Security of quantum position-verification limits Hamiltonian simulation via holography

We investigate the link between quantum position-verification (QPV) and holography established in [1] using holographic quantum error correcting codes as toy models. By inserting the “temporal” scaling of the AdS metric by hand via the bulk Hamiltonian interaction strength, we recover a toy model with consistent causality structure. This leads to an interesting implication between two topics in quantum information: if position-based verification is secure against attacks with small entanglement then there are new fundamental lower bounds for resources required for one Hamiltonian to simulate another.

Double holography in string theory

We develop the notion of double holography in Type IIB string theory realizations of braneworld models. The Type IIB setups are based on the holographic duals of 4d BCFTs comprising 4d N = 4 SYM on a half space coupled to 3d N = 4 SCFTs on the boundary. Based on the concrete BCFTs and their brane construction, we provide microscopic realizations of the intermediate holographic description, obtained by dualizing only the 3d degrees of freedom. Triggered by recent observations in bottom-up models, we discuss the causal structures in the full BCFT duals and intermediate descriptions. This confirms qualitative features found in the bottom-up models but suggests a refinement of their interpretation.

Causal diamonds, cluster polytopes and scattering amplitudes

The “amplituhedron” for tree-level scattering amplitudes in the bi-adjoint φ 3 theory is given by the ABHY associahedron in kinematic space, which has been generalized to give a realization for all finite-type cluster algebra polytopes, labelled by Dynkin diagrams. In this letter we identify a simple physical origin for these polytopes, associated with an interesting (1 + 1)-dimensional causal structure in kinematic space, along with solutions to the wave equation in this kinematic “spacetime” with a natural positivity property. The notion of time evolution in this kinematic spacetime can be abstracted away to a certain “walk”, associated with any acyclic quiver, remarkably yielding a finite cluster polytope for the case of Dynkin quivers. The A n–3 , B n–1 /C n–1 and D n polytopes are the amplituhedra for n-point tree amplitudes, one-loop tadpole diagrams, and full integrand of one-loop amplitudes. We also introduce a polytope D¯ n , which chops the D n polytope in half along a symmetry plane, capturing one-loop amplitudes in a more efficient way.

Subregion-subalgebra duality: Emergence of space and time in holography

In holographic duality, a higher dimensional quantum gravity system emerges from a lower dimensional conformal field theory (CFT) with a large number of degrees of freedom. We propose a formulation of duality for a general causally complete bulk spacetime region, called subregion-subalgebra duality, which provides a framework to describe how geometric notions in the gravity system, such as spacetime subregions, different notions of times, and causal structure, emerge from the dual CFT. Subregion-subalgebra duality generalizes and brings new insights into subregion-subregion duality (or equivalently entanglement wedge reconstruction). It provides a mathematically precise definition of subregion-subregion duality and gives an independent definition of entanglement wedges without using entropy. Geometric properties of entanglement wedges, including those that play a crucial role in interpreting the bulk as a quantum error correcting code, can be understood from the duality as the geometrization of the superadditivity of certain algebras. Using general boundary subalgebras rather than those associated with geometric subregions makes it possible to find duals for general bulk spacetime regions, including those not touching the boundary. Applying subregion-subalgebra duality to a boundary state describing a single-sided black hole also provides a precise way to define mirror operators. Published by the American Physical Society 2025

Identification of driver genes for critical forms of COVID-19 in a deeply phenotyped young patient cohort

The drivers of critical coronavirus disease 2019 (COVID-19) remain unknown. Given major confounding factors such as age and comorbidities, true mediators of this condition have remained elusive. We used a multi-omics analysis combined with artificial intelligence in a young patient cohort where major comorbidities were excluded at the onset. The cohort included 47 “critical” (in the intensive care unit under mechanical ventilation) and 25 “non-critical” (in a non-critical care ward) patients with COVID-19 and 22 healthy individuals. The analyses included whole-genome sequencing, whole-blood RNA sequencing, plasma and blood mononuclear cell proteomics, cytokine profiling, and high-throughput immunophenotyping. An ensemble of machine learning, deep learning, quantum annealing, and structural causal modeling were used. Patients with critical COVID-19 were characterized by exacerbated inflammation, perturbed lymphoid and myeloid compartments, increased coagulation, and viral cell biology. Among differentially expressed genes, we observed up-regulation of the metalloprotease ADAM9. This gene signature was validated in a second independent cohort of 81 critical and 73 recovered patients with COVID-19 and was further confirmed at the transcriptional and protein level and by proteolytic activity. Ex vivo ADAM9 inhibition decreased severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) uptake and replication in human lung epithelial cells. In conclusion, within a young, otherwise healthy, cohort of individuals with COVID-19, we provide the landscape of biological perturbations in vivo where a unique gene signature differentiated critical from non-critical patients. We further identified ADAM9 as a driver of disease severity and a candidate therapeutic target.

