beyond hydrocyclone separation is needed. The baseline levelized cost of wastewater disposal (LCWD) for hygroscopic wastewater recycling was estimated to be $\$ $3.69–$\$ $3.72 per m3 of plant blowdown. Capital cost was estimated to contribute over 54% to the LCWD, and parameters that impact capital cost such as the heat exchange coil material of construction and the tower’s wet-bulb approach temperature were identified as having the greatest impact on overall LCWD. A LCWD estimate prepared for the same application but using thermomechanical brine evaporation was almost 40% higher than that calculated for hygroscopic cooling, despite recovering distilled-quality water for reuse, while the LCWD for disposal-only, deep well injection was estimated to be 30% lower compared to hygroscopic wastewater recycling.

Business Case Analysis for Artificial Intelligence-Large Language Model Technology Integration

AI-assisted processes are expected to enhance operational efficiency and improve decision-making, supporting the long-term economic viability of nuclear power plants. However, detailed business analyses of AI-generated cost savings are rarely performed. Given the recent industry interest in Large Language Model (LLM), the U.S. Department of Energy (DOE) Light Water Reactor Sustainability (LWRS) Program has conducted a comprehensive business case analysis of LLM Artificial Intelligence (AI) implementation in nuclear plant engineering workflows. The research employed three complementary business case approaches to evaluate impact of an LLM, using three representative engineering processes as use-cases: Boric Acid Corrosion (BAC) Evaluations, Maintenance Rule Evaluations, and 10 CFR 50.59 Screenings. Through detailed workload analyses and structured interviews, the study quantified significant efficiency improvements ranging from 11% to 59% across these processes. The research further considers how these efficiency gains could translate into tangible reliability improvements through enhanced engineering capacity. Analysis of historical plant trip data indicates that enabling engineers to focus on proactive reliability activities could provide substantial financial benefits through avoided outages, potentially generating greater value than the direct efficiency improvements alone. By documenting successful applications, implementation challenges, and strategic opportunities, this research provides nuclear utilities with a practical framework for evaluating the value of AI technology to support long-term operations through advanced digital technologies.

Flight flutter testing of multi-jet aircraft

Extensive flight flutter tests were conducted by BAC on B-52 and KC-135 prototype airplanes. The need for and importance of these flight flutter programs to Boeing airplane design are discussed. Basic concepts of flight flutter testing of multi-jet aircraft and analysis of the test data will be presented. Exciter equipment and instrumentation employed in these tests will be discussed.

Environmental exposure effects on composite materials for commercial aircraft

Activities reported include completion of the program design tasks, resolution of a high fiber volume problem and resumption of specimen fabrication, fixture fabrication, and progress on the analysis methodology and definition of the typical aircraft environment. Program design activities including test specimens, specimen holding fixtures, flap-track fairing tailcones, and ground exposure racks were completed. The problem experienced in obtaining acceptable fiber volume fraction results on two of the selected graphite epoxy material systems was resolved with an alteration to the bagging procedure called out in BAC 5562. The revised bagging procedure, involving lower numbers of bleeder plies, produces acceptable results. All required laminates for the contract have now been laid up and cured. Progress in the area of analysis methodology has been centered about definition of the environment that a commercial transport aircraft undergoes. The selected methodology is analagous to fatigue life assessment.

Application of parametric weight and cost estimating relationships to future transport aircraft

A model comprised of system level weight and cost estimating relationships for transport aircraft is presented. In order to determine the production cost of future aircraft its weight is first estimated based on performance parameters, and then the cost is estimated as a function of weight. For initial evaluation CERs were applied to actual system weights of six aircraft (3 military and 3 commercial) with mean empty weights ranging from 30,000 to 300,000 lb. The resulting cost estimates were compared with actual costs. The average absolute error was only 4.3%. Then the model was applied to five aircraft still in the design phase (Boeing 757, 767 and 777, and BAC HS146-100 and HS146-200). While the estimates for the 757 and 767 are within 2 to 3 percent of their assumed break-even costs, it is recognized that these are very sensitive to the validity of the estimated weights, inflation factor, the amount assumed for nonrecurring costs, etc., and it is suggested that the model may be used in conjunction with other information such as RDT&E cost estimates and market forecasts. The model will help NASA evaluate new technologies and production costs of future aircraft.

IUE low-dispersion spectra of four luminous stars in symmetric nebulae

The stars and nebulae are HD 56925 in NGC 2359, + 60 deg 2522 in NGC 7635, AG Car in its nebula, and 209 BAC in M1-67. A discussion of known properties of these systems precedes the IUE data, which are tabulated with types and identifications of significant line features and velocities of ultraviolet-displaced absorption features. Weaver et al.'s theory of the interaction of a stellar wind with the ambient interstellar medium is applied to the combined observational data. This gives a table of self-consistent values for stellar terminal wind velocity, rate of mass loss, and wind power; nebular mass, radius, expansion velocity, and age; also ambient interstellar density and mass swept up from the interstellar medium by the wind. The relatively infrequent occurrence of such symmetric nebulae around young, massive stars is possibly related to the short lifetimes of the nebulae in comparison with stellar evolution lifetimes.

