Lyophilization of ASFV vaccine candidate ASFV-G-ΔI177L offers long term stability

Abstract For over a century African swine fever (ASF) has been causing outbreaks leading to devastating losses for the swine industry. The current pandemic of ASF has shown no signs of stopping and continues to spread causing outbreaks in additional countries. Currently control relies mostly on culling infected farms, and strict biosecurity procedures. Recently a vaccine, ASFV-G-ΔI177L was approved for use in Vietnam. In this study we evaluate the long-term stability of lyophilized ASFV-G-ΔI177L. Understanding the stability of different formulations of vaccines is information necessary for deployment of vaccines to ASF outbreak areas, particularly those that do not have a reliable well established cold chain to ensure conservation of vaccine quality. In this report, we determined that ASFV-G-ΔI177L, when lyophilized under specific conditions, is stable for up to one year at 4 °C, with similar vaccine titers after storage. Next-generation sequencing analysis also determined that lyophilization and long-term storage under these conditions had no effect on the genome of ASFV as the genome remained genetically identical to the original non-lyophilized form.

A global phylogenomic analysis of the shiitake genus Lentinula

Lentinula is a broadly distributed group of fungi that contains the cultivated shiitake mushroom, L. edodes . We sequenced 24 genomes representing eight described species and several unnamed lineages of Lentinula from 15 countries on four continents. Lentinula comprises four major clades that arose in the Oligocene, three in the Americas and one in Asia–Australasia. To expand sampling of shiitake mushrooms, we assembled 60 genomes of L. edodes from China that were previously published as raw Illumina reads and added them to our dataset. Lentinula edodes sensu lato (s. lat.) contains three lineages that may warrant recognition as species, one including a single isolate from Nepal that is the sister group to the rest of L. edodes s. lat., a second with 20 cultivars and 12 wild isolates from China, Japan, Korea, and the Russian Far East, and a third with 28 wild isolates from China, Thailand, and Vietnam. Two additional lineages in China have arisen by hybridization among the second and third groups. Genes encoding cysteine sulfoxide lyase ( lecsl ) and γ-glutamyl transpeptidase ( leggt ), which are implicated in biosynthesis of the organosulfur flavor compound lenthionine, have diversified in Lentinula . Paralogs of both genes that are unique to Lentinula ( lecsl 3 and leggt 5b) are coordinately up-regulated in fruiting bodies of L. edodes . The pangenome of L. edodes s. lat. contains 20,308 groups of orthologous genes, but only 6,438 orthogroups (32%) are shared among all strains, whereas 3,444 orthogroups (17%) are found only in wild populations, which should be targeted for conservation.

GAMMA SPECTRUM STABILIZATION FOR ENVIRONMENTAL RADIATION MONITORING STATIONS USING NAI(TL) DETECTOR

Abstract Gamma spectrum measured by an NaI(Tl) detector is known to be unstable with the in situ temperature. In the present work, an advanced method has been applied to stabilize the gamma spectrum measured by the NaI(Tl) detector at environmental radiation monitoring (ERM) stations. The method is based on experimental data obtained under controlled conditions in laboratory. In the temperature range from 4 to 45°C, the relative deviation of the peak positions within the stabilized gamma spectrum is less than 2%. To test this method in a real scenario, it has been integrated into the ERM station at the Military Institute of Chemical and Environmental Engineering in Hanoi, Vietnam. The results show that the proposed method is ready for a real application.

ASF Vaccine Candidate ASFV-G-ΔI177L Does Not Exhibit Residual Virulence in Long-Term Clinical Studies

African swine fever (ASF) is an important disease in swine currently producing a pandemic affecting pig production worldwide. Except in Vietnam, where two vaccines were recently approved for controlled use in the field, no vaccine is commercially available for disease control. Up to now, the most effective vaccines developed are based on the use of live-attenuated viruses. Most of these promising vaccine candidates were developed by deleting virus genes involved in the process of viral pathogenesis and disease production. Therefore, these vaccine candidates were developed via the genomic modification of parental virus field strains, producing recombinant viruses and reducing or eliminating their residual virulence. In this scenario, it is critical to confirm the absence of any residual virulence in the vaccine candidate. This report describes the assessment of the presence of residual virulence in the ASFV vaccine candidate ASFV-G-ΔI177L in clinical studies conducted under high virus loads and long-term observation periods. The results demonstrated that domestic pigs intramuscularly inoculated with 10 6 HAD 50 of ASFV-G-ΔI177L did not show the presence of any clinical sign associated with ASF when observed daily either 90 or 180 days after vaccination. In addition, necropsies conducted at the end of the experiment confirmed the absence of macroscopic internal lesions associated with the disease. These results corroborate the safety of using ASFV-G-ΔI177L as a vaccine candidate.