The executive disruption model of tinnitus distress: Model validation in two independent datasets using factor score regression

This study presents the executive disruption model (EDM) of tinnitus distress and subsequently validates it statistically using two independent datasets (the Construction Dataset: n = 96 and the Validation Dataset: n = 200). The conceptual EDM was first operationalised as a structural causal model (construction phase). Then multiple regression was used to examine the effect of executive functioning on tinnitus-related distress (validation phase), adjusting for the additional contributions of hearing threshold and psychological distress. For both datasets, executive functioning negatively predicted tinnitus distress score by a similar amount (the Construction Dataset: β = −3.50, p = 0.13 and the Validation Dataset: β = −3.71, p = 0.02). Theoretical implications and applications of the EDM are subsequently discussed; these include the predictive nature of executive functioning in the development of distressing tinnitus, and the clinical utility of the EDM.

Structural Break and Causal Analyses of U.S. Corn Use for Ethanol and Other Corn Market Variables

The causal basis for many of the relationships in models used to estimate the indirect effects of U.S. biofuels on global agricultural markets has not been adequately established. This paper addresses this gap by examining causal interactions among corn market variables through which the indirect effects of U.S. corn use for ethanol would be transmitted. Specifically, structural break and causal analyses of U.S. corn supply, uses, trade, and price are performed using quarterly data for marketing years 1986 to 2017. The structural break analysis identifies three breaks in corn use for ethanol that reflect the policy-driven evolution of U.S. corn ethanol production and other market factors. The causality analysis finds that U.S. corn use for ethanol is not a driver of the corn price and net corn exports. Changes in corn supply and domestic corn use are found to be the key factors in accommodating the large increase in corn use for ethanol between 2003 and 2010. These results mean that common assumptions linking U.S. corn ethanol production to large reductions in corn availability and exports, and higher global corn prices merit reconsideration.

Comparative Study of High-Resolution LysB29(Nε-myristoyl) des(B30) Insulin Structures Display Novel Dynamic Causal Interrelations in Monomeric-Dimeric Motions

The treatment of insulin-dependent diabetes mellitus is characterized by artificial supplementation of pancreatic β-cell ability to regulate sugar levels in the blood. Even though various insulin analogs are crucial for reasonable glycemic control, understanding the dynamic mechanism of the insulin analogs may help to improve the best-protracted insulin analog to assist people with type 1 diabetes (T1D) to live comfortably while maintaining tight glycemic control. Here, we present the high-resolution crystal structure of NN304, known as insulin detemir, to 1.7 Å resolution at cryogenic temperature. We computationally further investigated our crystal structure’s monomeric-dimeric conformation and dynamic profile by comparing it with a previously available detemir structure (PDB ID: 1XDA). Our structure (PDB ID: 8HGZ), obtained at elevated pH, provides electrostatically triggered minor movements in the equilibrium between alternate conformational substates compared to the previous structure, suggesting it might induce an intermediate state in the dissociation pathway of the insulin detemir’s hexamer:dihexamer equilibrium. Supplemented with orientational cross-correlation analysis by a Gaussian network model (GNM), this alternate oligomeric conformation offers the distinct cooperative motions originated by loose coupling of distant conformational substates of a protracted insulin analog that has not been previously observed.

What is (quantitative) system dynamics modeling? Defining characteristics and the opportunities they create

A clear definition of system dynamics modeling can provide shared understanding and clarify the impact of the field. We introduce a set of characteristics that define quantitative system dynamics, selected to capture core philosophy, describe theoretical and practical principles, and apply to historical work but be flexible enough to remain relevant as the field progresses. The defining characteristics are: (1) models are based on causal feedback structure, (2) accumulations and delays are foundational, (3) models are equation-based, (4) concept of time is continuous, and (5) analysis focuses on feedback dynamics. We discuss the implications of these principles and use them to identify research opportunities in which the system dynamics field can advance. These research opportunities include causality, disaggregation, data science and AI, and contributing to scientific advancement. Progress in these areas has the potential to improve both the science and practice of system dynamics.