IUE low-dispersion spectra of six luminous stars in symmetric nebulae

The stars and nebulae are HD 156738 and HDE 319703A, respectively centered in a pair of symmetric nebulae among the NGC 6334 group, AG Car in its nebula, HDE 250550 in nebula 8 of a catalog by Herbig, 209 BAC in Ml-67, and HD 89358 in NGC 3199. These include two O stars, two WN stars, an unstable B supergiant, and a ZAMS B star. Four of them are additions to a previous similar study, and the information about AG CAR and 209 BAC/Ml-67 is extended from that study. The objects are interpreted with Weaver et al.'s (1977) theory of the interaction of a stellar wind with the ambient interstellar medium, except where the short lifetime of the HDE 250550 nebula has forestalled such analysis. Spectral line identifications and types, and several parameters of mass loss, are tabulated. When the present mass loss rates are compared with previous results from other methods, there is an outstanding difference only for WN stars, since 1-3 x 10 to the -7th solar masses/year is derived here.

Effects of joints in truss structures

The response of truss-type structures for future space applications, such as Large Deployable Reflector (LDR), will be directly affected by joint performance. Some of the objectives of research at BAC were to characterize structural joints, establish analytical approaches that incorporate joint characteristics, and experimentally establish the validity of the analytical approaches. The test approach to characterize joints for both erectable and deployable-type structures was based upon a Force State Mapping Technique. The approach pictorially shows how the nonlinear joint results can be used for equivalent linear analysis. Testing of the Space Station joints developed at LaRC (a hinged joint at 2 Hz and a clevis joint at 2 Hz) successfully revealed the nonlinear characteristics of the joints. The Space Station joints were effectively linear when loaded to plus or minus 500 pounds with a corresponding displacement of about plus or minus 0.0015 inch. It was indicated that good linear joints exist which are compatible with errected structures, but that difficulty may be encountered if nonlinear-type joints are incorporated in the structure.

CN radical reactions with hydrogen cyanide and cyanogen - Comparison of theory and experiment

The method of laser photolysis/laser-induced fluorescence is used to obtain absolute rate constants for CN radical reactions with HCN and C2N2. The rate constants were found to be temperature-dependent in the range 300-740 K and pressure independent in the range 100-600 Torr. Rice-Remsperger-Kassel-Marcus theory for both reactions employing the transition state parameters obtained by the BAC-MP4 method are made. These calculations yielded reasonable results for the CN + HCN reaction, predicting both the temperature dependence and pressure independence. No pressure effect was observed in the pressure range 100-1000 Torr at temperatures below 900 K, confirming the experimental results.

Theoretical characterization of the potential energy surface for NH + NO

The potential energy surface (PES) for NH + NO was characterized using complete active space self-consistent field (CASSCF) gradient calculations to determine the stationary point geometries and frequencies followed by CASSCF/internally contracted configuration interaction (CCI) calculations to refine the energetics. The present results are in qualitative accord with the BAC-MP4 calculations, but there are differences as large as 8 kcal/mol in the detailed energetics. Addition of NH to NO on a (2)A' surface, which correlated with N2 + OH or H + N2O products, involves barriers of 3.2 kcal/mol (trans) and 6.3 kcal/mol (cis). Experimental evidence for these barriers is found in earlier works. The (2)A' surface has no barrier to addition, but does not correlate with products. Surface crossings between the barrierless (2)A' surface and the (2)A' surface may be important. Production of N2 + OH products is predicted to occur via a planar saddle point of (2)A' symmetry. This is in accord with the preferential formation of II(A') lambda doublet levels of OH in earlier experiments. Addition of NH (1)delta to NO is found to occur on an excited state surface and is predicted to lead to N2O product as observed in earlier works.

Anomalous Balmer continuum temperatures in the Orion Nebula

New long-slit spectra of the Orion Nebula in the near-ultraviolet were used to calculate the Balmer recombination temperature, T(Bac), from the Balmer discontinuity at 3646 A. The spatially resolved data show a decrease in temperature moving to the west of Theta(sup 1) Ori C, from 8400 K at a distance of 40 sec to a low of 2800 K at a distance of 220 sec. Such values are much lower than previously reported. The effect of scattered starlight on these results is calculated and shown to be less than 10%. Previous studies which found scattered light to be important at the discontinuity are in error. Such low temperatures and their impact on nebular physics and abundances are disconcerting and require further study.

Control Characteristics of Alcohol-Impaired Operators

Although the operation of vehicles like airplanes, cars, and bicycles involves a complex array of perceptual, decision and control activities, most accident statistics clearly show that intoxicated operators are a dominant cause of accidents, and not the difficulty of the task itself. This paper summarizes some recent research on the nature of the impairment of operator control under blood alcohol concentrations (BAC) up to above 0.16 percent. Alcohol toxicity is shown to be quite specific with respect to visual-motor functions involved in control of a vehicle, and experiments with a generalized workload task and special driving simulator show how these are reflected in terms of changes in operator control parameters such as response latency, gains, stability margins, and coherency.