African Swine Fever Vaccine Candidate ASFV-G-ΔI177L Produced in the Swine Macrophage-Derived Cell Line IPKM Remains Genetically Stable and Protective against Homologous Virulent Challenge

ASFV vaccine candidate ASFV-G-ΔI177L has been shown to be highly efficacious in inducing protection against challenges with the parental virus, the Georgia 2010 isolate, as well as against field strains isolated from Vietnam. ASFV-G-ΔI177L has been shown to produce protection even when used at low doses (10 2 HAD 50 ) and shows no residual virulence even when administered at high doses (10 6 HAD 50 ) or evaluated for a relatively long period of time (6 months). ASFV-G-ΔI177L stocks can only be massively produced in primary cell macrophages. Alternatively, its modified version (ASFV-G-ΔI177L/ΔLVR) grows in a swine-derived cell line (PIPEC), acquiring significant genomic modifications. We present here the development of ASFV-G-ΔI177L stocks in a swine macrophage cell line, IPKM, and its protective efficacy when evaluated in domestic pigs. Successive passing of ASFV-G-ΔI177L in IPKM cells produces minimal genomic changes. Interestingly, a stock of ASFV-G-ΔI177L obtained after 10 passages (ASFV-G-ΔI177Lp10) in IPKM cells showed very small genomic changes when compared with the original virus stock. ASFV-G-ΔI177Lp10 conserves similar growth kinetics in primary swine macrophage cultures than the original parental virus ASFV-G-ΔI177L. Pigs infected with 10 3 HAD 50 of ASFV-G-ΔI177Lp10 developed a strong virus-specific antibody response and were completely protected against the challenge with the parental virulent field isolate Georgia 2010. Therefore, IPKM cells could be an effective alternative for the production of ASFV vaccine stocks for those vaccine candidates exclusively growing in swine macrophages.

A Re-Evaluation of African Swine Fever Genotypes Based on p72 Sequences Reveals the Existence of Only Six Distinct p72 Groups

The African swine fever virus (ASFV) is currently causing a world-wide pandemic of a highly lethal disease in domestic swine and wild boar. Currently, recombinant ASF live-attenuated vaccines based on a genotype II virus strain are commercially available in Vietnam. With 25 reported ASFV genotypes in the literature, it is important to understand the molecular basis and usefulness of ASFV genotyping, as well as the true significance of genotypes in the epidemiology, transmission, evolution, control, and prevention of ASFV. Historically, genotyping of ASFV was used for the epidemiological tracking of the disease and was based on the analysis of small fragments that represent less than 1% of the viral genome. The predominant method for genotyping ASFV relies on the sequencing of a fragment within the gene encoding the structural p72 protein. Genotype assignment has been accomplished through automated phylogenetic trees or by comparing the target sequence to the most closely related genotyped p72 gene. To evaluate its appropriateness for the classification of genotypes by p72, we reanalyzed all available genomic data for ASFV. We conclude that the majority of p72-based genotypes, when initially created, were neither identified under any specific methodological criteria nor correctly compared with the already existing ASFV genotypes. Based on our analysis of the p72 protein sequences, we propose that the current twenty-five genotypes, created exclusively based on the p72 sequence, should be reduced to only six genotypes. To help differentiate between the new and old genotype classification systems, we propose that Arabic numerals (1, 2, 8, 9, 15, and 23) be used instead of the previously used Roman numerals. Furthermore, we discuss the usefulness of genotyping ASFV isolates based only on the p72 gene sequence.

