Effective Mechanisms for Improving Seed Oil Production in Pennycress (Thlaspi arvense L.) Highlighted by Integration of Comparative Metabolomics and Transcriptomics

Pennycress is a potentially lucrative biofuel crop due to its high content of long-chain unsaturated fatty acids, and because it uses non-conventional pathways to achieve efficient oil production. However, metabolic engineering is required to improve pennycress oilseed content and make it an economically viable source of aviation fuel. Research is warranted to determine if further upregulation of these non-conventional pathways could improve oil production within the species even more, which would indicate these processes serve as promising metabolic engineering targets and could provide the improvement necessary for economic feasibility of this crop. To test this hypothesis, we performed a comparative biomass, metabolomic, and transcriptomic analyses between a high oil accession (HO) and low oil accession (LO) of pennycress to assess potential factors required to optimize oil content. An evident reduction in glycolysis intermediates, improved oxidative pentose phosphate pathway activity, malate accumulation in the tricarboxylic acid cycle, and an anaplerotic pathway upregulation were noted in the HO genotype. Additionally, higher levels of threonine aldolase transcripts imply a pyruvate bypass mechanism for acetyl-CoA production. Nucleotide sugar and ascorbate accumulation also were evident in HO, suggesting differential fate of associated carbon between the two genotypes. An altered transcriptome related to lipid droplet (LD) biosynthesis and stability suggests a contribution to a more tightly-packed LD arrangement in HO cotyledons. In addition to the importance of central carbon metabolism augmentation, alternative routes of carbon entry into fatty acid synthesis and modification, as well as transcriptionally modified changes in LD regulation, are key aspects of metabolism and storage associated with economically favorable phenotypes of the species.

Improving the Precision of First-Principles Calculation of Parton Physics from Lattice Quantum Chromodynamics

Large momentum effective theory (LaMET) provides a general framework for computing the multi-dimensional partonic structure of the proton from first principles using lattice quantum chromodynamics (QCD). In this effective field theory approach, LaMET predicts parton distributions through a power expansion and perturbative matching of a class of Euclidean observables—quasi-distributions—evaluated at large proton momenta. Recent advances in lattice renormalization, such as the hybrid scheme with leading renormalon resummation, together with improved matching kernel that incorporates higher-loop corrections and resummations, have enhanced both the perturbative and power accuracy of LaMET, enabling a reliable quantification of theoretical uncertainties. Moreover, the Coulomb-gauge correlator approach further simplifies lattice analyses and improves the precision of transverse-momentum-dependent structures, particularly in the non-perturbative region. State-of-the-art LaMET calculations have already yielded certain parton observables with important phenomenological impact. In addition, the recently proposed kinematically enhanced lattice interpolation operators promise access to unprecedented proton momenta with greatly improved signal-to-noise ratios, which will extend the range of LaMET prediction and further suppress the power corrections. The remaining challenges, such as controlling excited-state contamination in lattice matrix elements and extracting gluonic distributions, are expected to benefit from emerging lattice techniques for ground-state isolation and noise reduction. Thus, lattice QCD studies of parton physics have entered an exciting stage of precision control and systematic improvement, which will have a broader impact for nuclear and particle experiments.

Systematic improvement of trial states in phaseless auxiliary-field quantum Monte Carlo

We extend the use of coupled cluster (CC) trial states in the phaseless auxiliary-field quantum Monte Carlo (AFQMC) method beyond single and double excitations to include both triple and quadruple excitations. With this AFQMC/CC hierarchy, we are able to systematically benchmark the method's performance on molecular systems as the quality of the trial is improved. Our results show that the phaseless AFQMC energy improves systematically and is typically significantly more accurate than the energy of the underlying trial state. However, the relative improvement compared to the trial CC energy decreases as we ascend the CC hierarchy. As the CC wavefunction is usually further approximated when used as an AFQMC trial, we also explore the relationship between the components of the CC wavefunction and the resulting AFQMC/CC error. Our results suggest that improving the representation of the CC wave function in the AFQMC trial does not always lower the bias even when it increases the fidelity of the trial with the exact ground state.

