Genomic DNA was isolated from soil and rhizosphere samples using the Zymo Quick-DNA fecal/soil microbe miniprep kit (catalog no. D6010) according to the manufacturer’s instructions (Zymo Research; Irvine, CA) with the modification of eluting in 100 uL elution buffer. Sample concentrations were quantified using the Qubit dsDNA HS assay kit (Thermo Fisher). For rhizosphere samples only, DNA was subsequently purified using Zymo’s ZR-96 DNA Clean & Concentrator kit (catalog no. D4024) to account for low DNA concentrations of these samples. . In each replicate block, there were five drip irrigation treatments (T1 = 100% normal irrigation, T2 = 56.25%, T3 = 37.5%, T4 = 18.75% and T5=no irrigation. On July 20, the strength of the drought treatments was increased: T1 remained at 100%, whereas T2 changed from 75% to 56.25%, T2 changed from 50% to 37.5%, and T4 changed from 25% to 18.75%. Sequencing was performed as described previously (Naylor, Fansler, et al. 2020). Sequences were amplified on the MiSeq platform (Illumina, San Diego, CA) using 16S primers (515F and 806R) specific to the V4 region. Raw sequence data was processed with the pipeline Hundo for amplicon quality control and annotation. Downstream statistical analyses on 16S datasets were performed using the program R and the packages ‘phyloseq’ and ‘vegan’.
Agrivoltaics integrates agricultural production with solar energy generation to address challenges related to land use, food security, and renewable energy development. This study provides the most comprehensive evaluation to date of global agrivoltaic research, aiming to classify the literature, identify strengths and gaps, and guide future work. We systematically screened over 3000 English-language publications through 2023 for relevant agrivoltaic publications. A total of 670 studies were categorized in the InSPIRE Data Portal across five agrivoltaic activities and multiple hierarchical themes, including physical, biological, technological, social, and crosscutting domains. We found that research was concentrated on crop production, microclimate dynamics, and PV performance, with gaps in areas like human health, wildlife, policy, and standardized methodologies. Although the U.S. emphasizes animal grazing and habitat-based systems in practice, most U.S.-based studies focused disproportionately on crop production. The analysis revealed uneven geographic and topical representation and highlighted a lack of integrated, interdisciplinary approaches. This study concludes that while agrivoltaic research has grown rapidly, more coordinated efforts could support standardized data collection, address overlooked ecological and social impacts, and align research focus with real-world system implementation, ultimately improving the scalability and successful deployment of agrivoltaic systems.
Genome-scale metabolic models (GEMs) enable the mathematical simulation of the metabolism of archaea, bacteria, and eukaryotic organisms. GEMs quantitatively define a relationship between genotype and phenotype by contextualizing different types of Big Data (e.g., genomics, metabolomics, and transcriptomics). In this review, we analyze the available Big Data useful for metabolic modeling and compile the available GEM reconstruction tools that integrate Big Data. We also discuss recent applications in industry and research that include predicting phenotypes, elucidating metabolic pathways, producing industry-relevant chemicals, identifying drug targets, and generating knowledge to better understand host-associated diseases. In addition to the up-to-date review of GEMs currently available, we assessed a plethora of tools for developing new GEMs that include macromolecular expression and dynamic resolution. Finally, we provide a perspective in emerging areas, such as annotation, data managing, and machine learning, in which GEMs will play a key role in the further utilization of Big Data.

Boron isotope systematics of planktonic foraminifera from core-top sediments and culture experiments have been studied to investigate the sensitivity ofδ11B of calcite tests to seawater pH. However, our knowledge of the relationship between δ11B and pH remains incomplete for many taxa. Thus, to expand the potential scope of application of this proxy, we report δ11B data for seven different species of planktonic foraminifera from sediment core tops. We utilize a method for the measurement of small samples of foraminifera and calculate the δ11B-calcite sensitivity to pH for Globigerinoides ruber, Trilobus sacculifer (sacc or without sacc), Orbulina universa, Pulleniatina obliquiloculata, Neogloboquadrina dutertrei, Globorotalia menardii, and Globorotalia tumida, including for unstudied core tops and species. These taxa have diverse ecological preferences and are from sites that span a range of oceanographic regimes, including some that are in regions of air–sea equilibrium and others that are out of equilibrium with the atmosphere. The sensitivity of δ11Bcarbonate to δ11Bborate (e.g., Δδ11Bcarbonate/Δδ11Bborate) in core tops is consistent with previous studies for T. sacculifer and G. ruber and close to unity for N. dutertrei, O. universa, and combined deep-dwelling species. Deep-dwelling species closely follow the core-top calibration fo rO. universa, which is attributed to respiration-driven microenvironments likely caused by light limitation and/or symbiont–host interactions. Our data support the premise that utilizing boron isotope measurements of multiple species within a sediment core can be utilized to constrain vertical profiles of pH andpCO2 at sites spanning different oceanic regimes, thereby constraining changes in vertical pH gradients and yielding insights into the past behavior of the oceanic carbon pumps.