Historical and projected future runoff over the Mekong River basin

The Mekong River (MR) crosses the borders and connects six countries, including China, Myanmar, Laos, Thailand, Cambodia, and Vietnam. It provides critical water resources and supports natural and agricultural ecosystems, socioeconomic development, and the livelihoods of the people living in this region. Understanding changes in the runoff of this important international river under projected climate change is critical for water resource management and climate change adaptation planning. However, research on long-term runoff dynamics for the MR and the underlying drivers of runoff variability remains scarce. Here, we analyse historical runoff variations from 1971 to 2020 based on runoff gauge data collected from eight hydrological stations along the MR. With these runoff data, we then evaluate the runoff simulation performance of five global hydrological models (GHMs) forced by four global climate models (GCMs) under the Inter-Sectoral Impact Model Intercomparison Project (ISIMIP). Furthermore, based on the best simulation combination, we quantify the impact of future climate change on river runoff changes in the MR. The result shows that the annual runoff in the MR has not changed significantly in the past 5 decades, while the establishment of dams and reservoirs in the basin visibly affected the annual runoff distribution. The ensemble-averaged result of the Water Global Assessment and Prognosis version 2 (WaterGAP2; i.e. GHM) forced by four GCMs has the best runoff simulation performance. Under Representative Concentration Pathways (RCPs; i.e. RCP2.6, RCP6.0 and RCP8.5), the runoff of the MR is projected to increase significantly (p<0.05); e.g. 3.81 ± 3.47 m 3 s -1 a -1 (9 ± 8 % increase in 100 years) at the upper reach under RCP2.6 and 16.36 ± 12.44 m 3 s -1 a -1 (13 ±10 % increase in 100 years) at the lower reach under RCP6.0. In particular, under the RCP6.0 scenario, the increase in annual runoff is most pronounced in the middle and lower reaches, due to increased precipitation and snowmelt. Under the RCP8.5 scenario, the runoff distribution in different seasons varies obviously, increasing the risk of flooding in the wet season and drought in the dry season.

Grid Interconnection and Renewables Deployment Related Air Quality and Human Health Benefits in Southeast Asia

The Association of Southeast Asian Nations (ASEAN) has increasingly focused on multilateral electricity trade, improved grid resiliency and modernization. These opportunities in the power sector have been studied through the ASEAN Interconnection Masterplan Studies, the most recent of which is the ASEAN Interconnection Masterplan Study III (AIMS III). AIMS III assessed four different scenarios of power system in the ASEAN countries that include a base scenario and three additional scenarios (called optimum RE, ASEAN RE, and High RE) considering different levels of deployment of renewable energy (RE) and cross-border generation and trade of electricity for three years (2025, 2030, 2040). In this work, we quantify the potential air quality and public health co-benefits of AIMS III scenarios. For a rapidly expanding, energy hungry region like SE Asia generation is power generation is expected to increase significantly. The AIMS III scenarios' capacity expansion modeling suggest that: 1) Generation nearly doubles from 2025 to 2040 in all four AIMS III scenarios; 2) Most of the increased generation is met by coal in all four AIMS III scenarios; 3) While renewables generation increases in all four AIMS III scenarios, the fraction of total generation (its share) generally decreases because of the much greater increase in non-renewable sources, mostly coal. Only in the High RE Target scenario do renewables represent a higher share in 2040 than in 2025, though even here it is at the expense of natural gas rather than coal; 4) As a result, emissions of gaseous and aerosol pollutants increase significantly in all scenarios. Because emissions increase in 2040 compared to 2025 for all four AIMS III scenarios, so do PM2.5 concentrations. Even for the High RE Target scenario (the scenario with highest share of renewable energy), compared to the Base scenario in 2025, in 2040 PM2.5 concentrations are higher. Compared to the Base scenario in the same years, the Optimum RE and ASEAN RE Target scenarios do not differ very much from the Base in terms of PM2.5 concentration. The High RE Target scenario, on the other hand, yields noticeably lower PM2.5 concentration. For example, in 2040 the population-weighted decrease in annual average PM2.5 concentration in the High RE Target scenario relative to the Base scenario is 0.5 ug m-3. Compared to the Base scenario in 2040, each of the alternative AIMS III scenarios is estimated to result in net reductions in power-sector air quality-related excess mortality in the ASEAN region. Yet there are a few countries for which the Optimum RE and ASEAN RE Target result in increases in PM2.5-related excess mortality (Thailand and Vietnam for the ASEAN RE Target scenario and Thailand for the Optimum RE scenario). However, for the High RE Target scenario, all countries benefit and find reductions in excess mortality resulting from the power sector scenarios modeled in AIMS III with regionwide mortality decreasing by 16,000 compared to the Base scenario in 2040. In summary, our analysis finds that changing power generation emissions is a crucial lever for improving public health in ASEAN member countries and provides a pathway for policymakers to make decision backed by a realistic power sector expansion and air quality analyses.

