Conformational Dynamics and Catalytic Backups in a Hyper-thermostable Engineered Archaeal Protein Tyrosine Phosphatase

Protein tyrosine phosphatases (PTPs) are a family of enzymes that play important roles in regulating cellular signaling pathways. The activity of these enzymes is regulated by the motion of a catalytic loop that places a critical conserved aspartic acid side chain into the active site for acid–base catalysis upon loop closure. These enzymes also have a conserved phosphate-binding loop that is typically highly rigid and forms a well-defined anion-binding nest. The intimate links between loop dynamics and chemistry in these enzymes make PTPs an excellent model system for understanding the role of loop dynamics in protein function and evolution. In this context, archaeal PTPs, which have often evolved in extremophilic organisms, are highly understudied, despite their unusual biophysical properties. We present here an engineered chimeric PTP (ShufPTP) generated by shuffling the amino acid sequence of five extant hyperthermophilic archaeal PTPs. Despite ShufPTP’s high sequence similarity to its natural counterparts, it presents a suite of unique properties, including high flexibility of the phosphate binding P-loop, facile oxidation of the active-site cysteine, mechanistic promiscuity, and, most notably, hyperthermostability, with a denaturation temperature likely >130 °C (>8 °C higher than the highest recorded growth temperature of any archaeal strain). Our combined structural, biochemical, biophysical, and computational analysis provides insight both into how small steps in evolutionary space can radically modulate the biophysical properties of an enzyme and showcases the tremendous potential of archaeal enzymes for biotechnology, to generate novel enzymes capable of operating under extreme conditions.

Conformal collider physics meets LHC data

The remarkably high energies of the Large Hadron Collider (LHC) have allowed for the first measurements of the shapes and scalings of multipoint correlators of energy flow operators, ⟨ Ψ | E ( n → 1 ) E ( n → 2 ) ⋯ E ( n → k ) | Ψ ⟩ , providing new insights into the Lorentzian dynamics of quantum chromodynamics (QCD). In this letter, we use recent advances in effective field theory to derive a rigorous factorization theorem for the light-ray density matrix, ρ = | Ψ ⟩ ⟨ Ψ | , inside high transverse momentum jets at the LHC. Using the light-ray operator product expansion, the scaling behavior of multipoint correlators can be computed from the expectation value of the twist-2 spin- J light-ray operators, O [ J ] , in this state, Tr [ ρ O [ J ] ] . We compute the light-ray density matrix at next-to-leading order, and combine this with results for the next-to-leading logarithmic scaling behavior of the correlators up to six-points, comparing with CMS open data. This theoretical accuracy allows us to resolve the quantum scaling dimensions of QCD light-ray operators inside jets at the LHC. Our factorization theorem for the light-ray density matrix at the LHC completes the link between recent developments in the study of energy correlators and LHC phenomenology, opening the door to a wide variety of precision jet substructure studies. Quark Matter at High Baryon Density, Conformality and Quarkyonic Matter

This paper discusses high-baryon-density quarkyonic matter in the context of recent observations concerning neutron stars and the qualitative reasons why quarkyonic matter explains certain features of the equation of state that arises from these observations. The paper then provides a qualitative discussion of the quarkyonic hypotheses, and the essential features of quarkyonic matter that explain the outstanding features of the equation of state.