The speciation of iodine in basalts has been investigated by combining in situ X-ray diffraction at high pressures and temperatures up to 4.9 GPa and 1600 °C, and Raman spectroscopy on recovered high pressure glasses at ambient conditions. Both methods point to iodine being oxidized in basalts, whether molten or quenched as glasses. Observed interatomic distances and Raman vibrational modes are consistent with iodine being dissolved as complex iodate groups alike polyiodates or periodates, not as IO$^-_3$ groups. Iodine speciation in basalts therefore seems to reflect a trend amongst halogens, with lighter chlorine bonding to network modifying cations, and bromine changing affinity from network modifying cations to oxygen anions under pressure. In the absence of a fluid aqueous phase, iodine could thus reach the Earth’s surface in basaltic magmas as an oxide, not as a reduced species.
The Molokai, Hawaii, Energy to Communities (E2C) Energyshed project represents a collaborative effort between the National Laboratory of the Rockies, Shake Energy Collaborative, the Molokai Clean Energy Hui, Sustainable Molokai, and Ho'ahu Energy Cooperative Molokai to advance Molokai's Community Energy Resilience Action Plan (CERAP). Supported by Hawaiian Electric Company and the Hawaii State Energy Office, the initiative aims to develop a community-defined portfolio of renewable energy solutions that enhance energy resilience while aligning with the Hawaiian Electric Integrated Grid Plan (IGP) and Molokai's energy goals. Phase 1 focused on technical analyses and community engagement to co-design feasible energy scenarios. Challenges such as grid upgrades, storage sizing, and inverter ride-through standards were addressed to align technical and operational requirements with community preferences. The project equips Molokai with actionable data and insights to implement energy initiatives while ensuring resilient and culturally informed solutions. Future efforts aim to finalize project designs, secure interconnection agreements, and deploy energy projects that reflect community priorities and technical feasibility.
This presentation contains results from work on an active NNSS Site-directed research and development (SDRD) project: NLV-019-20. It describes a new algorithm for "few-angle tomography", which can be used to reconstruct an approximation to an object from limited quantities or radiographs under specific conditions. This presentation is an initial documentation of research results thus-far, and we would like to share these methods with colleagues at Lawrence Livermore National Lab who have an expertise in the area of few-angle tomography. Sharing this presentation will be done via email and or teleconference amongst DoE personnel. In the future, we would also like to share these results with collaborators from Academia as well.
A method for measuring conductance of a material real-time during etching/milling includes providing a fixture having a socket for receiving the material. The socket is attached to a printed circuit board (PCB) mounted on one side of a plate that has at least one opening for providing ion beam access to the material sample. Conductive probes extend from the other side of the PCB to contact and span a target area of the material. A measurement circuit in electrical communication with the probes measures the voltage produced when a current is applied across the material sample to measure changes in electrical properties of the sample over time.
A multi electrode sensor that provides in-situ, real time measurements for molten salts and other process fluids such as real-time concentration and salt level measurements for nuclear systems such as molten salt reactors, nuclear reprocessing facilities utilizing molten salts and concentrated solar power systems. The sensor has multiple electrodes with unique lengths which are connected to a potentiostat. Measurements are taken when the electrodes are immersed in the process fluid.

Mass quantification using standoff gamma detection is a logical progression for MC&A applications in uranium processing facilities. While obtaining a ground truth from physical collection and weighing of masses is ideal and routine, it poses a logistical problem from both a standpoint of ALARA programs as well as interruptions to operations. The standard approach is the Generalized Geometry in Holdup (GGH) method that can apply for many cases where geometries of the holdup match simulated models; however, there are many cases where a model may not exist or a feature of the holdup geometry is such that it does not match a model particularly well.1 Gamma imaging at standoff distances and outside process equipment has potential to address these shortcomings and improve mass estimations while reducing personnel exposure; however, algorithms still require a significant degree of benchmarking to build sufficient confidence in algorithms before displacing GGH in some procedures. This paper presents the creation of a set of laboratory standards of depleted U3O8 in fixed size containers covering a range of masses and effective thicknesses up to the infinite thickness for the 185-keV line. These blocks are identically sized aluminum boxes loaded such that a thin, flat face is placed towards the imager, while the back side contains closed cell foam to apply pressure to keep the powder in a fixed position at the front face as the sources are manipulated for measurements. Included is an analysis of the mass estimation using a commercially available Compton imaging instrument where multiple geometries are created showing limitations in current methodologies.
