Enzymatic Properties of an Alkaline and Chelator Resistant Proportional to alpha-Amylase from the Alkaliphilic Bacillus sp. Isolate L1711

An alkaliphilic amylase producing bacterium, Bacillus sp. strain L1711, was selected among 13 soda lakes isolates. When grown at pH 10.5 and 370 C, strain L1711 produced multiple forms of amylases in the culture broth. One of these, BAA, was purified from the culture supernatant by QAE column chromatography and preparative native gel electrophoresis. The molecular weight of BAA was determined to be 51 kDa by denaturing gel electrophoresis. The pH optima for activity below and above 40 C were 9.5-10.0 and 7.0-7.5 respectively. BAA was stable in the pH range 6-11 and was completely inactivated at 55?C. The thermostability was not increased in the presence of Ca(2+). The enzyme was strongly inhibited by Ca(2+), Zn(2+), Mg(2+), Mn(2+), Ba(2+) and Cu(2+), whereas the presence of Na(+), Co2+ and EDTA (10 mM) enhanced enzymatic activity. The K(sub m) and specific activity of BAA on soluble starch were 1.9 mg/ml and 18.5 U/mg respectively. The main end products of hydrolysis were maltotetraose, maltose and glucose .

Enzymatic DNA molecules

The present invention discloses deoxyribonucleic acid enzymes--catalytic or enzymatic DNA molecules--capable of cleaving nucleic acid sequences or molecules, particularly RNA, in a site-specific manner, as well as compositions including same. Methods of making and using the disclosed enzymes and compositions are also disclosed.

Enzymatic Activity Detection via Electrochemistry for Enceladus

Electrochemical detection of biological molecules is a pertinent topic and application in many fields such as medicine, environmental spills, and life detection in space. Proteases, a class of molecules of interest in the search for life, catalyze the hydrolysis of peptides. Trypsin, a specific protease, was chosen to investigate an optimized enzyme detection system using electrochemistry. This study aims at providing the ideal functionalization of an electrode that can reliably detect a signal indicative of an enzymatic reaction from an Enceladus sample.

The properties of an ion selective enzymatic asymmetric synthetic membrane.

With the aid of a simple model membrane system, the properties of cellulose enzymes and of membrane selectivity and pump-like action are considered. The model is based on materials possibly present on a primitive earth, as well as on a membrane able to sort or concentrate these materials. An overview of the model membrane system is presented in terms of how it is constructed, what its properties are, and what to expect in performance characteristics. The model system is shown to be useful for studying the selective and in some cases accelerated transfer of nutrients and metabolites.

Characteristics of enzymatic induction provoked by chlordane

The effects of various stresses, such as restraint and lowering or raising of environmental temperature, in mice pretreated with chlordane were investigated. (Chlordane is an inhibitor of protein synthesis.) It was found that restraint or exposure to a cold environment for three hours mobilized the chlordane stored in the adipose tissue of mice.

Factors influencing the rate of non-enzymatic activation of carboxylic and amino acids by ATP

The nonenzymatic formation of adenylate anhydrides of carboxylic and amino acids is discussed as a necessary step in the origin of the genetic code and protein biosynthesis. Results of studies are presented which have shown the rate of activation to depend on the pKa of the carboxyl group, the pH of the medium, temperature, the divalent metal ion catalyst, salt concentration, and the nature of the amino acid. In particular, it was found that of the various amino acids investigated, phenylalanine had the greatest affinity for the adenine derivatives adenosine and ATP. Results thus indicate that selective affinities between amino acids and nucleotides were important during prebiotic chemical evolution, and may have played a major role in the origin of protein synthesis and genetic coding.

Enzymatic capacities of skeletal muscle - Effects of different types of training

Long-term adaptation mechanisms to maintain homeostasis at increased levels of exertion such as those caused by regular exercise are described. Mitochondrial changes have been found to be a result of endurance exercises, while mitochondrial responses to other types of exercise are small. Further discussion is devoted to long-term changes in glucose transport, hexokinase, phosphofructokinase, pyruvate kinase, and the increased sensitivity of an endurance trained muscle to insulin. Less lactate has been found to be produced by the skeletal muscles at the same work rate after adaptation to endurance exercise training, and the capacity for the flux of the two-carbon acetyl chain through the citric acid cycle increases in skeletal muscles in response to endurance training. Finally, endurance training is noted to result in glycogen sparing and an increase in the capacity to utilize fatty acids.

Non-enzymatic transcription of an oligodeoxynucleotide 14 residues long

Nonenzymatic synthesis of oligodeoxynucleotides up to 14 residues long from 2-MeImpG and 2-MeImpC mononucleotides was demonstrated. The synthesis is primed by 14-mer and 15-mer oligonucleotides d(C3GC3GC3GC2) and d(C3GC3GC3GC3) as templates. The predominant products are a series of 3-prime-5-prime-linked oligonucleotides, complementary to the template, ranging in length from GGC to GGCGGGCGGGCGGG. The 15-mer template directed the synthesis of the same family of products that were formed on the 14-mer template. This finding is explained by the preferential conversion of the dimer GG to GGC rather than to GGG. In the context of molecular evolution, these results suggest that the detailed kinetics of template-directed synthesis could form the basis for the selection of one replicating oligonucleotide from a family of closely related oligonucleotides.