Improved method for temporally interpolating radiosonde profiles in the convective boundary layer

A significantly improved technique for temporally interpolating radiosonde (RS) profiles of potential temperature and water vapor mixing ratio in the planetary boundary layer during daytime is introduced. The key innovation of this technique is its operation on a height grid normalized with the planetary boundary layer height. This study utilized a three-month dataset of three-hourly soundings from the Atmospheric Radiation Measurement Facility's Southern Great Plains site. The technique was evaluated for convective boundary layer cases, with the necessary boundary layer height data obtained from a ground-based infrared spectrometer. A total of 79 comparisons were conducted between reference soundings and interpolated profiles that did and did not employ height normalization. The results demonstrated a substantial improvement in the representation of interpolated profiles using the new technique, characterized by enhanced correlation, improved amplitude representation, and reduced bias for potential temperature, as well as improved correlation and reduced bias for water vapor mixing ratio.

Improving thermodynamic nudging in the E3SM Atmosphere Model version 2 (EAMv2): strategy and hindcast skills on weather systems

Nudging techniques are commonly employed to constrain atmospheric simulations toward observed states, facilitating model evaluation and sensitivity studies. However, if applied improperly – particularly to thermodynamic variables such as temperature and humidity – nudging can distort physical processes and introduce spurious biases, undermining the credibility of the simulations. This study presents an improved nudging implementation that applies vertically modulated tendencies to reduce adverse impacts on model physics. The framework is tested in version 2 of the Energy Exascale Earth System Model (EAMv2) using a suite of hindcast simulations nudged toward ERA5 reanalysis. We systematically evaluate the individual and combined effects of nudging wind, temperature, and humidity fields on the model's ability to represent large-scale atmospheric states and high-impact weather systems. Results show that the revised strategy – particularly when nudging temperature and humidity at selected levels – enhances hindcast skill by improving agreement with ERA5 without degrading the hydrological cycle or precipitation processes. Additional improvements in surface temperature, outgoing longwave radiation, and precipitation biases are achieved through targeted nudging of land-surface variables. The proposed approach strengthens the representation of large-scale conditions relevant to tropical cyclones, atmospheric rivers, and extratropical cyclones in the low-resolution EAMv2. These findings demonstrate that carefully designed thermodynamic nudging, especially of temperature and humidity, improves the realism of constrained simulations and broadens the utility of nudged EAMv2 for atmospheric modeling, machine learning, and high-impact weather research.

Development of improved pyroelectric detectors; Literature survey of pyroelectric materials and their characteristics

The object of this program is to improve the detectivity of the pyroelectric detector with the ultimate goal of operation at or near the temperature-noise limit. Two general areas of investigation are undertaken. The first is to improve responsivity through the use of new materials. The second is directed toward reduction of noise and will be effected with improved field effect transistor characteristics, and improved electroding of the pyroelectric material. The search for new materials has begun with a review of the literature on pyroelectric materials in several languages. The compiled data includes an extensive list of references. From this, several materials have already been selected for investigation. FETs are being obtained from various manufacturers, evaluated, and selected units will be tested with pyroelectric elements as complete detectors.

Compensator improvement for a large space vehicle.

This paper presents the theory and a technique for compensator improvement. Several definitions in regard to relative stability are presented along with some frequency response limitations and characteristics of a large space vehicle. A nonlinear programming algorithm for obtaining an improved solution for a strict constraint problem is developed and the necessary partial derivatives for applying the algorithm to compensator improvement are derived. Finally, for illustrating the effectiveness of the algorithm, the frequency response of a large space vehicle is improved.

Intelligibility improvement of analog communication systems using an amplitude control technique.

An amplitude control technique has been employed for use with analog voice communication systems, which improves low-level phoneme reception and eliminates the received noise between words and syllables. Tests were conducted on a narrow-band frequency-modulation simplex voice communication channel employing the amplitude control technique. Presented for both the modified rhyme word tests and the phonetically balanced word tests are a series of graphical plots of the tests' score distribution, mean, and standard deviation as a function of received carrier-to-noise power density ratio. At low received carrier-to-noise power density ratios, a significant improvement in the intelligibility was obtained. A voice intelligibility improvement of more than 2 dB was obtained for the modified rhyme test words, and a voice intelligibility improvement in excess of 4 dB was obtained for the phonetically balanced word tests.