Meeting NASA's Mission Through Commercial Partnerships

This paper examines novel approaches to furthering NASA's missions through the use of commercial partnerships. The exploration of space ha proven to be a costly endeavor requiring the development of new technologies at significant expense. One of the prime factors holding bac the robust development of space is insufficient investment in the technologies necessary to make it a reality. The key to success in bringin needed space development technologies to maturation lies in bringing technology investors together from government, industry and academia. aggressive road map for developing space will require a diverse set of interest to industry or other government agencies. By having each invest( contributing to the part of the technology development of interest to them development of space systems can be put together at a cost far below wl would be required to develop for a stand-alone effort. The NASA Space Partnership Division has been employing this technique to leverage a 30 million dollar NASA investment into at 100 million dollar advanced technology development effort focused on meeting NASA's mission needs.

A provisional regulatory gene network for specification of endomesoderm in the sea urchin embryo

We present the current form of a provisional DNA sequence-based regulatory gene network that explains in outline how endomesodermal specification in the sea urchin embryo is controlled. The model of the network is in a continuous process of revision and growth as new genes are added and new experimental results become available; see http://www.its.caltech.edu/~mirsky/endomeso.htm (End-mes Gene Network Update) for the latest version. The network contains over 40 genes at present, many newly uncovered in the course of this work, and most encoding DNA-binding transcriptional regulatory factors. The architecture of the network was approached initially by construction of a logic model that integrated the extensive experimental evidence now available on endomesoderm specification. The internal linkages between genes in the network have been determined functionally, by measurement of the effects of regulatory perturbations on the expression of all relevant genes in the network. Five kinds of perturbation have been applied: (1) use of morpholino antisense oligonucleotides targeted to many of the key regulatory genes in the network; (2) transformation of other regulatory factors into dominant repressors by construction of Engrailed repressor domain fusions; (3) ectopic expression of given regulatory factors, from genetic expression constructs and from injected mRNAs; (4) blockade of the beta-catenin/Tcf pathway by introduction of mRNA encoding the intracellular domain of cadherin; and (5) blockade of the Notch signaling pathway by introduction of mRNA encoding the extracellular domain of the Notch receptor. The network model predicts the cis-regulatory inputs that link each gene into the network. Therefore, its architecture is testable by cis-regulatory analysis. Strongylocentrotus purpuratus and Lytechinus variegatus genomic BAC recombinants that include a large number of the genes in the network have been sequenced and annotated. Tests of the cis-regulatory predictions of the model are greatly facilitated by interspecific computational sequence comparison, which affords a rapid identification of likely cis-regulatory elements in advance of experimental analysis. The network specifies genomically encoded regulatory processes between early cleavage and gastrula stages. These control the specification of the micromere lineage and of the initial veg(2) endomesodermal domain; the blastula-stage separation of the central veg(2) mesodermal domain (i.e., the secondary mesenchyme progenitor field) from the peripheral veg(2) endodermal domain; the stabilization of specification state within these domains; and activation of some downstream differentiation genes. Each of the temporal-spatial phases of specification is represented in a subelement of the network model, that treats regulatory events within the relevant embryonic nuclei at particular stages. Cryogenic Fluid Storage Technology Development: Recent and Planned Efforts at NASA

Recent technology development work conducted at NASA in the area of Cryogenic Fluid Management (CFM) storage is highlighted, including summary results, key impacts, and ongoing efforts. Thermodynamic vent system (TVS) ground test results are shown for hydrogen, methane, and oxygen. Joule-Thomson (J-T) device tests related to clogging in hydrogen are summarized, along with the absence of clogging in oxygen and methane tests. Confirmation of analytical relations and bonding techniques for broad area cooling (BAC) concepts based on tube-to-tank tests are presented. Results of two-phase lumped-parameter computational fluid dynamic (CFD) models are highlighted, including validation of the model with hydrogen self pressurization test data. These models were used to simulate Altair representative methane and oxygen tanks subjected to 210 days of lunar surface storage. Engineering analysis tools being developed to support system level trades and vehicle propulsion system designs are also cited. Finally, prioritized technology development risks identified for Constellation cryogenic propulsion systems are presented, and future efforts to address those risks are discussed.

Introduction of structural affinity handles as a tool in selective nucleic acid separations

The method is used for separating nucleic acids and other similar constructs. It involves selective introduction, enhancement, or stabilization of affinity handles such as single-strandedness in the undesired (or desired) nucleic acids as compared to the usual structure (e.g., double-strandedness) of the desired (or undesired) nucleic acids. The undesired (or desired) nucleic acids are separated from the desired (or undesired) nucleic acids due to capture by methods including but not limited to immobilized metal affinity chromatography, immobilized single-stranded DNA binding (SSB) protein, and immobilized oligonucleotides. The invention is useful to: remove contaminating genomic DNA from plasmid DNA; remove genomic DNA from plasmids, BACs, and similar constructs; selectively separate oligonucleotides and similar DNA fragments from their partner strands; purification of aptamers, (deoxy)-ribozymes and other highly structured nucleic acids; Separation of restriction fragments without using agarose gels; manufacture recombinant Taq polymerase or similar products that are sensitive to host genomic DNA contamination; and other applications.