HydroEcoLSTM: A Python package with graphical user interface for hydro-ecological modeling with long short-term memory neural network

Machine learning (ML) is emerging as a promising tool for modeling hydro-ecological processes due to the increasing availability of large environmental data. However, the use of ML requires sufficient programming knowledge due to a lack of a graphical user interface (GUI). In this study, we introduced a GUI package, named HydroEcoLSTM, with the long short-term memory network (LSTM) as the core model, that allows non-ML experts to utilize their domain knowledge to construct complex ML models. We demonstrated the functionalities of HydroEcoLSTM with two practical examples, including (1) predictions of streamflow in both gauged and ungauged catchments and (2) predictions of multiple outputs (i.e., streamflow and isotope transport from two catchments). The simulation results obtained in both case experiments are satisfactory. In the first example, the average Nash–Sutcliffe Efficiency (NSE) for streamflow simulation during the testing period is 0.79 while the application of the trained model in two assumed ungauged catchments also achieves the average NSE of 0.68. In the second example, the average NSE for streamflow and instream isotope simulation during the testing period is 0.71. Ultimately, applications of HydroEcoLSTM with real-world examples demonstrate its potential use for practical applications and research without requiring extensive coding skills.

Short‐Term Hourly Weather Forecasting Using PredRNN With Image Preprocessing

Global weather forecast models are vital tools with numerous applications, including public safety, agriculture, and transportation. Recent advancements in artificial intelligence (AI) and deep learning (DL) have shown the potential to enhance weather forecasting accuracy and speed. In this study, we developed a short-term hourly weather forecast framework with a wavelet transform function for data preprocessing and a spatiotemporal DL model, PredRNN, for predicting five surface atmospheric variables, including wind speed and direction, mean sea level pressure (MSLP), temperature, and precipitation. The framework demonstrated promising results. It produces global forecasts at 0.25° (∼25 km) with a 1-day lead time RMSE of 1.8 m/s for wind components, 180 Pa for MSLP, and 1.8 K for temperature. Although our model does not surpass state-of-the-art AI weather forecast models across all metrics, it outperforms these models in precipitation forecasting and wind prediction at short lead times and achieves comparable accuracy for MSLP. Its native hourly forecasting capability, together with training on widely accessible GPU hardware, contributes meaningfully to the advancement of accessible DL weather forecasting methods. Our work highlights the importance of integrating temporal components and data transformation techniques to improve the predictability and accuracy of weather forecasts.

Impact of Newly Measured 𝛽-Delayed Neutron Emitters around 78 Ni on Light Element Nucleosynthesis in the Neutrino Wind Following a Neutron Star Merger

Neutron emission probabilities and half-lives of 37 𝛽-delayed neutron emitters from 75 Ni to 92 Br were measured at the RIKEN Nishina Center in Japan, including 11 one-neutron and 13 two-neutron emission probabilities and six half-lives for the first time that supersede theoretical estimates. These nuclei lie in the path of the weak 𝑟 process occurring in neutrino-driven winds from the accretion disk formed after the merger of two neutron stars synthesizing elements in the 𝐴∼80 abundance peak. The presence of such elements dominates the accompanying kilonova emission over the first few days and have been identified in the AT2017gfo event, associated to the gravitational wave detection GW170817. Abundance calculations based on over 17,000 simulated trajectories describing the evolution of matter properties in the merger outflows show that the new data lead to an increase of 50%–70% in the abundance of Y, Zr, Nb, and Mo. This enhancement is large compared to the scatter of relative abundances observed in old very metal poor stars and thus is significant in the comparison with other possible astrophysical processes contributing to the light-element production. These results underline the importance of including experimental decay data for very neutron-rich 𝛽 -delayed neutron emitters into 𝑟 -process models.

Continuous Flow Photoelectrochemical Reactor with Gas Permeable Photocathode: Enhanced Photocurrent and Partial Current Density for CO 2 Reduction

Photoelectrochemical (PEC) CO 2 reduction using a photocathode is an attractive method for making valuable chemical products due to its simplicity and lower overpotential requirements. However, previous PEC processes have often been diffusion-limited leading to low production rates of the CO 2 reduction reaction, due to inefficient gas diffusion through the liquid electrolyte to the catalyst surface, particularly at high current densities. In this study, a gas-permeable photocathode in a continuous flow PEC reactor is incorporated, which facilitates the direct supply of CO 2 gas to the photocathode-electrolyte interface, unlike dark reaction-based flow reactors. This concept is demonstrated using Ag-TiO 2 on carbon paper, illuminated through a quartz window and flowing liquid electrolyte. CO 2 supply is managed via pressure and flow control on the non-illuminated side of the carbon paper. The photocurrent density is significantly influenced by the flow rates and pressure of CO 2 gas, and the electrolyte flow rates. Compared to the traditional H-cell, the continuous PEC flow reactor achieves ≈10-fold increase in CO faradaic efficiency, 30-fold increase in production rate and 16-fold increase in stability without catalyst modifications. This work provides essential insights into the design and application of continuous gas-liquid flow PEC reactor systems, highlighting their potential for other PEC reactions.