What can simulation test beds teach us about social science? Results of the ground truth program

The ground truth program used simulations as test beds for social science research methods. The simulations had known ground truth and were capable of producing large amounts of data. This allowed research teams to run experiments and ask questions of these simulations similar to social scientists studying real-world systems, and enabled robust evaluation of their causal inference, prediction, and prescription capabilities. We tested three hypotheses about research effectiveness using data from the ground truth program, specifically looking at the influence of complexity, causal understanding, and data collection on performance. We found some evidence that system complexity and causal understanding influenced research performance, but no evidence that data availability contributed. The ground truth program may be the first robust coupling of simulation test beds with an experimental framework capable of teasing out factors that determine the success of social science research.

Proteomic Profiling of the Substantia Nigra to Identify Determinants of Lewy Body Pathology and Dopaminergic Neuronal Loss

Proteinaceous aggregates containing ?-synuclein protein called Lewy bodies in the substantia nigra is a hallmark of Parkinson’s disease. The molecular mechanisms of Lewy body formation and associated neuronal loss remain largely unknown. To gain insights on proteins and pathways associated with Lewy body pathology, we performed quantitative profiling of the proteome. We analyzed substantia nigra tissue from 51 subjects arranged into three groups: cases with Lewy body pathology, Lewy body-negative controls with matching neuronal loss and controls with no neuronal loss. Using label-free LC-MS/MS approach we quantified the 2,963 most abundant proteins. Statistical testing for differential protein abundance, followed by pathway enrichment and Bayesian learning of the causal network structure were performed to identify likely drivers of Lewy body formation and dopaminergic neuronal loss. The identified pathways include (1) Arp2/3 complex-mediated actin nucleation; (2) synaptic function; (3) poly(A) RNA binding; (4) basement membrane and endothelium; and (5) hydrogen peroxide metabolic process. According to the data, the endothelial/basement membrane pathway is tightly connected with both pathologies and likely to be one of the drivers of the neuronal loss. The poly(A) RNA-binding proteins, including the ones relevant to other neurodegenerative disorders (e.g. TDP-43 and FUS) have strong inverse correlation with Lewy bodies and may reflect alternative mechanism of nigral neurodegeneration.

Back to the future: Causality on a moving braneworld

Brane observers executing appropriate motion through a partially compactified Lorentz invariant bulk spacetime, such as M 4 × S 1 , can send signals along the brane that are instantaneous or even travel backward in time. Nevertheless, causality in the braneworld remains intact. We establish these results, which follow from superluminal signal propagation reported in Greene et al. [Superluminal propagation on a moving braneworld, Phys. Rev. D 106, 085001 (2022).], through classical analysis and then extend our reasoning by examining quantum mechanical microcausality. Finally, one implication is the capacity for real time communication across arbitrarily large distances.

Quantum Mechanics of a Spherically Symmetric Causal Diamond in Minkowski Spacetime

We construct the phase space of a spherically symmetric causal diamond in ( d + 2 )-dimensional Minkowski spacetime. Utilizing the covariant phase space formalism, we identify the relevant degrees of freedom that localize to the d -dimensional bifurcate horizon and, upon canonical quantization, determine their commutators. On this phase space, we find two Iyer-Wald charges. The first of these charges, proportional to the area of the causal diamond, is responsible for shifting the null time along the horizon and has been well documented in the literature. The second charge is much less understood, being integrable for d ≥ 2 only if we allow for field-dependent diffeomorphisms and is responsible for changing the size of the causal diamond. Causal interaction in high frequency turbulence at the biosphere–atmosphere interface: Structural behavior

High-frequency (e.g., 10 Hz) eddy covariance measurements are typically used to estimate fluxes at the land–atmosphere interface at timescales of 15–60 min. These multivariate data contain information about the interdependency at high frequency between the interacting variables such as wind, humidity, temperature, and CO 2⁠ . We use data at 10 Hz from an eddy covariance instrument located at 25 m above agricultural land in the Midwestern US, which offers an opportunity to move beyond the traditional spectral analyses to explore causal dependency among variables. In this study, we quantify the structure of inter-dependencies of interacting variables at high frequency represented by a directed acyclic graph (DAG). We compare DAGs to investigate changes in structural differences in causal interactions. We then apply a distance-based classification and -means clustering approach to identify the evolution of the causal structure represented by a DAG. Our method selects an unbiased number of clusters of similar structures and characterizes the similarities and differences between them. We explore a range of dynamic behavior using data from a clear sky day and during a solar eclipse in 2017. Our results show well-defined clusters of similar causal dependencies as the system evolves. Furthermore, our approach provides a methodological framework to understand how causal dependence in turbulence manifests in high-frequency data when represented through a DAG.