Possibilities for improved supersonic inlet performance

The results from detailed large-scale inlet tests were examined to assess the penalties, in terms of vehicle cruise range, for boundary-layer bleed, less than ideal engine-face pressure recovery, inlet weight, and external cowl drag. The assessment suggested specific improvements in the design of the inlet system that may increase the range of a typical supersonic transport approximately 6.9%. While no single design improvement can account for a large increase in range, careful attention to each design detail can yield a substantial total improvement. Because of noise considerations, future engines for advanced supersonic transports may be approximately 50% larger than used for the present study, making the effects of improved inlet performance even more important.

Boron aluminum crippling strength shows improvement

Results are presented from an experimental program directed toward improving boron aluminum crippling strength. Laminate changes evaluated were larger filament diameter, improved processing, shape changes, adding steel-aluminum cross plies, reduced filament volume in corners, adding boron aluminum angle plies, and using titanium interleaves. Filament diameter and steel-aluminum cross plies have little effect on crippling. It is shown that better processing combined with appropriate shape changes improved crippling over 50 percent at both room temperature and 600 F. Tests also show that crippling improvements ranging from 20 to 40 percent are achieved using angle plies and titanium interleaves.

Improved turbine disk design to increase reliability of aircraft jet engines

An analytical study was conducted on a bore entry cooled turbine disk for the first stage of the JT8D-17 high pressure turbine which had the potential to improve disk life over existing design. The disk analysis included the consideration of transient and steady state temperature, blade loading, creep, low cycle fatigue, fracture mechanics and manufacturing flaws. The improvement in life of the bore entry cooled turbine disk was determined by comparing it with the existing disk made of both conventional and advanced (Astroloy) disk materials. The improvement in crack initiation life of the Astroloy bore entry cooled disk is 87% and 67% over the existing disk made of Waspaloy and Astroloy, respectively. Improvement in crack propagation life is 124% over the Waspaloy and 465% over the Astroloy disks. The available kinetic energies of disk fragments calculated for the three disks indicate a lower fragment energy level for the bore entry cooled turbine disk.

United States benefits of improved worldwide wheat crop information from a LANDSAT system

The value of worldwide information improvements on wheat crops, promised by LANDSAT, is measured in the context of world wheat markets. These benefits are based on current LANDSAT technical goals and assume that information is made available to all (United States and other countries) at the same time. A detailed empirical sample demonstration of the effect of improved information is given; the history of wheat commodity prices for 1971-72 is reconstructed and the price changes from improved vs. historical information are compared. The improved crop forecasting from a LANDSAT system assumed include wheat crop estimates of 90 percent accuracy for each major wheat producing region. Accurate, objective worldwide wheat crop information using space systems may have a very stabilizing influence on world commodity markets, in part making possible the establishment of long-term, stable trade relationships.

Optimizing airport runway improvement program - A dynamic programming approach

In order to reduce the air traffic delay in the terminal area, an immediate remedy is to increase airport capacity by an expansion of the existing runway system. The runway expansion program is often limited by budgetary constraints; the expensive facilities for a long-term improvement cannot be built at once. When a runway improvement strategy is being considered for a longer planning horizon, the investiment decision depends upon the interrelations of its composite periods. The problem, therefore, is to determine how time factor and investment decisions interact to yield an optimal improvement scheme that meets demand at a minimum cost. With this objective in mind, a dynamic programming methodology is employed to determine the optimal planning scheme. Also, an example runway improvement problem is tested to illustrate how a dynamic programming model is practical in actual application.

United States benefits of improved worldwide wheat crop information from a LANDSAT system overview

The value of improvements in worldwide information on wheat crops provided by LANDSAT was measured in the context of world wheat markets. These benefits were based on exiting LANDSAT technical goals and assumed that information would be made available to the United States and other countries at the same time. The benefits to the United States of such public LANDSAT information on wheat crops were found to be 174 million dollars a year on the average. The benefits from improved wheat crop information compare favorably with the annual system's cost of about $62 million. A detailed empirical sample demonstration of the effect of improved information was developed. The history of wheat commodity prices for 1971-72 was reconstructed and the price changes from improved vs. historical information were compared.