Near-Real-Time Statistical Analysis and Visualization of Streamflow from a Deep-Learning Rainfall-Runoff Model

Near-real-time (NRT) streamflow data are critical importance for timely water resources management. Here, we developed an open-source tool, FlowStats, for NRT streamflow analysis and visualization in Germany, based on NRT meteorological data from the German Weather Service and simulated streamflow from a long short-term memory neural network (LSTM). The LSTM model achieved very good overall performance, median NSE of 0.80 for the test period across 1,479 catchments. FlowStats provides options for deriving various streamflow statistics, from normal and abnormal streamflow detection to drought and flood analyses. An example analysis from FlowStats revealed widespread below-normal to extreme low-flow conditions across Germany from March to May 2025, which weakened from June to September 2025. Drought analysis for September 2025 highlighted severe to extreme drought conditions in northwestern Germany, while flood classifications indicated that high-flow events occurred in southwestern Germany. FlowStats can be used for various hydrological assessments to support water resources management.

The Value of Forecasters‐in‐the‐Loop in Real‐Time Flood Forecasting in the Age of Machine Learning

Machine learning (ML) applications in hydrological forecasting are increasingly prevalent and show great potential. However, many previous studies have only evaluated performance through reanalysis or retrospective simulations compared to simplified baselines. This study provides the first assessment of ML performance against actual operational forecasting systems operated by the California Nevada River Forecast Center (CNRFC), which combines the Community Hydrologic Prediction System (CHPS) with forecasters-in-the-loop. Results demonstrate that forecasters-in-the-loop systems consistently outperform ML models in both general forecasts and flood alerting across lead times up to 96 hr, even when ML models use observed forcings, while CNRFC operational process relies on biased weather forecasts. Our analysis reveals that forecaster expertise maintains forecast reliability despite inaccurate precipitation inputs, with human-guided systems showing superior performance degradation characteristics at extended lead times. These findings highlight the irreplaceable value of human expertise in operational forecasting and caution against overstating current ML capabilities in real-world applications.

Dual-precipitates enhance fatigue resistance in an additively manufactured high-entropy alloy

To accomplish the intense desire of high-strength materials for enhanced energy-efficiency, recent research applies a combined strategy of additive-manufacturing and precipitation-strengthening in high entropy alloys. In a context, Al 0.2 Co 1.5 CrFeNi 1.5 Ti 0.3 nanoprecipitation-strengthened system was developed, demonstrating very convincing strength and toughness. Moreover, additive-manufacturing facilitated additional strength by well-decorated cell-boundaries with blocky L2 1 precipitates and homogeneously distributed L1 2 precipitates. However, fatigue research of this alloy remained unexplored despite being the main precursor for structural applications. In this study low-cycle fatigue behavior of this alloy in both as-built and precipitation-strengthened (aged) conditions has been explored, combined with in-situ neutron diffraction investigation. Findings revealed a substantial cyclic-stress profile and a notable fatigue-life below ±0.50% strain-amplitude, exceeding 10 5 cycles at ±0.30% strain-amplitude. These demonstrate the potential to carry higher payloads with marked engineering-reliability. Residual-stress estimation revealed strain-compatibility between the matrix and L1 2 precipitate, indicating a crack-initiation immune interface. A comparative examination of dislocation character revealed shifting towards pure edge-character in aged alloy indicates precipitates promoted planar-slip during deformation.

AC electrical transport detection of magnetic properties of small, granular, and heterogeneous samples

Experimental constraints and sample limitations can preclude ideal measurements of electrical transport properties of materials. In such situations, AC electrical transport methods are often employed due to a significant increase in signal-to-noise ratio they can provide. However, dynamic effects that are not often accounted for may be present that may modify the signals in these measurements. In particular, dynamic filtering effects are prominent in small, granular, and heterogeneous materials. We demonstrate that a lock-in amplifier based circuit can distinguish between these DC transport and AC filtering effects. Here, we further demonstrate that this filtering can reveal distinct signatures of magnetic transitions while providing a measure of sample quality.
