Insights into the action of phylogenetically diverse microbial expansins on the structure of cellulose microfibrils

Microbial expansins (EXLXs) are non-lytic proteins homologous to plant expansins involved in plant cell wall formation. Due to their non-lytic cell wall loosening properties and potential to disaggregate cellulosic structures, there is considerable interest in exploring the ability of microbial expansins (EXLX) to assist the processing of cellulosic biomass for broader biotechnological applications. Herein, EXLXs with different modular structure and from diverse phylogenetic origin were compared in terms of ability to bind cellulosic, xylosic, and chitinous substrates, to structurally modify cellulosic fibrils, and to boost enzymatic deconstruction of hardwood pulp. Five heterogeneously produced EXLXs (Clavibacter michiganensis; CmiEXLX2, Dickeya aquatica; DaqEXLX1, Xanthomonas sacchari; XsaEXLX1, Nothophytophthora sp.; NspEXLX1 and Phytophthora cactorum; PcaEXLX1) were shown to bind xylan and hardwood pulp at pH 5.5 and CmiEXLX2 (harboring a family-2 carbohydrate-binding module) also bound well to crystalline cellulose. Small-angle X-ray scattering revealed a 20–25% increase in interfibrillar distance between neighboring cellulose microfibrils following treatment with CmiEXLX2, DaqEXLX1, or NspEXLX1. Correspondingly, combining xylanase with CmiEXLX2 and DaqEXLX1 increased product yield from hardwood pulp by ~ 25%, while supplementing the TrAA9A LPMO from Trichoderma reesei with CmiEXLX2, DaqEXLX1, and NspEXLX1 increased total product yield by over 35%. This direct comparison of diverse EXLXs revealed consistent impacts on interfibrillar spacing of cellulose microfibers and performance of carbohydrate-active enzymes predicted to act on fiber surfaces. These findings uncover new possibilities to employ EXLXs in the creation of value-added materials from cellulosic biomass.

Emergent Nanostructure and Ion Transport in Polyzwitterion/Polyanion Blends

We investigated blends of poly(1-(3-sulfonatopropyl)-2-vinylpyridinium) (P2VPPS) and poly(lithium (trifluoromethane)sulfonimide methacrylate) (poly(MTFSI)Li) at varying molar ratios to gain a mechanistic understanding of ionic conductivity in a miscible polyzwitterion/polyanion system. This dataset contains the raw numerical data corresponding to the figures in the manuscript. The data files include the following information: (1) Experimental Data – includes X-ray and neutron scattering measurements, broadband dielectric spectroscopy (BDS) data, extracted DC conductivity values, differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) results, extracted glass transition temperatures, etc. (2) CGMD Data – includes molecular dynamics (MD) trajectory files and computed structural correlations. All data files are organized/named according to the figure numbers in the manuscript.

Matrix characteristics and origin of lunar breccia samples no. 12034 and 12073

The transmission electron microscope was used to investigate the nature of the matrix in two samples of lunar breccia. This matrix is composed largely of tiny (0.5 microns) grains of unaltered glass which are plastically fitted against one another, and tightly welded to the clastic fragments with which they are in contact. The breccias have an open framework of angular clastic particles composed mainly of anorthositic, basaltic and glassy fragments. Most of the anorthositic fragments have been shock metamorphosed. Over half of the glassy clasts, and also the former diaplectic glass particles produced from shocked plagioclase, were partly or wholly devitrified before inclusion in the present breccias. It is concluded that each breccia was deposited from a hot basesurge cloud of impact debris, and the tiny bits of glass in the matrix condensed from rock that was volatilized by the heat of major meteorite impact.

Application of remote sensing to state and regional problems

The author has identified the following significant results. The Lowndes County data base is essentially complete with 18 primary variables and 16 proximity variables encoded into the geo-information system. The single purpose, decision tree classifier is now operational. Signatures for the thematic extraction of strip mines from LANDSAT Digital data were obtained by employing both supervised and nonsupervised procedures. Dry, blowing sand areas of beach were also identified from the LANDSAT data. The primary procedure was the analysis of analog data on the I2S signal slicer.
