Understanding local plant extinctions before it is too late: bridging evolutionary genomics with global ecology

Understanding evolutionary genomic and population processes within a species range is key to anticipating the extinction of plant species before it is too late. However, most models of biodiversity risk under global change do not account for the genetic variation and local adaptation of different populations. Population diversity is critical to understanding extinction because different populations may be more or less susceptible to global change and, if lost, would reduce the total diversity within a species. Here, two new modeling frameworks advance our understanding of extinction from a population and evolutionary angle: Rapid climate change-driven disruptions in population adaptation are predicted from associations between genomes and local climates. Furthermore, losses of population diversity from global land-use transformations are estimated by scaling relationships of species' genomic diversity with habitat area. Overall, these global eco-evolutionary methods advance the predictability – and possibly the preventability – of the ongoing extinction of plant species.

Nanoscale chromatin imaging and analysis platform bridges 4D chromatin organization with molecular function

Extending across multiple length scales, dynamic chromatin structure is linked to transcription through the regulation of genome organization. However, no individual technique can fully elucidate this structure and its relation to molecular function at all length and time scales at both a single-cell level and a population level. Here, we present a multitechnique nanoscale chromatin imaging and analysis (nano-ChIA) platform that consolidates electron tomography of the primary chromatin fiber, optical super-resolution imaging of transcription processes, and label-free nano-sensing of chromatin packing and its dynamics in live cells. Using nano-ChIA, we observed that chromatin is localized into spatially separable packing domains, with an average diameter of around 200 nanometers, sub-megabase genomic size, and an internal fractal structure. The chromatin packing behavior of these domains exhibits a complex bidirectional relationship with active gene transcription. Furthermore, we found that properties of PDs are correlated among progenitor and progeny cells across cell division.

A Mineral-Doped Micromodel Platform Demonstrates Fungal Bridging of Carbon Hot Spots and Hyphal Transport of Mineral-Derived Nutrients

Fungal species are foundational members of soil microbiomes, where their contributions in accessing and transporting vital nutrients is key for community resilience. To date, the molecular mechanisms underlying fungal mineral weathering and nutrient translocation in low-nutrient environments remain poorly resolved due to the lack of a platform for spatial analysis of biotic weathering processes.

From CAN to ROS: A Monitoring and Data Recording Bridge

The Controller Area Network (CAN) bus protocol is used in modern vehicles for sharing messages between several control units within a vehicle. CAN bus messages are encoded with unknown scheme and decoding these messages provide unlimited access to valuable information that is used in many autonomous vehicles applications. This paper proposes a ROS based package (CAN-to-ROS) for monitoring, recording, and real-time and offline decoding of CAN bus messages. The package is developed in the ROS framework to add modularity and ease of integration with other software, and it is written in C++ to guarantee speed of the execution during run-time. For realtime decoding of CAN bus data, CAN-to-ROS package used in conjunction with other library called Libpanda that provide access to CAN bus message from a vehicle. The package was evaluated and tested on a Raspberry Pi with real CAN bus data from a Toyota RAV4. The results confirm the capabilities of CAN-to-ROS package and resulted in using the package in other research projects.

Bridging the Gap: User-Centric Energy Monitoring for Policy-Driven Application Optimization in HPC Data Centers

Application energy optimization in HPC data centers face two critical gaps. Systematic methodologies that connect data center policies to application decisions and accessible monitoring tools that enable data-driven optimization. We address both gaps through two complementary pillars. First, we present a methodology based on extended weighted Energy Delay Product (EDP) to translate data center operational priorities and integrate energy considerations into the energy optimization workflow which starts from continuous monitoring through targeted optimization. Second, we present a user-space monitoring tool, Omnistat, that enables this methodology by providing developers with direct access to actionable energy telemetry. Through deployment on the Frontier supercomputer and case studies exploring performance-energy trade-offs, we show how these pillars help energy as an integral optimization target for developers as active participants in data center efficiency.

Solid-Binding Proteins: Bridging Synthesis, Assembly, and Function in Hybrid and Hierarchical Materials Fabrication

There is considerable interest in the development of hybrid organic–inorganic materials because of the potential for harvesting the unique capabilities that each system has to offer. Proteins are an especially attractive organic component owing to the high amount of chemical information encoded in their amino acid sequence, their amenability to molecular and computational (re)design, and the many structures and functions they specify. Genetic installation of solid-binding peptides (SBPs) within protein frameworks affords control over the position and orientation of adhesive and morphogenetic segments, and a path toward predictive synthesis and assembly of functional materials and devices, all while harnessing the built-in properties of the host scaffold. Furthermore, we review the current understanding of the mechanisms through which SBPs bind to technologically relevant interfaces, with an emphasis on the variables that influence the process, and highlight the last decade of progress in the use of solid-binding proteins for hybrid and hierarchical materials synthesis.

Bridging New Observational Capabilities and Process-Level Simulation: Insights into Aerosol Roles in the Earth System

The spatial distribution of ambient aerosol particles significantly impacts aerosol–radiation–cloud interactions, which contribute to the largest uncertainty in global anthropogenic radiative forcing estimations. However, the atmospheric boundary layer and lower free troposphere have not been adequately sampled in terms of spatiotemporal resolution, hindering a comprehensive characterization of various atmospheric processes and impeding our understanding of the Earth system. To address this research data gap, we have leveraged the development of uncrewed aerial systems (UAS) and advanced measurement techniques to obtain mesoscale spatial data on aerosol microphysical and optical properties around the U.S. Southern Great Plains (SGP) atmospheric observatory. Our study also benefits from state-of-the-art laboratory facilities that include three-dimensional molecular imaging techniques enabled by secondary ion mass spectrometry and nanogram-level chemical composition analysis via micronebulization aerosol mass spectrometry. Through our study, we have developed a framework for observation–modeling integration, enabling an examination of how various assumptions about the organic–inorganic components mixing state, inferred from chemical analysis, affect clouds and radiation in observation-constrained model simulations. By integrating observational constraints (derived from offline chemical analysis of the aerosol surface using collected samples) with in situ UAS observations, we have identified a prominent role of organic-enriched nanometer layers located at the surface of aerosol particles in determining profiles of aerosol optical and hygroscopic properties over the SGP observatory. Furthermore, we have improved the agreement between predicted clouds and ground-based cloud lidar measurements. This UAS–model–laboratory integration exemplifies how these new advanced capabilities can significantly enhance our understanding of aerosol–radiation–cloud interactions.

Feature issue introduction: laser driven inertial confinement fusion and bridging the gaps to inertial fusion energy systems

Major fusion research milestones have been achieved using laser driven inertial confinement fusion (ICF) in recent years, and these successes have ignited tremendous enthusiasm for inertial fusion energy (IFE). However, the complexity and difficulty of obtaining fusion ignition with a laser driver in a research setting are often underappreciated, as are the gaps to high driver efficiency, high repetition rates, and laser and target durability requirements needs for IFE. On the academic side, several new research laser systems have been constructed over the past few years, enabling researchers to probe the limits of ICF physics and engineering. This feature issue highlights the challenges and capabilities of laser research and development targeted towards advancing IFE.

Air Evacuation Bridge

Researchers at Los Alamos National Laboratory are researching and designing technology intended to help reduce the exposure of patients and medical staff to pathogens, such as SARS-CoV-2 while providing medical staff the dexterity and flexible work conditions necessary to conduct routine and complex patient care on infected patients. This investigative technology would potentially enable hospitals to adapt, utilize, and re-purpose existing and otherwise infrequently used equipment in the fight against COVID-19, thereby saving hospitals significant money, reducing exposure to staff and patients, and increasing the supply of critically needed devices at a time when supplies are scarce in the market place. A provisional patent has been filed on the technology and we are seeking a commercialization partner to license and further develop the product for commercial purposes, including processing the device through the FDA authorization or clearance process. Once FDA has cleared the product, at least one hospital system has expressed interest in the experimental test components for use in their hospitals.

Bridging Land-Surface Fluxes and Aerosol Concentrations to Triggering Convective Rainfall

During the period August 24-28, 2016, a delayed component the GO-Amazon Project, we launched nine altitude-controlled free balloons (CMET). Smaller than typical rawinsondes, CMET are equipped with altitude control, global communication via Iridium satellite, and aspirated sensors. The aims of our effort were to examine the interactions among convective boundary layer and dual river breeze circulations near the confluence of the Tapajos and Amazon Rivers in the eastern Basin. The week-long field campaign was timed to examine the reestablishment of the breeze circulations shortly after the passage of a strong instability line on August 22. Nine CMET were launched at the Curua-Una hydroelectric dam (2.8S; 54.3W), timed to encounter the Tapajos river breeze front by late afternoon. Soundings were made to establish the thickness of interface between the easterly trade and westerly Tapajos breeze circulation. Careful use of sounding strategies allowed these free balloons to track along the northerly channeled flow in the lowest 300 m above the River. Following the river encounter, balloons tracked to the west, sounding to describe the diurnal course of boundary layer in the forest west of the Tapajos River. The longest flight traveled more than 770 km over three days and twice rested overnight in the rain forest canopy. Ancillary data from surface climate and flux stations as well as the Santarem radiosonde, satellite images will be used to illustrate how the breeze circulations are seen near the surface and how they were disrupted by larger-scale events. Comparisons with HYSPLIT trajectories illustrate how sensitive real trajectories are to the refraction that the encounter with the breeze effects. A second effort was intensive analyses of data from the eddy flux tower at the Tapajós National Forest near Santarém Pará Brazil, with special emphasis on the role of clouds and canopy presence on light absorbance and photosynthetic activity.
